1.Comparison of Wild and Cultivated Alpiniae Oxyphyllae Fructus Based on Traditional Quality Evaluation

ObjectiveTo compare the differences between wild Alpiniae Oxyphyllae Fructus(WAOF) and cultivated Alpiniae Oxyphyllae Fructus(CAOF) through a traditional quality evaluation system for medicinal materials. MethodsA total of 10 batches of WAOF and 12 batches of CAOF samples were collected from various regions of Hainan province. Relevant analytical methods from the 2020 edition of the Pharmacopoeia of the People's Republic of China were employed to observe the characteristics of WAOF and CAOF, followed by microscopic identification, thin-layer chromatography(TLC) identification, moisture content(toluene method), total ash, acid-insoluble ash, water-soluble and alcohol-soluble extracts(hot dipping method), water-soluble protein, total polysaccharides and total flavonoids(ultraviolet spectrophotometry), and volatile oil content(method A under general rule 2204). The contents of five active components(protocatechuic acid, chrysin, kaempferol, tectochrysin and nootkatone) were quantified using ultra-performance liquid chromatography(UPLC), and the antioxidant activity was evaluated. Building upon traditional quality evaluation of AOF, quantitative measurements were conducted on its appearance traits including diameter, length, plumpness(diameter/length ratio), and color. Canonical correlation analysis was performed using SPSS 26.0 to explore relationships between appearance traits and intrinsic quality. ResultsNo significant differences were observed between WAOF and CAOF in microscopic observation, TLC identification, moisture content, protocatechuic acid content, kaempferol content, odor, or antioxidant activity measured by 2,2ʹ-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS) method. WAOF exhibited significantly higher levels in water-soluble extracts, alcohol-soluble extracts, total polysaccharide content, water-soluble protein content, 100-grain weight, length, and total color difference(ΔE*ab) compared to CAOF(P<0.01). In contrast, CAOF showed significantly higher levels of total ash, acid-insoluble ash, content of total flavonoids, volatile oil content, chrysin content, tectochrysin content, nootkatone content, diameter, plumpness, lightness(L*), red-green chromaticity(a*), yellow-blue chromaticity(b*), and antioxidant activity measured by 1,1-diphenyl-2-picrylhydrazyl(DPPH) method compared to WAOF(P<0.01). Correlation analysis between 7 phenotypic traits and 8 quality traits revealed that among the phenotypic traits, plumpness, L*, a*, and b* exerted significant influence on intrinsic quality. Among the quality traits, total flavonoids, volatile oils, nootkatone, chrysin, and tectochrysin contributed substantially to intrinsic quality. ConclusionPlumpness, L*, a*, and b* of AOF significantly influence its intrinsic quality, and higher values of these parameters indicate relatively superior intrinsic quality. The comprehensive quality evaluation reveals that CAOF samples collected in this study are superior to their wild counterparts.
2.Association between core symptoms and biological markers in children with autism spectrum disorders

Objective:
To explore the relationship between serum homocysteine (Hcy), interleukin-5 (IL-5), folate and core symptoms in children with autism spectrum disorders, so as to provide potential biomarkers for early diagnosis and intervention of diseases.
Methods:
A total of 106 children with autism spectrum disorders and 106 healthy children with matched sex and age in Lu an People s Hospital were enrolled as autism group and healthy group between May 2020 to December 2023. On the day of admission, levels of serum Hcy, IL-5 and folate were detected. The core symptoms in autism group were evaluated by Autism Behavior Checklist (ABC), Wechsler Preschool and Primary Scale of Intelligence-fourth edition(WPPSI-IV), Childhood Autism Rating Scale (CARS) and Social Responsiveness Scale (SRS). The levels of serum Hcy, IL-5 and folate in the two groups were compared by t- test. The relationship between serum Hcy, IL-5, folate and core symptoms in children with autism spectrum disorders was determined by Pearson correlation analysis.
Results:
The levels of serum Hcy and IL-5 in autism group were (7.48±0.32) μmol/L and (345.77±32.51) pg/mL, higher than those in healthy group [(6.11±0.54) μmol/L, (274.04±25.17) pg/mL], while folate level was lower than that in healthy group [(15.24±3.47) ng/mL, (22.51±4.69) ng/mL], the differences were statistically significant ( t =22.47, 17.96, 12.83, all P < 0.05 ). In autism group, levels of serum Hcy and IL-5 were positively correlated with scores of ABC, CARS and SRS ( r =0.29, 0.53 , 0.54; 0.45, 0.41, 0.50), while negatively correlated with WPPSI-IV score ( r =-0.28, -0.26)(all P <0.05). The folate level was negatively correlated with scores of ABC, CARS and SRS ( r =-0.55, -0.40, -0.25), while positively correlated with WPPSI-IV score ( r =0.41) (all P <0.05).
Conclusion
Children with ASD show elevated serum Hcy and IL-5 alongside decreased folate,and three markers correlate with core symptoms and intellectual level.
3.Association between depressive symptoms and risk of stroke and all-cause mortality in young and middle-aged adults

BackgroundCurrent studies on the relationship between depressive symptoms and the risks of stroke and all-cause mortality mostly focus on the elderly population or high-risk groups with chronic diseases. The long-term impact of depressive symptoms in young and middle-aged people on the risks of stroke and all-cause mortality still lacks systematic assessment, and the relevant evidence is insufficient. ObjectiveTo explore the association between depressive symptoms and the risks of stroke and all-cause mortality in young and middle-aged adults, thereby expanding the risk evidence in this age group and providing epidemiological basis for early identification of high-risk individuals and the development of targeted intervention strategies. MethodsBased on data from the National Health and Nutrition Examination Survey (NHANES) database (2009–2018), a total of 14 947 young and middle-aged respondents aged 18 to 64 were included as the research subjects. Logistic regression and restricted cubic spline (RCS) models were employed to examine the association between depressive symptoms and stroke. Cox proportional hazards model, RCS model, and Kaplan-Meier (K-M) survival curves were used to clarify the relationship between depressive symptoms and all-cause mortality risk. To verify the robustness of main findings, sensitivity analysis was conducted stratified by gender, hypertension, diabetes, hyperlipidemia, body mass index (BMI), and age. ResultsLogistic regression analysis revealed that compared with the non-depressed group, the risk of stroke in the mild depression group increased by 53.50% (OR=1.535, 95% CI: 1.147–2.053), while the moderate-to-severe depression group showed the highest risk with a 132.90% increase (OR=2.329, 95% CI: 1.733–3.129). RCS analysis indicated an approximately linear dose-response relationship between PHQ-9 score and the risk of stroke (Pfor non-linearity=0.089), with stroke risk increasing as PHQ-9 scores rose. As the PHQ-9 score increased, the risk of stroke also showed an upward trend. Cox regression analysis demonstrated that compared with the non-depressed group, the risk of all-cause mortality in the mild depression group increased by 48.80% (HR=1.488, 95% CI: 1.164–1.902), and the moderate-to-severe depression group had a 70.60% increased risk (HR=1.706, 95% CI: 1.310–2.221). RCS analysis showed a linear dose-response relationship between PHQ-9 scores and all-cause mortality risk (Pfor non-linearity=0.440). K-M curves indicated that as the severity of depressive symptoms increases, the risk of all-cause mortality gradually rises (P<0.05). Sensitivity analysis confirmed that the associations between depression symptoms and risks of stroke and all-cause mortality were not modified by gender, hypertension, diabetes, hyperlipidemia, BMI, or age, demonstrating robust findings. ConclusionAmong the young and middle-aged population, both mild and moderate-to-severe depression were associated with increased risks of stroke and all-cause mortality, with the moderate-to-severe depression group showed more pronounced risk elevations. A positive dose-response relationship was observed between depressive symptoms and these risks.
4.The efficacy and safety of zanubrutinib combined with BR regimen in B-cell chronic lymphoproliferative disorder

Objective To explore the efficacy and safety of a zanubrutinib-based combination regimen as first-line treatment for patients with B-cell chronic lymphoproliferative disorder (B-CLPD). Methods A retrospective analysis was performed on 26 patients with B-CLPD, including indolent B-cell non-Hodgkin lymphoma (B-iNHL), mantle cell lymphoma (MCL), and other unclassifiable B-CLPD, who were newly diagnosed and treated at the Department of Hematology, Zhongshan Hospital (Xiamen), Fudan University between January 2021 and January 2025. Patients were divided into two groups according to the treatment regimen: BR (bendamustine plus rituximab) and ZBR (zanubrutinib combined with BR regimen). Baseline characteristics, treatment responses, and the incidence of treatment-related adverse events (TRAE) were compared between the two groups. Results No statistically significant differences in baseline characteristics were observed between the two groups. All patients completed at least four cycles of treatment. There were no significant differences between the BR and ZBR groups in partial response rate (46.2% vs 53.8%), complete response rate (53.8% vs 15.4%), overall response rate (100.0% vs 84.6%), or TRAE incidence. After the first cycle, the lymphocyte count was higher in the ZBR group than in the BR group (P=0.002); no other statistically significant differences in other hematologic parameters were observed at any treatment cycle. Conclusions In patients with B-iNHL, MCL, and other unclassifiable B-CLPD, the ZBR regimen demonstrates similar safety and response rates to the BR regimen. These findings support further investigation into the clinical value of ZBR as first-line treatment regimen in a limited course for indolent lymphoid malignancies.
5.Contamination levels of perfluorinated compounds in water bodies of key river basins in the Yangtze River Delta Region in 2023

ObjectiveThis study aimed to provide a scientific basis for the safety management of drinking water by assessing the contamination levels of 21 per- and polyfluoroalkyl substances (PFASs) in water samples collected from the Yangtze River Delta region. MethodsSolid-phase extraction coupled with ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) was employed to determine 21 PFASs in 6 source water samples, 20 treated water samples, and 60 tap water samples collected from 5 administrative districts in the Yangtze River Delta region across four seasons in 2023. The Kruskal-Wallis H test was used to compare differences in PFAS concentrations among the three types of water samples, followed by post hoc pairwise comparisons with Bonferroni correction. Removal efficiencies of target compounds were also calculated. ResultsAmong the 86 samples, 14 PFASs were detected, with 7 compounds detected in 100% of samples. The total concentrations of PFASs in source water, treated water, and tap water ranged from 63.85 to 190.54 ng·L-¹, 55.75 to 774.36 ng·L-¹, and 40.72 to 689.80 ng·L-¹, respectively. The compounds with the highest median concentrations were perfluorobutanoic acid (PFBA) (40.19, 15.20, and 14.05 ng·L-¹), perfluorohexanoic acid (PFHxA) (29.69, 32.62, and 34.24 ng·L-¹), perfluorooctanoic acid (PFOA) (17.65, 39.45, and 36.83 ng·L-¹), and potassium perfluorobutanesulfonic acid (PFBS) (16.99, 24.38, and 23.89 ng·L-¹). 6.Xixintang-medicated Serum Regulates Aβ25-35-induced Polarization of BV-2 Microglial Cells

ObjectiveTo investigate the effects of Xixintang (XXT)-medicated serum on the amyloid β-protein (Aβ)25-35-induced polarization of BV-2 microglial cells by a cell experiment and uncover the potential mechanisms of this formula in the prevention and treatment of Alzheimer's disease (AD), thus providing scientific evidence for the clinical application of XXT. MethodsBV-2 microglial cells were subcultured. The optimal concentrations of XXT-medicated serum and Aβ25-35 were determined via the cell-counting kit-8 (CCK-8) assay. The cell experiment was carried out with the following groups: blank control, model (Aβ25-35 at 40 μmol·L-1), XXT-medicated serum (Aβ25-35 at 40 μmol·L-1 + 10% XXT-medicated serum), and blank serum (Aβ25-35 at 40 μmol·L-1 + 10% blank serum). After 24 hours of cell incubation, immunofluorescence was used to detect the expression of ionized calcium-binding adaptor molecule 1 (IBA1), CD16/32, and CD206. Real-time PCR was performed to measure the mRNA levels of CD206, CD163, inducible nitric oxide synthase (iNOS), and arginase 1 (Arg-1). Enzyme-linked immunosorbent assay (ELISA) was employed to quantify the levels of nerve growth factor (NGF), iNOS, and Arg-1. The nitric oxide (NO) concentration was determined via the nitrate reductase method. ResultsCompared with the blank control group, the model group showed increased expression of IBA1 and CD16/32 (P<0.01), decreased expression of CD206 (P<0.05), upregulation in the mRNA level (P<0.01) and content (P<0.05) of iNOS, downregulation in the mRNA levels of CD206, CD163, and Arg-1 (P<0.05, P<0.01), lowered levels of Arg-1 and NGF (P<0.05), and an elevation in the NO level (P<0.05). Compared with the model group, the XXT-medicated serum group exhibited reduced expression of IBA1 and CD16/32 (P<0.05, P<0.01) and increased expression of CD206 (P<0.01). Both the content and mRNA level of iNOS were downregulated (P<0.05, P<0.01), while the mRNA levels of CD206, CD163, and Arg-1 were upregulated (P<0.01) in the XXT-medicated serum group. In addition, the XXT-medicated serum group showed elevated levels of Arg-1 and NGF (P<0.05) and a lowered level of NO (P<0.05). The blank serum group showed no statistically significant differences in the measured parameters compared with the model group. ConclusionThe XXT-medicated serum can inhibit the polarization toward the M1 phenotype and promote the polarization toward the M2 phenotype, exerting anti-inflammatory and neurotrophic effects.

Radiation-induced heart disease(RIHD)is a severe complication in patients with thoracic cancer undergoing radiotherapy,with important impacts on long-term survival among cancer survivors.There is an urgent need to investigate the pathogenesis of RIHD and to develop effective therapeutic agents,and the establishment of high-quality RIHD animal models is pivotal to addressing these issues.This review summarizes the critical factors to consider in establishing RIHD animal models,including species selection,radiation type,dosage,fractionation,and target fields,and modeling timeline,along with the evaluation method and success criteria.We also consider the potential pathogenic mechanisms underlying RIHD,including DNA damage,oxidative stress,inflammatory responses,mitochondrial dysfunction,renin-angiotensin-aldosterone system activation,and myocardial fibrosis,as well as their interrelationships.These insights provide a comprehensive reference framework for constructing RIHD animal models and advancing mechanistic investigations into this condition.

In 2025,the International Association of Pancreatology(IAP),in collaboration with the American Pancreatic Association,European Pancreatic Club,Indian Pancreas Club,and Japan Pancreas Society,released the International Association of Pancreatology revised guidelines on acute pancreatitis 2025.This edition represents a comprehensive revision of the 2013 guidelines,based on high-quality evidence accumulated over the past decade,particularly randomized controlled trials.The guidelines encompass 18 key areas-including pain management,fluid therapy,nutritional support,management of infected necrosis,complication control,discharge and follow-up,and recurrence prevention-offering a total of 96 recommendations that emphasize individualized treatment.These updates provide important guidance for standardizing clinical practice and improving outcomes in acute pancreatitis,while also indicating future research directions such as the development of targeted therapies.However,some recommendations remain limited by lower evidence quality,uncertain applicability in specific clinical settings,and insufficient consideration of economic burden and cost-effectiveness.
9.Trends in disease burden of aortic aneurysm attributable to high body mass index in China based on GBD data

Background and Aims:Aortic aneurysm,a major cardiovascular disease with high mortality and disability rates,has emerged as a critical global public health challenge.Elevated body mass index(BMI)has been confirmed as an independent risk factor for aortic aneurysm.However,the long-term trends and heterogeneity of the disease burden attributable to high BMI in China-across sex,age,and region-remain insufficiently studied.This study,based on the Global Burden of Disease(GBD)database,aimed to analyze the changes in the disease burden of high BMI-related aortic aneurysm in China from 1990 to 2021 and compare these trends with those in global and socio-demographic index(SDI)-stratified regions.Methods:Data from GBD 2021 were used to extract mortality rates and disability-adjusted life years(DALY)due to aortic aneurysm attributable to high BMI in China from 1990 to 2021.Age-standardized mortality rate(ASMR)and age-standardized DALY rate(ASDR)were calculated.Long-term trends were assessed using the estimated annual percentage change(EAPC)and Joinpoint regression models.An autoregressive integrated moving average model was applied to project trends from 2022 to 2036.Results:During the study period,deaths and DALY from aortic aneurysms attributable to high BMI in China increased nearly sevenfold.ASMR rose from 1 to 3 per 100 000 population(EAPC=3.91),and ASDR increased from 23 to 74 per 100 000(EAPC=4.11),both showing a marked upward trend.Males consistently bore a higher burden across all age groups,particularly among those aged≥65 years.Decomposition analysis revealed that the increased burden in males was mainly driven by epidemiological improvements,while that in females was primarily attributable to population aging.Compared with the United States and global trends,China exhibited a faster increase in high BMI-related aortic aneurysm burden,which is projected to remain elevated over the next 15 years.Conclusion:The disease burden of aortic aneurysm attributable to high BMI continues to rise in China,with pronounced differences across sex and age.Targeted weight management,health interventions,and early screening strategies are urgently needed for high-risk populations to curb the upward trend and optimize public health resource allocation.
10.Optimization of external contract management for laboratory animal technology services in medical research institutes

The demand for laboratory animals has recently been increasing steadily,leading to a rise in the number of external contracts for laboratory animal technology services issued by medical research institutes.Improving the management of these contracts has thus become an important focus.This review highlights five major issues in the current management of external laboratory animal technology service contracts in research institutes:(1)insufficient awareness of the legal risks associated with contracts;(2)a lack of standardized contract templates,with ambiguous or missing terms;(3)a disconnect between the departments responsible for contract execution and management;(4)insufficient professional qualifications among contract management staff;and(5)the unique nature of managing contracts for animal experimentation.This review offers several possible recommendations to address these challenges:(1)strengthen the establishment of legal teams within research institutes and raise awareness of legal risks in contracts;(2)improve the contract management system and increase staff motivation for more effective management;(3)create an electronic information management platform for external laboratory animal technology service contracts;and(4)provide enhanced professional training for staff involved in managing these contracts.Hope medical research institutes aim to implement these suggestions in their practices to optimize the management of laboratory animal technology service contracts in the future.This will allow them to better support social scientific research and contribute to the advancement of life sciences research and technological innovations.
