1.The impact of the Guangzhou voluntary blood donation privilege certificate on blood donation behavior

Objective: To evaluate the causal effects and population heterogeneity of Guangzhou′s Voluntary Blood Donation Privilege Certificate Policy on blood donation behavior, and to provide empirical evidence for optimizing blood management policies. Methods: Using an interrupted time series (ITS) design, we analyzed 30 quarters of blood collection data from Guangzhou Blood Center (from July 2018 to December 2025). Taking the third quarter of 2021 as the intervention node, we constructed ordinary least squares (OLS) regression models incorporating level and trend effects, and conducted stratified analyses by gender, age group, previous donation frequency, and per-donation volume. Results: The tiered incentive policy demonstrated significant heterogeneous effects. Apheresis donations showed sustained growth, with 2 units of component blood exhibiting a significant trend effect (β=304.58, P<0.001); high-frequency donors (11+ times) demonstrated sustained growth trends in apheresis donations. For whole blood, immediate effects were significant for donors with 1-2 previous donations (β=4 537.55, P<0.001) and 3-10 previous donations (β=2 159.69, P<0.05); high-frequency donors (11-30 times) showed sustained growth trends (P<0.01). Stratified by per-donation volume, 400 mL whole blood demonstrated a significant immediate effect (β=4 391.01, P<0.1), while 200 mL whole blood showed a significant declining trend (β=-893.24, P<0.01). Conclusion: The tiered incentive policy effectively enhanced blood donation participation while maintaining the donors’ altruistic motivations. Young and middle-aged adults primarily drove apheresis donations. The policy demonstrated immediate incentive effects on low-frequency whole blood donors and long-term retention effects on high-frequency donors. Per-donation volume showed an "upward migration" trend, optimizing the blood collection structure. Differentiated outreach, conversion and retention strategies are recommended to further strengthen blood supply security.
2.Relationship between Polymorphisms of Vitamin D Receptor Gene TaqI rs731236 and BsmI rs1544410 and Benign Paroxysmal Positional Vertigo

Objective To investigate the relationship between vitamin D receptor(VDR)gene TaqI rs731236 and BsmI rs1544410 polymorphisms and benign paroxysmal positional vertigo(BPPV),the relationship and clinical significance of BPPV.Methods A total of 263 patients with confirmed BPPV admitted to Dongguan People's Hospital from January 2021 to October 2023 were retrospectively selected,and another 100 healthy subjects in the same period were selected as controls.Single nucleotide polymorphisms(SNPs)at the locus of TaqI rs731236 and BsmI rs1544410 gene locus of the were detected by fluorescence quantitative PCR.Single nucleotide polymorphisms(SNP)at TaqI rs731236 and BsmI rs1544410 were detected by fluorescence quantitative PCR.The genetic balance of TaqI and BsmI genes was tested by Hardy-Weinberg balance.Genotype and allele distribution were compared between the two groups.The correlation between TaqI and BsmI genotype and clinical features of BPPV was analyzed.The risk factors of BPPV were analyzed by Logistic regression.Results Compared with the clinical data of the two groups,the levels of diabetes(22.05%),hypertension(28.90%),blood uric acid(345.38±36.69μmol/L),cholesterol(4.98±0.68mmol/L)and low density lipoprotein(3.48±0.67mmol/L)in BPPV group were significantly higher than those in control group(11.00%,15.00%,328.76±32.24μmol/L,4.36±0.57mmol/L,3.07±0.62mmol/L),and the serum 25(OH)D level(62.78±8.69nmol/L)was significantly lower than that in control group(97.65±9.54nmol/L),and the differences were statistically significant(t/χ2=3.982~33.233,all P<0.05).Hardy-Weinberg test showed that the genotype distribution of the SNP locus of TaqI and BsmI genes conforms s to the law of genetic equilibrium and is representative of the population(χ2=1.406~2.851,all P>0.05).The frequencies of AG genotype(53.99%)and A allele(57.41%)at TaqI rs731236 in BPPV group were significantly higher than those in control group(44.00%,48.00%),while the frequencies of GG genotype(15.59%)at TaqI rs731236 in BPPV group were significantly lower than those in control group(30.00%),the frequencies of CC genotype(32.32%)and C allele(54.56%)at BsmI rs1544410 locus in BPPV group were significantly higher than those in control group(18.00%,38.00%),while the frequencies of TT genotype(23.19%)and T allele(45.44%)at BsmI rs1544410 locus were significantly lower than those in control group(42.00%,62.00%),and the differences were statistically significant(χ2=9.589,5.185;14.603,15.900,all P<0.05).BPPV patients carrying AG and AA genotypes at TaqI rs731236 and CT and CC genotypes at BsmI rs1544410 had higher blood uric acid and cholesterol levels and lower serum 25(OH)D levels,and the differences were statistically significant(t=4.256~12.742,all P<0.05).The AG genotype and A allele at TaqI rs731236,CC genotype and C allele at BsmI rs1544410 were one of the independent risk factors for BPPV(Wald χ2=8.651,7.269;5.010,6.793,all P<0.05).Conclusion Carrier of AG genotype and A allele at TaqⅠ rs731236 and CC genotype and C allele at BsmI rs1544410 are closely associated with blood uric acid,cholesterol and serum 25(OH)D levels,and insrease the risk of BPPV.The polymorphism of VDR gene is an independent risk factor for BPPV.
3.LncRNA GUSBP11 regulates malignant biological behaviors of gastric cancer cells through the miR-339-5p/MDM2 axis

Objective:To investigate the effect of long non-coding RNA glucuronidase β pseudogene 11(GUSBP11)regulating miR-339-5p/mouse two-minute homolog 2(MDM2)axis on the proliferation,migration,and invasion of gastric cancer AGS cells.Methods:Cancerous and adjacent tissues from 25 gastric cancer patients who underwent surgical treatment at Foshan Hospital of Traditional Chinese Medicine Affiliated to Guangzhou University of Chinese Medicine from December 2023 to June 2024 were collected.Gastric cancer AGS cells and normal gastric mucosal epithelial GES-1 cells were routinely cultured.The control plasmids and knockdown plasmids were transfected into AGS cells using transfection reagents,dividing the cells into Ctrl group,sh-NC group,sh-GUSBP11 group,sh-GUSBP11+anti-NC group,and sh-GUSBP11+anti-miR-339-5p group.The mRNA expression of GUSBP11,miR-339-5p,and MDM2 in gastric cancer tissues and cells of each group was detected by qPCR.A dual-luciferase reporter gene assay was used to detect the targeting relationship between GUSBP11 or MDM2 and miR-339-5p.EdU staining,scratch healing assay,and Transwell chamber assay were adopted to assess the proliferation,migration,and invasion abilities of AGS cells,respectively.WB assay was used to measure the protein expression of CDK1,MMP-2,and MMP-9 in AGC cells.The effects of GUSBP11 knockdown on tumor growth were examined through AGS cell xenograft experiments.Results:The mRNA expression of GUSBP11 and MDM2 were significantly upregulated in gastric cancer tissues and cells(both P<0.05),while miR-339-5p was significantly downregulated(P<0.05).A targeting relationship was found between GUSBP11 and miR-339-5p,as well as between MDM2 and miR-339-5p.Knockdown of GUSBP11 in AGS cells significantly inhibited MDM2 protein expression and promoted miR-339-5p expression,while inhibition of miR-339-5p promoted MDM2 protein expression.GUSBP11 knockdown significantly inhibited the proliferation,migration,and invasion of AGS cells,while inhibition of miR-339-5p reversed this effect.GUSBP11 knockdown significantly inhibited the protein expression of CDK1,MMP-2,and MMP-9,and inhibition of miR-339-5p reversed this effect.Furthermore,GUSBP11 knockdown significantly inhibited the growth of AGS cell xenografts.Conclusion:GUSBP11 is highly expressed in gastric cancer tissues and cells,and knocking down GUSBP11 expression may inhibit malignant biological behaviors of gastric cancer cells through regulating the miR-339-5p/DM2 axis.

In recent years,the problem of population aging in China has been continuously intensifying.The mechanism of aging and its prevention and treatment have gradually become research hotspots.The successful establishment of aging animal models is an important part of aging-related research,and behavioral experiments can effectively evaluate these models.This article reviews the methods of constructing aging animal models from different aspects,analyzes the advantages and disadvantages of various behavioral evaluation experiments and their comparisons,with the aim of providing theoretical references for aging-related research.

Acute Gelsemium poisoning is a systemic disease primarily affecting the central nervous system and respiratory symptoms caused by the ingestion of a substantial amount of Gelsemium within a short period. It manifests as sudden onset and rapid progression, primarily caused by accidental ingestion due to misidentification, and posing significant health risks. The compilation of the Technical Plan for Emergency Health Response to Acute Gelsemium Poisoning describes in detail the specialized practice and technical requirements in the process of handling acute Gelsemium poisoning, including accident investigation and management, laboratory testing and identification, in-hospital treatment, and health monitoring. The guidelines clarify key procedures and requirements such as personal protection, investigation elements, etiology determination, medical rescue, and health education. The key to acute Gelsemium poisoning investigation lies in promptly identifying the toxin through exposure history, clinical manifestations, and sample testing. Because there is no specific antidote for Gelsemium poisoning, immediate removal from exposure, rapid elimination of the toxin, and respiratory monitoring are critical on-site rescue measures. Visual identification of food or herbal materials, followed by laboratory testing to determine Gelsemium alkaloids in samples is a rapid effective screening method. These guidelines offer a scientific, objective, and practical framework to support effective emergency responses to acute Gelsemium poisoning incidences.
7.Correlation analysis of central venous oxygen saturation-related indexes at different time points with low cardiac output syndrome in children after congenital heart disease correction surgery

Objective To explore the correlation between central venous oxygen saturation(ScvO2)-re-lated indexes at different time points and the occurrence of low cardiac output syndrome(LCOS)after con-genital heart disease(CHD)correction surgery.Methods A total of 73 children who underwent CHD correc-tion surgery in this hospital from July 1st,2021 to July 1st,2024 were selected as the research subjects.The clinical data,preoperative conditions,and postoperative conditions of the children were collected.The ScvO2 and arterial lactate(Lac)levels of the children at different time points(the 1st,6th,12th,and 24th hours after surgery)were monitored,and the ScvO2/Lac at different time points and the change rate of ScvO2 in different time periods were calculated.The correlation between ScvO2-related indexes and LCOS after CHD correction surgery was analyzed.Results ScvO2 at the 6th hour after surgery,ScvO2 at the 12th hour after surgery,Sc-vO2/Lac at the 12th hour after surgery,the change rate of ScvO2 from the 1st to the 24th hour after surgery,the change rate of ScvO2 from the 6th to the 12th hour after surgery,and the change rate of ScvO2 from the 12th to the 24th hour after surgery were independent influencing factors of LCOS occurrence after CHD cor-rection surgery(P<0.05).There was a negative correlation between ScvO2 at the 12th hour after surgery,ScvO2/Lac and LCOS occurrence after CHD correction surgery(r=-0.543,-0.523,P<0.05).The area under the curve(AUC)of ScvO2 at the 12th hour after surgery for predicting LCOS occurrence after CHD correction surgery was 0.938(95%CI:0.865-1.000);the AUC of ScvO2/Lac at the 12th hour after surgery for predicting LCOS occurrence after CHD correction surgery was 0.922(95%CI:0.851-0.994).Conclusion ScvO2 and ScvO2/Lac at the 12th hour after surgery have good predictive potential for LCOS occurrence af-ter CHD correction surgery.
8.Construction and application of nursing quality evaluation index system for percutaneous radiofrequency ablation of liver cancer under the background of disease diagnosis related groups

Objective Based on the background of disease diagnosis related groups(DRGs)in public hospitals to construct the nursing quality evaluation index system for percutaneous radiofrequency ablation(PRFA)of liver cancer so as to improve the nursing quality for PRFA of liver cancer.Methods Using three-dimensional quality model and through interview,literature analysis,two rounds of Delphi expert letter inquiry and analytic hierarchy process,the evaluation standard of PRFA nursing quality of liver cancer was established,and its effect was validated in clinical practice.Results The evaluation index system of PRFA nursing quality for liver cancer was constructed,which included 3 first-level indexes,9 second-level indexes and 22 third-level indexes.After clinical application,the incidence of intraoperative and postoperative moderate-severe pain,the incidence of high fever,and the 24-hour number of times required care in the intervention group were lower than those in the control group(P<0.05),the incidence of postoperative Ⅲ degree vomiting in the intervention group was remarkably lower than that in the control group(P<0.01),and the average hospitalization days in the intervention group were less than those in the control group(P<0.05),and the average hospitalization expense in the intervention group was lower than that in the control group,but the difference was not statistically significant(P>0.05).Conclusion The PRFA nursing quality evaluation index system for liver cancer is scientific and practical,which is helpful for improving the outcome of patients and promoting the quality of nursing.

10.Value of prealbumin-to-total bilirubin ratio in predicting the short-term prognosis of patients with HBV-related acute-on-chronic liver failure

ObjectiveTo investigate the value of prealbumin-to-total bilirubin (PA/TBil) ratio on admission in predicting 90-day mortality or liver transplantation in patients with HBV-related acute-on-chronic liver failure (HBV-ACLF), as well as the effect of its combination with Model for End-Stage Liver Disease (MELD) score on the predictive performance for short-term prognosis. MethodsA retrospective analysis was performed for the clinical data of 216 HBV-ACLF patients who were admitted to Department of Infectious Diseases in the First Affiliated Hospital of Soochow University from April 2020 to May 2025, and the patients were followed up for 3 months. According to the outcome, the patients were divided into survival group with 104 patients and death/transplantation group with 112 patients. The Kolmogorov-Smirnov test was used to check whether the continuous data was in accordance with the normal distribution; the two-independent-samples t test was used for comparison of normally distributed continuous data between two groups, while the Mann-Whitney U test was used for comparison of continuous data with skewed distribution between two groups. The chi-square test was used for comparison of categorical data between two groups. Univariate and multivariate binary Logistic regression analyses were used to investigate the influencing factors for prognosis, and the receiver operating characteristic (ROC) curve was used to analyze the performance of each indicator in predicting the prognosis of ACLF. The area under the ROC curve (AUC) was calculated, and the Delong test was used for comparison of AUC. ResultsA total of 216 patients were enrolled in this study, with a 90-day survival rate of 48.15% (104/216). Compared with the death/transplantation group, the survival group had significantly higher platelet count, lymphocyte count, albumin, and PA/TBil ratio (all P<0.05) and significantly lower age, white blood cell count, neutrophil count, prothrombin time, international normalized ratio, aspartate aminotransferase, total bilirubin, creatinine, and MELD score (all P<0.05). The multivariate Logistic regression analysis showed that age (odds ratio [OR]=1.05, 95% confidence interval [CI]: 1.02 — 1.09, P<0.001), PA/TBil ratio (OR=0.16, 95%CI: 0.05 — 0.46, P<0.001), and MELD score (OR=1.09, 95%CI: 1.01 — 1.17, P=0.024) were independent influencing factors for 90-day prognosis in HBV-ACLF patients. PA/TBil ratio and MELD score used alone or in combination had an AUC of 0.760, 0.779, and 0.811, respectively, in predicting the prognosis of HBV-ACLF patients, and PA/TBil ratio combined with MELD score had a better AUC than PA/TBil ratio or MELD score used alone (Z=-2.058 and 2.017, both P<0.05). ConclusionBoth serum PA/TBil ratio and MELD score can effectively predict the prognosis of patients with HBV-ACLF, and a combination of the two indicators had a better predictive performance than MELD score alone, which provides an important reference for clinical risk stratification management and timely intervention.
