1.Rapid Discrimination of Processing Degree of Wine-processed Chuanxiong Rhizoma Based on Intelligent Sensory Technology and Multivariate Statistical Analysis

ObjectiveTo explore the changes in color, odor and chemical components during wine-processing of Chuanxiong Rhizoma(CR), identify differential markers, and provide a basis for standardizing the process and establishing quality standards. MethodsFifteen batches of CR samples from 4 producing areas were collected. Colorimeter and electronic nose were used to detect the color changes and odor components of CR before and after wine-processing. Multivariate statistical methods including partial least squares-discriminant analysis(PLS-DA), principal component analysis(PCA), discriminant factor analysis(DFA) and Fisher discriminant analysis were applied to identify wine-processed CR at different processing stages and establish discriminant models, and differential components were screened out based on variable importance in the projection(VIP) value1. Then, high performance liquid chromatography(HPLC) was employed to detect the content changes of four components(ferulic acid, senkyunolide I, senkyunolide A and ligustilide) during the processing stages. ResultsThe differences of wine-processed CR at various stages were primarily reflected in color parameters L*(brightness value), a*(red-green value) and b*(yellow-blue value). Based on chromaticity differences, the color reference ranges were established for moderately processed CR, including L* of 46.75-48.24, a* of 5.37-6.07 and b* of 20.32-21.70. In odor analysis, DFA revealed significant differences among processing stages, and 11 odor markers were identified, with four differential markers(4-hydroxy-3-butylphthalide, isopropyl butyrate, L-limonene and 1-methoxyhexane) based on VIP values. HPLC results showed that there was no significant difference of the four components except for ligustilide in wine-processed CR at different stages. ConclusionThis study achieved rapid identification of wine-processed CR with different processing degrees by electronic sensory technology and differential component content detection, with discrimination accuracy rates of 92.4% and 93.272% for color and odor, respectively. This paper also established the reference ranges of main colorimetric parameters for wine-processed CR at different stages, and four differential components were screened out, providing a basis for standardizing the processing of wine-processed CR and establishing quality standards for this decoction pieces.

3.Effects of inflammation on serum hepcidin and iron metabolism related parameters in patients with type 2 diabetes mellitus:a meta-analysis

OBJECTIVE:Disorders in iron metabolism increase the risk of type 2 diabetes mellitus.Hepcidin play an important role in maintaining iron homeostasis in the body,but its level increases with increased inflammation.Changes in hepcidin and iron homeostasis and the extent of their association with inflammation in people with and without type 2 diabetes mellitus are unknown.Meta-analysis was used to evaluate the effect of inflammation on serum hepcidin and iron metabolism related parameters in patients with type 2 diabetes mellitus.METHODS:CNKI,PubMed,Web of Science and EBSCOhost databases were searched by computer to collect observational studies related to inflammatory index and hepcidin in patients with type 2 diabetes mellitus.The search time was from September 1,2000 to September 30,2024.Three researchers independently screened the literature,extracted data and evaluated the quality of the included literature.Meta-analysis was performed by Review Manager 5.3,Stata 17.0 and GraphPad Prism 8.0.2 software.RESULTS:A total of 15 articles(17 studies)involving 3 159 participants,including 1 357 patients with type 2 diabetes mellitus,were included.Meta-analysis results showed that compared with the control group,patients with type 2 diabetes mellitus had higher levels of serum hepcidin[standardized mean difference(SMD)=0.35,95%confidence interval(CI)(0.05,0.65),P<0.05],serum ferritin(SMD=0.49,95%CI(0.21,0.78),P<0.01)and serum transferrin(SMD=0.19,95%CI(0.00,0.37),P<0.05).Subgroup analysis results indicated that inflammation had a significant effect on serum hepcidin(SMD=0.76,95%CI(0.17,1.34),P<0.05)and serum ferritin(SMD=0.77,95%CI(0.06,1.47),P<0.05)in patients with type 2 diabetes mellitus.CONCLUSION:Hepcidin concentration is positively correlated with type 2 diabetes mellitus.Inflammation is one of the risk factors of type 2 diabetes mellitus.Early prevention of inflammation has certain significance in preventing iron metabolism disorder in patients with type 2 diabetes mellitus.

Objective: Intrathyroid thymic carcinoma(ITTC) is a rare thyroid tumor that lacks typical clinical manifestations and imaging features, making preoperative diagnosis challenging.The primary treatment for ITTC is radical surgery; however, the effectiveness of adjuvant radiotherapy and chemotherapy post-surgery is not well-established. This paper presents a case of ITTC , analyzing the clinical data and correlating it with the literature to explore the clinical manifestations, diagnostic approach, treatment, and prognosis of ITTC.

6.Disrupting calcium homeostasis and glycometabolism in engineered lipid-based pharmaceuticals propel cancer immunogenic death.

Homeostasis and energy and substance metabolism reprogramming shape various tumor microenvironment to sustain cancer stemness, self-plasticity and treatment resistance. Aiming at them, a lipid-based pharmaceutical loaded with CaO2 and glucose oxidase (GOx) (LipoCaO2/GOx, LCG) has been obtained to disrupt calcium homeostasis and interfere with glycometabolism. The loaded GOx can decompose glucose into H2O2 and gluconic acid, thus competing with anaerobic glycolysis to hamper lactic acid (LA) secretion. The obtained gluconic acid further deprives CaO2 to produce H2O2 and release Ca2+, disrupting Ca2+ homeostasis, which synergizes with GOx-mediated glycometabolism interference to deplete glutathione (GSH) and yield reactive oxygen species (ROS). Systematical experiments reveal that these sequential multifaceted events unlocked by Ca2+ homeostasis disruption and glycometabolism interference, ROS production and LA inhibition, successfully enhance cancer immunogenic deaths of breast cancer cells, hamper regulatory T cells (Tregs) infiltration and promote CD8+ T recruitment, which receives a considerably-inhibited outcome against breast cancer progression. Collectively, this calcium homeostasis disruption glycometabolism interference strategy effectively combines ion interference therapy with starvation therapy to eventually evoke an effective anti-tumor immune environment, which represents in the field of biomedical research.
7.CDH17-targeting CAR-NK cells synergize with CD47 blockade for potent suppression of gastrointestinal cancers.

Gastrointestinal (GI) cancers are a leading cause of cancer morbidity and mortality worldwide. Despite advances in treatment, cancer relapse remains a significant challenge, necessitating novel therapeutic strategies. In this study, we engineered nanobody-based chimeric antigen receptor (CAR) natural killer (NK) cells targeting cadherin 17 (CDH17) for the treatment of GI tumors. In addition, to enhance the efficacy of CAR-NK cells, we also incorporated CV1, a CD47-SIRPα axis inhibitor, to evaluate the anti-tumor effect of this combination. We found that CDH17-CAR-NK cells effectively eliminated GI cancers cells in a CDH17-dependent manner. CDH17-CAR-NK cells also exhibit potent in vivo anti-tumor effects in cancer cell-derived xenograft and patient-derived xenograft mouse models. Additionally, the anti-tumor activity of CDH17-CAR-NK cells is synergistically enhanced by CD47-signal regulatory protein α (SIRPα) axis inhibitor CV1, likely through augmented macrophages activation and an increase in M1-phenotype macrophages in the tumor microenvironment. Collectively, our findings suggest that CDH17-targeting CAR-NK cells are a promising strategy for GI cancers. The combination of CDH17-CAR-NK cells with CV1 emerges as a potential combinatorial approach to overcome the limitations of CAR-NK therapy. Further investigations are warranted to speed up the clinical translation of these findings.
8.AConvLSTM U-Net: a multi-scale jaw cyst segmentation model based on bidirectional dense connection and attention mechanism.

OBJECTIVES:
We propose a multi-scale jaw cyst segmentation model, AConvLSTM U-Net, which is based on bidirectional dense connections and attention mechanisms to achieve accurate automatic segmentation of mandibular cyst images.
METHODS:
A dataset consisting of 2592 jaw cyst images was used. AConvLSTM U-Net designs a MBC on the encoding path to enhance feature extraction capabilities. A DPD was used to connect the encoder and decoder, and a bidirectional ConvLSTM was introduced in the jump connection to obtain rich semantic information. A decoding block based on scSE was then used on the decoding path to enhance the focus on important information. Finally, a DS was designed, and the model was optimized by integrating a joint loss function to further improve the segmentation accuracy.
RESULTS:
The experiment with AConvLSTM U-Net for jaw cyst lesion segmentation showed a MCC of 93.8443%, a DSC of 93.9067%, and a JSC of 88.5133%, outperforming all the other comparison segmentation models.
CONCLUSIONS
The proposed algorithm shows a high accuracy and robustness on the jaw cyst dataset, demonstrating its superior performance over many existing methods for automatic segmentation of jaw cyst images and its potential to assist clinical diagnosis.

9.Protective Effect and Mechanism of Cynarin on Septic Cardiomyopathy in Mice

Objective To explore the effect and potential mechanisms of cynarin on septic cardiomyopathy.Methods Seventy-eight male C57BL/6mice were divided into three groups using a random number table method:control group,model group,and treatment group,with 26mice in each group.The treatment group received daily intraperitoneal injections of cynarin at a dose of 25 mg/(kg·d)for 7days,while control group and model group received daily intraperitoneal injections with the same amount of saline.One hour after the in-traperitoneal injection of cynarin on the 7th day,the model group and treatment group were intraperitoneally injected with lipopolysaccha-ride(LPS)at a dose of 10mg/kg to induce septic cardiomyopathy.12hours later,6mice were randomly selected from each group to test cardiac function,and the remaining mice were monitored for 7-day survival.Hematoxylin-eosin staining was used to observe the histo-logical structure of mouse myocardium;Enzyme linked immunosorbent assay were used to detect the levels of inflammatory factors,mark-ers of myocardial injury,markers of myocardial oxidative stress;determination of tissue iron content by the colourimetric method;real-time fluorescence quantitative polymerase chain reaction(RT-qPCR)was used to detect the mRNA levels of myocardial inflammatory factors;Terminal Deoxynucleotidyl Transferase-Mediated dUTP Nick End Labeling(TUNEL)staining was used to detect the apoptosis of cardiac myocytes;Dihydroethidium(DHE)staining was used to detect the levels of reactive oxygen species;and Western blot was used to detect the protein expression levels of B-cell lymphoma-2(Bcl-2),Bcl-2-Associated X(Bax),glutathione peroxidase 4(GPX4),recombinant solute carrier family 7,member 11(SLC7A11),ferritin heavy chain 1(FTH1),transferrin receptor(TFR),kelch-like ECH associated protein 1(Keap1),nuclear factor erythroid 2-related factor 2(Nrf2)protein expression levels.Results Compared with the control group,mice in the model group had lower left ventricular ejection fraction and short-axis shortening rate,higher lactate dehydrogenase and creatine kinase-MB,higher content and mRNA expression of inflammatory factors,decreased survival,significant myocardial pathological damage and increased apoptotic cells,enhanced oxidative stress,inhibition of myocardial GPX4,SLC7A11,FTH1 and Nrf2 expression,and up-regulation of TFR,Keap1 expression,and increased iron content(P<0.05);Compared with the model group,mice in the treatment group had increased survival rate,significantly improved cardiac dysfunction and myocardial injury,and reduced inflammatory injury,apoptosis and oxidative stress,as well as up-regulated expression of GPX4,SLC7A11,FTH1 and Nrf2,suppressed expression of TFR and Keap1,and decreased iron content(P<0.05).Conclusion Cynarin may alleviate inflam-mation,apoptosis and oxidative stress by inhibiting Keap1/Nrf2-mediated ferroptosis,and protect septic cardiomyopathy.
10.Novel Retrograde Intramedullary Nail on Medial Femur Condyle for Internal Fixation of Distal Femur Type A Fracture:A Finite Element Analysis

Objective To compare the biomechanical characteristics of novel retrograde intramedullary nail,common femoral retrograde intramedullary nail,and medial femoral condyle locking plate for treating distal femur type A fractures by finite element analysis,and study the advantages of retrograde intramedullary nail on medial femoral condyle.Methods A novel retrograde intramedullary nail on medial femur condyle was designed.CT scan was performed on lower limb bones of a male volunteer,and a three-dimensional(3D)model of the femur was established.The model was then segmented to create models of distal femur type A2 and A3 fractures.The 3D models of internal fixator were established.The small and standard retrograde intramedullary nail on medial femoral condyle,common femoral retrograde intramedullary nail,locking plate on medial femoral condyle were used for internal fixation on A2 type fracture.A3 type fracture was fixed with one of the above internal fixators combined with lateral condylar plate respectively.The 600 N axial load and 4 N·m torsional load were applied to the models.The displacement and stress of femurs,the displacement and stress of internal fixators,the micro-momentum between the internal fixator and the bone,the relative displacement of the two fracture broken ends were observed in each group.Results Among the A2 and A3 type fractures under axial loads,the correponding standard group and corresponding standard combined group had the smallest peak displacement and peak stress of internal fixation.Among the A2 and A3 type fractures under torsional loads,the correponding standard group and corresponding standard combined group had the smallest peak displacement of the femur and internal fixation.The relative displacement of the two broken ends in the standard combined group was the smallest.Conclusions Compared with femoral medial condylar locking plate and common retrograde intramedullary nail,this novel retrograde intramedullary nail on medial femur condyle has a mechanical advantage of reducing stress concentration and reducing the risk of internal fixation failure,and it can be used alone for distal femur type A2 fracture,or in combination with the lateral condylar plate to fix distal femur type A3 fracture.
