Objective:To analyze the expression of myosin light chain phosphatase target subunit-1 (MYPT1), protein kinase C-activated phosphatase inhibitor 17 (CPI17), and protein phosphatase 1 catalytic subunit (PP1C) in the biliary epithelium of children with pancreaticobiliary maljunction (PBM).Methods:A total of 22 children with PBM and biliary dilatation from the Children's Hospital of Soochow University from March 2015 to December 2017 were collected as the PBM group ( n=22), including 8 males and 14 females, with an age of 4.2 (2.8, 6.3) years. 21 patients who underwent cholecystectomy for gallstones at the First Affiliated Hospital of Soochow University were collected as the control group ( n=21), including 9 males and 12 females, with an age of 32.2 (23.8, 48.3) years. The expression of tight junction-related proteins pMLC, CPI17, PP1C, and MYPT1 in the PBM group and the control group was analyzed by immunohistochemical staining combined score. Western blot was used to detect the protein expression of MYPT1, PP1C, MLC, pCPI17, CPI17, and pMYPT1 in the PBM group and the control group. Results:The immunohistochemical staining combined score showed that the expression of pMLC protein in the PBM group was higher than that in the control group, while the expression of MYPT1, CPI17, and PP1C proteins was lower than that in the control group, and the differences were statistically significant (all P<0.05). The Western blot detection results showed that the expression values of pMYPT1 (Thr696), pCPI17 (Thr38), MLC, CPI17, MYPT1, and PP1C proteins in the PBM group were (1.187±0.140), (0.455±0.038), (0.181±0.013), (0.246±0.016), (0.459±0.054), and (0.202±0.032), respectively, while those in the control group were (0.238±0.048), (0.144±0.025), (3.268±0.278), (1.462±0.214), (1.138±0.189), and (0.853±0.120), respectively. The expression of MLC, CPI17, MYPT1, and PP1C proteins in the PBM group was lower than that in the control group, while the expression of pMYPT1 and pCPI17 proteins was higher than that in the control group, and the differences were statistically significant (all P<0.05). Conclusion:The increased expression of pMYPT1 and pCPI17 and the decreased expression of PP1C in children with PBM may be related to the disruption of tight junctions in the biliary epithelium of these children.

Objective To identify the effects of parenteral nutrition (PN) on the gene expressions of rat intestinal inflammatory cytokincs and receptors and to explore the role of these changes in PN-related intestinal impairment. Methods Totally 12 male Sprague-Dawley rats were equally divided into the control group and the PN group. A silastic catheter was inserted into the right jugular vein of each rat. No food or water was administered to the PN group except for a continuous 24-hour PN infusion through the silastic catheter in the jugular vein. The control group, while being regularly fed, was administered with an infusion of normal saline through the silastic catheter in thc jugular vein. After 7 days, intestinal tissues were taken for electron microscopy and real-time PCR array to analyze thc microstructure change in rat intestine and thc gene expressions of inflammatory cytokines and their receptors. Results Electron microscopy revealed atrophy of microvillus, engorgement of mitochondria, cell-cell junction breakage, and several apoptotic bodies in the PN group and normal intestinal microstructure in the control group. Compared with the control group, the PN group showed an up-regulation in the gene expressions of interferon γ, interleukin-1 receptor type I , interlcukin-8 receptor type b and a down-regulation in the gene expressions of CC chemokine ligand 17 (CCL17) , CCL19, CCL21, CCL22, CCL9, CXC chemokine receptor 3, CC chemokine receptor 3 ( CCP3 ), CCR7, CCR5, C-reactive protein, and interleukin-10. Conclusions PN influences the gene expressions of rat intestinal inflammatory cytokincs and receptors. The expression of cytokine interferon γ increases and that of interleukin-10 declines, and the expressions of CCL19, CCL21, CXC chemokine receptor 3, CCR3,CCR7, and CCR5 decline. The alterations of these genes may be associated with the impairment of intestinal immune and mechanical functions.

Objective To investigate the effect of anomalous pancreaticobiliary ductal union on the injuries of pancreas.Methods 10 health cats were treated by a 4～6 cm incision in biliary and pancreatic duct adjacent to duodenum, and interrupted suture was made to induce the model of anomalous pancreaticobiliary ductal union.The pancreatic tissues were harvested during model induction and 6 months after model induction,and histopathological changes were evaluated by electron microscopic examination and the level of maleic dialdehyde (MDA) were determined.Results Seven out of 10 cats survived surgery and remained alive for 6 months.6 months later,the pancreas appeared to be in the color of dark red and hyperemia,edema was present.Dilated pancreatic duct could be found in three of them.Light microscopic examination suggested there was inflammatory cell infiltration in the pancreas in 3 cats;mesenchymal vascular proliferation and white cell adhesion was present in the pancreas in 4 cats.Electron microscopic examination suggested rough endoplasmic reticulum expanding in the shape of pool,swelling mitochondria disaggregated and increased in number,Golgi complex became well developed.The preoperative level of MDA in pancreatic tissue was (1.23±0.7 ) nmol/mg prot,which increased to (2.90±1.9 ) nmol/mg prot 6 months later with statistically significant difference (F=4.80,P=0.0215 ).Conclusions This animal model demonstrated that anomalous pancreaticobiliary ductal union may cause injury to the pancreas,and antioxidant treatment may alleviate this injury.
