1.Exploring the therapeutic effect of cornuside on diabetic retinopathy in rats based on the ROS/TXNIP/NLRP3 signaling pathway

Objective To explore the therapeutic effect of cornuside on diabetic retinopathy(DR)in rats and ana-lyze the acting mechanism of the reactive oxygen species(ROS)/thioredoxin interacting protein(TXNIP)/NOD-like recep-tor thermal protein domain associated protein 3(NLRP3)signaling pathway in this process.Methods A total of 56 suc-cessfully modeled DR rats were randomly divided into the model group,the cornuside 20 mg·kg-1 group,the cornuside 40 mg·kg-1 group,the calcium dobesilate 5.8 mg·kg-1 group,with 14 rats in each group;while,meanwhile another 14 healthy rats were selected as the control group.After the corresponding intervention of rats in each group,retinal tissue in-flammation,oxidative stress indicators,fasting blood glucose(FBG),and angiogenic factor levels were detected by the en-zyme-linked immunosorbent assay(ELISA).The hematoxylin-eosin(HE)staining and terminal deoxynucleotidyl transfer-ase dUTP nick end labeling(TUNEL)staining were used to observe retinal histopathology and retinal cell apoptosis,re-spectively.The mRNA expression of ROS,TXNIP,and NLRP3 in the retinal tissue was detected by the quantitative fluores-cence polymerase chain reaction(PCR).The apoptosis of retinal cells and the protein expression of the ROS/TXNIP/NL-RP3 signaling pathway were detected by Western blotting.Results Compared with the control group,the model group showed disordered arrangement of retinal cells and a significant decrease in the cell number,accompanied by nuclear con-densation and edema;interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),malondialdehyde(MDA),FBG,angiopoie-tin-1(Ang-1),vascular endothelial growth factor(VEGF),retinal cell apoptosis rate,ROS,and the mRNA and protein ex-pression levels of TXNIP and NLRP3 increased significantly,while superoxide dismutase(SOD)and B cell lymphoma-2(Bcl-2)protein expression levels decreased significantly(all P<0.05).Compared with the model group,the arrangement of retinal cells in the cornuside 20 mg·kg-1 group,the cornuside 40 mg·kg-1 group,and the calcium dobesilate 5.8 mg·kg-1 group gradually became normal,the number of retinal cells increased,and the nuclear condensation and edema were relieved;IL-6,TNF-α,MDA,FBG,Ang-1,VEGF,retinal cell apoptosis rate,ROS,and the mRNA and protein expression level of TXNIP and NLRP3 decreased significantly,while the protein expression level of SOD and Bcl-2 increased signifi-cantly(all P<0.05).In the intergroup comparison of the pathological damage of retinal tissue and the improvement degree of the above quantitative indexes in DR rats,the cornuside 40 mg·kg-1group was superior to the cornusin 20 mg·kg-1 group(all P<0.05);the calcium dobesilate 5.8 mg·kg-1 group was superior to the cornusin 20 mg·kg-1 group,but infe-rior to the cornuside 40 mg·kg-1 group(all P<0.05).Conclusion Cornuside can mitigate retinal inflammation,oxi-dative stress,and pathological damage in DR rats and inhibit blood glucose,retinal angiogenesis,and cell apoptosis.The acting mechanism of cornuside may be related to the inhibition of the ROS/TXNIP/NLRP3 signaling pathway.

3.Study on the Expression of AIM2 in PBMC of Patients with Advanced Gastric Cancer and Its Relationship with Intestinal Microbiota and the Efficacy of Targeted Therapy

Objective To investigate the relationship between the expression of absent in melanoma 2(AIM2)gene in peripheral blood peripheral blood mononuclear cell(PBMC)of patients with advanced gastric cancer(GC)and the intestinal flora and the efficacy of targeted therapy.Methods 83 patients with advanced GC who were treated from January to December 2023 were all given Trastuzumab/Nebuliumab combined with conventional chemotherapy for 4 weeks/course and 3 continuous courses of treatment to evaluate the efficacy of targeted therapy.The abundance of intestinal flora was analyzed using 16S rDNA sequencing technologies.Quantitative real-time PCR(qRT-PCR)was used to detect AIM2 mRNA expression levels in PBMC,which were divided into a high expression group and a low expression group according to the median digit.Pearson correlation analysis was used to investigate the relationship between AIM2 expression and the relative abundance of intestinal flora,the efficacy of targeted therapy and clinical features.Results At the phylum level,the relative abundance of Bacteroidetes(0.70±0.15)and Proteobacteria(0.71±0.21)in the high expression group of AIM2 was significantly lower than that in the low expression group(0.81±0.17,0.80±0.16),and the relative abundance of Firmicutes(0.73±0.12)was significantly higher than that in the low expression group(0.64±0.08),and the differences were statistically significant(t=3.108,2.210,4.061,all P<0.05).At the genus level,the relative abundance of Bacteroides(0.13±0.02)in the high expression group of AIM2 was significantly lower than that in the low expression group(0.19±0.04),and the relative abundance of Enterococcus(0.31±0.08 vs 0.12±0.05)and Escherichia coli(0.18±0.04 vs 0.10±0.02)was significantly higher than that in the low expression group(0.12±0.05,0.10±0.02),and the differences were statistically significant(t=8.472,153.462,11.722,all P<0.05).Pearson correlation showed that at phylum level,AIM2 expression was positively correlated with the relative abundance of Firmicutes(r=0.598,P<0.05),and negatively correlated with the relative abundance of Bacteroides and Proteobacteria(r=-0.641,-0.520,all P<0.05).At the genus level,AIM2 expression was positively correlated with the relative abundance of Enterococcus and Escherichia coli(r=0.529,0.577,all P<0.05),and negatively correlated with the relative abundance of Bacteroides(r=-0.574,P<0.05).Among the 83 patients with advanced GC,17 cases were effective and 66 cases were ineffective,and the Overall response rate(ORR)was 20.48%.The lesion diameter(5.73±0.74cm)and the proportion of cTNM stage IV(53.85%)in AIM2 high expression group were significantly lower than those in low expression group(6.08±0.51cm,75.00%),and the targeted therapy ORR(30.77%)was higher than that in low expression group(11.36%),with statistical significance(t/χ2/Z=2.477,6.558,4.780,all P<0.05).Conclusion The expression of AIM2 in PBMC of advanced GC patients is closely related to the imbalance of intestinal flora and the effect of targeted therapy.Detection of AIM2 expression is conducive to guiding clinical screening of patients who benefit from targeted therapy.
4.Association between different physical activity and bone mineral density at various skeletal sites

Objective:To investigete the relationship between different physical activity(PA)and bone mineral density(BMD)at various skeletal sites,providing evidence for the prevention and improvement of bone health.Methods:Cross-sectional data from the National Health and Nutrition Examination Survey(NHANES)from 2007 to 2018 was used,and 7 692 participants aged≥20 were included.Multivariable linear regression model was used to analyze the relationship between different PA and BMD at the femoral neck,total hip,and lumbar spine across different genders.Statistical modeling employed a sequential variable inclusion approach:Model 1 included age,race/ethnicity,and body mass index,while Model 2 further incorporated calcium intake,marital status,smoking status,dietary inflammatory index,and serum phosphate levels.Results:PA showed statistically significant differences across age,gender,body mass index,race,marital status,smoking,and dietary inflammatory index(P<0.05).Among females,low-intensity PA showed a positive association with femoral neck BMD in both Model 1 and Model 2(P<0.05),but no significant correlation was found between low-intensity PA and total hip BMD(P>0.05).High-intensity PA exhibited an inverse association with femoral neck and total hip BMD(P<0.05),though this was only observed in Model 1.No significant association was detected between different PA intensities and lumbar spine BMD(P>0.05).Among males,high-intensity PA showed a positive correlation with lumbar spine BMD(P<0.05),but this association was only significant in Model 2.No significant association were observed between low-intensity PA and lumbar spine BMD in either Model 1 or Model 2(P>0.05).Additionally,no significant association were found between different PA intensities and femoral neck or total hip BMD(P>0.05).Conclusions:The relationship between PA and BMD varies by sex and skeletal site.Low-intensity PA is positively associated with femoral neck BMD in females,while high-intensity PA is positively correlated with lumbar spine BMD in males.
5.Application value of radiomics in predicting radiation-induced injury in the era of precise radiotherapy

Radiotherapy is one of the primary treatment modalities for tumors. In recent years, advancements in radiotherapy technology have enabled radiation to reach target areas with greater precision. However, radiation damage to normal tissue remains unavoidable. Traditional normal tissue complication probability (NTCP) models are widely used to predict such injuries, but they often fail to account for individual and tissue heterogeneity. As an emerging technology, radiomics can provide more comprehensive biological information and shows promise in predicting radiation-induced damage. This article reviews the application value of radiomics in predicting damage to organs at risk in the head and neck, thorax, abdomen, and pelvis.
6.Research status of degradable magnesium-based alloy guided bone regeneration membrane on stomatol-ogy

An ideal guided bone regeneration(GBR)membrane usually needs to possess good biocompatibility,appropriate me-chanical properties,reasonable degradability and clinical manageability.However,traditional biological barrier membranes cannot meet clinical requirements such as high mechanical strength and controllable degradation simultaneously,so it is necessary to de-velop a new type of guided bone regeneration membrane material.Degradable magnesium-based alloy guided bone regeneration membranes have the characteristics and potential to become new guided bone regeneration membrane materials.This review intro-duces the research history and field applications of magnesium-based alloys,as well as the advantages and limitations of degradable magnesium-based alloy GBR membranes.Finally,it presents the applications and prospects of degradable magnesium-based alloy GBR membranes in various oral medicine fields,providing a scientific basis for their clinical transformation.

Objective:To investigate the incidence of shoulder work-related musculoskeletal disorders (WMSDs) among occupational population in China, and to explore their intrinsic association with personal and work-related factors.Methods:In April 2024, 73497 valid questionnaires of the Chinese version of the Musculoskeletal Disorders Electronic Questionnaire were retrospectively analyzed from June 2018 to December 2023 in 22 provinces and 29 key industries in China, and the general information, occurrence of WMSDs and related risk factors of key occupational populations in different regions in China were collected. By using Chi-square test and confirmatory factor analysis, the relationship between shoulder fatigue and pain in key occupational groups and individual factors, work type, work posture and work organization was discussed, and the internal relationship was analyzed based on structural equation model.Results:Higher incidence of shoulder fatigue and pain were associated with female, lack of physical exercise, uncomfortable working posture and neck leaning forward ( P<0.05). Structural equation model analysis showed that work type, work posture and work organization were strongly correlated ( r=0.58, 0.55). Work organization and work type were strongly correlated with shoulder fatigue ( r=0.65) and moderately correlated with shoulder fatigue ( r=0.21). Shoulder fatigue was moderately associated with shoulder pain ( r=0.40). Individual factors, work type, work posture and shoulder fatigue could directly affect shoulder pain ( OR=0.07, -0.09, 0.17 and 0.40), and work type and work posture could also indirectly affect shoulder pain through shoulder fatigue ( OR=0.08, 0.03). Work organization only indirectly affected shoulder pain through shoulder fatigue ( OR=0.26) . Conclusion:The main influencing factor of shoulder pain is shoulder fatigue, followed by work posture and individual factors. Structural equation model can better reflect the complex relationship between work type, work posture and work organization and shoulder WMSDs. Improving work posture and work organization may be an effective way to control the influence of shoulder fatigue on shoulder pain.

Objective:To explore the structural relationship between WMSDs in the upper limbs and various risk factors in the occupational population in China, based on a large sample epidemiological survey and structural equation analysis, and to establish a structural equation model, so as to lay a foundation for the prevention and control of such diseases.Methods:The Chinese version of the Musculoskeletal Disorders Electronic Questionnaire was used to conduct a nationwide survey on the prevalence of WMSDs in the upper extremity. Six factors related to WMSDs in the upper extremity were extracted by the classification standard of adverse ergonomic factors and their source and confirmatory factor analysis, including work organization, work type, upper extremity work posture, individual factors, upper extremity fatigue and upper extremity WMSDs. The structural equation analysis was carried out and the structural equation model was established.Results:The incidence of WMSDs and fatigue in the upper limbs was 24.44% and 43.76%, respectively. The adjusted structural equation model fitting indicators were generally up to the standard (GFI=1.000, AGFI=1.000, RMSEA=0.043, NFI=0.808, TLI=0.784) . The four exogenous latent variables of work organization, work type, upper limb work posture and individual factors were correlated. There was a strong positive correlation between job type and upper limb work posture ( r=0.865) , a moderate positive correlation between work organization and job type and upper limb work posture ( r=0.570, 0.490) , and a weak negative correlation between individual factors and the other three exogenous latent variables. Upper limb work posture and individual factors had direct effects on upper limb WMSDs, and the effect coefficients were 0.10 and 0.06, respectively. Upper limb fatigue played a mediating role between work organization, work type, upper limb work posture and upper limb WMSDs. The effect coefficient was 0.46, and the composition ratios of indirect effects were 100.0%, 100.0%, and 38.3%, respectively. The direct path effect of upper limb work posture, individual factors and upper limb WMSDs was weaker than the mediating path through upper limb fatigue. Conclusion:When carrying out the prevention and control of upper limbWMSDs, it is necessary to comprehensively consider the pathogenesis path of upper limb muscle fatigue and upper limb WMSDs caused by work organization, work type, and upper limb work posture, so as to provide theoretical reference for improving the prevention and control level of such diseases.

Body weight abnormalities, including overweight, obesity, and underweight, have become a dual public health challenge in Chinese adults: overweight and obesity lead to a variety of chronic complications, while underweight increases the risks of malnutrition, sarcopenia, and organ dysfunction. To systematically address these issues, multidisciplinary experts in endocrinology, sports science, nutrition, and psychiatry from various regions have held multiple weight management seminars. Based on the latest epidemiological data and clinical evidence, they expanded the guideline to include assessment and intervention strategies for underweight, in addition to the core content of obesity management. This guideline outlines the etiological mechanisms, evaluation methods, and multidimensional management strategies for overweight and obesity, covering key areas such as diagnosis and assessment, medical nutrition therapy, exercise prescription, pharmacological intervention, and psychological support. It is intended to provide a scientific and standardized approach to weight management across the adult population, aiming to curb the rising prevalence of obesity, mitigate complications associated with abnormal body weight, and improve nutritional status and overall quality of life.
