1.Effect of miR-196b-5p in adipose-derived stem cell exosomes on burn wound healing in rats

BACKGROUND:Currently,miR-196b-5p has been found to play a role in cell proliferation,migration and inhibition of scar hyperplasia,but there is a lack of relevant studies on whether it plays a role in the process of wound healing.OBJECTIVE:To investigate the effect of miR-196b-5p in adipose stem cell-derived exosomes on burn wound healing.METHODS:The models of deep second degree skin burn in SD rats were established and randomly divided into four groups:blank control group,exosome group,agomiR-196b-5p group,and exosome+antagomiR-196b-5p group,with 10 rats in each group.PBS,adipose-derived stem cell-derived exosomes,miR-196b-5p agonist,and miR-196b-5p inhibitor were injected around the wound according to different groups.Wound healing was observed immediately after injury and on days 7,14,and 21 after injury.On day 7 after surgery,hematoxylin-eosin staining was used to observe the expression of inflammation in the wound surface.On day 14 after suregery,Masson staining was used to observe the expression of collagen and immunohistochemical staining was used to observe the expression of CD31 in the wound surface.On day 7 after surgery,western blot assay was performed to detect the expression of α-smooth muscle actin and type Ⅰ collagen in the wound surface.RESULTS AND CONCLUSION:(1)The wound healing was faster in the agomiR-196b-5p group,while it was slower in the blank control group and the exosome+antagomiR-196b-5p group.(2)Compared with the blank control group and the exosome+antagomiR-196b-5p group,the wound infiltration of inflammatory cells was less in the exosome group and the agomiR-196b-5p group,and the expression of CD31 was significantly increased(P<0.01).(3)Compared with the blank control group and the exosome+antagomiR-196b-5p group,the expression levels of α-smooth muscle actin and type Ⅰ collagen were increased in the exosome group and the agomiR-196b-5p group(P<0.05).These findings indicate that miR-196b-5p in adipose stem cell-derived exosomes can promote burn wound healing in rats.

3.Construction and identification of synovial tissue conditional Grk2 knockout mice

Aim To construct and analyze the genotype of G protein-coupled receptor kinase 2(GRK2)conditional knockout mice in synoviocytes,and to provide an animal model for stud-ying the function of GRK2 in synoviocytes.Methods Grk2flox/+mice were bred to generate Grk2flox/flox mice,Grk2flox/flox mice were bred to Col1a1-iCre+mice,Grk2flox/+Col1a1-iCre+mice were bred to Grk2flox/flox mice.Grk2flox/flox Col1a1-iCre+mice were ob-tained as target mice.DNA was extracted and amplified by PCR to identify the genotype.Western blot was used to verify the effect of Grk2 knockout in synovium,liver and kidney tissues.HE staining was used to detect the effects of Grk2 conditional knockout in synovial cells on ankle synovium,liver and kidney tissues.Multiple immunofluorescence was used to detect GRK2 expression in synovial cells.Results The results of gene iden-tification showed that Grk2flox/flox Col1a1-iCre+mice had both Flox and Col1a1-iCre genotypes.Western blot results showed that GRK2 expression decreased in synovial tissues of Grk2flox/flox Col1a1-iCre+mice,but there was no significant change in the expression of GRK2 in liver and kidney tissues.HE staining showed that Grk2flox/flox Col1a1-iCre+mice had no significant pathological changes in the ankle synovium,liver and kidney.The results of multiple immunofluorescence showed that GRK2 expression in synovial cells of Grk2flox/flox Col1a1-iCre+mice de-creased.Conclusion Grk2 conditional knockout mice in syno-viocytes are successfully constructed and identified,which pro-vides an animal model for further study of the role of GRK2 in synovial-related diseases.
4.Effect of adipose-derived stem cell exosome on fibroblasts and wound healing

Objective To investigate the effect of adipose-derived stem cell exosome(ADSC-Exo)on the proliferation and migration of fibroblasts as well as on wound healing of deep second degree burns in rats.Methods Fibroblasts were cultured with ADSC-Exo or in phosphate-buffered saline(PBS).The effect of ADSC-Exo on the proliferation and migration of fibroblasts was observed using the CCK-8 assay and scratch test.Forty Sprague-Dawley(SD)rats were selected to construct a deep second-degree burn model and divided into the exosome and control groups.The exosome group was injected with the ADSC-Exo solution around the wound,and the control group was injected with an equal amount of PBS solution.Wound healing was observed in both groups.Inflammatory infiltration was observed using hematoxylin and eosin(HE)staining,and wound angiogenesis was observed using CD31 immunohistochemical staining.Results The CCK-8 assay revealed that ADSC-Exo promoted fibroblast proliferation in a concentration-dependent manner.The results of the scratch test showed that ADSC-Exo significantly enhanced the migratory ability of fibroblasts(P<0.000 1).The wound healing results showed that ADSC-Exo injection promoted burn wound healing in rats(P<0.000 1).Pathological analysis indicated that inflammatory cell infil-tration was less and CD31 expression was higher in the ADSC-Exo group than in the control group(P<0.01).Conclusion ADSC-Exo promoted the proliferation and migration of fibroblasts,reduced wound inflammation,promoted neovascularization,and enhanced wound healing.
5.Development and reliability and validity testing of the Symptom Assessment Scale for Patients Undergoing Bladder Irrigation Chemotherapy

Objective:To develop the Symptom Assessment Scale for Patients Undergoing Bladder Irrigation Chemotherapy and evaluate its reliability and validity.Methods:Based on the theory of unpleasant symptoms, a preliminary version of the Symptom Assessment Scale for Patients Undergoing Bladder Irrigation Chemotherapy was developed through literature analysis, semi-structured interviews, and the Delphi method. After revising certain items in the pre-survey, convenience sampling was used to select patients who underwent bladder instillation chemotherapy in the Department of Urology Surgery of three ClassⅢ Grade A hospitals in Yunnan Province from January to July 2024 as research subjects to test the reliability and validity of the scale.Results:A total of 168 questionnaires were distributed, and 162 valid questionnaires were collected, with a valid response rate of 96.429% (162/168). The Symptom Assessment Scale for Patients Undergoing Bladder Irrigation Chemotherapy covered two areas of symptom severity and symptom distress, comprising five dimensions and 27 items. The Cronbach's α coefficient for the total scale was 0.953, and the split-half reliability coefficient was 0.806. Exploratory factor analysis revealed that the four common factors for symptom severity contributed to 73.196% of the cumulative variance, while the single common factor for symptom distress accounted for 68.285% of the cumulative variance. Confirmatory factor analysis revealed that all indicators met the fit criteria, indicating that the model possessed good goodness-of-fit. The content validity index at the scale level was 0.940, while the content validity index at the item level ranged from 0.833 to 1.000.Conclusions:The Symptom Assessment Scale for Patients Undergoing Bladder Irrigation Chemotherapy developed in this study demonstrates good reliability and validity, and is suitable for evaluating symptoms in patients undergoing bladder infusion chemotherapy.
6.Study on the Relationship between the Changes of Four Indexes Related to Plasma Ferroptosis and the Prognosis after TACE in Patients with Hepatocellular Carcinoma

Objective To analyze the relationship between the expression of ferroptosis markers in the tumor microenvironment(TME)and the prognosis of transcatheter arterial chemoembolization(TACE)for hepatocellular carcinoma(HCC).Methods This prospective observational study included 100 HCC patients who received TACE treatment at Langfang Hospital of Traditional Chinese Medicine from March 2019 to June 2021 as the study subjects.The levels of 8-isoprostaglandin F2α(8-iso-PGF2α),4-hydroxy-2-nonenal(4-HNE),8-hydroxy-2'deoxyguanosine(8-OH-dG)and hepcidin in plasma were evaluated by ELISA kit at baseline(1 day before TACE),1 day after TACE and 4~8 weeks.The changes of ferroptosis related markers during TACE treatment were compared.The difference between the level of 8-iso-PGF2α,4-HNE and the baseline 1 day after TACE treatment was recorded as △8-iso-PGF2α,△4-HNE.Results Compared with the baseline,the levels of 8-iso-PGF2α and 4-HNE increased significantly and the level of hepcidin decreased significantly one day after TACE treatment,and the differences were statistically significant(t=8.03,16.29,2.92,all P<0.05).Compared with 1 day after treatment,the levels of 8-iso-PGF2α,4-HNE decreased and the level of 8-OH-dG increased at 4~8 weeks after TACE treatment,and the differences were statistically significant(t=9.12,17.17,2.63,all P<0.05).Multivariate COX analysis showed that △8-iso-PGF2α,△4-HNE and 8-iso-PGF2α 1 day after TACE treatment were independent factors affecting the overall survival after TACE(Wald χ2=5.205,13.801,6.054,all P<0.05).The survival time of patients with △4-HNE>2.01 μg/ml was significantly longer than that of patients with △4-HNE≤2.01 μg/ml(Log-rank=5.718,P=0.017),and that of patients with△8-iso-PGF2α>1.75ng/ml was sig-nificantly longer than that of patients with△8-iso-PGF2≤1.75ng/ml(Log-rank=4.163,P=0.041).Conclusion The prognosis of HCC patients who are in a state of high ferroptosis(4-HNE and 8-iso-PGF2 increased)at 1 day after TACE treatment is better,which indicate that ferroptosis mediated HCC death induced by TACE treatment.
7.Analysis on psychosocial factors for early puberty in children

Objective:To investigate the effects of psychosocial factors on early puberty and its sex specific differences.Methods:Bengbu Yuhui Longitudinal Study of Children Development Cohort, consisting of 1 162 students in grades 1-3 (524 girls) and their parents, was established in April 2021 in Bengbu, Anhui Province. The parents were surveyed by questionnaire to collect the information about their children's 56 items of psychosocial factor exposures, including 5 dimensions: major life events, early life adversity, interpersonal/relational conflict, family economic status and daily life troubles. Pubertal growth and physical examinations were conducted by trained professionals at baseline, the 1 st (May 2022), 2 nd (October 2022), 3 rd (May 2023), and 4 th (October 2023) follow-up surveys. In this study, the pubertal development status data were from the 4 th follow-up examination. Fianally, the valid data on pubertal development of 1 105 children were obtained, including the data from 510 girls (46.15%). Multinomial logistic regression model was used to analyze the association between psychosocial factors and early puberty, and the final predictors of early puberty were explored by using four models [least absolute shrinkage and selection operator (LASSO), multiple logistic regression, ridge regression, and Bayesian]. Results:The detection rates of early puberty in boys and girls were 13.60% and 16.30%, respectively. The median of intra-group absolute correlation of psychosocial factor exposure (0.042-0.066) was generally higher than that of inter-group among girls (0.031-0.059), while no similar diference was found among boys. LASSO model showed that moving house, being raised by someone other than parents, sexual abuse, peer conflict, high monthly household income, and high exposure to electronics on school days were predictors of early puberty in girls, while moving house and high monthly household income were the predictors of early puberty in girls in all four models (LASSO, multiple logistic regression, ridge regression, and Bayesian). Among boys, only a transient loss from family was a predictor of early puberty.Conclusion:Early puberty in children can be predicted by psychosocial factors, and there are gender-specific differences.
8.Development and validation of an innovative minimally invasive rotary-cutting surgical system for axillary osmidrosis

Objective To develop an innovative minimally invasive rotary-cutting surgical system for axillary osmidrosis,and conduct clinical validation.Methods The design concept,technical principles and system composition of the innovative minimally invasive rotary-cutting surgical system for axillary osmidrosis were introduced.A total of 73 patients(146 axillae)with axillary osmidrosis were enrolled as subjects,and underwent surgery using the newly developed surgical system.Clinical validation of the system was performed by evaluating postoperative scarring,odor elimination rate,postoperative complication incidence,and patient satisfaction.Results The study demonstrated favorable clinical outcomes in the following aspects:postoperative scarring,odor elimination rate,postoperative complication incidence,and patient satisfaction.Conclusion The minimally invasive rotary-cutting surgical system for axillary osmidrosis is rationally designed.The rotary-cutting puncture device is safe,effective,minimally invasive,and convenient for axillary osmidrosis surgery,warranting further clinical validation and widespread application.
9.Phages in human health and gut microbiota transplantation therapy

Phages, prokaryotic viruses widely present in the human, are a crucial component of the gut microbiome. They play a significant role in human health and the development of diseases. Emerging evidence indicates that phages can interact with bacteria to affect their abundance, metabolism, and antibiotic resistance, thereby influencing the balance of the gut microbiota. In addition, phages also contribute to the gut immune response, and can become dysregulated in a range of immune-related diseases. Gut phages also carry important roles in fecal microbiota transplantation (FMT) for disease treatment. Phages can target specific bacterial members and communities, thereby reduce the risk of bacterial infections or the presence of bacteria, and maintain the stability of the gut microbiome. However, gut phageome research is still in its infancy and additional basic and clinical researches are required to evaluate its species composition, mechanisms of pathogenicity or protection, as well as its efficacy and safety.
10.The mechanism of serpinA3N alleviates retinal neural injury in diabetic mice by inhibiting Müller cell inflammation

Objective To investigate the mechanism of serine protease inhibitor A3N(SerpinA3N)in alleviating reti-nal neural injury in diabetic mice by inhibiting Müller cell inflammation.Methods Thirty-six db/db mice(72 eyes)were randomly divided into the db/db group,the db/db+SerpinA3N-overexpressing adeno-associated virus(AAV-SerpinA3N)group,and the db/db+empty vector adeno-associated virus(NC-SerpinA3N)group,with 12 mice in each group.Twelve age-matched healthy male littermate mice were randomly selected as the healthy control group(db/m group).Mice in each group were sacrificed 4 weeks after the corresponding treatments.Immunofluorescence staining was used to detect the co-localization of SerpinA3N and glial fibrillary acidic protein(GFAP)in the mouse retina.Hematoxylin-eosin(HE)staining was used to observe histopathological changes in the retinal tissue.TUNEL staining was used to detect the apoptosis of reti-nal ganglion cells(RGCs).Immunohistochemistry was used to detect GFAP expression.ELISA was used to measure the levels of inflammatory factors[interleukin(IL)-1 β,IL-6,IL-18,and tumor necrosis factor-α(TNF-α)]in the retinal tis-sue.The predicted target genes of SerpinA3N were imported into the STRING database to construct a protein-protein inter-action(PPI)network.The highest-scoring target was selected based on the scores for molecular docking.Western blot was used to detect the expression levels of SerpinA3N,nuclear factor kappa B(NF-κB),and spleen focus-forming virus proviral integration oncogene(Spi1)proteins in the retinal tissue.Results Immunofluorescence staining showed co-lo-calized expression of SerpinA3N and GFAP in the retinal tissue.Compared with the db/m group,the db/db,db/db+NC-SerpinA3N,and db/db+AAV-SerpinA3N groups showed decreased retinal thickness and RGC count,and increased number of TUNEL-positive cells,relative GFAP-positive expression intensity,levels of all inflammatory factors,and expression lev-els of NF-κB and Spi1 proteins,while SerpinA3N protein expression was decreased(all P<0.05).Compared with the db/db group,the db/db+AAV-SerpinA3N group showed increased retinal thickness and RGC count,and decreased number of TUNEL-positive cells,relative GFAP-positive expression intensity,levels of all inflammatory factors,and expression levels of NF-κB and Spi1 proteins,while SerpinA3N protein expression was increased(all P<0.05).Compared with the db/db+AAV-SerpinA3N group,the db/db+NC-SerpinA3N group showed decreased retinal thickness and RGC count,and increased number of TUNEL-positive cells,relative GFAP-positive expression intensity,levels of all inflammatory factors,and ex-pression levels of NF-κB and Spi1 proteins,while SerpinA3N protein expression was decreased(all P<0.05).The PPI re-sults indicated an interaction between SerpinA3N and Spi1.Molecular docking results showed that Spi1 could form hydro-gen bonds with residues surrounding SerpinA3N.Conclusion Overexpression of SerpinA3N can inhibit Müller cell in-flammation and ameliorate retinal neural injury in diabetic mice,and the mechanism may be associated with the inhibition of the Spi1/NF-κB signaling pathway.
