Objective To explore the electroencephalography(EEG)characteristics of patients with knee osteoarthritis(KOA)which are different from those of healthy subjects during dynamic balance tests,and study the connection and influence between brain activity and KOA.Methods Fifteen patients with KOA and 17 healthy subjects performed the timed up and go test(TUGT)and five times sit-to-stand test(5STS).The EEG characteristics of the 4 frequency bands,namely δ(0.5-4 Hz),θ(4-8 Hz),α(8-14 Hz)and β(14-30 Hz)of 9 electrodes including CZ,C3,C4 in the central region,PZ,P3,P4 in the parietal lobe,and OZ,O1,O2 in the occipital lobe of the cerebral cortex of the subjects were recorded.Time and frequency domain analysis were conducted.Results During 5STS,the time-domain amplitudes of electrodes C4,P3,and P4 in the δ band,electrodes C4,P3,and P4 in the θ band,and electrodes P3 and P4 in the β band of KOA patients were significantly higher than those of healthy subjects(P<0.05);the power of EEG signals of electrodes P3,P4,and O2 in the β band was significantly higher than that of healthy subjects.During TUGT,except that the time-domain amplitude of electrode PZ in the θ band of KOA patients was significantly lower than that of healthy subjects(P<0.05),there was no significant difference in all electrodes in other frequency bands;the power of EEG signals of electrodes C3,CZ,C4,PZ,P4,O1,OZ,O2 in the δ band and electrodes CZ,P3,PZ,and OZ in the α band and electrodes CZ,P4,OZ,and O2 in the β band was significantly lower than that of healthy subjects(P<0.05).Conclusions Compared with healthy subjects,the activity in the parietal lobe area of the brain of KOA patients during 5STS was stronger,while the brain activity in the central region,parietal lobe,and occipital lobe areas of KOA patients during TUGT was weaker than that of healthy subjects.This study may provide some references for clinicians in the prevention and treatment of KOA.

Objective To observe the abnormal brain activity in patients with migraine without aura(MwoA)with MRI.Methods Fifty MwoA patients(MwoA group)and 46 healthy volunteers(control group)were prospectively enrolled.Functional MRI(fMRI)was performed to observe the dynamic regional homogeneity(dReHo)of brain regions,then brain regions with differences of dReHo value between groups were extracted,and correlations with clinical scales were analyzed.Results Compared with control group,dReHo values of surrounding cortex of bilateral calcarine fissure and right middle occipital gyrus increased,of right middle temporal gyrus,right middle frontal gyrus and left cuneus decreased in MwoA group(GRF correction,voxel level all P＜0.005,cluster level all P＜0.05).The weight analysis showed that brain regions with top 3 absolute weight values were surrounding cortex of right calcarine fissure,left cuneus and right middle occipital gyrus.dReHo value of surrounding cortex of left calcarine fissure in MwoA patients was negative correlated with self-rating depression scale(SDS)(r=-0.28,P=0.04).Conclusion Abnormalities in the upward transmission pathway of visual information existed in MwoA patient,especially in surrounding cortex of right calcarine fissure,left cuneus and right middle occipital gyrus.

Objective:To investigate the application effect of injecting ozone water into the dorsal root ganglion combined with skin lesions in the acute stage of herpes zoster.Methods:A prospective study was conducted on 90 patients with acute lesions of herpes zoster in the chest and abdomen who were admitted to the outpatient department of Changsha Central Hospital from February 2021 to April 2024. According to the random number table method, they were divided into two groups, each with 45 cases. The control group received oral administration of famciclovir, pregabalin, weak laser skin lesion illumination, and local application of mupirocin. The observation group received 10 ml of 25 μ g/ml ozone water injected into the dorsal root ganglion under ultrasound guidance on the basis of the control group, and 20 ml of 25 μg/ml ozone water injected subcutaneously into the skin lesion. Two groups of local lesions were compared in terms of the cessation of blister growth, local scab formation, and significant pain relief time. The changes in pain Numeric Rating Scale (NRS) score and the proportion of viral inclusions in the blisters during the treatment were also analyzed. The sleep quality and quality of life scores, as well as complications during the treatment process, were analyzed after 10 days of treatment in both groups.Results:The observation group had a shorter time for the cessation of local lesion blister growth, local scab formation, and significant pain relief compared to the control group (all P<0.05). After 48 hours and 10 days of treatment, the pain NRS score of both groups was lower than before treatment (all P<0.05), and the pain NRS score of the observation group was lower than that of the control group at the same time after 48 hours and 10 days of treatment (all P<0.05). After 48 hours and 10 days of treatment, the proportion of viral inclusions in the blisters of both groups was lower than before treatment (all P<0.05), and the observation group had a lower proportion of viral inclusions in the blisters than the control group at the same time (all P<0.05). After 10 days of treatment, the sleep quality and quality of life scores of the observation group were better than those of the control group (all P<0.05). The total incidence of complications such as local infections, insomnia, palpitations, and local numbness during the treatment process of the observation group was significantly lower than that of the control group ( P<0.05). Conclusions:Subcutaneous injection of ozone water into the dorsal root ganglion and lesion site of patients with acute stage herpes zoster can inhibit the growth of local blisters, promote scab formation, reduce the level of viral inclusions in blisters, effectively alleviate patient pain, improve their sleep quality and quality of life, and reduce the occurrence of complications.

Clinical characteristics were analyzed retrospectively in a patient with empty sella and adrenal adenoma with regard to the elaborated diagnosis and treatment.Although empty sella syndrome alone was common,clinical combination with adrenal adenoma was rarely reported.It was difficult to diagnose due to complex symptoms and hormone levels.This case could help increase awareness of the disease and accumulate the experience in diagnosis and treatment.

Objective To evaluate the cancer risk among the inhabitants around a certain nuclear facility of Gansu Province after operation for nearly 50 years.Methods Specially developed forms were filled by countryside doctors in the villages 30 km on the leeward or at the lower reaches of Yumen City,Gansu Province where a nuclear facility had operated for nearly 50 years,to register the relevant data of those who died of solid cancer and leukemia during the period from 1 January 2004 to 31 December 2008 based on the relevant data and household survey.A town 90 km away from the nuclear facility with 4181 people were regarded as control area.Results The crude mortality and standardized mortality rate(SMR) of cancer were 95.51×10-5 and 93.56×10-5.respectively.The crude mortality and SMR of leukemia were 2.44×10-05 and 2.22×10-05, respectively.and the crude mortality and SMR of cancer were 106.88×10-05, and 89.31×10-5, respectively.The crude mortality and SMR of leukemia were 2.44×10-05 and 2.22×10-5, respectively in the investigated area,and the crude mortality of leukemia was 3.56×10-5 (only one leukemia cage died)in the control area in the same time.The cancer and leukemia mortality rates of the investigated area were similar to those of control area.Conclusions Nuclear facility does not significantly increase the cancer mortality among the residents living nearby.

Objective To construct two DNA vaccines encoding Gag-Env fusion protein and Tat-Rev-Integrase(C-half)-Vif-Nef fusion protein derived from the first HIV-1 CRF01_AE isolate(AE2f) in Chi-na and to evaluate the immunogenicity in mice. Methods Two DNA vaccines were constructed by inserting the codon optimized and synthesized gag-env fusion gene and tat-rev-integrase(c-half)-vif-nef fusion gene de-rived from AE2f into mammalian expression vector pDRVISV1. 0, the generated DNA vaccines were desig-nated as pSVAE/GE and pSVAE/TRIVN, respectively, and their in vitro expression were determined by Western blot with transfected 293T cells. Mice were i. m. immunized with either pDRVI1.0 as mock control, pSVAE/GE or pSYAE/TRIVN for 4 times at two-week interval. Two weeks following the final im-munization, cellular responses to pool of HIV-1 Env, Gag, Tat, Rev, Intergrase, Vif and Nef peptides were evaluated by ELISPOT assay. Results The construction of DNA vaccine pSVAE/GE and pSVAE/TRIVN was validated by restriction enzyme digestion and bidirectional sequencing. Western blot showed a specific band at molecular mass 220×10~3 in lane of pSVAE/GE transfeeted 293T cell and a specific band at 95×10~3 in the lane of pSVAE/TRIVN. Both DNA vaccines mounted significant specific T cell responses with (3010 ± 566) SFC/10~6 splenocytes for DNA vaccine pSV AE/GE and (948 ± 737) SFC/10~6 spleno-cytes for DNA vaccine pSVAE/TRIVN, whereas the mock control of pDRVISV1.0 only raised marginal T cell responses. Conclusion Both pSVAE/GE and pSVAE/TRIVN were capable of expressing the inserted fusion immunogen genes and able to elicit vigorous cellular immune responses, therefore, these DNA vac-cines are highly immunogenic.

Objective To evaluate the efficacy and safety of domestic olmesartan in treatment of mild to moderate essential hypertension in comparison with losartan. Methods Two hundred and thirty-seven patients with mild to moderate essential hypertension were enrolled in a randomized, double-blind, multi-center, paralleded and active-controlled trial, and were divided into olmesartan group (olmesartan 20 mg + losartan 50 mg placebo) and losartan group (losartan 50 mg + olmesartan 20 mg placebo) for a 8-week therapy. Four weeks after treatment, dosages of drugs were doubled in patients with seated diastolic blood pressure ≥90 mmHg (1 mmHg =0.133 kPa). All patients were followed up every two weeks, and the efficacy and adverse effects were observed. Another 32 patients with mild to moderate essential hypertension were enrolled and given olmesartan only, and ambulatory blood pressure monitoring was performed before and 8 weeks after treatment. Results Compared with those before treatment, both systolic blood pressure and diastolic blood pressure significantly decreased in olmesartan group and losartan group 8 weeks after treatment [(15.2 ±13.3) mmHg and (19.5 ±11.8) mmHg, respectively for systolic blood pressure (P <0.001); (15.9 ±7.48) mmHg and (16.2 ± 5.95) mmHg, respectively for diastolic blood pressure (P < 0.01) ], while there was no significant difference between these two groups (P > 0.05). There was no significant difference in total effective rate and incidence of adverse effect between these two groups (86.9% vs 93.7% and 7.63% vs 5.88% , P > 0.05) . Ambulatory blood pressure monitoring demonstrated that trough to peak ratios of systolic blood pressure and diastolic blood pressure were 86% and 71%, respectively. Conclusion Domestic olmesaratan provides an effective, safe and long action in the treatment of mild to moderate essential hypertension.

Objective To discuss the effect of skill training for staff working in washing rooms of sterilization and supply center after application of centralized management model.Methods The training objects included all the working staff in the washing rooms and the training procedure covered four steps：preparation,discussion,implementation and evaluation.The evaluation indices involved level of post skill,cleaning and sterilization effect of instruments and satisfaction degree of staff.Results After training the level of post skills were increased dramatically(P＜0.01).The cleaning and sterilization effect of instruments was also improved(P＜0.01).96％ of staff considered their post ability elevated after training.Conclusion After skill training for staff working in washing rooms,the post skill and cleaning and sterilization quality of instruments were both enhanced.It proved valuable for guarantee of cleaning and sterilization quality of medical instruments and improvement of professional skills for staff working in washing rooms of sterilization and supply center.

A 1 270 bp full-length cDNA fragment was obtained from the Schistosoma japonicum (Chinese strain) adult cDNA library after the '3' and 5' ends of the incomplete expression sequence tag (EST) of hypoxanthine-guanine phosphoribosyltransferase of Schistosoma japonicum (SjHGPRT) were amplified by the anchored PCR with 2 pairs of primer that were designed according to the published incomplete SjHGPRT EST and the sequence of multiclone sites of library λgt1 1 vector. Sequence analysis indicated that this fragment, with an identity of 82% to hypoxanthine-guanine phosphoribosyltransferase ofSchistosoma mansoni (SmHGPRT), contained a complete open reading frame(ORF). The deduced amino acid sequence showed 83% identity to that of SmHGPRT. This fragment was cloned into the prokaryotic expression vector pQE30, and subsequently sequenced and expressed in Escherichia coli. SDS-PAGE revealed that M of the recombinant protein was about 28 ku. Western-blot analysis showed that the recombinant protein was recognized by the polyclonal antisera from rabbits immunized with Schistosoma japonicum adult worm antigen. Mice vaccinated with recombinant protein revealed significant worm burden, liver eggs per gram (LEPG), fecal eggs per gram (FEPG) and intrauterine eggs of the female worms reduction percentage, compared with the controls. Taken together, the SjHGPRT full-length cDNA can be cloned and expressed in E. coli as a recombinant protein that elicited immunity against the challenge infection with Schistosoma japonicum, indicating its potential as a partia1 protection vaccine candidate.
