Objective:To investigate auricular acupressure combined with massage-tapping exercises on body indicators, blood lipids, and liver function in patients with obesity-related fatty liver disease. Methods:This retrospective study included 78 patients with obesity-related fatty liver disease admitted to Hangzhou Hospital of Traditional Chinese Medicine from April to December 2023. The patients were divided into two groups based on different treatment methods: the conventional group (36 patients receiving conventional interventions) and the combined group (36 patients receiving auricular acupressure combined with massage-tapping exercises in addition to conventional interventions). The total response rates, physical indicators (body weight, body mass index), blood lipid levels (triglycerides, total cholesterol, high-density lipoprotein cholesterol, and low-density lipoprotein cholesterol), and liver function indicators (aspartate aminotransferase, alanine aminotransferase, and gamma-glutamyltransferase were recorded and compared between the two groups. Results:The total response rate in the combined group was 94.87% (37/39), which was significantly higher than the 76.92% (30/39) in the conventional group ( χ2 = 5.19, P < 0.05). After intervention, the combined group showed lower body weight [(70.42 ± 8.32) kg vs. (75.84 ± 8.46) kg], lower body mass index [(24.32 ± 1.26) kg/m2 vs. (25.48 ± 1.30) kg/m2], lower triglycerides [(1.62 ± 0.42) mmol/L vs. (2.94 ± 0.78) mmol/L], lower total cholesterol [(5.24 ± 0.54) mmol/L vs. (6.72 ± 0.66) mmol/L], lower low-density lipoprotein cholesterol [(2.14 ± 0.46) mmol/L vs. (2.78 ± 0.50) mmol/L], lower aspartate aminotransferase [(35.32 ± 4.38) U/L vs. (40.16 ± 5.14) U/L], lower alanine aminotransferase [(31.05 ± 4.20) U/L vs. (37.24 ± 4.56) U/L], and lower gamma-glutamyltransferase [(43.35 ± 8.74) U/L vs. (57.62 ± 9.04) U/L] levels compared with the conventional group. The combined group showed higher high-density lipoprotein cholesterol level compared with the conventional group [(1.85 ± 0.26) mmol/L vs. (1.26 ± 0.30) mmol/L]. All differences were statistically significant ( t = 2.85, 4.00, 9.31, 10.84, 5.88, 4.48, 6.24, 7.09, 9.28, all P < 0.05). Conclusions:The combination of auricular acupressure and massage-tapping exercises is effective in treating obesity-related fatty liver disease in patients. This approach can regulate physical indicators, improve lipid metabolism, and enhance liver function, making it a method worth promoting in clinical practice.

Objective To explore the effective substances that specifically bind to RSV-F protein in the water extract of Forsythia suspensa, and verify its effect. Methods Surface plasmon resonance(SPR)affinity fishing technology was used to enrich, regenerate and recover the active components that specifically bind to RSV-F protein in the water extract of Forsythia suspensa. The recovered samples were analyzed and identified by UHPLC-MS/MS,and the active components that stably bind to F protein were screened by molecular docking, molecular dynamics simulation and SPR affinity detection, and then returned to animal experiments for pharmacodynamics verification.BALB/c mice were randomly divided into normal group, model group, positive control group, protocatechuic acid group and verbascoside group. In the experimental group,RSV was dripped into the nose to make models. Each drug group began to be given intragastric intervention on the day of modeling, and samples were taken after 4 days.HE staining was used to observe the pathological changes of lung tissue, and ELISA was used to detect the changes of inflammatory factors in lung tissue. Results A variety of active components in the aqueous extract of Forsythia suspensa could bind to RSV-F protein, among which the results of verbascoside and protocatechuic acid were more stable. In vivo pharmacodynamics experiments showed that the lung tissue of the model group had obvious pathological changes and the level of inflammatory factors increased, and the pathological state was improved after the intervention of verbascoside and protocatechuic acid compared with the normal group. Conclusion Forsythia suspensa can interfere with the expression of RSV-F protein through many active components such as verbascoside and protocatechuic acid, which may be the material basis of its anti-RSV.

Objective:
To construct a Family with sequence similarity 50 member A(FAM50A) gene knockout mouse insulinoma pancreatic β-cell line Beta-TC-6 using CRISPR/Cas9 gene editing technology and to prepare polyclonal antibodies specifically recognizing FAM50A.
Methods:
Two guide RNAs(sgRNAs) targeting the FAM50A gene were designed, and a recombinant plasmid expressing blue fluorescent protein(BFP) was constructed for gene knockout. The successfully constructed plasmid was transfected into Beta-TC-6 cells, and BFP-positive single cells were isolated for clonal expansion. The expanded monoclonal cell lines were genotyped by Sanger sequencing, and FAM50A protein expression was assessed by Western blot.Purified human recombinant FAM50A protein was used to immunize New Zealand rabbits for the preparation of a polyclonal antibody. The specificity of the prepared antibody was then validated using the successfully established FAM50A knockout cell line.
Results:
A monoclonal cell line with a successful knockout of the FAM50A gene was identified.Sanger sequencing confirmed base deletions at the target site.Western blot analysis showed a complete absence of FAM50A protein expression in this cell line. The prepared polyclonal antibody successfully recognized endogenous murine FAM50A protein in wild-type Beta-TC-6 cells and in hTERT-RPE1 cells overexpressing human FAM50A-GFP fusion protein, while no signal was detected in the FAM50A knockout cells.
Conclusion
This study successfully established a FAM50A gene knockout Beta-TC-6 cell model and generated a FAM50A polyclonal antibody, providing powerful tools for future research.

Objective:
To investigate hepatocellular carcinoma ( HCC ) risk factors in hepatitis B e antigen (HBeAg)-negative cirrhotics , and to develop and validate a predictive model using these indicators .
Methods:
A total of 649 hospitalized patients with HBeAg-negative hepatitis B cirrhosis and HBeAg-negative primary HCC were enrolled . Patients were randomly divided into a modeling group (n = 298) and a validation group (n = 351) at a 7 ∶3 ratio . Logistic regression analysis was used to screen for independent predictors of HCC occurrence . A predic- tive model was constructed and validated using receiver operating characteristic ( ROC) curves . The clinical net benefit of the prediction model was assessed via decision curve analysis .
Results:
Univariate analysis showed sig- nificant statistical differences between the modeling and validation groups in serum alanine aminotransferase (ALT) , aspartate aminotransferase ( AST) , triglycerides ( TG) , gamma-glutamyl transferase ( GGT) , red blood cell count (RBC) , hemoglobin (Hb) , platelet count (PLT) , international normalized ratio (INR) , alpha-feto- protein (AFP) , serum calcium (Ca2 + ) , serum cholinesterase (CHE) , and HBV DNA levels . Multivariate logistic regression analysis identified AST , GGT , Hb , PLT , Ca2 + , CHE , and HBV DNA as independent influencing fac- tors for HCC occurrence (P < 0. 05) , with OR (95% CI) of 1 . 002 ( 1 . 000 - 1 . 005) , 1 . 006 ( 1 . 003 - 1 . 008) , 0. 994 (0. 988 - 0. 999) , 0. 984 (0. 981 - 0. 988) , 9. 624 (3 . 821 - 24. 245 ) , 0. 999 (0. 987 - 0. 999) , and 7. 530 (4. 143 - 13 . 687) , respectively. A nomogram prediction model was established based on these seven indi- cators . The area under the ROC curve was 0. 936 in the modeling group and 0. 941 in the validation group . Cali- bration curves demonstrated high predictive accuracy of the nomogram.
Conclusion
AST , GGT , Hb , PLT , Ca2 + , CHE , and HBV DNA are independent risk factors for HCC development in patients with HBeAg-negative hepatitis B-related cirrhosis . The established non-invasive prediction model exhibits good discriminative ability and clinical utility , providing an experimental basis for early detection and preventive screening of HCC in this patient population .
