Objective: To analyze the compliance status of physical fitness indicator and lifestyle characteristics among secondary vocational school students, as well as their associations, so as to provide a reference for physical fitness promotion among secondary vocational school students. Methods: Using a convenience sampling method, a questionnaire survey and physical fitness tests were conducted from September to December 2024 among 5 141 secondary vocational school students recruited from 16 secondary vocational schools across 8 provinces/municipalities in China, including Fujian, Chongqing, Guangdong, Guangxi, Hubei, Jiangxi, Shanghai and Zhejiang. Data on lifestyle factors were collected using a self designed questionnaire combined with standardized scales, including dietary habits, unhealthy behaviors, and 24 hour behaviors (physical activity, screen time, and sleep behavior). Multivariable Logistic regression model was used to analyze the associations of individual lifestyle behaviors and clustering of unhealthy lifestyle behaviors with non-compliance in physical fitness indicators among secondary vocational school students. Results: The compliance rates for waist circumference, vital capacity, grip strength, standing long jump, 30 second sit ups, 20 meter shuttle run, 50 meter dash, sit and reach, 20 second side steps among secondary vocational school students were 55.9%, 89.6%, 81.8%, 79.4%, 77.8%, 61.4%, 87.0%, 12.6%, and 63.5%, respectively, with a non-overweight/obesity rate of 78.9%. The reporting rates of daily breakfast consumption, milk consumption, and vegetable consumption were 58.9%, 18.3%, and 37.5%, respectively. The reporting rates of frequent takeout consumption, frequent sugar sweetened beverage consumption, smoking, alcohol drinking, insufficient moderate to vigorous physical activity (MVPA), excessive screen time, and insufficient sleep were 30.0%, 43.8%, 8.4%, 15.8%, 42.9%, 90.8%, and 42.6%, respectively. Multivariable Logistic regression analysis showed that secondary vocational school students with insufficient MVPA had higher risks of non-compliance in vital capacity, grip strength, standing long jump, 30 second sit ups, 20 meter shuttle run, 50 meter dash, and 20 second side steps ( OR =1.22- 1.60 ); secondary vocational school students who consumed breakfast daily had lower risks of non-compliance in vital capacity and overweight/obesity ( OR =0.81, 0.85); daily vegetable consumption was associated with a lower risk of vital capacity non-compliance ( OR =0.72); frequent takeout consumption was associated with an increased risk of non-compliance in the sit and reach test ( OR =1.28); smoking was associated with increased risks of non-compliance in the 20 meter shuttle run and 20 second side steps ( OR =1.38, 1.58) (all P <0.05). Regarding the clustering of unhealthy behaviors, secondary vocational school students with 4- 5 clustered unhealthy behaviors had higher risks of non-compliance in waist circumference and standing long jump; those with≥6 clustered behaviors had higher risks of non-compliance in standing long jump, 20 meter shuttle run, and 20 second side step ( OR = 1.17-1.36, all P <0.05). Conclusions The clustering of unhealthy behaviors is negatively associated with physical fitness status among secondary vocational school students. Secondary vocational schools should prioritize the promotion of comprehensive lifestyle management to improve the physical fitness levels of secondary vocational school students.

Objective:To explore the prognostic value of long non-coding RNA (lncRNA) AL445524.1 in hepatocellular carcinoma (HCC) and its regulatory effect on liver cancer cells.Methods:Based on the TCGA database, bioinformatics analysis was conducted to assess the expression levels of AL445524.1 in HCC. Using the median expression level of AL445524.1 as the cut-off value, 343 HCC patients were divided into high expression group and low expression group. Survival analysis was performed using Kaplan-Meier curves. Univariate and multivariate Cox proportional hazards regression models were employed to analyze the relationship between AL445524.1 expression, other clinical characteristics, and patients' prognosis, identifying independent risk factors for the prognosis of HCC patients. Based on the results of the multivariate analysis, patients with complete data were randomly divided into a testing set ( n=215) and a training set ( n=92) in a 7∶3 ratio using a random number table method, and a nomogram prognostic prediction model for HCC was constructed. Receiver operator characteristic (ROC) curves were plotted to calculate the area under the curve (AUC) to analyze and validate the predictive performance of the model. Functional enrichment analysis was conducted on the differentially expressed genes regulated by AL445524.1. The expression level of AL445524.1 in liver cancer cells was detected using RT-PCR. Human hepatoma cell lines HCCLM3 were divided into si-AL445524.1-1, si-AL445524.1-2, si-AL445524.1-3 and si-NC groups. The CCK-8 assay was used to assess cell proliferation capability; cell scratch and Transwell assays were performed to evaluate cell migration and invasion abilities; and flow cytometry was utilized to detect cell apoptosis. Results:The expression of AL445524.1 in liver cancer tissues was significantly higher than that in para-carcinoma tissues (4.38±1.26 vs. 2.08±0.45, t=24.58, P<0.001). Kaplan-Meier survival analysis showed that the 5-year overall survival (OS) rate for HCC patients in high AL445524.1 expression group ( n=170) was 37.37%, and that of patients in low AL445524.1 expression group ( n=173) was 58.38%, with a statistically significant difference ( χ2=8.83, P=0.003). Multivariate analysis showed that tumor recurrence ( HR=2.58, 95% CI: 1.64-4.07, P<0.001), clinical stage ( HR=2.49, 95% CI: 1.63-3.81, P<0.001), and AL445524.1 expression ( HR=1.23, 95% CI: 1.06-1.41, P=0.010) were independent factors affecting the prognosis of HCC patients. A nomogram prognostic prediction model was constructed based on tumor recurrence, clinical stage, and AL445524.1 expression, with the model risk score calculated as: risk score=0.774×tumor recurrence+0.753×clinical stage+0.231×AL445524.1. The prediction model C-index values of 0.726, 0.660, and 0.678 in the training set, testing set, and overall set, respectively. ROC curve analysis showed that the AUC values for the 1-year OS rates in the training set, testing set, and overall set were 0.746, 0.630, and 0.684, respectively; the AUC values for the 3-year OS rates were 0.778, 0.736, and 0.743; and the AUC values for the 5-year OS rates were 0.794, 0.760, and 0.774. In the training set, the test set and the overall set, the predictive performance of the model score in predicting the 5-year OS rate of patients was superior to the individual predictions of AL445524.1 expression, clinical stage and tumor recurrence alone (all P<0.05). The 5-year overall survival (OS) rates of high-risk group ( n=154) and low-risk group ( n=153) were 33.54% and 77.73%, respectively, with a statistically significant difference ( χ2=28.97, P<0.001). GO and KEGG enrichment analysis suggested that the differential expression of AL445524.1 genes in high and low expression groups was mainly related to lipid metabolism and oxidation. GSEA analysis showed that the oxidative phosphorylation pathway was significantly enriched in HCC patients with high expression of AL445524.1. In vitro experiments showed that AL445524.1 expression was higher in liver cancer cells (1.97±0.14) compared to normal liver cells (1.00±0.10), with a statistically significant difference ( t=11.62, P=0.007). Silencing AL445524.1 could significantly inhibit the proliferation, migration, and invasion of liver cancer cells and promote apoptosis. In the CCK-8 proliferation experiment, the A450 values of the si-NC, si-AL445524.1-1, si-AL445524.1-2, and si-AL445524.1-3 groups after 24 hours were 0.433±0.012, 0.377±0.020, 0.383±0.020, and 0.423±0.005, respectively, with a statistically significant difference ( F=20.51, P<0.001). Additionally, the cell proliferation in the si-AL445524.1-1, si-AL445524.1-2, and si-AL445524.1-3 groups was lower than that in the si-NC group (all P<0.001). The cell scratch assay showed that the scratch healing rates of the above 4 groups were 33.60%±6.15%, 21.60%±4.30%, 26.40%± 4.60%, and 27.30%±2.60%, respectively, with a statistically significant difference ( F=42.71, P<0.001). The scratch healing rates of the si-AL445524.1-1, si-AL445524.1-2, and si-AL445524.1-3 groups were also significantly lower than that of the si-NC group (all P<0.001). Transwell migration and invasion experiments revealed that the number of migrated cells in the above 4 groups were 293.50±14.80, 110.50±10.28, 132.44±5.57, and 115.25±8.66, respectively, with a statistically significant difference ( F=374.16, P<0.001). The number of migrated cells in the si-AL445524.1-1, si-AL445524.1-2, and si-AL445524.1-3 groups were significantly lower than that in the si-NC group (all P<0.001). For the invasion assay, the number of invaded cells in the above 4 groups were 247.00±9.49, 119.00±5.57, 153.25±5.85, and 163.67±5.51, respectively, with a statistically significant difference ( F=218.14, P<0.001). The number of invaded cells in the si-AL445524.1-1, si-AL445524.1-2, and si-AL445524.1-3 groups were also significantly lower than that in the si-NC group (all P<0.001). Flow cytometry showed that the apoptosis rates of the above 4 groups were 1.70%±0.08%, 2.17%±0.11%, 2.38%±0.08%, and 2.02%±0.27%, respectively, with a statistically significant difference ( F=29.36, P<0.001). The apoptosis rate in the si-AL445524.1-1, si-AL445524.1-2, and si-AL445524.1-3 groups were significantly higher than that in the si-NC group (all P<0.001) . Conclusion:AL445524.1 can serve as a prognostic marker for HCC. The AL445524.1-related prognostic prediction model demonstrates high accuracy in predicting the 5-year OS rate of HCC patients. Patients with high AL445524.1 expression have poorer survival prognosis. Silencing AL445524.1 can inhibit liver cancer cell proliferation, migration, and invasion while promoting apoptosis, which may be related to cellular lipid metabolism or oxidative phosphorylation.

Objective:To explore the effect of Jian-Pi-Zhi-Dong decoction on autonomous activity and dopamine (dopamine, DA) synaptic vesicle protein expression in the striatum of Tourette syndrome (TS) model rats.Methods:The 4-week-old male SD rats were used to establish the TS model by intraperitoneal injection of N-aminodipropionate. Thirty successfully modeled rats were randomly divided into model group, Chinese medicine group, and tiberide group according to random number table method, with 10 rats in each group. And another 10 rats with matched body mass were selected as the control group. Rats in Chinese medicine group were given Jian-Pi-Zhi-Dong decoction solution (1.6 g/100 g) and the rats in tiapride group were given sulfate tiapride suspension (2.1 mg/100 g), while rats in control group and model group were given an equal volume of 0.9% sodium chloride solution, once a day for 4 weeks.The number of autonomous activities in rats was determined by autonomous activity programmer. ELISA was used to detect the level of DA in the striatum of rats and the expression of dopamine transporter (DAT).Vesicular monoamine transporter-2 (VMAT2) and α-synuclein (α-syn) were measured by Western blot.SPSS 25.0 software was used for data analysis. Multiple group comparisons were performed using one-way ANOVA, non-parametric test and repeated measures ANOVA.Results:Comparing the number of autonomous activities among the 4 groups, the interaction effect between time and group was significant ( F=184.354, P<0.001). At the 1-4 weeks of gavaging, the numbers of autonomic activities in the model group were more than those in the control group (all P<0.05).While the numbers of autonomic activities in Chinese medicine group and tiapride group were less than those in the model group (all P<0.05). Moreover, the numbers of autonomic activities in Chinese medicine group and tiapride group from 1 to 4 weeks were less than those after model making (all P<0.05). The Western blot results showed significant differences in the relative expression of α-syn ( H=29.098), DAT ( F=54.632) and VMAT2 ( H=18.982) among the 4 groups (all P<0.001). The expression levels of α-syn protein in Chinese medicine group and tiapride group were both lower than that in the model group (0.39(0.36, 0.51), 0.39(0.36, 0.50), 0.62(0.50, 0.70)) (both P<0.05). The expression level of DAT protein in Chinese medicine group was higher than that in the model group and lower than that in tiapride group ((0.37±0.06), (0.26±0.07), (0.49±0.09)) (both P<0.05). And the expression level of VMAT2 protein in Chinese medicine group had no significant difference compared with that in the model group ( P>0.05).The ELISA results showed significant differences in DA content of striatum among the 4 groups ( F=75.370, P<0.001). The level of DA in the model group was higher than that in the control group ((7.65±0.72) ng/L, (3.71±0.59) ng/L, P<0.05). The levels of DA in Chinese medicine group ((3.92±0.81) ng/L) and tiapride group ((4.40±0.53) ng/L) were lower than that in the model group (both P<0.05), and the difference between Chinese medicine group and tiapride group was not significant ( P>0.05). Conclusion:Jian-Pi-Zhi-Dong decoction can relieve the tic symptoms of the model rats with TS, and its mechanism may be related to inhibiting the excessive release of α-syn, improving the expression of DAT and VMAT2, improving the DA synaptic vesicle circulation, and reducing the DA content in the synaptic space of the brain.

ObjectiveTo investigate the possible mechanism of Buzhong Yulin Decoction （补中愈淋汤） in the prevention and treatment of recurrent urinary tract infection. MethodsThe mouse models of recurrent urinary tract infection were established by uropathogenic Escherichia coli （UPEC） strain UTI89 by bladder perfusion， and the successful mouse models were randomly divided into a model group， an antibiotic group， and a low- and high-dose Buzhong Yulin Decoction group， with six mice in each group. In addition， 5 C57BL/6 mice without modelling were taken as blank group. The low- and high-dose Buzhong Yulin Decoction groups received 0.1 ml/10 g of decoction by gavage， with concentrations of 1.3 g/ml and 5.2 g/ml， respectively； the antibiotic group received 0.1 ml/10 g of levofloxacin hydrochloride solution with 5 mg/ml by gavage； the blank and model groups received 0.1 ml/10 g of distilled water by gavage. Each group was gavaged once a day for 7 consecutive days. The total number of urine marks， the number of central urine marks， and the total urine volume of the urine marks were observed by the urine marking test； HE staining was used to observe the histopathological changes in the bladder of mice； serum levels of the inflammatory factors interleukin 1β （IL-1β）， interleukin 6 （IL-6） and tumour necrosis factor α （TNF-α） were detected by ELISA； the morphology of the epithelial cells of bladder was observed by scanning electron microscopy； immunofluorescence assay to detect bladder tissue anti-UroPlakin 3A protein level and UPEC bacterial load； the spread plate method to detect urinary bacterial load and bacterial load of bladder epithelial cells； RT-PCR method to detect Ras-related protein Rab-11A （RAB11A） and Ras-related protein Rab-27B （RAB27B） mRNA level in bladder tissue； immunoblotting to detect microtubule-associated protein 1 light chain3 （LC3） and P62 protein levels in bladder tissue. ResultsCompared with the blank group， the bladder epithelial cell layers were lost and showed abnormal morphology in mice of the model group； bladder tissue UroPlakin 3A protein and RAB11A and RAB27B mRNA levels reduced， the total number of urine marks， the number of central urine marks， bladder tissue UPEC bacterial load， urinary bacterial load， bacterial load in bladder epithelial cells， serum IL-1β， IL-6， and TNF-α levels， and LC3 and P62 protein levels in bladder tissue all elevated （P＜0.05 or P＜0.01）. Compared with the model group， the bladder epithelial cell layers were intact and the morphology of epithelial cells were regular in the low- and high-dose Buzhong Yulin Decoction groups； the average surface area of bladder epithelial cells reduced， the levels of UroPlakin 3A protein and RAB11A and RAB27B mRNA in bladder tissues elevated， and total number of urine marks， the number of central urine marks， bladder tissue UPEC bacterial load， urinary bacterial load， bacterial load in bladder epithelial cells， serum IL-1β， IL-6， and TNF-α levels， and P62 protein levels in bladder tissue all reduced （P＜0.05 or P＜0.01）， but LC3 protein levels showed no statistically significant （P＞0.05）. In the antibiotic group， the bladder epithelial cells were partially missing and the morphology of epithelial cells was abnormal. Compared with the antibiotic group， the average surface area of the bladder epithelial cells in the mice increased in the low- and high-dose Buzhong Yulin Decoction groups， the bacterial load of the bladder epithelial cells decreased， and the P62 protein level of the bladder tissue decreased （P＜0.05）. When comparing between the low- and high-dose Buzhong Yulin Decoction groups， the differences in each index were not statistically significant （P＞0.05）. ConclusionBuzhong Yulin Decoction may prevent and treat recurrent urinary tract infection by repairing the bladder mucosal barrier， increasing RAB11A and RAB27B level and enhancing autophagy in bladder tissues， thereby facilitating bacterial clearance from bladder epithelial cells and reducing the bacterial load of bladder epithelial cells.

OBJECTIVE To construct a double helix model for evaluating the job competency of licensed pharmacists in retail pharmacies， and to provide a reference for building a talented team of licensed pharmacists and promoting the high-quality development of pharmaceutical care. METHODS Based on the competency iceberg model and double helix structure theory， literature analysis and the Delphi method were adopted to explore the model indicators； questionnaires were designed to survey licensed pharmacists in retail pharmacies from Shandong Province. The exploratory factor analysis was used to correct the model indicators and establish a double helix model for evaluating the job competency of licensed pharmacists in retail pharmacies， which was examined by validation factor analysis. RESULTS The recovery rate of the questionnaires in the two rounds of expert consultation was greater than 90%， the expert authority coefficients were both more than 0.86， and the overall Kendall’s W values were 0.288 and 0.510， respectively； after the correction of the exploratory factor analysis， the double helix model for evaluating the job competency of licensed pharmacists in retail pharmacies was established， which contained 4 first-level indexes such as occupational literacy， occupational knowledge， occupational competency and internal motivation， and 23 second-level indexes. The model contained occupational knowledge and occupational competency in the explicit competency chain， occupational literacy and internal motivation in the implicit competency chain， and policy support， incentive mechanism， education and training as the hydrogen bonds connecting the two main chains. The validation factor analysis showed that the model’s goodness of fit index， comparative fit index， and normed fit index all exceeded 0.9； the reliability of the combination of the four first-level indexes ranged from 0.89 to 0.95； the average variance extracted ranged from 0.58 to 0.75， and Cronbach’s α coefficients for both the overall model and the first-level indicators were all greater than 0.84. CONCLUSIONS The constructed double helix model for evaluating the job competency of licensed pharmacists in retail pharmacies is scientific， reasonable and practical.

Objective: To assess the health risk associated with drinking water in Hangzhou from 2016 to 2017,and to provide evidence for the safety of drinking water . Methods: The monitoring data of 5 genetic toxic substances（arsenic,hexavalent chromium,cadmium,chloroform,tetrachloromethane）and 13 body toxic substances（lead,mercury,selenium,cyanide,fluoride,nitrate,iron,ammonia nitrogen,manganese,copper,zinc,aluminum,volatile phenol）from 36 source water samples,36 finished water samples and 288 tap water samples in the main urban areas of Hangzhou were collected from 2016 to 2017. The health risk of drinking water containing the chemical pollutants mentioned above were assessed based on the evaluation models recommended by United States Environmental Protection Agency . Results: The concentrations of 5 genetic toxic substances and 13 body toxic substances in source water,finished water and tap water were all within the reference limits issued by Standards for Drinking Water Quality（GB 5749—2006）. The carcinogenic risk,non-carcinogenic risk and total health risk caused by the chemical pollutants in the source water were 2.18×10-5/a,7.75×10-9/a and 2.18×10-5/a. The carcinogenic risk,non-carcinogenic risk and the total health risk caused by the chemical pollutants in the finished water were 1.08×10-5/a,3.70×10-9/a and 1.08×10-5/a. The carcinogenic risk,non-carcinogenic risk and total health risk caused by the chemical pollutants in the tap water were 1.96×10-5/a,3.61×10-9/a and 1.96×10-5/a. The carcinogenic risk and total health risk caused by chemical pollutants ranged from high to low in the source water,tap water and finished water. The non-carcinogenic risks ranged from high to low in the source water,finished water and tap water . Conclusion The health risks of 18 chemical pollutants in drinking water in Hangzhou were at a low level,with the greater carcinogenic risk than the non-carcinogenic risk. Hexavalent chromium had the highest carcinogenic risk,while fluoride and aluminum had the highest non-carcinogenic risk.

Objective:To investigate the prognostic significance of preoperative neutrophil-lymphocyte ratio(NLR)combined with tu-mor-associated neutrophils(TANs)in the stroma of gastric cancer tissues of patients.Methods:One hundred and twenty-six gastric cancer tissue samples from patients enrolled in Changhai hospital from June 2006 to May 2011 were divided into four groups accord-ing to NLRs in preoperative peripheral blood combined with high or low infiltration of neutrophils in gastric cancer tissues.The 5-year survival of the four groups was then compared,and their correlations with clinicopathologic features and prognosis were analyzed.Re-sults:High NLRs in peripheral blood combined with low infiltrating TANs in gastric tissues was associated with lower differential grade (P<0.001)and larger tumor size(P=0.026).Of the four groups,patients with high NLR in peripheral blood combined with low infiltrat-ing TANs in gastric cancer tissues demonstrated the lowest survival rates,whereas those with low NLR and high infiltrating TANs had highest survival rates, and this difference was statistically significant (P<0.05). Univariate and multivariate Cox regression analyses showed that high NLR in peripheral blood combined with low infiltrating TANs in gastric cancer tissues(P<0.05)was an independent factor indicating poor prognosis.Conclusions:NLR in preoperative peripheral blood combined with infiltrating TANs in gastric cancer tissues can be used as a prognostic indicator for patients with gastric cancer,especially high NLR in preoperative peripheral blood com-bined with low infiltrating TANs in gastric cancer tissue indicates poor prognosis.

Objective: To evaluate the relationship between platelet distribution width(PDW) and the extent of coronary artery disease and 2-year outcome in patients received percutaneous coronary artery intervention(PCI) because of stable coronary artery disease(SCAD). Methods: We consecutively enrolled 4 293 patients who received PCI because of SCAD in Fuwai Hospital from Jan 2013 to Dec 2013, patients were followed up for 2 years. Patients were divided into three groups according to tertiles values of PDW as follows: PDW≤11.4%(1 402 patients), 11.4%12.9% (1 450 patients). Major adverse cardiovascular and cerebrovascular events (MACCE) were defined as the occurrence of death, myocardial infarction, target vessel revascularization, intra stent thrombosis and stroke during follow-up. Multivariable logistic regression was used to evaluate the relationship between PDW and the extent of CAD. Multivariable Cox regression was used to evaluate the relationship between PDW and prognosis of SCAD patients. Results: PDW was associated with diabetes mellitus, body mass index, red cell distribution width, mean platelet volume (MPV), platelet counts and glycosylated haemoglobin (P<0.05), but not associated with age, sex, estimated glomerular filtration rate (P>0.05). PDW was not correlated with the extent of CAD(P=0.990), SYNTAX score(P=0.721), no-reflow phenomenon after PCI(P=0.978). Multivariable logistic regression also showed no relationship between PDW and extent of CAD (OR=0.994, 95%CI 0.961-1.029, P=0.73). PDW was found to be an independent risk factor of 2-year cardiac death (HR=1.242, 95%CI 1.031-1.497, P=0.022), but was not an independent risk factor of all-cause death and MACCE. Conclusions PDW is not related with the extent of coronary artery disease. PDW is an independent risk factor of 2-year cardiac death, but is not an independent risk factor of all-cause death and MACCE in this patient cohort.
