1.The expression and function of MST1 in prostate cancer

ObjectiveTo investigate the expression level of mammalian sterile 20-like kinase 1 (MST1) in prostate cancer (PCa) tissues, its biological functions, and its regulatory effects on androgen receptor (AR) and vascular endothelial growth factor (VEGF) expression. MethodsImmunohistochemistry was performed to detect MST1 expression in 77 prostate cancer tissues and 34 benign prostatic hyperplasia (BPH) tissues, and the relationship between MST1 expression and clinicopathological parameters was analyzed. MST1-overexpressing C4-2 and LNCaP cell lines were constructed. CCK-8 assay, Transwell migration assay, and angiogenesis assay were conducted to evaluate the effects of MST1 on cell proliferation, migration, and angiogenesis. The expression changes of AR and VEGF were detected by qRT-PCR and Western blot. ResultsMST1 expression was significantly lower in PCa tissues compared with BPH tissues (P=0.012). Low MST1 expression was significantly associated with tumor metastasis (P=0.033). MST1 overexpression inhibited C4-2 cell proliferation and migration, with transmembrane migrated cells reduced by 53.6% (P<0.01). MST1 overexpression decreased the mRNA and protein expression levels of AR and VEGF (P<0.05). Angiogenesis assay demonstrated that the length of new vessel structure was significantly reduced (P<0.05). ConclusionMST1 shows a low expression level in PCa tissues and is associated with tumor metastasis. MST1 exerts tumor-suppressive effects in prostate cancer by inhibiting cell proliferation, migration, and angiogenesis, as well as downregulating the expression of AR and VEGF, indicating its potential as a therapeutic target for prostate cancer.
2.Advances in research on fine motion control of prosthesis fingers with brain-computer interface

The deficiency of fingers due to various reasons leads to a certain degree of loss of full or part hand functions. Physical and mental health of patients are seriously affected, and patients have varying degrees of reduced quality of life. Prosthesis fingers play an important role in completing the body shape and enhancing patients’ self-confidence and self-esteem. However, how to make prosthesis fingers perform coordinated movements and restore complete functions is a crucial problem that urgently needs to be solved. This paper reviews the methods of brain-computer interface controlled fine finger movements and elaborates on the origin, current situation, and advancements of the development of this technology, laying a foundation for subsequent research, with the expectation of helping patients solve the problems arising from the insufficiency or absence of finger functions.
3.Changes in glucose metabolism and intestinal flora in patients with type 2 diabetes mellitus after high-intensity intermittent exercise

BACKGROUND:Exercise has a regulatory effect on intestinal flora and glucose metabolism,but the effects of high-intensity intermittent exercise on intestinal flora and glucose metabolism in patients with type 2 diabetes mellitus are unclear. OBJECTIVE:To investigate the effects of high-intensity intermittent exercise on glucose metabolism and intestinal flora in patients with type 2 diabetes mellitus. METHODS:Eleven patients with type 2 diabetes mellitus were recruited,among which,two were lost to the follow-up and nine were finally enrolled.High-intensity intermittent exercise intervention was conducted 3 times per week for 6 continuous weeks.Fasting blood and fecal samples were collected before and after the intervention.Glucose metabolism indexes were detected in the blood samples,and intestinal flora was detected in the fecal samples.Changes in glucose metabolism indexes and intestinal flora indexes of the patients with type 2 diabetes mellitus before and after the intervention were compared. RESULTS AND CONCLUSION:After 6 weeks of high-intensity intermittent exercise intervention,fasting blood glucose and glycosylated serum protein levels in patients were significantly reduced(P<0.05),and fasting insulin,although not significantly changed,was decreased compared with before intervention.Alpha diversity analysis showed that the diversity(Shannon index),richness(Chao index)and coverage(Coverage index)did not change significantly.Venn diagrams showed that the relative abundance of Bacteroidetes,Actinobacteria,Proteobacteria,and Fusobacteria in the intestinal flora of the patients increased,and the relative abundance of Firmicutes decreased,and a significant decrease was seen in Ruminococcus_torques and Ruminococcus_gnavus in the Firmicutes,which were both positively correlated with the abnormalities of the glycemic metabolism-related indicators,as well as with other disease development.All these findings indicate that high-intensity intermittent exercise intervention has an improvement effect on the glycemic metabolism-related indexes of patients with type 2 diabetes mellitus,and the abundance of beneficial flora in the intestinal tract increases,and the abundance of harmful flora decreased,enhancing the stability of the intestinal flora in patients.
4.Respiratory syncytial virus vaccine based on bacterial outer membrane vesicle

Objective:To analyze the protective effect of a respiratory syncytial virus (RSV) vaccine based on bacterial outer membrane vesicle (OMV) in mice.Methods:The pre-fusion protein (preF) of RSV was linked to the surface of OMV through the transmembrane protein cytolysin A (ClyA) to form the nanovaccine OMV-preF. The morphological characteristics of OMV and OMV-preF were observed under a transmission electron microscope. OMV-preF was intramuscularly injected into BALB/c mice and the elicited humoral and cellular immune responses were evaluated. The protective effect of OMV-preF was assessed by challenging the immunized mice with RSV Long strain. One-way analysis of variance and Tukey test were used for statistical analysis.Results:The results showed that preF was stably expressed in OMV, and both OMV-preF and OMV exhibited a double-layer vesicle structures under the microscope. OMV-preF could significantly activate the cellular and humoral immune responses in mice, causing a significant increase in CD8 + T cells and CD19 + B cells as well as a significant increase in the serum level of specific IgG. The neutralizing antibodies produced in the immunized mice could significantly inhibit the replication of RSV Long strain in vivo. Conclusions:The nanovaccine OMV-preF can induce high-level humoral and cellular immune responses, and the antibodies produced following immunization can effectively inhibit viral replication. This study provides a new strategy for RSV subunit vaccines.
5.Effects of surgical pleth index and nociception index on the consumption of remifentanil and postoperative recovery in children undergoing adenotonsillectomy

Objective To investigate the appropriate indicators for monitoring pediatric nociceptive stimu-lation,this study compared the SPI and NOX,two dual-parameter nociceptive stimulation monitors based on different principles,in terms of their effects on remifentanil consumption and postoperative recovery in pediatric adenotonsil-lectomy.Methods Children aged 3~12 years who were scheduled to undergo adenotonsillectomy under general anesthesia with endotracheal intubation were randomly assigned to the conventional group(Group R,n=19),the SPI group(Group S,n=19),and the NOX group(Group N,n=18)according to the type of nociceptive stimu-lation monitor used.All children were subjected to routine fasting.The depth of anesthesia was monitored using a BIS monitor,and the remifentanil infusion rate was adjusted according to heart rate,SPI,or NOX values to maintain the index within the range of 30~50.After surgery,all children were transferred to the Post-Anesthesia Care Unit(PACU)with the tracheal catheter in place until they recovered.During the operation,the consumption of anes-thetics such as remifentanil was recorded.Postoperatively,pain and agitation scores,the incidence of agitation at different time points,the duration of anesthesia,the surgical time,the time to extubation,and the length of stay in the recovery room were measured.Additionally,postoperative adverse reactions and perioperative vital signs were documented.Results In comparison with Group R,in Group N,the intraoperative consumption of remifentanil and the agitation score during the recovery period were significantly reduced.Conversely,in Group S,both of(P<0.05).There were no significant disparities in the FLACC score,the incidence of agitation,and the extubation time among the three groups.Conclusions The NOX index can serve as a quantitative metric for monitoring nonci-ceptive stimulation during pediatric adenotonsillectomy.This index has the potential to decrease the intraoperative consumption of opioids and the residence time in the recovery room.

Objective To investigate the clinical characteristics,diagnosis and treatment of ileal injury secondary to percutaneous nephrolithotomy percutaneous nephrolithotomy(PCNL).Methods The diagnosis and treatment of a patient were reviewed,and relevant literature were retrieved.Results The patient was a 41-year-old male,who underwent stage PCNL(initial percutaneous nephrostomy,followed by secondary PCNL)due to right ureteral calculi with severe hydronephrosis.On postoperative day 1,he developed abdominal distension and pain.Abdominal X-ray revealed subdiaphragmatic free gas,and CT showed pelvic and abdominal fluid and gas accumulation,suggesting peritonitis due to intestinal perforation.Emergency exploratory laparotomy identified a 3 mm×3 mm ileal perforation approximately 30 cm from the ileocecal valve,which was repaired surgically.The patient recovered well and was discharged after one week,with no discomfort reported during a 6-month follow-up.Conclusion The clinical features of ileal injury secondary to PCNL include early postoperative abdominal distension,pain and peritonitis.Diagnosis relies on clinical manifestations,abdominal X-ray and CT,with surgical exploration if necessary.Conservative management under vigilant observation can be cautiously adopted for localized injuries,while surgical repair is required for peritonitis or failed conservative therapy.

Ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation, has been recently identified as a key role in Parkinson's disease (PD) pathogenesis. In particular, lipid peroxidation-mediated ferroptosis is considered a key event leading to the death of dopaminergic neurons. This article reviews the role of lipid peroxidation-mediated ferroptosis in PD and its involved key signaling pathways, and explores the related targeted therapeutic strategies, with the aim of providing new ideas for targeted treatment of PD.
