1.Hepatitis B cirrhosis comorbid with Staphylococcus lugdunensis bloodstream infection: A case report

As a coagulase-negative staphylococcus, Staphylococcus lugdunensis has stronger virulence and pathogenicity than other Staphylococcus bacteria and is comparable to Staphylococcus aureus. In recent years, there has been a gradual increase in the number of related case reports, A patient with hepatitis B cirrhosis and Staphylococcus lugdunensis bloodstream infection was admitted to our department and was followed up for more than 2 years, and the patient was analyzed in terms of clinical features, pathogenicity, invasion, and the application of related antibiotics.
2.(+)-Strebloside induces Non-Hodgkin lymphoma cell death through the STEAP3-Mediated Ferroptosis and MAPK pathway.

(+)-Strebloside, a significant bioactive compound isolated from the roots of Streblus asper Lour., demonstrates inhibitory effects against multiple malignancies. However, its specific function and underlying mechanistic pathways in Non-Hodgkin lymphoma (NHL) remain unexplored. This investigation sought to elucidate the role and potential mechanisms of (+)-strebloside-induced NHL cell death. The results demonstrated that (+)-strebloside significantly induced apoptosis and ferroptosis in NHL cells, including those from Raji cell-derived xenograft models. Mechanistic analyses revealed that (+)-strebloside enhanced six-transmembrane epithelial antigen of prostate 3 (STEAP3)-induced ferroptosis in NHL, and STEAP3 inhibition reduced the proliferation-inhibitory effects of (+)-strebloside. Furthermore, (+)-strebloside suppressed NHL proliferation through the mitogen-activated protein kinase (MAPK) pathway, and extracellular signal-regulated kinase (ERK) inhibition diminished the proliferation-inhibitory activity induced by (+)-strebloside. These findings indicate that (+)-strebloside presents promising therapeutic potential for NHL treatment.

3.Mass spectrometry of HBV peptides derived from hepatitis B virus polymerase and X protein presented by B lymphoblastoid cells

Objective:To apply mass spectrometry as well as bioinformatics techniques to analyze HBV peptides derived from Pol and X proteins presented by human immortalized B lymphocytes(BLCLs),and to screen for effective T-cell epitopes,which lays the foundation for the development of therapeutic vaccines.Methods:The group has constructed a genome-wide expression plasmid containing 1.2-fold HBVC2 isoforms and transfected it into BLCLs by electro-transfection,isolated and identified HBV peptides by LC-MS/MS,bioinformatically predicted peptide sequences in terms of sensitization,antigenicity,toxicity,HLA molecular affinity,and the ability of peptide HLA-class Ⅰ complex to bind to specific T cells,and retrieved reported sequences.Results:HBV peptides from lysates of five immortalized B cells(BLCLs-1 to BLCLs-5)carrying HBVC2 subtype-expressing recombinants were analyzed,and 141 peptides with sequence lengths of no less than 8 amino acid residues(≥8 aa)were successfully isolated and identified,of which 133 were derived from Pol,and 8 from X proteins.The 141 HBV peptides were analyzed for sensitization,antigenicity and toxicity,and 50 antigenic,non-toxic and sensitizing HBV peptides(47 from Pol and 3 from X protein)were screened for affinity analysis with HLA-class Ⅰ and Ⅱ molecules and for prediction of the binding ability of the peptide HLA-class Ⅰ complexes to specific T cells.Among these peptides,37 had affinity to the corresponding genotypes of HLA-class Ⅰ and Ⅱ molecules.Finally,we obtained 37 peptides with antigenic,nontoxic,and sensitizing properties with IC50<500 nmol/L to HLA-class Ⅰ and Ⅱ molecules,which have affinitied to the corresponding genotypes but have not been reported,and can be continued to be explored as potential epitopes.Finally,15 HBV peptide hotspot core regions were formed by intra-and inter-strain common,contained,overlapping or neighboring sequence analysis of 141 peptides,with 7 core region sequences with antigenic,nontoxic,and sensitizing properties restricted to the corresponding geno-types.Peptides with affinity IC50<500 nmol/L had an affinity core sequence overlap of no less than 8 amino acids,and can be priori-tized for candidate T cell epitopes for subsequent studies.Conclusion:Peptides derived from Pol and X proteins have been successfully isolated and identified,and potential T cell epitopes have been screened.
4.The Effectiveness and Safety of Aromatase Inhibitors Combined with r-hGH in Improving Final Adult Height in Late-Pubertal Boys: A Retrospective Cohort Study

To investigate the efficacy and safety of aromatase inhibitors (AI) combined with recombinant human growth hormone (r-hGH) in improving the final height (FH) of boys in late puberty. Clinical data of pubertal boys with growth deceleration, consecutively admitted to the Department of Child Growth and Development, Zhangzhou Affiliated Hospital of Fujian Medical University between February 2017 and December 2022, were collected. According to different treatment regimens, the patients were divided into the GH group, the AI+GH group, and the control group. The GH group received r-hGH monotherapy[dose 0.18-0.2 U/(kg·d)] until the growth velocity was less than 0.5 cm/3 months. The AI+GH group received anastrozole 1 mg/d or letrozole 2.5 mg/d combined with r-hGH[dose 0.18-0.2 U/(kg·d)]; AI was discontinued after 1 year, while r-hGH was continued until the growth velocity was less than 0.5 cm/3 months. The control group did not receive any height-promoting drugs. All patients were followed up outpatient every 3 months until FH was reached, monitoring growth development indicators and adverse reactions. The efficacy and safety were compared between the GH group and the AI+GH group. A total of 69 pubertal boys meeting the inclusion and exclusion criteria were enrolled, including 28 in the GH group, 17 in the AI+GH group, and 24 in the control group. There were no statistically significant differences among the three groups in terms of height, bone age, height standard deviation score for bone age (HtSDSBA), target height (TH), and predicted adult height for bone age (PAHBA) (all Combination therapy with AI and r-hGH can effectively improve the FH of boys in late puberty, but adverse reactions require close monitoring during treatment.
5.TruScreen Combined With High-Risk Human Papillomavirus Testing vs Thinprep Cytology Test Combined With High-Risk Human Papillomavirus Testing for Cervical Cancer Screening:A Comparative Clinical Study

Objective To compare the clinical value of TruScreen,an artificial intelligence-based real-time cervical cancer screening system,combined with high-risk human papillomavirus(hr-HPV)test and Thinprep cytology test(TCT)combined with hr-HPV test in cervical cancer screening.Methods A total of 297 women undergoing cervical cancer screening at Panzhihua Central Hospital between June 2020 and December 2023 were enrolled.All participants underwent hr-HPV testing,TCT,and TruScreen test.The diagnostic performance of TruScreen combined with hr-HPV test and TCT combined with hr-HPV test for low-grade squamous intraepithelial lesion positive(LSIL+)and high-grade squamous intraepithelial lesion positive(HSIL+)cervical lesions was evaluated.Results Among the 297 enrolled women,pathology analysis identified 128(43.10%)LSI+cases and 67(22.56%)HSIL+cases.Additionally,there were 110(37.04%)HPV16/18 positive cases,177(59.60%)cases with TCT results≥atypical squamous cells of undetermined significance(ASCUS),and 176(59.26%)cases with abnormal TruScreen results.The area under the curve(AUC)of TruScreen combined with hr-HPV test in diagnosing LSIL+and HSIL+cervical lesions was higher than that of TCT combined with hr-HPV test(P<0.05).Conclusion TruScreen combined with hr-HPV test demonstrates superior performance in cervical cancer screening compared with TCT combined with hr-HPV test.TruScreen combined with hr-HPV test may serve as an alternative to conventional cytology-based methods for cervical cancer screening in China.
6.Analysis on the Regional Differences and Equity of High Quality Health Resource Allocation in Primary Healthcare Institutions

Objective:To analyze the regional differences and equity of the high-quality health resource allocation in primary healthcare institutions,and provide decision-making references for improving the equity of high-quality health resource allocation in primary healthcare institutions among provinces in China.Methods:The entropy weight method is used to determine the weights of indicators.Breakpoint method,Gini coefficient and grey correlation method are applied to conduct the classification,equity analysis and influencing factor analysis on the allocation levels of 31 provinces.Results:The Gini coefficients of the supply-demand ratio index in 2011,2016 and 2021 were 0.34,0.33 and 0.34,respectively.Population density,education level,education investment level,public service level,per capita GDP,regional income level and average altitude were the key influencing factors.Conclusion:There are significant differences in the high-quality health resource allocation among community-level medical and health care institutions in different provinces of China,and there are certain issues with equity.It is necessary to comprehensively consider multiple influencing factors to promote the balanced development of high-quality health resource allocation in community-level medical and health care institutions in China.
7.Application of α-cyanoacrylate medical glue for mesenteric fissure closure during laparoscopic radical resection of colorectal cancer

Background and Aims:Whether to close mesenteric fissures during laparoscopic radical resection of colorectal cancer remains controversial.Traditional suture closure is technically demanding and may injure mesenteric vessels.This study aimed to evaluate the safety and efficacy of using α-cyanoacrylate medical glue to close mesenteric fissures during laparoscopic colorectal cancer surgery.Methods:A retrospective analysis was conducted on patients who underwent laparoscopic radical resection of colorectal cancer in the Department of Colorectal Surgery,the Sixth Affiliated Hospital of Sun Yat-sen University,from January 2022 to December 2023.Seventy-eight patients who received intraoperative α-cyanoacrylate glue closure of mesenteric fissures were included as the observation group,and 74 patients without fissure closure were selected as the control group using the propensity score matching method.Perioperative parameters,postoperative recovery,and complications were compared between the two groups.Results:No significant differences were observed in baseline characteristics or main intraoperative variables between groups(all P>0.05).The observation group had significantly less ascitic drainage within 3 days after operation[(203.14±116.44)mL vs.(384.53±243.89)mL,P<0.01]and shorter postoperative gas passage,defecation,and drainage tube removal times(all P<0.01).The incidence of postoperative complications and intestinal obstruction was comparable between groups(all P>0.05).Multivariate analysis showed that intraoperative application of α-cyanoacrylate glue was an independent promoting factor for intestinal exhaust within 3 days after surgery(OR=5.739,P=0.000).Conclusion:The use of α-cyanoacrylate medical glue for closing mesenteric fissures during laparoscopic radical resection of colorectal cancer is safe and feasible.It effectively reduces postoperative ascitic drainage and accelerates bowel recovery,offering a simple and reliable alternative to traditional suture closure.
