1.Value of Fetuin-A, Fetuin-B, and insulin resistance index in predicting nonalcoholic fatty liver disease

ObjectiveTo investigate the value of serum Fetuin-A and Fetuin-B combined with Homeostasis Model Assessment of Insulin Resistance (HOMA-IR) in predicting nonalcoholic fatty liver disease (NAFLD). MethodsA total of 120 patients with NAFLD who attended Department of Gastroenterology, The First Affiliated Hospital of Dali University, from June 2020 to June 2021, and 120 healthy individuals who underwent physical examination at Physical Examination Center during the same period of time were enrolled as subjects, and clinical data were collected from all subjects. The serum levels of Fetuin-A and Fetuin-B were measured. The independent-samples t test or the Mann-Whitney U test was used for comparison of continuous data between two groups, and the chi-square test was used for comparison of categorical data between two groups; the multivariate Logistic regression analysis was used to assess the risk factors for NAFLD. The receiver operating characteristic (ROC) curve was plotted to evaluate the predictive efficacy of Fetuin-A and Fetuin-B combined with HOMA-IR in NAFLD patients. ResultsCompared with the healthy control group, the NAFLD group had significantly higher levels of body mass index, systolic blood pressure, diastolic blood pressure, alanine aminotransferase, aspartate aminotransferase, fasting blood glucose, fasting insulin, triglycerides, HOMA-IR, Fetuin-A, and Fetuin-B (all P<0.05). The multivariate Logistic regression analysis showed that Fetuin-A (odds ratio [OR]=1.010, 95% confidence interval [CI]: 1.001 — 1.020, P<0.05), Fetuin-B (OR=1.113, 95%CI: 1.021 — 1.214, P<0.05), and HOMA-IR (OR=24.053, 95%CI: 2.624 — 220.470, P<0.05) were independent risk factors for NAFLD. The ROC curve analysis showed that Fetuin-A, Fetuin-B or HOMA-IR alone had an area under the ROC curve (AUC) of 0.637 (95%CI: 0.551 — 0.722), 0.853 (95%CI: 0.796 — 0.912), and 0.837 (95%CI: 0.763 — 0.912), respectively, and Fetuin-A combined with Fetuin-B, Fetuin-A combined with HOMA-IR, and Fetuin-B combined with HOMA-IR had an AUC of 0.853 (95%CI: 0.795 — 0.911), 0.843 (95%CI: 0.770 — 0.916), 0.922 (95%CI: 0.877 — 0.967), respectively, while the combination of these three indicators had an AUC of 0.922 (95%CI: 0.877 — 0.966). ConclusionFetuin-A and Fetuin-B have a certain value in predicting NAFLD, and Fetuin-B combined with HOMA-IR tends to have a higher predictive value.
2.Effects of lncRNA RP11-770J1.3 and TMEM25 expression on paclitaxel resistance in human breast cancer cells.

OBJECTIVETo investigate the effects of long non-coding RNA(lncRNA) RP11-770J1.3 and transmembrane protein 25 (TMEM25) on paclitaxel resistance in human breast cancer MCF-7/PR cell line.
METHODSThe expression of lncRNA RP11-770J1.3 and TMEM25 in human breast cancer MCF-7(paclitaxel sensitive) and MCF-7/PR(paclitaxel resistant) cells were detected by quantitative RT-PCR. The synthetic interfering fragments of lncRNA RP11-770J1.3 and TMEM25 were transfected into MCF-7/PR cells. Sulforhodamine B assay was used to detect the sensitivity of MCF-7/PR cells to paclitaxel after interference of lncRNA RP11-770J1.3 and TMEM25. The expression of multidrug-resistance genes and proteins were detected by qRT-PCR and Western blot, respectively.
RESULTSlncRNA RP11-770J1.3 and TMEM25 were highly expressed in MCF-7/PR cells, and were significantly down-regulated after transfection of synthetic interfering fragments. Down-regulation of lncRNA RP11-770J1.3 and TMEM25 enhanced the sensitivity of MCF-7/PR cells to paclitaxel, and inhibited the expression of MRP, BCRP and MDR1/P-gp (all<0.05). Such effects were more significant when lncRNA RP11-770J1.3 and TMEM25 were both down-regulated (all<0.05).
CONCLUSIONSlncRNA RP11-770J1.3 and TMEM25 are highly expressed in MCF-7/PR cells, and the down-regulation of lncRNA RP11-770J1.3 and TMEM25 can enhance paclitaxel sensitivity in MCF-7/PR cells.
3.CXC chemokine receptor 4 regulates breast cancer cell cycle through S phase kinase associated protein 2.

OBJECTIVETo investigate the effect of CXC chemokine receptor 4 (CXCR4) on cell cycle of breast cancer and its molecular mechanisms.
METHODSThe expression of CXCR4 and S phase kinase associated protein 2 (Skp2) was detected by real-time fluorescence quantitative PCR (fqRT-PCR) and Western blot in breast cancer cells. The expression of signal proteins and the downstream genes of Skp2 was detected by Western blot. The effect of CXCR4, PI3K/Akt pathway inhibitor LY294002 and ERK pathway inhibitor U0126 on cell cycle of breast cancer was detected by propidium iodide staining.
RESULTSSkp2 was significantly down-regulated in CXCR4-downregulated cells and up-regulated in CXCR4-upregulated cells. CXCR4 also regulated the expression of Skp2 and other downstream genes by signaling protein. The proportion of cells in G/Gphase increased and that in S phase declined in CXCR4-downregulated cell, and the effect was more significant when combined with the use of LY294002 or U0126.
CONCLUSIONSCXCR4 can affect cell cycle and inhibit the proliferation of breast cancer cells by regulating Skp2 gene expression through PI3K/Akt and ERK signaling pathway.
4.Inhibiting effects of gambogenic acid in combination withmiR-218 on cervival cancer HeLa cells and its mechanisms

Aim To study the inhibitory effects of gambogenic acid in combination with miR-218 on cervical cancer HeLa cells and its mechanisms.Methods Eukaryotic expression vector of miR-218(pmi8-218) was transfected into HeLa cells.Transcript levels of miR-218 were quantified by real-time quantitative PCR.HeLa cells were incubated with different concentrations of gambogenic acid alone or in combination with pmiR-218.The cell growth inhibiting ratio of HeLa cells was assessed by MTT assay.Cell apoptosis was measured by fluorescence activated cell sorting.The expression levels of Bcl-2, Bax and E-cadherin were measured by Western blot and qRT-PCR.Results Levels of miR-218 transcript significantly increased in pmiR-218 transfected HeLa cells.Overexpression of miR-218 may enhance the sensitivity of HeLa cells to gambogenic acid.Over expression of miR-218 could enhance the effect of gambogenic acid on inhibition cell proliferation, promoting apoptosis of HeLa cells.pmiR-218 could enhance the regulation of Bax expression and decrease the expression of Bcl-2 in HeLa cells.Conclusions Over expression of miR-218 may enhance the sensitivity of HeLa cells to gambogenic acid;miR-218 can enhance the effect of gambogenic acid on inhibition cell proliferation and promote the apoptosis of HeLa cells, and the mechanism may be related to down-regulation of Bcl-2/Bax expression.
5.Influence of data quality on early warning sensitivity of syndromic surveillance system based on medical institutions

Objective To evaluate the influence of data quality on the sensitivity of early warning syndromic surveillance system based on medical institutions in Qianjiang,Hubei province and explore the relationship between data quality and sensitivity of early warning of the system.Methods The delay reporting rate and underreporting rate were calculated for the evaluation of the data quality.Data obtained from semi-synthetic simulated outbreak and area under the curve (AUC) were used in combination to test the sensitivity of early warning of various models and select the optimal model.Time-series generalized additive model (GAM) was used to analyze the curve fitting and threshold effect between data quality and early warning sensitivity of the system.Results A total of 179 905 cases were reported from April 1,2012 to January 31,2014,in which 8 744 were not reported timely (16.45%).Averagely 416 reporting were delayed in each month.There were 2 566 cases which were underreported (4.83%).Compared with other early warning models,i.e.Cumulative Sum (CUSUM),Shewhart,Exponentially Weighted Moving Average (EWMA),Early Aberration Reporting System (EARS-3C),the MA model had the maximum area under the curve (AUC=0.93),and the difference was significant (P<0.001).The early warning sensitivity ranged from 84.89% to 97.25% during the operation period of the syndromic surveillance system.Underreporting had influence on early warning sensitivity,when underreporting rate was over 2.78%,the sensitivity would decrease obviously.No obvious associations were observed between the delay reporting rate and early warning sensitivity of the system.Conclusion The data quality had influence on the early warning sensitivity of the syndromic surveillance system based on medical institution in Qianjiang.In the context of this study,underreporting had the main influence on the sensitivity of early warning.
6.Analysis of the results of patch test in 192 patients with hand eczema

Objective To investigate the common allergens in the patients with hand eczema.Methods From November 2014 to March 2016,the patients with hand eczema were tested by the patch test kit of daily life series.Results The results of the patch test of 192 patients with hand eczema were collected.Allergens were detected in 178 (92.71%) cases.The top 5 allergens were nickel chloride (23.96%),cobalt chloride (18.75%),aromatic compounds (17.19%),nickel sulfate (16.67%) and thimerosal (13.54%).The positive rates of kappa mixture,aromatic compounds,tertiary butyl phenolic resin in males were 16.88%,14.29%,11.69%,respectively,which were higher than those (5.22%,4.35%,3.48%) in females.Conclusion Nickel chloride,cobalt chloride,aromatic compounds,nickel sulfate and thimerosal are common allergens in patients with hand eczema.

8.Nursing of patients undergoing skin tissue biopsy on head and face

Objective To summarize the experience in nursing patients undergoing skin biopsy on head and face . Method One hundred and twenty patients with head facial dermatosis undergoing skin tissue biopsy were nursed from August 2012 to October 2013. Result All operation for biopsy were completed smoothly, with the cure rate at the first phase 100.0%, healing time 5~7 days, averaged (7.0 ± 3.0)d and no complications. Conclusion For those patients having undergone facial skin tissue biopsy, the nursing measures like psychological nursing, health education before operation, preoperative relief of mental disorder, postoperative wound nursing and health education, prevention of complications are critical for their recovery.
9.Curative effect of 308nm excimer laser on child vitiligo and nursing measures

Objective To explore the curative effect of 308 nm excimer laser on child vitiligo and nursing measures. Method Fourty-eight children with vitiligo were treated with 308 nm excimer laser and nursed with targeted psychological treatment, skin and diet nursing.Result After treatment,6 of them got recovered(12.5%),24 remarkably improved(50%),15 slightly improved(31.2%)and 3 ineffectively treated(6.2%).Conclusions 308 nm excimer laser is effective for the treatment of child vitiligo.High quality nursing can ensure and improve the efficiency of the 308 nm excimer laser.
10.Nursing care of patients with cosmetic dermatitis receiving patch test

Objective To summarize key points of nursing patients with cosmetic dermatitis during patch test.Method One hundred and forty one patients with contact dermatitis induced by cosmetics underwent patch test and the nursing care was performed. Results The top five allergens based on the positive rate ranked in descending order were potassium dichromate(43?97%),nickel sulfate (43?26%),cobalt chloride(36?17%),mixture of hydroxy benzoic acid(23?40%)and fragrance mix(20?57%).The positive suspicious cosmetics ranked in ascending order included body spray(80?00%),freckle cream(56?00%),liquid products(38?89%),cleaning supplies (31?58%)and skin care cream(25?62%).Conclusion During the cosmetic patch test for the patients with contact dermatitis,the nurses should conduct mental care,carefully observe the patients and make instructions to them on cosmetic health knowledge.
