1.Effects of Estradiol tablet/Estradiol and dydrogesterone tablet combined with Dan’e fukang decoction on post-operative indicators in patients with high risk of endometrial basal layer injury after induced abortion

OBJECTIVE To investigate the effects of Estradiol tablet/Estradiol and dydrogesterone tablet combined with Dan’e fukang decoction on postoperative recovery, sex hormone levels, uterine artery blood flow, and endometrium in patients with high risk of endometrial basal layer injury after induced abortion, and evaluate the safety of drug use. METHODS A retrospective analysis was conducted on the clinical data of 244 patients with high risk of endometrial basal layer injury who underwent artificial abortions in our hospital between January 2022 and December 2023. They were divided into control group (120 cases) and observation group (124 cases) according to postoperative medication plan. All patients received anti-infective treatment after induced abortion. Based on it, control group was given Estradiol tablet/Estradiol and dydrogesterone tablet, and observation group was given Estradiol tablet/Estradiol and dydrogesterone tablet+Dan’e fukang decoction, for consecutive 28 d. The time of vaginal bleeding, abdominal pain, return of menstruation, and vaginal bleeding volume were observed in 2 groups; serum sex hormone levels [serum follicle stimulating hormone (FSH), luteinizing hormone (LH), estradiol, and progesterone], uterine artery blood flow parameters [peak systolic blood flow velocity (PSV), pulsatility index (PI), and resistance index (RI)] and endometrial thickness were compared between the two groups before and after treatment. The occurrence of complications and adverse reactions in patients were recorded. RESULTS After treatment, the duration of vaginal bleeding, vaginal bleeding volume, duration of abdominal pain, and menstrual cycle recovery time in observation group were significantly shorter or less than the control group (P<0.05). After treatment, both groups exhibited a significant increase in endometrial thickness, serum FSH and LH levels, and PSV, compared to before treatment, and the observation group was significantly thicker/higher than the control group (P<0.05); the levels of estradiol and progesterone, as well as the values of PI and RI, were significantly lower than those before treatment, and the observation group was significantly lower than the control group (P<0.05). The total incidence of postoperative complications in observation group was significantly lower than control group (P<0.05), but there was no significant difference in the total incidence of adverse reactions between the 2 groups (P>0.05). CONCLUSIONS Estradiol tablet/Estradiol and dydrogesterone tablet combined with Dan’e fukang decoction can accelerate the postoperative recovery of patients with high risk of endometrial basal layer injury after induced abortion, promote endometrial growth, regulate serum sex hormone levels, improve uterine artery blood flow, reduce postoperative complications, and have good safety.
2.Evaluation of the efficacy and safety of cryopreserved deglycerolized red blood cells infusion based on propensity score matching method

[Objective] To compare the efficacy and safety of deglycerolized red blood cells (DRBC) and suspended red blood cells (SRBC) based on the propensity score matching (PSM) method, so as to provide evidence for the rational use of DRBC resources in clinical practice. [Methods] A total of 89 patients who received DRBC transfusion and 2 916 patients who received SRBC transfusion in our hospital from January 2023 to September 2024 were included. A 1∶1 nearest neighbor PSM was used to balance covariates such as gender, age, and body mass index (BMI). The changes of hemoglobin (Hb), red blood cell (RBC) count, hematocrit (HCT), and inflammatory markers such as white blood cell (WBC) count, neutrophil (NE) count, C-reactive protein (CRP), and Interleukin-6(IL-6) in the last 72 hours after transfusion were analyzed by SPSS 26.0 and R software to evaluate clinical efficacy and transfusion safety. [Results] The baseline of the two groups was balanced after PSM (P>0.05). There was no significant difference in the total effective rate between the DRBC group (80.9%) and the SRBC group (86.5%) (P>0.05). In the SRBC group, WBC (×10
/L) increased from 9.634±6.742 to 10.147±6.835, CRP (mg/dL) increased from 5.468±4.647 to 6.174±6.114, and IL-6(pg/mL) decreased from 213.733±587.191 to 157.255±552.626. In the DRBC group, WBC (×10
/L) decreased from 11.123±7.880 to 11.011±8.549, CRP (mg/dL) decreased from 5.729±4.761 to 5.326±4.466, and IL-6(pg/mL) decreased from 238.806±639.060 to 152.255±266.558. Compared with the before treatment, the differences between the SRBC group and DRBC group were not statistically significant (P>0.05). Among all patients included in the statistics, the overall incidence of transfusion adverse reactions was 0.205% (6/2 916) in the SRBC group, and no adverse reactions occurred in the DRBC group. The incidence in the SRBC group was higher than that in the DRBC group. [Conclusion] Based on PSM analysis, there was no significant difference in the efficacy and safety of DRBC transfusion compared with SRBC transfusion, which can provide evidence-based support for routine application.
3.Effect of Wenpi tongluo kaiqiao formula against neuronal necroptosis in mice with Alzheimer’s disease and its mechanism

OBJECTIVE To investigate the effects and mechanism of Wenpi tongluo kaiqiao formula (WPTL) against neuronal necroptosis in Alzheimer’s disease (AD) mice based on the Z-DNA binding protein 1 (ZBP1)/mixed lineage kinase domain-like protein (MLKL) signaling pathway. METHODS Forty APP/PS1 transgenic AD mice were randomly divided into model group, WPTL low-dose (WPTL-L) group (10.4 g/kg, calculated by the raw medicine), WPTL high-dose (WPTL-H) group (20.8 g/kg, calculated by the raw medicine) and donepezil hydrochloride group (3 mg/kg), with 10 mice in each group; another 10 C57BL/6J mice were selected as normal control group. Intragastric administration, once a day, for 30 consecutive days. Twenty-four hours after the last administration, Morris water maze test was performed to evaluate learning and memory abilities; the pathological morphology of hippocampal tissues was observed; the serum levels of tumor necrosis factor-α (TNF-α) and interleukin-4 (IL-4) were determined; the expressions of amyloid precursor protein (APP), Tau protein, and ZBP1/MLKL signaling pathway-related proteins in hippocampal tissues were detected; the positive expression of phosphorylated receptor-interacting protein kinase 3 (p-RIPK3) in the neurons of hippocampal tissues and mRNA expression of ZBP1 were measured in hippocampal tissues. RESULTS Compared with normal control group, the escape latency of mice in model group was prolonged significantly on day 3 to 5 (P<0.05), the times of crossing platform reduced significantly (P<0.05), and obvious pathological changes were observed in the hippocampal tissue. The level of TNF- α, the expressions of APP, p-Tau and ZBP1, the phosphorylation levels of RIPK1, RIPK3 and MLKL, the fluorescence intensity of p-RIPK3 as well as the mRNA expression of ZBP1 were significantly increased (P<0.05), while the serum level of IL-4 was decreased significantly (P<0.05). Compared with model group, above indexes were reversed significantly in administration groups (P<0.05), and pathological damage of hippocampal tissue was alleviated. CONCLUSIONS WPTL can inhibit the ZBP1/MLKL signaling pathway, reduce neuronal necroptosis in AD mice, and inhibit inflammatory responses, thereby improving learning and spatial memory abilities in AD mice.

[摘 要] 目的:肝细胞癌(HCC)是肝脏最常见的原发性恶性肿瘤。早期HCC诊出率低,多数患者就诊时已经是晚期,预后极差,因此亟须探索有效的HCC早期诊断标志物及干预治疗靶点。癌症/睾丸抗原1A(CTAG1A)在多种肿瘤中异常表达并且具有强免疫原性,但其在HCC中的表达特性及免疫原性尚不明确。本研究旨在鉴定HCC组织和细胞中CTAG1A的表达趋势和免疫原性,为HCC早期诊断提供新的生物标志物,为临床免疫治疗提供新的潜在靶点。方法:通过转录组芯片筛选10对极早期HCC(BCLC 0期HCC)肿瘤与癌旁组织的差异表达基因谱。利用RT-qPCR在独立大样本(BCLC 0、A、B、C期HCC组织及癌旁组织, n = 149)及多种肝癌细胞系中验证CTAG1A的表达。通过生物信息学工具(IEDB数据库的TepiTool和Swiss Model)预测CTAG1A的MHC-Ⅰ型和MHC-Ⅱ型抗原表位。采用固相多肽合成法合成候选肽段,经HPLC纯化及质谱验证后,使用IFN-γ酶联免疫斑点法(ELISpot)检测9例HCC患者外周血单个核细胞(PBMC)对各个肽段的特异性T细胞反应。临床样本来源于2015至2022年海军军医大学第三附属医院(东方肝胆外科医院)收治的HCC患者,所有样本的采集使用均获患者知情同意,并经东方肝胆外科医院伦理委员会审查批准(EHBHKY2015-01-017),严格按照相关要求和伦理规定严格执行。采用SPSS 30.0软件进行统计学处理,通过ROC曲线评估诊断效能。结果:转录组芯片筛选结果显示,CTAG1A在极早期HCC(BCLC 0期HCC)中表达显著上调(|FC| = 99.16,P < 0.000 1),临床独立样本验证显示其在各分期HCC中均呈现出高表达趋势且在早期HCC中呈现出较佳的诊断效能(BCLC 0期HCC AUC = 0.6893,敏感性 = 85.71%;BCLC A期HCC AUC = 0.8229,敏感性 = 83.33%);进一步的分析结果显示,CTAG1A在多种肝癌细胞系中的表达显著高于相对正常肝细胞系(P < 0.001)。与甲胎蛋白(AFP)相比,CTAG1A在BCLC 0期和A期HCC中的诊断效能更优(AFP在早期HCC中ROC曲线分析无显著差异,P > 0.05)。生物信息学工具预测发现CTAG1A含8个MHC-Ⅰ型和4个MHC-Ⅱ型抗原表位。IFN-γ ELISpot实验显示,12条合成肽段能不同程度地诱导HCC患者PBMC的特异性T细胞反应。结论:CTAG1A在HCC早期即显著高表达,且具有多表位免疫原性,可能能够激活CD8⁺和CD4⁺ T细胞,提示其具有作为HCC免疫治疗靶点的潜力,可望为开发基于mRNA疫苗或过继性细胞疗法的联合免疫治疗策略提供新的方向。相较于AFP,CTAG1A在早期HCC中展现出更优的诊断效能,提示其作为HCC早期诊断标志物的可能。
5.Association of dining locations with nutritional status among Chinese children aged 6-17 years

Objective:
To analyze the association of eating dining locations and their association with nutritional status among Chinese children aged 6-17 years,so as to provide reference for guiding children s reasonable diet.
Methods:
Stratified random cluster sampling was used to select children aged 6 to 17 years from 28 cities and rural areas of 14 provinces in East, North, Central, South, Southwest, Northwest, Northeast of China, and a total of 52 535 children were included in the study from 2019 to 2021. Information including dining locations, demographic characteristics, dietary intakes and physical activity were collected through a questionnaire survey. Fasting body height and weight were measured in the morning. Unordered multiclass Logistic regression analysis was conducted to assess the relationship between dining locations and nutritional status in children.
Results:
Regarding children s dining locations, 66.3% ate breakfast at home,25.8% ate breakfast at school,7.9% ate breakfast outside (small dining tables, restaurants, stalls, etc.); 67.7% ate dinner at home,29.0% ate dinner at school,3.3% ate dinner outside; and 63.6% ate lunch at school,30.8% ate lunch at home,5.7% ate lunch outside. The prevalence rates of overweight/obesity and undernutrition were 28.6% and 9.3%, respectively. The adjusted multiclass Logistic regression analysis (controlling for age, region, parental education, household income, total energy intake, and moderate-to-vigorous physical activity) demonstrated that, compared to eating at home, school based breakfast and dinner consumption was associated with significantly lower overweight/obesity risks for both genders (boys: breakfast OR =0.70, 95% CI =0.65-0.75; dinner OR =0.80, 95% CI = 0.74- 0.86; girls: breakfast OR = 0.89 , 95% CI = 0.82-0.96; dinner OR =0.88, 95% CI =0.81-0.95), whereas eating lunch away from home significantly increased overweight/obesity risks (boys: OR =1.32, 95% CI =1.17-1.48; girls: OR =1.43, 95% CI =1.26- 1.62 ), with all associations being statistically significant ( P <0.05). After adjusting for confounding factors, boys who ate breakfast away from home showed a significantly reduced risk of undernutrition ( OR =0.80,95% CI =0.66-0.97), while those consuming lunch away from home had an increased risk ( OR =1.26, 95% CI =1.01-1.57) ( P <0.05).
Conclusions
The choice of dining locations for children is becoming more diverse, and a relatively high proportion of children eat meals outside the home and at school. Eating out have a higher risk of malnutrition for children. School feeding may be beneficial to children s physical health.

Background:
and Purpose Symptomatic intracranial hemorrhage (sICH) following endovascular thrombectomy (EVT) is a severe complication associated with adverse functional outcomes and increased mortality rates. Currently, a reliable predictive model for sICH risk after EVT is lacking.
Methods:
This study used data from patients aged ≥20 years who underwent EVT for anterior circulation stroke from the nationwide Taiwan Registry of Endovascular Thrombectomy for Acute Ischemic Stroke (TREAT-AIS). A predictive model including factors associated with an increased risk of sICH after EVT was developed to differentiate between patients with and without sICH. This model was compared existing predictive models using nationwide registry data to evaluate its relative performance.
Results:
Of the 2,507 identified patients, 158 developed sICH after EVT. Factors such as diastolic blood pressure, Alberta Stroke Program Early CT Score, platelet count, glucose level, collateral score, and successful reperfusion were associated with the risk of sICH after EVT. The TREAT-AIS score demonstrated acceptable predictive accuracy (area under the curve [AUC]=0.694), with higher scores being associated with an increased risk of sICH (odds ratio=2.01 per score increase, 95% confidence interval=1.64–2.45, P<0.001). The discriminatory capacity of the score was similar in patients with symptom onset beyond 6 hours (AUC=0.705). Compared to existing models, the TREAT-AIS score consistently exhibited superior predictive accuracy, although this difference was marginal.
Conclusions
The TREAT-AIS score outperformed existing models, and demonstrated an acceptable discriminatory capacity for distinguishing patients according to sICH risk levels. However, the differences between models were only marginal. Further research incorporating periprocedural and postprocedural factors is required to improve the predictive accuracy.

BACKGROUND:How to repair bone defect has been a clinical problem for a long time.The effective ingredients of traditional Chinese medicine have good biological activity and therapeutic effect,and the combination of effective ingredients of traditional Chinese medicine and tissue engineering materials has a broad prospect in the field of bone repair.The combination of different effective ingredients of traditional Chinese medicine and scaffolds has similarities in their functional relationships. OBJECTIVE:To collect the cases of the combinations of effective ingredients of traditional Chinese medicine and scaffolds,then analogize tissue engineering scaffolds and effective ingredients of traditional Chinese medicine into two types of traditional Chinese medicine that generate compatibility relationships based on the inspiration of the compatibility of seven emotions and summarize the relationship between the two based on their functional relationships. METHODS:Relevant articles from January 1998 to January 2024 were searched in PubMed and China National Knowledge Infrastructure(CNKI),using English search terms"traditional Chinese medicine,Chinese medicine,traditional Chinese medicine monomers,bone defect,bone repair,bone tissue engineering,tissue engineering,scaffold"and Chinese search terms"traditional Chinese medicine,effective ingredients of traditional Chinese medicine,traditional Chinese medicine monomers,bone tissue engineering,bone tissue engineering scaffold,scaffold,tissue engineering,bone defect,bone repair."A total of 88 articles were included for review and analysis. RESULTS AND CONCLUSION:(1)Both tissue engineering scaffold materials and active ingredients of traditional Chinese medicine have been widely used in the field of bone repair.Although they have obvious advantages in osteogenesis,there are still many shortcomings.Many studies are dedicated to preparing composite materials from the two,hoping to exert a detoxification and synergism through the interaction between the two.(2)Some drugs and materials can promote each other in osteogenesis,antibacterial,and promoting angiogenesis,enhancing their original effects.Inspired by the traditional concept of prescription compatibility,this article summarized it as a"Mutual promotion"relationship and provided examples to support it.(3)Some drugs can enhance the strength of materials,while some materials can achieve sustained release and controlled release effects,increase drug loading and stability,or achieve targeted delivery of drugs loaded on them.The article summarized this unilateral enhancement effect as a"Mutual assistance"relationship.(4)The combination of some traditional Chinese medicine and materials can reduce the toxic side effects of the other party.The article summarizes this detoxification relationship as"Mutual restraint and detoxification."(5)The article provided a new perspective on traditional Chinese medicine composite scaffolds,inspired by the seven emotions compatibility relationship and based on the classification of action relationships.It introduced traditional Chinese medicine concepts into the field of tissue engineering,providing new research ideas for subsequent researchers of composite scaffolds,and providing certain convenience in material selection and matching.
9.6-Gingerol Induced Apoptosis and Cell Cycle Arrest in Glioma Cells via MnSOD and ERK Phosphorylation Modulation

6-gingerol, a bioactive compound from ginger, has demonstrated promising anticancer properties across various cancer models by inducing apoptosis and inhibiting cell proliferation and invasion. In this study, we explore its mechanisms against glioblastoma multiforme (GBM), a notably aggressive and treatment-resistant brain tumor. We found that 6-gingerol crosses the blood-brain barrier more effectively than curcumin, enhancing its potential as a therapeutic agent for brain tumors. Our experiments show that 6-gingerol reduces cell proliferation and triggers apoptosis in GBM cell lines by disrupting cellular energy homeostasis. This process involves an increase in mitochondrial reactive oxygen species (mtROS) and a decrease in mitochondrial membrane potential, primarily due to the downregulation of manganese superoxide dismutase (MnSOD). Additionally, 6-gingerol reduces ERK phosphorylation by inhibiting EGFR and RAF, leading to G1 phase cell cycle arrest. These findings indicate that 6-gingerol promotes cell death in GBM cells by modulating MnSOD and ROS levels and arresting the cell cycle through the ERFR-RAF-1/MEK/ ERK signaling pathway, highlighting its potential as a therapeutic agent for GBM and setting the stage for future clinical research.
