OBJECTIVE To improve the inventory turnover rate of traditional Chinese medicine (TCM) decoction pieces in hospitals and achieve refined inventory management for cost reduction and efficiency improvement. METHODS Inventory data of TCM decoction pieces in our hospital from 2022 to 2023 were collected. Based on the sale amount and demand coefficient of variation respectively, the ABC classification method and XYZ analysis method were adopted for classification. The two classification results were combined through a cross matrix to divide them into three-level management priorities: level Ⅰ (AX, AY, BX), level Ⅱ (AZ, BY, CX), and level Ⅲ (BZ, CY, CZ). Differentiated inventory strategies were formulated for each level, and core indicators such as turnover rate and inventory cost before and after inventory management optimization were compared. RESULTS There were 58 types (14.22%) in level Ⅰ, 85 types (20.83%) in level Ⅱ, and 265 types (64.95%) in level Ⅲ. After inventory management optimization, the average inventory value of TCM decoction pieces decreased by 341 800 yuan, the average turnover rate increased by 73.03%, and the average turnover days decreased by 4.98 days. The differences in all indicators before and after optimization were statistically significant ( P <0.01). CONCLUSIONS The ABC-XYZ analysis method can break through the single-dimensional limitation of the traditional ABC classification method, adapt to characteristics such as the strong seasonality of TCM decoction pieces, significantly optimize the inventory structure, improve turnover efficiency, and reduce operating costs. It provides a feasible plan for the refined and scientific management of hospital TCM warehouses.

Objective To determine the expression of FOXM1 and PLK1 in colorectal cancer tissues and their relationship with clinicopathological characteristics and prognosis of patients. Methods Sixty patients who underwent surgical resection of colorectal cancer were retrospectively selected. Colorectal cancer tissues and adjacent tissues (>5 cm from the margins of colorectal cancer tissues) were collected. Immunohistochemistry, Western blot, and qRT-PCR analyses were used to detect the expression levels of FOXM1 and PLK1 in colorectal cancer tissues. Human colon cancer HCT-116 cells were treated with FOXM1 inhibitor FDI-6, and the effect of downregulating FOXM1 on PLK1 expression levels was investigated by Western blot and qRT-PCR. Results FOXM1 and PLK1 were highly expressed in the cytoplasm of colorectal cancer cells, and the positive expression rate was significantly higher than those in adjacent tissues (P<0.05). FOXM1 expression was closely related to the degree of differentiation, TNM stage, lymph node metastasis, and invasion depth (all P<0.05). PLK1 expression was closely related to TNM stage, lymph node metastasis, and invasion depth (all P<0.05). The expression levels of FOXM1 and PLK1 in colorectal cancer tissues were positively correlated (rs=0.373, P=0.003). Western blot and qRT-PCR results showed that the expression level of PLK1 decreased significantly after inhibition of FOXM1 expression. Patients with either FOXM1 or PLK1 expression alone, or with neither expressed, had significantly longer survival time and more favorable prognosis than those with FOXM1 and PLK1 co-expression. Conclusion FOXM1 and PLK1 are highly expressed in colorectal cancer tissues. FOXM1 may promote colorectal cancer through PLK1, and its high expression suggests poor prognosis of patients and may be a potential target for colorectal cancer.

Intelligent acupuncture-moxibustion medical equipment is an important force in promoting the inheritance, innovation, and modernization of acupuncture-moxibustion. This paper reviews the development status of intelligent acupuncture-moxibustion medical equipment and related new technologies, as well as the challenges faced. It is found that, with the advancement of technologies such as big data and artificial intelligence, acupuncture-moxibustion medical equipment has shown characteristics of greater precision, miniaturization, intelligence, and portability. However, deficiencies remain in areas such as standardization and regulation, including relatively low rates of effective transformation and a lack of innovation in research outcomes. Therefore, there is an urgent need to formulate corresponding strategies: improving the development of relevant standards for intelligent acupuncture-moxibustion medical equipment, encouraging the integration of medicine and engineering, cultivating interdisciplinary talents, and strengthening the protection of invention patents. It is necessary to establish a demand-oriented pathway connecting "equipment development, equipment evaluation, product formation" through multiple stages such as talent training and research project initiation, thereby promoting the modernization and standardization of intelligent acupuncture-moxibustion medical equipment and supporting the revitalization of traditional medicine.

Objective:To develop a quality control model for anal sphincter ultrasound images and validate its diagnostic performance across multiple centers. Methods:A retrospective analysis was conducted on anal sphincter ultrasound images from seven medical centers in China between May 2019 and June 2022. A total of 7 040 images from 3 116 patients were included and divided into a training set(4 912 images)and a validation set(2 128 images). The images were classified as standard or non-standard images by three experts. Three models were developed based on different image feature extraction methods:a single-branch model,a multi-branch weighted model, and a multi-branch ensemble model. The diagnostic performance of each model was evaluated using the area under the ROC curve(AUC),sensitivity, specificity, accuracy, positive predictive value, and negative predictive value, respectively. The optimal model was selected and compared with the performance of 4 doctors with varying experience levels. Sixty days later, the images with the assistance of the model's output were reassessed by the doctors to evaluate its impact on manual quality control.Results:① Among the 3 models, the multi-branch ensemble model demonstrated the highest AUC and sensitivity, with an AUC of 0.966(95% CI=0.958 - 0.974),a sensitivity of 91.83%,and a specificity of 91.41%. This model was named M quality. ② M quality's AUC was slightly lower than that of Senior A and B(0.966 vs. 0.976,0.976,and P<0.05),its sensitivity was slightly lower than that of Senior A(91.83% vs. 95.61%, P<0.001)but comparable to Senior B(91.83% vs. 92.89%, P=0.315),its specificity was slightly lower than Senior A and B(91.41% vs. 94.44%,98.18%,and P<0.05). However,M quality significantly outperformed Junior A and B in AUC and sensitivity(AUC:0.966 vs. 0.850,0.818;sensitivity:91.83% vs. 84.90%,61.46%;all P<0.001),its specificity was higher than that of Junior A(91.41% vs. 80.28%, P<0.001)but lower than that of Junior B(91.41% vs. 95.96%, P<0.001). ③ With model assistance,Senior B's sensitivity(92.89% vs. 94.20%, P=0.001)and Senior A's specificity(94.44% vs. 96.56%, P<0.001)improved significantly. Junior A and B showed significant improvements in AUC and sensitivity(AUC:0.931 vs. 0.850,0.914 vs. 0.818;sensitivity:91.83% vs. 84.90%,89.53% vs. 61.46%;all P<0.001). After model assistance,Junior A's specificity increased(93.62% vs. 80.28%, P<0.001),while Junior B's specificity decreased(91.60% vs. 95.96%, P=0.013). Conclusions:This study develops a quality control model for anal sphincter ultrasound images with robust diagnostic performance, approaching the level of seniors. The model significantly enhances the image quality assessment capabilities of juniors, demonstrating promising clinical application potential.

Chemotherapy is currently the mainstay of systemic management for triple-negative breast cancer (TNBC), but chemoresistance significantly impacts patient outcomes. Our research indicates that Doxorubicin (Dox)-resistant TNBC cells exhibit increased glycolysis and ATP generation compared to their parental cells, with this metabolic shift contributing to chemoresistance. We discovered that ALKBH3, an m1A demethylase enzyme, is crucial in regulating the enhanced glycolysis in Dox-resistant TNBC cells. Knocking down ALKBH3 reduced ATP generation, glucose consumption, and lactate production, implicating its involvement in mediating glycolysis. Further investigation revealed that aldolase A (ALDOA), a key enzyme in glycolysis, is a downstream target of ALKBH3. ALKBH3 regulates ALDOA mRNA stability through m1A demethylation at the 3'-untranslated region (3'UTR). This methylation negatively affects ALDOA mRNA stability by recruiting the YTHDF2/PAN2-PAN3 complex, leading to mRNA degradation. The ALKBH3/ALDOA axis promotes Dox resistance both in vitro and in vivo. Clinical analysis demonstrated that ALKBH3 and ALDOA are upregulated in breast cancer tissues, and higher expression of these proteins is associated with reduced overall survival in TNBC patients. Our study highlights the role of the ALKBH3/ALDOA axis in contributing to Dox resistance in TNBC cells through regulation of ALDOA mRNA stability and glycolysis.
