Objective :
To characterize MMP12 as a pan-cancer marker and assess its screening value as a tumor serum marker.
Methods :
Bioinformatics tools such as GEPIA2,GSCA,cBioPortal,and GeneMANIA were used to analyze the pan-cancer features of MMP12 in TCGA datasets, including encompassing differential gene expression analysis, prognostic analysis,DNA methylation analysis, gene structural variation analysis and immune microenvironment analysis.Furthermore, serum samples we collected from patients with lung adenocarcinoma, breast invasive carcinoma, esophageal squamous cell carcinoma, stomach adenocarcinoma, liver hepatocellular carcinoma, and healthy individuals.ELISA was used to detect MMP12 expression in serum, and the screening performance was evaluated using the area under the ROC curve.Additionally,we followed up 28 ESCC patients and compared serum MMP12 levels between 19 patients with disease progression and 9 patients with stable disease.
Results:
The pancancer feature analysis revealed a significant negative correlation between MMP12 mRNA expression and its promoter DNA methylation(P<0.05),as well as a positive correlation with gene copy number variations(P<0.05).MMP12 mRNA expression was up-regulated in 14 cancer tissues compared to normal tissues next to cancer(P<0.05) and was associated with poor prognosis of cancer patients(P<0.05).Immunocorrelation analysis showed that MMP12 was significantly associated with immunity, infiltration of stromal cells, tumor mutational burden(TMB) and microsatellite instability(MSI)(P<0.05).ROC curve analysis indicated that MMP12 could serve as a potential biomarker for screening lung adenocarcinoma, breast invasive carcinoma, esophageal squamous cell carcinoma, stomach adenocarcinoma, and liver hepatocellular carcinoma. In a 30-month follow-up study of esophageal squamous cell carcinoma patients, the expression of MMP12 was higher in the disease progression group than that in the stable group.
Conclusion
MMP12 serves as a potential prognostic and screening marker of pan-cancer.

Radiotherapy can cause varying degrees of damage to surrounding normal tissues while treating tumors. In recent years, single-cell RNA sequencing (scRNA-Seq), an emerging technology in modern biology, has gradually played a significant role in research on radiation-induced injuries. Researchers have delved into a variety of radiation-induced brain injuries, including radiation-induced brain injury (RIBI), cardiovascular injury, lung injury, intestinal injury, skin injury, and hematopoietic injury. Their studies provide novel perspectives for revealing the mechanisms behind radiation-induced injuries, assessing the degree of injury, and exploring potential therapies. This study reviews the advances in the application of scRNA-seq to research on radiation-induced injuries.

The Janus kinase/signal transducers and activators of transcription (JAK-STAT) control natural killer (NK) cells development and cytotoxic functions, however, whether long non-coding RNAs (lncRNAs) are involved in this pathway remains unknown. We found that miR155HG was elevated in activated NK cells and promoted their proliferation and effector functions in both NK92 and induced-pluripotent stem cells (iPSCs)-derived NK (iPSC-NK) cells, without reliance on its derived miR-155 and micropeptide P155. Mechanistically, miR155HG bound to miR-6756 and relieved its repression of JAK3 expression, thereby promoting the JAK-STAT pathway and enhancing NK cell proliferation and function. Further investigations disclosed that upon cytokine stimulation, STAT3 directly interacts with miR155HG promoter and induces miR155HG transcription. Collectively, we identify a miR155HG-mediated positive feedback loop of the JAK-STAT signaling. Our study will also provide a power target regarding miR155HG for improving NK cell generation and effector function in the field of NK cell adoptive transfer therapy against cancer, especially iPSC-derived NK cells.

Objective To explore the efficacy of 4R technology combined with prone five-direction cer-vical muscle strength training for neck type cervical spondylosis. Methods A total of 112 patients with neck and shoulder pain who visited the Affiliated Shapingba Hospital of Chongqing University and the Shapingba District Traditional Chinese Medicine Hospital in Chongqing from January to November 2024 were selected as research subjects. They were randomly assigned using a random number table method into an observation group and a control group, with 56 patients in each group. The observation group received 4R technology com-bined with prone five-direction cervical muscle strength training, while the control group received conventional rehabilitation treatment. Both interventions lasted for 4 weeks.Differences in Visual Analogue Scale(VAS)scores,Neck Disability Index(NDI)scores, and cervical range of motion in flexion, extension, lateral bending, and rota-tion were assessed before treatment and at 1 and 6 months after treatment for both groups. Results At 1 and 6 months of treatment,VAS scores in both groups decreased compared to before treatment, with the observation group lower than the control group, and the difference was statistically significant(P<0.05).At 1 and 6 months of treatment,NDI scores in both groups decreased compared to before treatment, with the observation group lower than the control group, and the difference was statistically significant(P<0.05).At 1 and 6 months of treatment, cervical flexion, cervical extension, and cervical lateral flexion in both groups increased compared to before treatment, and cervical rotation in the observation group increased compared to before treatment.Cervical extension and cervical rotation in the observation group were greater than those in the con-trol group, and only at 6 months of treatment was cervical lateral flexion in the observation group greater than in the control group, with all differences statistically significant(P<0.05).Conclusion The 4R technology combined with prone five-direction cervical muscle strength training can effectively improve cervical-type cer-vical spondylosis.

Objective To evaluate the safety, effectiveness and feasibility of robotic-assisted kidney transplantation (RAKT). Methods Clinical data of 16 patients who underwent kidney transplantation were collected. Among them, 8 recipients received RAKT (RAKT group) and 8 cases underwent open kidney transplantation (OKT) with the contralateral kidney from the same donor (OKT group). Perioperative status and the recovery of renal allograft function were compared between two groups. Results All patients successfully completed the surgery. In the RAKT group, no patient was converted to open surgery. The operation time in the RAKT group was longer than that in the OKT group (P=0.015). No significant differences were observed in the serum creatinine levels before surgery and upon discharge between two groups (both P>0.05). In the OKT group, one recipient developed delayed graft function (DGF), and the remaining recipients did not experience perioperative complications. No significant difference was noted in the short-term recovery of renal allograft function between two groups (P>0.05). Conclusions Postoperative recovery of the recipients in the RAKT group is equivalent to that of their counterparts in the OKT group. RAKT is a safe and effective procedure for the team expertise in kidney transplantation.
