Objective To explore the feasibility of phase-contrast CT for three-dimensional visualization of in vitro intrahepatic vessels and necrotic foci in mouse model of acute liver injury(ALI).Methods Six clean grade male wild-type C57BL/6 mice were randomly divided into model group and control group(each n=3).The mice in model group were fed with overdose of acetaminophen to induce ALI,while those in control group were fed with the same amount of distilled water. After 24 h,the mice were all sacrificed, the livers were harvested and then fixed and dehydrated.Phase-contrast CT images of in vitro liver were acquired, sectional and three-dimensional images were reconstructed. The effect of phase-contrast CT for displaying the outline of liver and internal vessels and necrotic foci were observed, the maximum diameter and volume of necrotic foci were quantitatively analyzed, and the results of phase-contrast CT were compared with pathological findings. Results The original projection and sectional images of phase-contrast CT clearly showed the outline of in vitro liver and internal vasculatures.Necrotic foci within the liver were found in model group, but not in control group. The findings of phase-contrast CT corresponded accurately to those of pathology.Three-dimensional reconstruction images of phase-contrast CT clearly displayed intrahepatic portal vein system and hepatic vein system in both groups, and discontinuous punctate necrotic foci within the liver were found in model group, mainly distributed around hepatic vein, with a median maximum diameter of 18.50 μm and a median volume of 5 870.11 μm3,but was not observed in control group. Conclusion Phase-contrast CT could be used in three-dimensional visualization of intrahepatic vessels and necrotic foci of in vitro liver in mouse model of ALI.

Objective:To evaluate the clinical value of an obstetric intelligent assistant in predicting postpartum hemorrhage (PPH) after vaginal delivery. Methods:This retrospective cohort study included 4 832 women who delivered vaginally at ≥26 weeks of gestation at the Third Affiliated Hospital, Guangzhou Medical University between May 2023 and April 2025. Participants were categorized into PPH (382 cases, blood loss ≥500 ml within 24 h after delivery) and non-PPH groups (4 450 cases). Using traditional statistical methods combined with machine learning approaches, including support vector machines and extreme gradient boosting, supplemented with deep learning techniques, we developed a novel artificial neural network model—the obstetric intelligent assistant. This model provides a refined classification of PPH occurrence and estimated blood loss volume. The obstetric intelligent assistant integrates 70 antenatal and intrapartum risk factors through hospital information system interfacing to generate visualized risk probability outputs. Predictive performance was compared between the obstetric intelligent assistant and four conventional prediction tools (Chinese Labor Room Traffic Light System; Association of Women's Health, Obstetric and Neonatal Nurses; American College of Obstetrics and Gynecology Safe Motherhood Initiative; and California Maternal Quality Care Collaborative prediction tools) using receiver operating characteristic curve.Results:(1) For antenatal prediction, the obstetric intelligent assistant achieved an area under the curve of 0.826 (95% CI: 0.774-0.838), with sensitivity of 0.794 and specificity of 0.712, while the four conventional prediction tools showed area under the curve ranging from 0.569 to 0.586. (2) For intrapartum prediction, the obstetric intelligent assistant achieved an area under the curve of 0.786 (95% CI: 0.751-0.820), with sensitivity of 0.837 and specificity of 0.762, whereas the conventional tools showed area under the curve between 0.600 and 0.613. Conclusion:The obstetric intelligent assistant demonstrates superior performance in predicting PPH compared to conventional prediction tools.

To develop vaccines for the prevention and control of porcine circovirus 2d genotype(PCV2d)and pseudorabies virus(PRV),the PCV2d ORF2 gene was amplified by PCR,and cloned into the BamH Ⅰ site of PRV transfer plasmid pG vector harboring the enhanced green fluorescent protein(EGFP)gene. The resulting recombinant transfer plasmid pG-PCV2d-EGFP was transfect-ed into ST cells infected with the three gene deleted PRV variant strain gE-/g-/TK-PRV NY to generate a recombinant virus rPRV-PCV2d-EGFP+,and then the EGFP gene was knocked out to harvest the rPRV-PCV2d using gene-editing technology termed CRISPR/Cas9 system. The recom-binant virus rPRV-PCV2d had similar genetic stability to the parental PRV as indicated by PCR and one-step growth curve test, and the expression of PCV2d capsid(Cap)protein was validated by Western blot.In animal experiment, higher PCV2-specific ELISA antibodies and detectable PCV2-specific neutralizing antibodies could be elicited in mice immunized with the recombinant vi-rus rPRV-PCV2d compared to commercial PCV2 inactivated vaccine.rPRV-PCV2d significantly re-duced the PCV2d loads in tissues such as the heart, liver and spleen of mice following virulent PCV2d challenge.Moreover,rPRV-PCV2d elicits PRV-specific immune responses in mice and can prevent PRV virulent infection in mice, indicating the recombinant virus rPRV-PCV2d has strong immunogenicity.

Macrophages are important immune cells involved in innate immunity, with significant heterogeneity and polarization.Macrophages can polarize into different phenotypes(mainly M1 and M2)under stimulation by various factors in the microenvironment, leading to different roles and functions.Macrophages play an important role in the pathophysiological mechanism of renal ischemia-reperfusion injury(RIRI).An excessive immune response will inevitably lead to tissue damage.M1 macrophages are pro-inflammatory cells involved in the clearance of pathogens, while M2-type macrophages have anti-inflammatory effects and participate in the repair and remodeling of renal tissue after RIRI.The balance between these macrophage phenotypes is thus an important factor affecting the outcome and treatment of RIRI.This review considers the pathophysiologic mechanism of macrophages in RIRI and the latest treatments from the perspective of macrophage polarization, with the aim of supporting further studies of the function of macrophage polarization in RIRI and adjusting the macrophage polarization process to improve RIRI treatment strategies.

Objective:To verify mechanism of Dunhuang Ruicao ointment in frostbite treatment through network pharmacology and experiment. Methods:Targets of Dunhuang Ruicao ointment in intervention of frostbite was screened through network pharmacology.Dunhuang Ruicao ointment was used to treat rat model of frostbite.Effect of drug was observed and results of network pharmacological enrichment analysis were verified. Results:A total of 239 active drug components,186 disease targets and 36 common targets were screened.KEGG pathway enrichment screened out 96 pathways, mainly enriched in Toll-like receptor signaling pathway,IL-17 signaling pathway, tumor necrosis factor signaling pathway,AGE-RAGE signaling pathway,Th17 cell differentiation pathway, suggesting Toll-like receptor signaling pathway may play an important role in treatment of frostbite by Dunhuang Ruicao ointment.Animal experiments showed that compared with model group, skin frostbite of rats treated with different doses of Dunhuang Ruicao ointment group was significantly improved, wound healing rate was increased, swelling rate was decreased, wound tissue epithelialization was obvious, inflammatory infiltration was reduced, angiogenesis was increased, and collagen fiber content was increased.mRNA and protein expressions of TLR4,MyD88 and NF-κB p65 in local skin lesions were decreased to different degrees, and middle-dose group had the best effect. Conclusion:Dunhuang Ruicao ointment in treatment of frostbite has characteristics of multiple components, multiple targets, multiple pathways synergy, probably playing a role in treatment of frostbite by inhibiting TLR4/MyD88/NF-κB signaling path-way activated to reduce frostbite rats serum proinflammatory media release, reduce inflammation, improve rate of tissue remodeling, promote healing of frostbite, providing a theoretical basis for further study of complex mechanism of Dunhuang Ruicao ointment in treatment of frostbite.
