1.Methodological establishment of red blood cell lysis method for handling Rh typing double group samples

Objective: To establish an accurate and rapid typing method for Rh typing of samples from patients who have received recent blood transfusions by utilizing the difference in osmotic fragility between fresh and old red blood cells. Methods: A lysing solution suitable for destroying old RBCs was prepared. Sixty-one samples collected in our hospital in 2024 with Rh typing of double groups were treated with the lysing solution to remove the old allogeneic red blood cells while preserving the patient's own fresh red blood cells, followed by repeat Rh typing tests. Results: For 61 samples with Rh typing in double groups, 41 were accurately detected identified through the red blood cell lysis method, yielding an identification rate of 67.21%. No significant difference was observed compared to the detection rate of the commonly used capillary centrifugation modified method (χ
=0.103, P>0.05). Conclusion: The red blood cell lysis method provides a novel and rapid experimental approach for clinical use in processing Rh-typed samples that are of double groups, thereby offering a basis for Rh compatibility blood transfusion.
2.Summary of the best evidence for cognitive function improvement in Alzheimer's disease patients

Objective:To retrieve and summarize the best evidence for cognitive function improvement in Alzheimer's disease patients, so as to provide reference for clinical practice.Methods:Following the "6S" evidence model, guidelines, expert consensus, clinical decision, and systematic reviews on cognitive function improvement in Alzheimer's disease patients were systematically retrieved from domestic and foreign databases and websites. The search keywords included "Alzheimer's disease, cognitive function, improvement". Evidence-based nursing methods were used to evaluate the quality of literature and extract evidence. The search period was from July 1, 2000 to July 31, 2024.Results:A total of 15 articles were included, including five guidelines, four expert consensus, two clinical decision, and four systematic reviews. Sixteen pieces of best evidence were summarized from four aspects of evaluation and identification, pharmacological intervention, non-pharmacological intervention, and caregiver support.Conclusions:Patients with Alzheimer's disease require timely identification of symptoms related to cognitive impairment and assessment using standardized scales. Multiple intervention methods, such as pharmacological intervention, non-pharmacological intervention, and caregiver support, should be used to enhance patients' quality of life. Evidence application should be tailored to each patient's specific circumstances through individualized selection and adjustment to ensure the effectiveness and scientific rigor of cognitive function improvement strategies, thereby facilitating the translation of optimal evidence into clinical practice.
3.Correlations between post-doctoral talent cultivation and high-quality development of public hospitals

Objective To explore the role of postdoctoral talent cultivation in high-quality development of public hospi-tals.Methods The data of 165 postdoctoral fellows were analyzed from Zhongshan Ophthalmology Center,Sun Yat-sen Univer-sity over the past decade.The analysis involved examining the entire management process from recruitment to completion,inclu-ding the improvement of relevant systems,evaluation standards,implementation of hierarchical management,mentor-student in-teractions,and academic exchanges,as well as studying the quality enhancement of the postdoctoral talent pool,research output,and employment analysis.Results The standardized management of whole postdoctoral cultivation process and the continuous improvement of systems significantly enhanced the achievements of postdoctoral fellows in research,publications,and commer-cialization of the scientific and research findings.For instance,from 2018 to 2022,the number of National Science Foundation-funded programs that the postdoctoral fellows obtained accounted for 26.7%of the total number,while the number of provincial and ministerial funded programs accounted for 73.3%.In 2020,the proportion of postdoctoral researchers who obtained the Na-tional Natural Science Foundation of China's Youth Program was 80%of the total.In 2021,the proportion of postdoctoral fellows who published papers with an impact factor of above 10 as the first author or co-first author was 45.83%.Conclusion The training of high-level medical postdoctoral talents is crucial for high-quality development of public hospitals,improving interdisci-plinary adaptability of postdoctoral fellows and collaborative mentorship awareness.
4.Morita therapy for misophonia: a case report

Misophonia is a psychophysiological and behavioral disorder characterized by an individual's low tolerance to specific sounds, leading to intense negative emotional experiences and physiological responses. Currently, there is no standardized and universally effective treatment for misophonia in clinical practice worldwide. This article reports the case of an 18-year-old male patient with misophonia who showed poor response to sertraline combined with exposure and response prevention therapy. Subsequently, the patient received 8 weeks of Morita therapy (once a week, 50 minutes per session), with symptomatic improvement. By presenting this case, we explore the potential efficacy of Morita therapy in treating misophonia, aiming to provide a reference for its clinical management. 5.The role of BRD4 in HPV16 virus replication in cervical tissue and cells

Abstract:
To explore the relationship between the replication-associated bromodomain protein 4 ( BRD4)and human papillomavirus (HPV) 16(HPV16) viral load in cervical squamous cell carcinoma and CIN Ⅰ tissues , confirm the effects of BRD4 degradation agent MZ1 on viral load .
Methods :
Thirty HPV16-positive cervi- cal cancer specimens and 30 non-cervical cancer specimens were collected , and the viral load of the samples was detected by real-time fluorescence quantitative PCR , and the expression of BRD4 was analyzed by immunohisto- chemistry and Western blot.
Results :
The viral load was higher in the samples of cervical cancer group than in the samples of non-cancer group , the difference is statistically significant ( P < 0. 01) . Immunohistochemistry results showed that the expression of BRD4 were significantly higher in cervical cancer specimens than in noncancerous specimens , the difference is statistically significant (P < 0. 05) . BRD4 expression was significantly and positively correlated with high viral loads , the difference is statistically significant (P < 0. 001) . the BRD4 degradation agent MZ1 significantly reduced the viral load , the difference is statistically significant (P < 0. 01) .
Conclusion
BRD4 may be involved in the replication of HPV16 virus , and BRD4 degradation agent MZ1 can inhibit the replication of HPV16 virus .
6.Modulation of osteogenic differentiation in mouse osteoprogenitor cells by curved microgrooves with varied roughness on polycaprolactone surfaces

Objective:To construct curved microgrooves with gradient surface roughness on polycaprolactone (PCL) members by regulating alkali etching time and to investigate the synergistic effect of surface roughness and curved microgrooves on the in vitro osteogenic differentiation of mouse pre-osteoblasts (MC3T3-E1), aiming to determine the optimal PCL surface modification strategy. Methods:Soft lithography and melt-casting techniques were used to fabricate PCL membranes with regularly arranged curved microgrooves (CMP). Alkali etching was performed for 24, 48, and 72 h. Groups: smooth PCL (control), CMP (curved microgrooves only), CMP-24 h, CMP-48 h, CMP-72 h (CMP etched for 24, 48, 72 h, respectively). Surface physicochemical properties were characterized: surface morphology was observed by scanning electron microscopy (SEM), surface roughness was measured by atomic force microscopy (AFM), and surface hydrophilicity was evaluated by contact angle measurement. MC3T3-E1 cells were cultured in vitro. Cell adhesion, proliferation, and osteogenic differentiation were assessed using cell counting (CCK-8), immunofluorescence staining, alkaline phosphatase (ALP) and Alizarin red staining with quantification. The mRNA expression levels of osteogenesis-related genes [ALP, collagen type Ⅰ (COL-1), Runt-related transcription factor 2 (RUNX-2), osteocalcin (OCN), osteopontin (OPN)] were detected by real-time fluorescence quantitative PCR (RT-qPCR). Results:Curved microgrooves were successfully fabricated on PCL membranes. Alkali treatment improved surface hydrophilicity and increased roughness. The CMP-72 h group exhibited the best hydrophilicity (contact angle: 33.2°±5.5°), with significant differences compared to all other groups (all P<0.05). The CMP-72 h group showed the highest roughness [(59.966±4.729) nm], which was significantly different from all other groups (all P<0.05). CCK-8 results on day 5 showed that both curved microgrooves and roughness promoted cell proliferation: CMP-24 h (0.292±0.003) and CMP-72 h (0.383±0.004) were significantly higher than those in the smooth group (0.270±0.005) (all P<0.05). Immunofluorescence staining revealed that curved microgrooves induced significant contact guidance of cells; this effect weakened with increasing etching time. ALP and Alizarin red staining indicated the deepest osteogenic staining in the CMP-48 h group. Both ALP activity (0.013 021±0.000 032) and Alizarin red quantification (0.290±0.003) were highest in the CMP-48 h group, significantly different from all other groups (all P<0.05). RUNX-2 expression in CMP-24 h and CMP-48 h groups (1.845±0.087 and 1.837±0.027, respectively) was significantly higher than in other groups (all P<0.05), with no significant difference between these two groups ( P>0.05). CMP-48 h group exhibited the highest mRNA expression of all osteogenic genes tested, specifically ALP (2.194±0.028), COL-1 (1.983±0.024), OCN (7.644±0.156), and OPN (2.648±0.031), all significantly greater than other groups (all P<0.05). Conclusions:Both curved microgrooves and surface roughness modification enhance the in vitro osteogenic differentiation of cells on PCL membranes. Among the tested strategies, alkali etching of curved microgrooves for 48 hours (CMP-48h) provided the optimal enhancement of osteogenic capability for MC3T3-E1 cells and represented a promising surface modification strategy for future PCL membranes.
7.Analysis of acoustic characteristics of speech in Parkinson's disease patient recorded via smartphones

Objective To investigate the acoustic characteristics of speech in Parkinson's disease(PD)pa-tients compared to healthy individuals using smartphone-recorded speech,laying the foundation for future intelligent cloud-based PD detection.Methods A total of 81 PD patients and 63 healthy controls were randomly selected to re-cord their speech using Huawei,Xiao mi,and Apple smartphones through the"Active Health"screening app from the National Key R&D Program.Using the Praat library in Python,parameters such as mean fundamental frequen-cy(Mean F0),minimum fundamental frequency(Min F0),maximum fundamental frequency(Max F0),formants(F1,F2,F3,F4),shimmer,noise-to-harmonics ratio(NHR),and jitter were extracted and compared.Results In PD patients,jitter and shimmer were significantly higher in the/ba/,/bi/,/bu/,and/bo/vowels compared to the control group,while Mean F0 and Max F0 was significantly lower in/bi/and/bo/.Min F0 was significantly higher in/bi/and/bu/for PD patients.No significant differences were found in F1,F2,F3,or F4 between PD pa-tients and controls in/ba/,/bi/,/bo/and/bu/.Conclusion Quantitative analysis of smartphone-recorded speech can detect speech changes in PD patients,providing a solid foundation for the development of a cloud-based PD de-tection system.
8.Impact of tubeless technique on male sexual function after transurethral ureteroscopic lithotripsy

Objective To investigate the impact of tubeless technique on male sexual function after transurethral ureteroscopic lithotripsy.Methods 82 male patients from January 2023 to June 2023 with transethral ureteroscopic lithotripsy were selected as the study objects.Prior to surgery,39 male patients without double J tube indwelling after lithotripsy were randomly selected as the tubeless group and 43 male patients with double J tube indwelling were selected as the indwelling group by drawing lots.General clinical data and postoperative complications(lumbago,hematuria,irritation sign of bladder and fever)of the two groups were compared.The International Index of Erectile Function-5(IIEF-5)score,sexual life quality questionnaire(SLQQ)score and premature ejaculation diagnostic tool(PEDT)score of the two groups were statistically analyzed before surgery,1 month after surgery(before excutation in indwelling group),3 months and 6 months after surgery.Results Compared with the indwelling group,patients in the tubeless group had significantly fewer cases of lumbago,hematuria,and irritation sign of bladder 1 month after surgery,and the difference was statistically significant(P<0.05),but there was no significant difference in postoperative fever between the two groups(P>0.05).There were no statistically significant differences in the IIEF-5 score,SLQQ score and PEDT score between the two groups before the operation,3 months after the operation and 6 months after the operation(P>0.05).One month after the operation,the IIEF-5 score and SLQQ score of the tubeless group were significantly higher than those of the indwelling group,while the PEDT score was lower than that of the indwelling group,the differences were statistically significant(P<0.01).There were no statistically significant difference in the IIEF-5 score,SLQQ score and PEDT score at different time points in the tubeless group(P>0.05).The IIEF-5 score and SLQQ score of the indwelling group one month after the operation were significantly lower than those before the operation,while the PEDT score was significantly higher than that before the operation,the IIEF-5 score and SLQQ score of the indwelling group 3 and 6 months after the operation were significantly higher than those at 1 month after the operation,and the PEDT score was significantly lower than that at 1 month after the operation,the differences were statistically significant(P<0.01).There were statistically significant differences in the IIEF-5 score,SLQQ score and PEDT score at different time points in the indwelling group(P<0.05).Conclusion The tubeless technique after ureteroscopy has a relatively small impact on male sexual function and can significantly improve the quality of life of patients,alleviate lumbago,hematuria and irritation sign of bladder.It is worthy of clinical promotion and application.
9.Establishment of RAA detection method for infectious laryngotracheitis virus

The aim of this study is to establish a rapid,efficient,and sensitive method for detecting the infectious laryngotracheitis virus(ILTV).The DNA of ILTV was extracted and used as a tem-plate to develop a recombinant enzyme-mediated isothermal amplification(RAA)fluorescence de-tection method for ILTV through optimization of conditions,sensitivity analysis,and repeatability assessment.Additionally,the nucleic acids of avian influenza virus(AIV),IBV,and Newcastle dis-ease virus(NDV)were detected to verify the specificity of this method.Finally,this method was applied to analyze 59 clinical samples collected from multiple large-scale chicken farms in Hebei Province,and the results were compared with those obtained from real-time fluorescence quantifi-cation(qPCR)and PCR methods according to national standards.The results showed that the RAA detection method established in this study had a reaction system of 25.0 μL buffer,2.1 μL primer,0.6 μL probe,5.0 μL magnesium acetate,and 5.0 μL template.The reaction temperature was 39 ℃ and the amplification time was within 20 minutes.The sensitivity of this method was 101 copies/μL,and the specificity detection was 100%.Testing of 59 clinical samples showed that 17 were detected positive by both RAA fluorescence and qPCR,and 12 were detected by PCR,and the detection rate of RAA(fluorescence)was consistent with real-time fluorescence quantification and qPCR,which was higher than that of the PCR assay.The research results indicate that the RAA fluorescence method has a short detection time,good specificity and sensitivity,and can be used for rapid detection of ILTV.
