1.Relationship of three-dimensional power Doppler ultrasonographic parameters and endocrine profile in different symptoms of patients with polycystic ovary syndrome

Objective To study the relationship of three-dimensional power Doppler ultrasonographic parameters and endocrine profile in different symptoms of patients with polycystic ovary syndrome(PCOS).Methods One hundred and forty nine women with PCOS were divided into two groups,which included obese PCOS(OB-PCOS) group and non-obese PCOS (NOB-PCOS) group.The ultrasonic parameters such as follicle number,ovarian average diameter,ovarian volume,stromal volume,follicle volume,vascularization index(Ⅵ),flow index(FI),vascularization flow index(VFI) were measured and compared.Serum levels of luteinizing hormone (LH),follicle stimulating hormone (FSH),progesterone (P),estradiol (E2),testosterone(T),prolactin (PRL),sex hormone binding globulin (SHBG),free androgen index (FAI),fasting plasma glucose (FPG),fasting insulin (FINS),homeostasis model assessment-IR(HOMA-IR) were also measured and compared.The correlation of the ultrasonic parameters and hormonal factors were analyzed.Results The follicle number,ovarian average diameter,ovarian volume,stromal volume,follicle volume,FI and VFI,FINS,HOMA-IR,FAI of OB-PCOS were significantly higher than those of NOB-PCOS (P <0.01 or 0.05),the FSH,SHBG were significantly lower than those of NOB-PCOS (P <0.05 or 0.01).In OB-PCOS group,the follicle number was significantly associated with FSH(r =0.771,P <0.01).The ovarian volume,stromal volume,FI and VFI were significantly associated with HOMA-IR(r =0.412,0.842,0.389,0.415,P <0.05 or 0.01),FI was significantly associated with FAI (r =0.812,P <0.01).In NOB-PCOS group,the follicle number,ovarian volume were significantly associated with FAI(r =0.472,0.552,P <0.05)..Conclusions There are some different characters in ultrasonography and endocrine parameters between obese and non-obese PCOS patients.

Polycystic ovary syndrome (PCOS) is a complex hormonal disorder and one of the most common reproductive endocrinology abnormalities in women. Recently, many studies have been conducted assessing Chinese herbal medicine as an alternative treatment for women with PCOS, it is, therefore, worthwhile to analyze and observe the curative effects of traditional Chinese medicine treatment in PCOS.
3.Clinical analyses for 136 patients of moderate and severe ovarian hyperstimulation syndrome

Objective To investigate the high risk factors, clinical manifestation and therapy strategy of ovarian hyperstimulation syndrome (OHSS) in assisted reproduction techniques (ART). Methods Clinical data of 136 cycles of OHSS patients after in vitro fertilization-embryo transfer (IVF-ET) from 2005 to 2008 were analyzed retrospectively. Results (1) A large number of follicles stimulated, luteal phase supported with hCG and pregnancy were high risk factors to induce OHSS. (2)Most of OHSS occurred on the 3~(rd) day to 10~(th) day after administration of medicine to induce ovulation. (3) Longer disease process, more severe symptoms and larger amount albumin used for treatment of OHSS were commonly observed in pregnant patients with OHSS compared with non-pregnant ones. (4) Human serum albumin through intravenous dripping was one of the most successful treatment as plasma expanders through restoration of effective blood volume; paracentesis was necessary for critical OHSS patients with severe ascites and hydrothorax. Conclusions OHSS is a serious complication of controlled ovarian hyperstimulation, which deserves proper prevention and treatment.
4.The inhibitory effects of Pinella's ingredients on the growth of cervical cancer cell lines

Objective To investigate the effects of Pinella's ingredients on the viability of cells, morphology, microstructure, cell cycle and apoptosis in human cervical cancer cell lines. Methods The β-sitosterol and/or total protein of Pinella were incubated at different concentrations with cervical cancer cell line SiHa. The effects of β-sitosterol and/or total protein of Pinella on the viability of cells were tested by MTT assay. The effects of β-sitosterol on morphology, microstructure, cell cycle and apoptosis were studied by phase-contrast microscope, electron microscope and flowcytometry, respectively. Results β-sitosterol could obviously inhibit the viability of SiHa cells in a time-and dose-dependent manner. The total protein of Pinella had no effects on SiHa cells' viability. 20 μmol/L β-sitosterol induced the accumulation of SiHa cells in S phase in the cell cycle. And the percents of apoptosis and necrosis increased. The morphology and microstructure of SiHa cells changed significantly after treated with 20 μmol/L β-sitosterol. Conclusions The total protein of Pinella had little influence on the viability of cervical cancer cells SiHa. The viability of SiHa cells could be suppressed by β-sitosterol. β-sitosterol could induce the accumulation of cells in S phase and the percent of apoptosis and necrosis. The morphology and microstructure changed significantly after treated with β-sitosterol. Therefore, β-sitosterol might be a prospect safe and low toxicity anti-cervical cancer drug.

Objective:To study the effect of Kidney-replenishing herb on the indoleamine 2,3-dioxygenase(IDO) of decidual macrophages.Methods:RT-PCR was applied to analyze the IDO mRNA transcription of decidue;Western blot was applied to analyze the expression of IDO protein on decidual macrophages;ELISA was applied to determinate the IL-10 and IFN-? concentration in culture supernates of decidual macrophages and lymphocytes; high-performance liquid chromatography was applied to determinate the kynurenine and tryptophan concentration in cell culture supernates, the ratio of kyn/try was used to determinate IDO activity.Results:The decidual IDO mRNA transcription of normal pregnancy is higher than that of inevitable abortion; the inhibitor of IDO make the balance of Th cytokine depart from Th2; the serum made from herbal medicine improve the expression of IDO protein and IDO activity, and resume the balance of Th cytokine.Conclusion:Appropriate IDO mRNA transcription and IDO activity of decidue is necessary for normal pregnancy;Kidney-replenishing herb improve IDO activity to appropriate value.

Objective:To study the immunologic and endocrinologic mechanism of Chinese herbs in the treatment of abortion Methods:SD rats injected with bromocriptine 0 125 mg/d subcutaneously(sc)during day 6~8 of pregnancy were used as aborting model Uteruses of the rats treated with or without Chinese herbs were taken out at day 12 of pregnancy for counting embryoes and determining mRNA levels of Th1/Th2 type cytokines in decidua by semiquantity RT PCR Results:The insufficiency of PRL and P in serum during gestation was related to embryo miscarriage remarkably A predominant Th1 response at the maternal fetal interface is harmful to pregnancy Aborting rate induced by bromocriptine was decreased after treating with Chinese herbs Chinese herbs can increase serum PRL and P , then trigger a predominant Th2 response in the model Conclusion:With it's effect of increasing PRL and P in serum and the Th2 bias response at the maternal fetal interface, Chinese herbs can reverse the abortion effect of bromocriptine
8.Comparison of the binding characteristics of insulin receptors on ovary, adrenal and hepatic plasma membrane from rats

Objective To compare the binding characteristics of insulin receptors (IR) on ovary,adrenal and hepatic plasma membrane from rats. Methods The number and affinity of IR was detected by radioligand binding assay. Results Scatchard plot analyses showed that the receptor numbers for high affinity sites are 7. 359?10/mg protein, 8. 029? 10/mg protein and 6. 440?10/mg protein of liver,ovarian and adrenal plasma membrane with KD of 6.147?107 M-1, 1. 528?10, M-1, and 1. 010?107 M-1 that. The receptor numbers for low affinity sites are 2. 403?10/mg protein,2. 212?10/mg protein,and 2. 257?10/mg protein respectively with KD of 2. 920?10M-1,2. 008?10 M-1 and 0. 433?10sM-1. Conclusion There are abundent IR on ovary and adrenal just as many as those on liver, although the KD are smaller. It suggests that insulin may play an important regulatory role in ovary and adrenal.
9.Clinical Observation on Treatment of Advanced Menorrhea with QingJing Capsule

Objective To investigate the effect of Qingjing Capsule (QC) in treating advance menorrhea (AM) and to evaluate its safety. Methods Double- blind, double- imitating, parallel- controlled and randomized trial was performed in 63 cases of AM with deficiency- heat syndrome.Thirty- two cases was in QC group(Group A) and other 31 in Gujing pill group(Group B).The menstrual cycle, clinical symptoms and signs, basal body temperature (BBT), serum levels of estradiol (E2) and progesterone and endometrium biopsy were observed to evaluate the effects of QC. Results The short- term markedly- effective rate was 90.63% and 77.42% and the short- term effective rate was 96.88% and 93.55% in Group A and Group B respectively. The long- term markedly- effective rate was 83.87% and 67.74% and the long- term effective rate was 93.55% and 80.65% in Group A and Group B respectively. 10.Expression of suppressors of cytokine signaling in mater-fetal interface of human first trimester gestation

Objective:To investigate the mRNA and protein expression of suppressors of cytokine signaling in human villa and decidua of first trimester gestation.Methods:We first obtained villous tissues and decidual tissues from 38 women who had undergone selective termination at 7-9 weeks of normal gestation.The transcripts of SOCS1,SOCS2,SOCS3 in villous tissues and decidual tissues were detected by using semi-quantitative reverse transcriptase polymerase chain reaction.The mRNA of SOCS1,SOCS2,SOCS3 from cultured trophoblasts and decidual stromal cells were detected.Also the protein expression of 35 pregnant women were detected by western blot.Immunohistochemistry analysis revealed the location of SOCS1,SOCS2,SOCS3 in materno-fetal interface.IL-10 and IFN-? secreted by cells in vitro were detected by ELISA.Results:The expression of SOCS1,SOCS2,SOCS3 were found in mater-fetal interface,while positive expression of SOCS3 in villi/decidua was 73.7%/71.1%,and that of SOCS2 was 50.0%/39.5%,then SOCS1 expression was 34.2%/31.6%.The protein expression of SOCS1,SOCS2,SOCS3 were similar to the transcription level.SOCS1,SOCS2,SOCS3 were mainly expressed in villi and decidual stroma.Trophoblasts and decidual stromal cells were not found the expression of SOCS1,but the low level mRNA of SOCS2,SOCS3 without serum,and IL-10 secreted by them increased significantly(P
