2.Effects of ROCK-siRNA transfection on Ang II-induced endothelial cell senescence and endothelial microparticles.

Objective To investigate the effects of ROCK-siRNA transfection on endothelial cell senescence and endothelial microparticles (EMPs) induced by angiotensin II (Ang II). Methods Human umbilical vein endothelial cells (HUVECs) were treated with Ang II (1.0 μmo/L) to induce cellular senescence models, followed by transfection with ROCK-siRNA. The cells were divided into four groups: control group, model group, negative transfection control group (Ang II combined with NC-siRNA), and ROCK-siRNA transfection group (Ang II combined with ROCK-siRNA). Cellular senescence was assessed by SA-β-Gal staining. EMP levels in cell supernatants and intracellular reactive oxygen species (ROS) levels were assessed using flow cytometry. The expression levels of silenced information regulator 1(SIRT1) and p53 protein in each group were analyzed by Western blotting. Results Following ROCK-siRNA transfection, the number of senescent cells induced by Ang II was significantly reduced, accompanied by decreased CD31+ EMP levels and suppressed intracellular ROS levels. Meanwhile, the expression levels of SIRT1 were up-regulated, while the expression levels of p53 were down-regulated. Conclusion Silencing ROCK expression suppresses EMP release, reduces ROS generation, regulates the expression of SIRT1 and p53, and ultimately attenuates Ang II-induced endothelial cell senescence.

Tumor deposits(TD)is one of the independent risk factors for poor prognosis of colorectal cancer.The diagnosis of TD primarily relied on postoperative pathology,and precise preoperative assessment was difficult to achieve.With the development of medical imaging,many imaging techniques and analyzing methods had been applied to explore TD in colorectal cancer.The progresses in imaging research of TD in colorectal cancer were reviewed in this article.
4.METTL3 Regulates the Proliferation,Migration,and Invasion of Uveal Melanoma Cell via Modifying m6A Methylation

[Objectives]To investigate the effect of METTL3 on the malignant biological behaviors of uveal melanoma cells and to verify whether this effect is related to m6A methylation.[Methods]Uveal melanoma cell models with METTL3 knockdown,overexpression,and point mutations at m6A-related catalytic sites were constructed via lentivirus transfection.Transwell assays were used to assess cell migration and invasion;CCK8 assays were used to measure cell proliferation and flow cytometry was used to analyze apoptosis and cycle changes.[Results]The proliferation,migration and invasion abilities of C918 and MUM-2B cells with METTL3 knockdown were significantly decreased(P<0.001),while the apoptosis rate was increased.The proportion of cells in G1 phase significantly increased,whereas the proportion in the S phase significantly decreased.Cells overexpressing METTL3 showed significantly enhanced proliferation,migration and invasion abilities(P<0.001),along with a decreased apoptosis rate.In C918 cells,the proportion of cells in G1 phase decreased significantly,while the proportion in S phase increased significantly.The cell cycle distribution of MUM-2B cells did not change remarkably.Following point mutation of m6A-related catalytic sites,cell proliferation,migration and invasion decreased,and the apoptosis rate increased.In MUM-2B cells,the percentage of cells in G1 phase significantly increased;the percentage in S phase significantly decreased and the percentage in G2 phase slightly decreased.In C918 cells,the percentage of G1 phase cells significantly increased,with no significant changes in the proportions of S and G2 phases.[Conclusions]The proliferation,invasion and metastasis of uveal melanoma cells were positively correlated with the expression of METTL3,while the apoptosis rate was negatively correlated.Changes in METTL3 levels differentially affect the cell cycles in different cell lines.The effects of METTL3 on the proliferation,migration and invasion of uveal melanoma cells are related to m6A methylation modification.

7.Research progress on combined treatment of physical exercise and anti-hypertensive drugs for hypertension in elderly people

Hypertension has a high prevalence in China, and the predominant reliance on drug treatment consumes enormous medical resources.Physical exercise is the only widely-accepted nonpharmacologic therapy for hypertension and is potentially cost-effective.How to incorporate exercise into a prescription for rational treatment of hypertension is a topic that needs to be addressed.This review examines the research progress on the treatment of hypertension combining physical exercise with anti-hypertensive drugs in elderly people, aiming to provide some information and options to promote personalized treatment strategies for hypertension in elderly people.
8.Development of an experience scale for patients with chronic wound debridement and dressing

Objective:To develop and evaluate a medical experience assessment scale for debridement and dressing change in chronic wound patients in China, and to provide a reference for improving hospital service quality.Methods:Based on the framework of hospital consumer assessment of healthcare providers and systems survey (HCAHPS) in the United States, a preliminary draft of the scale was formed through a literature review and qualitative interviews with 12 chronic wound patients (7 males and 5 females aged 58.1±12.3 years). Five experts were invited for content validity testing, and 191 chronic wound patients (111 males and 80 females aged 53.5±19.1 years) were selected to evaluate the internal consistency reliability, half reliability, retest reliability, and structural validity of the scale.Results:The Chronic Wound Patient Debridement and Dressing Experience Scale covered 5 dimensions with 30 sub-items and 2 comprehensive evaluation items, including demand response, good and friendly communication, professional trust, optimization of the medical treatment process, and encouragement of patient participation. The scale Cronbach′s α coefficient was 0.967 and ranged from 0.890 to 0.962 for each dimension. The overall retest reliability of the scale was 0.940 and ranged from 0.895 to 0.940 for each dimension. The overall half reliability of the scale was 0.923 and ranged from 0.834 to 0.935 for each dimension. 5 factors were extracted, with a cumulative variance contribution rate of 82.061%.Conclusions:Based on the HCAHPS framework in the United States, the Chronic Wound Debridement and Dressing Experience Scale developed has high reliability and validity, and can be used to evaluate the dressing change experience of patients with chronic wounds. It has clinical practice significance for dressing change in chronic wounds.
9.Role of Ferroptosis in Non-small Cell Lung Cancer and Progress of Traditional Chinese Medicine Intervention

Non-small cell lung cancer(NSCLC)is one of the malignant tumors with high morbidity and mortality worldwide.Ferroptosis is a new type of programmed cell death caused by abnormal accumulation of iron-dependent reactive oxygen species(ROS)leading to lipid peroxidation.It involves the balance between iron metabolism,lipid metabolism,oxy-gen free radical reaction and lipid peroxidation.Recent studies have found that ferroptosis is closely related to the occurrence and development of NSCLC.Due to the emergence of chemotherapy resistance and radiotherapy resistance in the treatment of NSCLC,there is an urgent need to develop new effective drugs and treatment strategies.Traditional Chinese medicine has unique advantages in the prevention and treatment of NSCLC due to its multi-targets and minimal side effects.In this review,we summarize the mechanism of ferroptosis in NSCLC,and discuss the research status of active ingredients of traditional Chinese medicine,single-herb traditional Chinese medicine and Chinese herbal compounds in the intervention of NSCLC through ferroptosis,in order to provide a new theoretical basis for the research of ferroptosis pathway and the prevention and treatment of NSCLC by targeted ferroptosis of traditional Chinese medicine.
10.Application of Radiomics in Classification and Prediction of Benign and Malignant Lung Tumors.

Aiming at the lack of quantitative evaluation methods in clinical diagnosis of lung cancer, a classification and prediction model of lung cancer based on Support Vector Machine (SVM) was constructed by using radiomics method. Firstly, the definition and processing flow of radiomics were introduced. The experimental samples were selected from 816 lung cancer patients on LIDC. Firstly, ROI was extracted by central pooling convolution neural network segmentation method. Then, Pyradiomics and FSelector feature selection models were used to extract features and reduce dimension. Finally, SVM was used to construct the classification and prediction model of lung tumors. The predictive accuracy of the model is 80.4% for the classification of benign and malignant pulmonary nodules larger than 5 mm, and the value of the area under the curve (AUC) is 0.792. This indicates that the SVM classifier model can accurately distinguish benign and malignant pulmonary nodules larger than 5 mm.
