1.Eucommia ulmoides promotes alveolar bone formation in ovariectomized rats

BACKGROUND:Eucommia ulmoides has a certain osteogenic effect,which can promote the proliferation and differentiation of osteoblasts.However,it is unclear whether Eucommia ulmoides has effects on alveolar bone formation and Wnt/β-Catenin signaling pathway. OBJECTIVE:To investigate the mechanism by which Eucommia ulmoides promotes alveolar bone formation in ovariectomized rats based on the Wnt/β-Catenin signaling pathway. METHODS:Sixty female Sprague-Dawley rats were selected and randomly divided into five groups:blank control group,sham-operation group,model group,low-dose group Eucommia ulmoides group,and high-dose Eucommia ulmoides group,with twelve rats in each group.Osteoporosis animal models were constructed by bilateral oophorectomy in the model group and the low-dose and high-dose Eucommia ulmoides groups.The sham-operation group underwent the same method to remove adipose tissue of equal mass around the bilateral ovaries.Three months after surgery,the low-and high-dose Eucommia ulmoides groups were given 2.1 g/kg/d and 4.2 g/kg/d Eucommia ulmoides by gavage,respectively.The sham-operation group and model group were given the same amount of physiological saline by gavage.After 12 weeks of drug intervention,the changes in alveolar bone mass of rats in each group were observed through Micro-CT;hematoxylin-eosin staining was used to observe the pathological structural changes of alveolar bone in rats;enzyme linked immunosorbent assay was used to detect the expression levels of alkaline phosphatase and osteocalcin in the serum of rats;western blot was used to detect the expression levels of β-Catenin and Frizzled9 receptor proteins in the alveolar bone of rats;and real-time fluorescence quantitative PCR was used to detect the expression of osteocalcin,Runt-related transcription factor 2(Runx2),alkaline phosphatase,β-catenin,and frizzled9 mRNAs in alveolar bone tissues of rats. RESULTS AND CONCLUSION:Compared with the blank control group,bone volume fraction,trabecular number,trabecular thickness,and bone mineral density were reduced in the model group(P<0.05),and trabecular separation was elevated(P<0.05).Pathological observation showed that the arrangement of trabeculae was disordered and irregular,the trabeculae were thinned or broken,and the marrow cavity was enlarged in the model group,with a significant reduction in bone volume;the level of alkaline phosphatase in the serum was increased(P<0.05),and the level of osteocalcin was decreased(P<0.05);mRNA expression of alkaline phosphatase,osteocalcin,Runx2,β-catenin,and frizzled9 were decreased(P<0.05);protein expression of β-Catenin and Frizzled9 was decreased(P<0.05).Compared with the model group,the low-and high-dose Eucommia ulmoides groups showed an increase in bone volume fraction,trabecular number,trabecular thickness,and bone mineral density(P<0.05)and a decrease in trabecular separation(P<0.05).In the low-and high-dose Eucommia ulmoides groups,bone trabeculae were slightly aligned and thickened,with a significant increase in bone mass.Compared with the model group,the serum level of alkaline phosphatase was reduced(P<0.05)and the serum level of osteocalcin was elevated(P<0.05)in the low-and high-dose Eucommia ulmoides groups.Compared with the model group,the mRNA expression of alkaline phosphatase,osteocalcin,Runx2,β-catenin,and frizzled9 were increased in the low-and high-dose Eucommia ulmoides groups(P<0.05).Compared with the model group,the protein expression of Frizzled9 was increased in the low-dose Eucommia ulmoides group(P<0.05),while the protein expression of β-Catenin and Frizzled9 was increased in the high-dose Eucommia ulmoides group(P<0.05).Compared with the low-dose Eucommia ulmoides group,the high-dose Eucommia ulmoides group had a more significant improvement in the above indexes.To conclude,Eucommia ulmoides can effectively promote the alveolar bone formation,and its mechanism of action might be related to the activation of the Wnt/β-catenin signaling pathway.
2.Identification of monoclonal antibodies against spike protein of severe fever with thrombocytopenia syndrome virus based on immune repertoire sequencing

Objective To obtain monoclonal antibodies against the glycoprotein Gn of severe fever with thrombocytopenia syndrome virus(SFTSV) based on immune repertoire sequencing and identify the biological characteristics.Methods Using purified SFTSV-Gn as bait, virus-specific B cells were captured from B cell library of immunized female BALB/c mice. Based on 10 × immune repertoire sequencing and bioinformatics analysis, the antibody sequences with potential binding activity were selected and constructed into eukaryotic expression vector pcDNA3.4. The monoclonal antibodies were expressed and purified, and identified for the biological activity.Results Fourteen antibodies were selected from 3 544 captured B cells, and the functional verification showed that 12 antibodies could bind to SFTSV-Gn glycoprotein, among which C9 and 9/10 antibodies had higher binding ability. Western blot, indirect immunofluorescence assay(IFA) and flow cytometry proved that they specifically recognized SFTSV, and the effective concentration was as low as 2 μg/mL.Conclusion The monoclonal antibodies against SFTSV-Gn glycoprotein were successfully obtained, which lays a foundation for the development of rapid diagnostic reagents for SFTSV.
3.Regulation of Gastrointestinal Tumor Stem Cells by Traditional Chinese Medicine: A Review

Gastrointestinal tumors (GTs), including colorectal cancer, gastric cancer, liver cancer, pancreatic cancer, and esophageal cancer, are increasing in incidence worldwide and have become one of the major diseases threatening human health. Tumor stem cells (TSCs), an undifferentiated subpopulation within tumor tissues, possess biological characteristics such as self-renewal, multidirectional differentiation, high tumorigenicity, and resistance to radiochemotherapy. They play an important role in the occurrence, progression, recurrence, and metastasis of GTs and have increasingly become a research hotspot in GT treatment. Although modern medicine has made remarkable progress, there remain many problems in therapeutic approaches targeting TSCs. In this context, traditional Chinese medicine (TCM), with its favorable safety profile and multi-target mechanisms, has shown potential advantages and value in regulating TSCs. It can reduce TSC drug resistance, enhance the sensitivity of tumor cells to chemotherapeutic agents, inhibit tumor growth and metastasis, and has shown unique advantages in improving the quality of life and prolonging the survival of GT patients. Studies have found that active components of Chinese medicine, such as terpenoids, polyphenols, flavonoids, glycosides, and quinones, and Chinese medicine compound formulas, including Zuojin pills, Sijunzi decoction, Biejiajian pills, and Xuanfu Daizhe decoction, can inhibit TSCs-related signaling pathways such as the Notch signaling pathway, the Wnt/β-catenin signaling pathway, the phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) signaling pathway, and the Hippo signaling pathway. They also reduce the expression of TSC surface markers, including sex-determining region Y-box 2 (SOX2), sex-determining region Y-box 9 (SOX9), octamer-binding transcription factor 4 (OCT4), prominin-1 (CD133), cluster of differentiation 44 (CD44), cluster of differentiation 24 (CD24), and thyroid transmembrane protein 1 (CD90), thereby hindering TSC differentiation, accelerating their metabolic processes, improving the tumor microenvironment, and consequently inhibiting GT growth. This study collects and analyzes recent research on the regulation of TSCs by TCM in the treatment of GT, aiming to provide a new theoretical basis for tumor therapy with TCM, expand its application in the comprehensive treatment of GT, and offer new therapeutic ideas and methods for clinical practice.
4.Qualitative study on the behavioral intentions of pediatric nurses towards the use of vascular visualization devices in light of the theory of planned behavior

Objective:To explore the behavioral intentions of pediatric nurses towards using vascular visualization devices in light of the theory of planned behavior.Methods:This was a qualitative study. From March to November 2023, 20 pediatric nurses from Peking University First Hospital were selected using purposive sampling for semi-structured interviews. The interview data were analyzed and themes were extracted using Colaizzi 7-step analysis method.Results:The behavioral intentions of pediatric nurses towards the use of vascular visualization devices could be summarized into nine themes based on the theory of planned behavior. Attitudes towards behavior included positive evaluations, negative evaluations, and unclear information. Subjective norms involved peers' attitudes as well as expectations and pressures from patients. Perceived behavioral control encompassed workload, level of knowledge, more limited equipment performance, and external resource support.Conclusions:The behavioral intentions of pediatric nurses towards the use of vascular visualization devices are influenced by their attitudes towards the behavior, subjective norms, and perceived behavioral control. Nursing managers should conduct standardized training on vascular visualization devices to enhance nurses' knowledge and skills, while also valuing the sources of various supports to create a supportive environment for the use of vascular visualization devices. This will improve the usage experience of pediatric nurses and promote the use of vascular visualization devices in pediatric nursing practice.

Premature ovarian insufficiency(POI),also known as"ovarian insufficiency",has an incidence of 1%~5%.The incidence has been on the rise in recent years,seriously affecting women's physical and mental health and quality of life.At present,the cause and mechanisms of POI are still unclear,and the method and applications of model construction are also confusing.Most models have some shortcomings in pertinence and stability.The limitations greatly limit research into the clinical diagnosis and treatment of POI.This paper summarizes and discusses the etiology and pathogenesis of POI and the construction of POI animal models to provide a comprehensive reference for those studying POI.
6.Lithium carbonate-induced distal renal tubular acidosis: a case report and literature review

Antipsychotics, lithium preparations can cause a variety of renal side effects, most of which occur insidiously. The paper reports a 46-year-old female patient developing fatigue and soft paresis after taking lithium carbonate for 17 years. Laboratory tests showed hypokalemia, distal renal tubular acidosis (dRTA), and renal calculus. After discontinuation of lithium carbonate, partial remission of hypokalemia and dRTA were observed. Combined with literature review, in addition to dRTA, the renal side effects of lithium preparations also include acute toxic kidney injury, nephrogenic diabetes insipidus and various glomerulopathy.

Replantation of traumatic amputation in children has its own characteristics. This consensus primarily focuses on the issues related to the treatment of traumatically amputated limb injuries in children. Organised along a timeline, the consensus summarises domestic and international clinical experiences in emergency care and injury assessment of traumatic limb amputation limbs, indications and contraindications for replantation surgery, principles and procedures of replantation surgery, postoperative medication and management, as well as rehabilitation in children. The aim of this consensus is to propose standardise the treatment protocols for limb replantation for children therefore to serve as a reference for clinical practitioners in medical practices, and further improve the treatment and care for the traumatic limb amputations in children.

Objective In the context of rapid development and widespread application of new generation information technologies represented by big data,artificial intelligence,the Internet of Things,and 5 G,a hospital has initiated the construc-tion and application of an integrated smart emergency medical information management system project for"Pre-Hospital Emergen-cy+Emergency Department+Specialized Disease Center."Methods T This paper primarily discusses the construction of the"Pre-Hospital Emergency+Emergency Department+Specialized Disease Center"integrated smart emergency medical information management system project,summarizing and exploring the implementation of a new efficient emergency treatment model charac-terized by"hospital admission upon boarding"and"no waiting for emergency care."Results A comprehensive integrated sys-tem covering pre-hospital emergency,triage,in-hospital treatment,and five major centers has been established.This system aims to optimize the treatment process,enhance treatment efficiency,and create a better medical experience for patients.Conclusion The paper discusses the optimization and improvement of the emergency medical information platform under the new circum-stances.
10.Relationship between haptoglobin gene polymorphism and disease severity and susceptibility in senile vascular dementia patients

Objective To explore the relationship between haptoglobin gene polymorphism and disease se-verity and susceptibility in senile vascular dementia patients.Methods A total of 80 patients with senile vas-cular dementia admitted to the hospital from February 2018 to February 2023 were selected as the vascular de-mentia group,and 80 stroke patients with non-vascular dementia admitted to the hospital during the same pe-riod were selected as the control group.The genotype distribution and allele frequency of haptoglobin gene were measured using polymerase chain reaction with sequence-specific primers,and their relationship with the severity and susceptibility of vascular dementia patients was analyzed.Results The proportion of history of hyperlipidemia and diabetes mellitus and the levels of total cholesterol and triglyceride in vascular dementia group were higher than those in control group,the differences were statistically significant(P<0.05).The distribution of genotypes was in Hardy-Weinberg equilibrium(P>0.05).The frequency of haptoglobin 2-2 genotype and haptoglobin 2 allele in vascular dementia group were higher than those in control group,and the differences were statistically significant(P<0.05).There were significant differences in the scores of mini-mental state examination and hachinski ischaemic score among patients with vascular dementia with different haptoglobin genotypes(P<0.05).Multivariate Logistic regression analysis showed that the carrier of hapto-globin 2-2 genotype and the carrier of haptoglobin 2 allele were independent risk factors for vascular dementia(P<0.05).Conclusion Haptoglobin 2-2 genotype and haptoglobin 2 allele distribution frequency are associ-ated with the occurrence of vascular dementia after stroke,and those with high frequency of haptoglobin 2-2 genotype and haptoglobin 2 allele distribution suffer a severe disease,which can provide reference for early i-dentification and assessment of vascular dementia.
