1.Shear wave viscoelastography for differentiating lung peripheral inflammatory masses and malignant tumors

Objective To observe the value of shear wave viscoelastography(SWV)for differentiating lung peripheral inflammatory masses and malignant tumors.Methods Conventional gray-scale ultrasound and SWV were prospectively performed in 70 patients with lung peripheral inflammatory mass or malignant tumor.The patients were divided into malignant group(n=42)and inflammatory group(n=28)according to pathological results.Clinical and ultrasonic data,including the maximum diameter of lesions,the mean Young's modulus(Emean),mean viscosity(Vmean),and mean dispersion slope(Dmean)were compared between groups.Receiver operating characteristic curves of ultrasonic parameters being significantly different between groups were drawn,and area under the curves(AUCs)were calculated to evaluate the efficacy of each parameter for differentiating lung peripheral inflammatory mass or malignant tumor.Results In malignant group,the maximum diameter and Emean of lesions were both higher,while Vmean and Dmean of lesions were both lower than those in inflammatory group(all P<0.05).Vmean and Dmean of lesions had moderately/good efficacy for differentiating lung peripheral inflammatory mass or malignant tumor(AUC=0.843,0.866),both better than that of conventional ultrasound and Emean(AUC=0.673,0.685)(all P<0.05).The combination of Emean,Vmean and Dmean had good efficacy for differentiating lung peripheral inflammatory masses and malignant tumors,with AUC of 0.874.Conclusion The viscous parameters of SWV could effectively differentiating lung peripheral inflammatory masses and malignant tumors.
2.Construction of risk model of liver injury related to parenteral nutrition in patients with severe acute pancreatitis

Objective:To construct a risk model of liver injury related to parenteral nutrition (PNALD) in patients with severe acute pancreatitis (SAP).Methods:The clinical data of 176 SAP patients admitted to the 971 Hospital of Navy from January 2019 to August 2021 were retrospectively collected. According to whether PNALD occurred or not, the patients were divided into liver injury group ( n=33) and non-liver injury group ( n=143). Multivariate logistic regression was used to analyze the influencing factors of PNALD in SAP patients. Then decision tree model and multivariate logistic regression model were established based on the screened risk factors. Hosmer and Lemeshow Test calibration curves were used to calibrate the two models, and receiver operating characteristic curve (ROC) was drawn and area under the curve (AUC) was calculated to compare the prediction efficiency of the two models. Results:Drinking history (history of alcohol intake), serum albumin / globulin ratio ≤1.45, prothrombin time (PT)≥18.52 s, PT activity ≤48.96, activated partial thromboplastin time (APTT) ≥45.91 s were all risk factors for PNALD. The ROC curve of the multivariate logistic regression model and the decision tree model was drawn, and calculated AUC of the two models was 0.851 and 0.906, respectively; the sensitivity was 79.6% and 80.8%, respectively; the specificity was 80.5% and 79.6%, respectively; and the Youden index was 0.601 and 0.604, respectively, with good consistency.Conclusions:Low serum albumin/globulin ratio and PT activity, high PT and APTT are all risk factors for PNALD. The PNALD prediction model based on the above risk factors has high specificity and sensitivity.
3.Improvement effects of tilianin on atherosclerosis model mice and its mechanism study

OBJECTIVE To s tudy the impr ovement effects of tilianin on the atherosclerosis (AS)model mice and its potential mechanism. METHODS Eight C 57BL/6J mice were taken as the normal group. Forty ApoE-/- mice were randomly divided into model group ,tilianin low-dose ,medium-dose and high-dose groups [ 2.1,3.5,7.0 mg/(kg·d)] and simvastatin group [positive control drug ,3.5 mg/(kg·d)],with 8 mice in each group. Normal group was given normal diet ,and other groups were given high-lipid diet to induce AS model. At the same time ,normal group and model group were given normal saline intragastrically , administration groups were given relevant drug intragastrically ,once a day ,for 12 consecutive weeks. The levels of TC ,TG, LDL-C,HDL-C,Ox-LDL,IL-1β,IL-6,MCP-1 and TNF-α in plasma were determined. The pathomorphological changes of the aorta in mice were observed. The positive rate of ICAM- 1,VCAM-1 and PCNA in the aorta were determined. mRNA expressions of MMP- 2,MMP-9,TGF-β1,Smad2 and Smad 3 as well as protein expressions of TGF-β1,Smad2/3 and p-Smad 2/3 were also determined in aorta of mice. RESULTS Compared with normal group ,the plasma levels of TC ,TG,LDL-C,Ox-LDL,IL-1β, IL-6,MCP-1 and TNF-α in model group were increased significantly(P<0.01),while HDL-C level was significantly reduced (P<0.01). Lipid plaques were formed in the aorta ,and the plaque area was large and caused severe stenosis of the lumen. mRNA expressions of MMP- 2,MMP-9,TGF-β1,Smad2 and Smad 3 as well as positive rate of ICAM- 1,VCAM-1,PCNA and protein expression TGF-β1,Smad2/3,and p-Smad 2/3 in the aorta were significantly increased (P<0.01). Compared with model group , most of above indexes of medication groups were improved significantly (P<0.05 or P<0.01). CONCLUSIONS Tilianin can inhibit the activation of TGF-β1/Smads signaling pathway and then inhibit the proliferation of vascular smooth muscle cells ,reduce , inflammation and regulate lipid metabolism to inhibit the No.81960766) formation of AS.
4.Study on Protective Mechanism of Dracocephalum moldavica Total Flavonoids against Myocardial Ischemia- reperfusion Injury in Rats Based on AMPK/SIRT 1/PGC-1α Signaling Pathway

OBJECTIVE:To s tudy the effects of Dracocephalum moldavica total flavonoids (TFDM)on AMPK/SIRT 1/PGC-1α signaling pathway ,and to explore the mechanism of its protective effect on myocardial ischemia reperfusion injury (MIRI)rats. METHODS:Totally 50 healthy male SD rats were randomly divided into sham operation group ,model group ,TFDM group [ 60 mg/(kg·d),by extract] ,Compound C+TFDM group [ig administration of 60 mg/(kg·d)TFDM+intravenous injection of 250 μg/kg Compound C(AMPK inhibitor )via tail vein 15 min before reperfusion] ,EX-527+TFDM group [ig administration of 60 mg/ (kg·d)TFDM+ip injection of 5 mg/kg EX- 527(SIRT1 inhibitor)20 min before reperfusion] ,with 10 rats in each group. They were given relevant medicine intragastrically ,once a day ,for consecutive 7 days. After last ig administration ,sham operation group underwent sham operation ,other 4 groups were established MIRI model by ligating left anterior descending coronary artery , ischemia for 30 min and reperfusion for 2 h. After reperfusion ,the myocardial histopathological changes were observed by HE staining;RP-HPLC method was used to determine the contents of ATP ,ADP,AMP and NAD + in cardiac tissue. mRNA expressions of AMPK ,SIRT1 and PGC- 1α were detected by quantitative real-time PCR assay. Western blotting assay was the expressions of SIRT 1 and PGC- 1α protein in myocardium. RESULTS: Compared with sham operation group , model group showed myocardial fib ers arranged disorder and horizontal stripes disappearance ,cell swelling burst and necrosis ,and nuclei deformation displacement ;the contents of ATP and NAD+,mRNA expression of AMPK ,SIRT1 and PGC- 1α,protein expression of SIRT 1 and PGC- 1α in cardiac tissue were decreased significantly (P<0.05 or P<0.01);the contents of ADP and AMP ,the phosphorylation level of AMPK protein were increased significantly (P<0.01). Compared with model group ,myocardial pathological morphology were improved significantly in TFDM group ;the contents of ATP and NAD + in cardiac tissue ,mRNA expression of AMPK ,SIRT1 and PGC- 1α,the phosphorylation level of AMPK protein ,the protein expression of SIRT 1 and PGC- 1α were increased significantly(P<0.05 or P< 0.01),while the contents of ADP and AMP were decreased significantly (P<0.01). Compared with TFDM group ,improvement effects of Compound C + TFDM group and EX- 527 + TFDM group on above indexes were reversed (P<0.05 or P<0.01). CONCLUSIONS:TFDM may play a protective role on myocardium by activating AMPK/SIRT 1/PGC-1α signaling pathway and regulating energy metabolism.
5. Rapid detection of variola virus by revolution transcription loop mediated isothermal amplification method

Objective:
To establish the loop-mediated isothermal amplification (LAMP) method for detection of variola virus.
Methods:
One set of primers were designed for recognizing 5 distinct sequences on variola virus-specific gene HA. To optimize the reaction temperature and primers screening, and the sensitivity and specificity of this method for variola virus (VARV) detection were evaluated.
Results:
Rapid detection of variola virus by LAMP assay was completed within 60 min at 63 ℃. The sensitivity of LAMP with detection limits of 1 pg/μl was 10 times higher than that of PCR, that is, the LAMP sensitivity was 3.37×105 copies/μl, and the PCR sensitivity was 3.37×106 copies/μl. and the result of 2 kinds of other virus were negative, showing that it had a good specificity.
Conclusions
The method reported here demonstrates a potential and valuable means for detection of VARV. The LAMP assay is suitable for wide-area sample screening and on-site detection of variola virus in grassroots units, for on-site and primary quarantine, medical units for rapid diagnosis.
6.Study on Detection Value of Platelet Activation Indexes and Platelet Parameters in Patients with Hepatitis B-induced Cirrhosis

Objective:To study the detection value of platelet activation indexes and platelet parameters in patients with hepatitis B-induced cirrhosis.Methods:40 patients with hepatitis B-induced cirrhosis in our hospital from November 2015 to August 2016 were selected as the observation group.40 healthy subjects went through physical examination during the same period were chosen as the control group.The platelet activation indexes and platelet parameters of subjects with same age in two groups were detected and compared.The detection results of the two items in patients with different stages of hepatitis B-induced cirrhosis were compared,too.Results:The PLT of observation group was lower than that of control group,while the other platelet parameters and platelet activation indexes were all higher than those of control group.The detection results of two items in patients with different stages of hepatitis B-induced cirrhosis were with significant differences,all P<0.05.Conclusion:The platelet activation indexes and platelet parameters in patients with hepatitis B-induced cirrhosis are obviously abnormally expressed.There are significant differences among the detection results in patients with different stages.Therefore,surveillance of those indexes in patients with hepatitis B-induced cirrhosis should be strengthened.
7.Pollution characteristics of aerosols and variations of polycyclic aromatic hydrocarbons in the cabins of a certain submarine during submergence

Objective To investigate the pollution caused by the aerosols and variations of polycyclic aromatic hydrocarbons (PAHs) in a certain submarine cabins,and analyze the pollution source of PAHs,then to evaluate the toxicology of the contaminated submarine air.Methods Andersen cascade sampler was used to collect atmospheric particulate matter samples,during 6 days of continuous submergence of the submarine.The composition and variation of PAHs were analyzed by gas chromatography-mass spectrometry (GC-MS).Research on the main pollution sources of PAHs was made by using principal component analysis.Results Aerosol pollution in various cabins of the submarine was quite serious,furthermore it displayed the feature of pollution by different particle sizes.Particle size distribution analysis revealed that the 8-man cabin,meeting room,command room,torpedo room and engine room were of a unimodal size distribution,and the galley and hot air engine room displayed 3 relatively high peaks.Total PAHs were detected,with the concentration scope from 33.1 ng/m3 to 90.8 ng/m3 and with the 3-ring and 4-ring as the primary components.Total TEQ concentration ranged from 0.077 ng/m3 to 0.251 ng/m3.Conclusions Aerosol pollution in the cabins of the submarine was serious,with the cooking mist and volatile components of fuel as the main sources of PAHs and electrical appliances and laundry detergents as secondary source of pollution.
