1.Research progress in signaling pathways related to acupuncture regulation of post-stroke depression

The signaling pathways involved in acupuncture intervention in post-stroke depression (PSD) mainly include cAMP signaling pathway, MAPK signaling pathway, PI3K/Akt/mTOR signaling pathway, NF-κB signaling pathway, CREB signaling pathway, and CaMK signaling pathway. A variety of regulatory networks are formed between these signaling pathways, which play a key role in reducing inflammation, inhibiting apoptosis, improving neuroplasticity, promoting angiogenesis, and protecting neurons.
2.Roxadustat attenuates heat stress-induced apoptosis and senescence in renal tubular epithelial cells by upregulating HIF-1α

Objective To investigate the effect and underlying mechanism of roxadustat on apoptosis and senescence of renal tubular epithelial cell line HK-2 induced by heat stress.Methods After HK-2 cells were treated with different concentrations of roxadustat(10,20,30,40 and 50 μmol/L)for 24 h,CCK-8 assay was used to determine the optimal intervention concentration of roxadustat.HK-2 cells were divided into 4 groups(n=3):control group,roxadustat group(30 μmol/L,24 h),heat-stress group(43 ℃,2 h),and heat-stress+roxadustat group(30 μmol/L roxadustat treatmnet for 24 h followed by heat-stress 2 h).Cell viability was detected by CCK-8 assay.Expression of hypoxia-inducible factor-1α(HIF-1α),Cleaved Caspase-3,p16 and p21 at protein level was detected by Western blotting.Immunofluorescence assay was employed to observe the distribution of HIF-1α.β-galactosidase staining kit was utilized to detect SA-β-Gal activity.TUNEL staining was used to measure cell apoptosis.Results The highest cell viability was observed in the cells after 30 μmol/L roxadustat treatment.Heat stress resulted in a significant decrease in cell viability(P<0.05),elevated protein levels of HIF-1α,Cleaved Caspase-3,p16 and p21(P<0.05),enhanced SA-β-Gal activity(P<0.05)and increased percentage of TUNEL-positive cells(P<0.05)when compared with the cells in the control group.In comparison with the heat-stress group,the heat-stress+roxadustat group showed significant decrease in the protein levels of Cleaved Caspase-3,p16 and p21(P<0.05),reduced activity of SA-β-Gal[(65.44±5.00)%vs(77.15±2.61)%,P<0.05]and decreased percentage of TUNEL-positive cells[(16.73±2.20)%vs(46.40±13.87)%,P<0.05],but increase in cell viability[(86.33±4.51)%vs(66.33±8.50)%,P<0.05]as well as HIF-1α protein expression(P<0.05).Furthermore,immunofluorescence assay showed that HIF-1α was mainly distributed in the nucleus and perinucleus.Conclusion Roxadustat attenuates heat stress-induced apoptosis and senescence of renal tubular epithelial cells by upregulating HIF-1α.
3.Effect of maxillary expansion combined with orofacial myofunctional therapy on the position of the tongue of children with mouth breathing.

Objective:This study aimed to investigate the change of the position of the tongue before and after combined treatment of maxillary expansion and orofacial myofunctional therapy in children with mouth-breathing and skeletal class Ⅱmalocclusion. Methods:A total of 30 children with skeletal class Ⅱ malocclusion and unobstructed upper airway were selected. The 30 children were divided into mouth-breathing group(n=15) and nasal-breathing group(n=15) and CBCT was taken. The images were measured by Invivo5 software. The measurement results of the tongue position of the two groups were analyzed by independent samples t-test. 15 mouth-breathing children with skeletal class Ⅱ malocclusion were selected for maxillary expansion and orofacial myofunctional therapy. CBCT was taken before and after treatment, the measurements were analyzed by paired sample t test with SPSS 27.0 software package. Results:The measurement of the tongue position of the mouth-breathing and nasal-breathing groups were compared, the differences were statistically significant(P<0.05). The measurement of the tongue position showed significant difference after the combined treatment of maxillary expansion and orofacial myofunctional therapy in children with mouth-breathing and skeletal class Ⅱmalocclusion(P<0.05). Conclusion:Skeletal class Ⅱ malocclusion children with mouth-breathing have low tongue posture. The combined treatment of maxillary expansion and orofacial myofunctional therapy can change the position of the tongue.

4.Effects of auricular acupressure therapy on postoperative pain and early rehabilitation of patients with total knee arthroplasty

Objective:To explore the effect of auricular acupressure therapy on postoperative pain and early rehabilitation of patients with total knee arthroplasty.Methods:From March to June 2021, 68 patients who underwent unilateral total knee arthroplasty for the first time under general anesthesia combined with peripheral nerve block in Liaocheng People's Hospital were selected by convenient sampling. According to the random number method, the patients were divided into observation group (35 cases) and control group (33 cases) . Both groups received routine pain management and early postoperative rehabilitation nursing after operation, and the Chinese medicine doctors pasted the Wangbuliuxing seed adhesive tape at the auricular points one day before operation. The control group did not take any pressing operation. The observation group was pressed at the time of 1 day before operation to 3 days after operation, 3 times a day. The pressing frequency was 60 times/min, and the pressing time was 1 minute per acupoint. The dosage of analgesics used immediately after leaving the Anesthesia Recovery Room and at 4, 24, 48 and 72 hours after operation, as well as the visual analog pain scores of rest and activity at each follow-up time point were recorded and compared between the two groups. The knee range of motion of the two groups on the second, the fourth and seventh days after operation was compared.Results:The dosage of patient-controlled intravenous analgesia analgesics in the observation group were lower than those in the control group at 24, 48 and 72 hours after operation, with statistically significant differences ( P<0.05) . The pain score of the patients in the observation group at rest 24 hours after operation was lower than that in the control group, and the pain scores of the patients in the active state at 24, 48 and 72 hours after operation were lower than those in the control group, and the differences were statistically significant ( P<0.05) . The knee range of motion in the observation group were higher than those in the control group on the second, fourth and seventh days after operation, and the differences were statistically significant ( P<0.05) . Conclusions:Auricular acupressure therapy can reduce the dosage of analgesics used after total knee arthroplasty, relieve the degree of pain, and promote the early recovery of patients.
5.Research status and development trends in fat transplantation: bibliometrics and visual analysis

Objective:Using bibliometrics to study the international literature on fat transplantation in recent 10 years, and to explore the research status, hot spots and development trend in this field.Methods:Literatures related to fat transplantation research from 2011 to 2020 were searched through the core collection of Web of Science database, and the literatures were screened strictly according to inclusion and exclusion criteria. Statistical analysis was conducted on the year, journal, first author, number of articles published by countries and institutions, and distribution of disciplines by bibliometrics. Citespace5.7.R5W software was used for co-citation analysis of the included literatures. Cooccurrence analysis and emergent word analysis were also carried out on the keywords in the literature. VOSviewer1.6.16 software was used to perform visual clustering of terms in the title and abstract of the included literature to reveal hot topics and research frontiers.Results:A total of 4 901 papers were included, and the number of annual publications continued to increase from 2011 to 2020. The top three publications during the study period were Plastic and Reconstructive Surgery (320 papers), Aesthetic Surgery Journal (171 papers), and PLoS One (113 papers). The first authors were Rafael Denadai (13 papers), Gentile Pietro(12 papers), Khouri Roger(6 papers). The most productive countries and institutions in this field were the United States (1 646 papers), China (853 papers), and Italy (383 papers). The organizations with the largest number of publications were the University of Pittsburgh (96 papers), Shanghai Jiao Tong University (78 papers), and Milan University (63 papers). There were 87 research categories in the literature related to fat transplantation, of which surgery was involved mostly. Visual analysis of Cites Pace5.7.R5W software showed that 4 901 references had established a co-citation network consisting of 381 nodes, 608 links and 12 clusters. There were 69 599 keywords in the literature, and the top five keywords from high to low were "transplantation" "adipose tissue" "stem-cell" "tissue" , and "obesity" . In the literature, the keywords with the highest intensity of outburst was "outcome" , and the keywords with the longest duration of outburst was "cartilage" . In recent years, the five keywords with outburst were "liver disease" "trial" "stromal vascular fraction" "outcome" , and " Akkermansia muciniphila" . VOSviewer1.6.16 software visualization clustering result showed that the application of fat transplantation in plastic surgery, the source of adipose tissue and its survival mechanism in vivo, and the laboratory research on fat regeneration were the three hot spots in the field of fat transplantation technology. Conclusions:The research on fat transplantation is on the rise in the world, and research institutions at home and abroad are conducting in-depth exploration on fat transplantation. Recent research focuses on improving the long-term survival rate of fat transplantation, and the research level is gradually in-depth at the molecular level.

7.An automatic VMAT planning method for primary liver cancer radiotherapy based on predicting the feasibility DVH

Objective:To establish an automatic planning method using volumetric-modulated arc therapy (VMAT) for primary liver cancer (PLC) radiotherapy based on predicting the feasibility dose-volume histogram (DVH) and evaluate its performance.Methods:Ten patients with PLC were randomly chosen in this retrospective study. Pinnacle Auto-Planning was used to design the VMAT automatic plan, and the feasibility DVH curve was obtained through the PlanIQ dose prediction, and the initial optimization objectives of the automatic plan were set according to the displayed feasible objectives interval. The plans were accessed according to dosimetric parameters of the planning target volume and organs at risk as well as the monitor units. All patients′ automatic plans were compared with clinically accepted manual plans by using the paired t-test. Results:There was no significant difference of the planning target volume D 2%, D 98%, D mean or homogeneity index between the automatic and manual plans ((58.55±2.81) Gy vs.(57.98±4.17) Gy, (47.15±1.58) Gy vs.(47.82±1.38) Gy, (53.14±0.95) Gy vs.(53.44±1.67) Gy and 1.15±0.05 vs. 1.14±0.07, all P>0.05). The planning target volume conformity index of the manual plan was slightly higher than that of the automatic plan (0.77±0.08 vs. 0.69±0.06, P<0.05). The mean doses of normal liver, V 30Gy, V 20Gy, V 10Gy, V 5Gy and V< 5Gy of the automatic plan were significantly better than those of the manual plan ((26.68±11.13)% vs.(28.00±10.95)%, (29.96±11.50)% vs.(31.89±11.51)%, (34.88±11.51)% vs.(38.66±11.67)%, (45.38±12.40)% vs.(50.74±13.56)%, and (628.52±191.80) cm 3vs.(563.15±188.39) cm 3, all P<0.05). The mean doses of the small intestine, the duodenum, and the heart, as well as lung V 10 of the automatic plan were significantly less than those of the manual plan ((1.83±2.17) Gy vs.(2.37±2.81) Gy, (9.15±9.36) Gy vs.(11.18±10.49) Gy, and (5.44±3.10) Gy vs.(6.25±3.26) Gy, as well as (12.70±7.08)% vs.(14.47±8.11)%, all P<0.05). Monitor units did not significantly differ between two plans ((710.67±163.72) MU vs.(707.53±155.89) MU, P>0.05). Conclusions:The automatic planning method using VMAT for PLC radiotherapy based on predicting the feasibility DVH enhances the quality for PLC plans, especially in terms of normal liver sparing. Besides, it also has advantages for the protection of the intestine, whole lung and heart.
8.Analysis of self-efficacy and influencing factors of male nurses

Objective:To investigate and analyze the current situation and influencing factors of professional decision-making self-efficacy of male nurses.Methods:Convenience sampling was used to select 133 male nurses from September to October 2020 in Shenzhen City as the research objects. The Self-efficacy Scale for Career Decision-making was used to conduct self-evaluation, career information collection, career goal selection, career planning formulation, and job-selection problem resolution.Results:Attitudes towards nursing majors, academic qualifications, reasons for applying for nursing majors, monthly family income, whether it is an only child, family residence, work status, number of job changes, mother′s education level, and married or not were the factors that affect male nurses′ professional self-efficacy ( t values were -1.989-12.523, F values were 7.476-325.316, P<0.05 or 0.01). Conclusion:Career decision-making self-efficacy has a good guiding role in the career selection, development and planning of male nurses. Medical units should formulate reasonable training methods or related training for new male nurses entering the clinic, so as to increase male nurses′ recognition of their occupations, thereby enhancing them Career decision-making self-efficacy.
9.Screening of interacting proteins of idiopathic gonadotropin-releasing hormone deficiency pathogenic gene RNF216.

OBJECTIVE:
To screen proteins interacting with ring finger protein 216(RNF216) through yeast two hybrid experiment, and further clarify the role of RNF216 in the pathogenesis of gonadotropin-releasing hormone deficiency.
METHODS:
A recombinant expression vector pGBKT7-RNF216 was constructed and transformed into yeast Y2HGold, which was hybridized with a human cDNA library in order to screen proteins interacting with RNF216. The interaction was verified in yeast Y2HGold.
RESULTS:
A recombinant expression vector pGBKT7-RNF216 was successfully constructed and expressed in yeast Y2HGold. Filamin B (FLNB) was identified by yeast two hybrid experiment, and their interaction was verified in yeast Y2HGold.
CONCLUSION
An interaction between FLNB and RNF216 was identified through yeast two hybrid experiment. RNF216 may affect the proliferation and migration of GnRH neurons by regulating FLNB or FLNB/FLNA heterodimers.

10.Diagnosis of two cases from one family with Joubert syndrome caused by novel mutations of TCTN1 gene by whole exome sequencing.

OBJECTIVE:
To explore the pathogenesis of two fetuses from one family affected with Joubert syndrome (JS).
METHODS:
Whole exome sequencing was employed to screen potential mutations in both fetuses. Suspected mutations were verified by Sanger sequencing. Impact of intronic mutations on DNA transcription was validated by cDNA analysis.
RESULTS:
Two novel TCTN1 mutations, c.342-8A>G and c.1494+1G>A, were identified in exons 2 and 12, respectively.cDNA analysis confirmed the pathogenic nature of both mutations with interference of normal splicing resulting in production of truncated proteins.
CONCLUSION
The genetic etiology of the family affected with JS has been identified.Above findings have enriched the mutation spectrum of TCTN1gene and facilitated understanding of the genotype-phenotype correlation of JS.
