1.Fragaria Indica ethanolic extracts: bioactive compounds identification, ADMET analysis and putative inhibitors of bla-TEM beta-lactamase from foodborne bacterial pathogens

Fragaria indica stem, leaf, and root extract was used to assess the antimicrobial activity against bla-TEM
beta-lactamase producing Shigella flexneri, Salmonella typhi, and Escherichia coli. Ethanolic extracts
of F. indica leaves exhibited a maximum zone of inhibition of 20 ± 1.2 mm (p < 0.001), 18 ± 1.4 mm
(p < 0.001), and 16 ± 1.5 mm (p < 0.001) against E. coli, S. flexneri, and S. typhi, respectively. To identify
different compounds, the bioactive ethanolic leaf extracts were further processed for HPLC, GC-MS,
ADMET, and docking analysis. In the ethanolic leaf extract, 19 phenolic compounds were identified
in HPLC analysis. Three phenolic compounds, including Isorhamnetin-3-rutinoside, 5, 7-Dihydroxy-4’
-methoxyflavone, and Luteolin, were for the first time reported in F. indica. In GC-MS analysis, sixteen
compounds were identified. From the literature search, among the identified compounds, AlphaHimachalene, 1,3-Diphenyl-1-((trimethylsilyl)oxy)-1(Z)-heptene, 1H-Benzocycloheptene, Bicyclo[5.2.0]
nonane, cis-1,1,3,4-Tetramethylcyclopentane, Bicyclo[2.2.2]octane-cis-2,3-diyl Carbonate, BetaHimachalene and 2-methylene-4,8,8-trimethyl-4-vinyl were reported for the first time in F. indica. Two
compounds, including Alpha-Himachalene and Bicyclo[2.2.2]octane-cis-2,3-diyl carbonate, having
good water solubility, drug likeness, and medicinal characteristics, were selected for docking analysis.
Docking of Alpha-Himachalene and Bicyclo[2.2.2]octane-cis-2,3-diyl carbonate showed interaction
with Pro-26, Phe-22, and Ala-23 residues of bacterial bla-TEM beta-lactamase. Similarly, Bicyclo[2.2.2]
octane-cis-2,3-diyl carbonate interacts with amino acid residues of Ala-235 and Ser-233 of bla-TEM
beta-lactamase. Further studies will help to characterize the anti-infective compounds against bla-TEM
producing foodborne bacterial pathogens.
2.Effectiveness of cephalosporins Microbiology in hydrolysis and inhibition of Staphylococcus aureus and Escherichia coli biofilms

Objective:
The study examined the efficacy of various generations of cephalosporins against biofilms developed by pathogenic S. aureus and E. coli.
Methods:
The development of biofilms by both bacteria was assessed using petri-plate and microplate methods. Biofilm hydrolysis and inhibition were tested using first to fourth generations of cephalosporins, and the effects were analyzed by crystal violet staining and phase contrast microscopy.
Results:
Both bacterial strains exhibited well-developed biofilms in petri-plate and microplate assays. Cefradine (first generation) showed 76.78% hydrolysis of S. aureus biofilm, while significant hydrolysis (59.86%) of E. coli biofilm was observed by cefipime (fourth generation). Similarly, cefuroxime, cefadroxil, cefepime, and cefradine caused 78.8%, 71.63%, 70.63%, and 70.51% inhibition of the S. aureus biofilms, respectively. In the case of E. coli, maximum biofilm inhibition (66.47%) was again shown by cefepime. All generations of cephalosporins were more effective against S. aureus than E. coli, which was confirmed by phase contrast microscopy.
Conclusions
and Relevance: Cephalosporins exhibit dual capabilities of hydrolyzing and inhibiting S. aureus and E. coli biofilms. First-generation cephalosporins exhibited the highest inhibitory activity against S. aureus, while the third and fourth generations significantly inhibited E. coli biofilms. This study highlights the importance of tailored antibiotic strategies based on the biofilm characteristics of specific bacterial strains.
4. β-Islet cell regeneration potential of Mirabilis jalapa in hyperglycemic rats

Objective: To investigate the role of Mirabilis jalapa root extracts in restoration of glucose homeostasis in alloxan-induced hyperglycemic Wistar albino rats. Methods: Experimental hyperglycemic rats were treated daily with 200 and 400 mg/kg of Mirabilis jalapa extracts after initial fasting for 6 h. Two-hour postprandial glucose and changes in body weight were monitored during treatment. After 14 d, the rats were sacrificed and blood was collected for biochemical assessment of serum glucose and insulin levels, lipid profile, and oxidative stress markers. Histopathological examinations of harvested pancreas were also carried out. Results: Mirabilis jalapa root extracts at 200 and 400 mg/kg increased the body weight of hyperglycemic rats. Postprandial glucose levels of the extract-Treated hyperglycemic groups progressively declined during treatment compared with the untreated hyperglycemic control group (P<0.05). The lipid profile indices of the untreated negative control group were significantly elevated (P<0.05), which were reversed by treatment with Mirabilis jalapa extracts. The remarkable increases in antioxidant enzyme activities and a significant decrease in malondialdehyde levels were observed in the hyperglycemic group treated with Mirabilis jalapa extracts. Mirabilis jalapa extracts also significantly increased serum insulin levels (P<0.05). In addition, histopathological examinations of the pancreas revealed a significant cell population within the islet nests of the extract-Treated hyperglycemic groups. Conclusions: Mirabilis jalapa extract can restore glucose homeostasis and show hypoglycemic and hypolipidemic effects in hyperglycemic rats. Further studies are needed to verify the active components of the plant and the underlying mechanism of action in the future.
5.Arterial Spin Labelling-Based Blood-Brain Barrier Assessment and Its Applications

The brain relies on the blood-brain barrier (BBB) for the selective absorption of nutrients and the exclusion of other big molecules from the circulating blood. Therefore, the integrity of BBB is critical to brain health, and assessing BBB condition is of great clinical importance. BBB is often examined using exogenous tracers that can travel across the BBB, but the tracers might cause severe side effects. To avoid the use of external tracers, researchers have used magnetically labeled arterial blood as the endogenous tracer to assess the water permeability of BBB as a surrogate index of BBB. This paper reviews the three major types of arterial spin labelling-based BBB water permeability assessment techniques and their applications in brain diseases such as Alzheimer’s disease.
6.Gastrointestinal parasitic infections of buffaloes (Bubalus bubalis) in Sarawak Borneo: Prevalence, risk factors, and farming practices

The objectives of this study were to investigate the prevalence and associated risk factors for gastrointestinal (GI) parasites in buffaloes from various areas of Sarawak, and to assess current management practices of GI parasites among farmers. Faecal samples were collected from 15 farms and 129 animals, as well as data on farm and animal-based characteristics. A total of 129 faecal samples were examined for GI parasites using a modified McMaster and sedimentation. Association between potential risk factors and the prevalence of GI parasites was investigated using Chi-square statistic. The prevalence of Paramphistomum sp., strongyles, and coccidia were 75.2% (95% CI±7.5), 52.7% (95% CI±8.6) and 48.1% (95% CI±8.6), respectively. Farms which had a grazing area less than 50 acres in size had significantly higher prevalence of strongyles (70.5%, χ² = 8.34, P = 0.004) and paramphistomes (88.6%, χ² = 6.46, P = 0.01) relative to farms with a larger grazing area (43.5% and 68.2%, respectively). Prevalence of strongyles was lower in farms that did not implement a cut- and-carry system (45.6%, χ² = 4.17, P = 0.04) in comparison to those that did (64%). The prevalence of paramphistomes was higher on farms with more than 40 animals (80.6%, χ² = 3.18, P = 0.05) relative to farms with fewer animals. The majority of farmers surveyed (67.9%) showed awareness of GI parasite infection and reported that they recognized the associated symptoms. Most farmers practised deworming, and ivermectin was the most commonly used anthelminthic (60.4%); only 1.9% of farmers used albendazole. Overall this study revealed a high prevalence of GI parasites in buffalo in Sarawak. Although farmers report they are aware of parasitic diseases, further education is still required. This could include how they can successfully implement on-farm changes to reduce the prevalence of GI parasites in their herds.
7.A Review and Description of Acetabular Impaction Bone Grafting: Updating the Traditional Technique

Restoring acetabular bone loss in revision hip arthroplasty is a major challenge for the orthopaedic surgeon. This paper discusses the traditional cemented technique of impaction bone grafting as applied to the acetabulum, as well as the evolution of the technique to employ uncemented implants. Some of the recent published literature regarding these techniques is reviewed and the personal experiences of the senior author with these techniques are also reported.

9.Prevalence of Toxoplasma gondii Antibodies in Stray Dogs from Various Locations in West and East Malaysia

Toxoplasmosis is caused by an obligate intracellular protozoan parasite; Toxoplasma gondii, which is one of the most important zoonotic parasite worldwide. In dogs, the sexual reproductive cycle of T. gondii is lacking, and the animals are not widely consumed as food, but they are vital in the mechanical transmission of the parasite. However, there is no present data on the exposure of stray dogs to T. gondii in Malaysia. The objective of this serological survey was to determine the prevalence of T. gondii antibodies (IgG) and associated factors in stray dogs in East and West Malaysia. Antibodies to T. gondii were determined in serum samples from 222 stray dogs from 6 different states in East and West Malaysia (Peninsular Malaysia) using an Indirect ELISA. The seroprevalence for T. gondii was 23.4% (Confidence interval: CI 17.8-29.2%). Stray dogs from Selangor and Kuala Lumpur had the highest seroprevalence (32.4%; CI 13.2-45.5%) and lowest in those from Penang and Kedah (12.5%; CI 1.3-23.5%). Gender and breed were not associated with T. gondii seropositivity. However, adult dogs were more likely to be seropositive for T. gondii (OR=2.89; CI 1.1-7.7) compared with younger dogs. These results revealed that T. gondii is prevalent in stray dogs in the studied areas in Malaysia, and indicative of the level of environmental contamination of this parasite especially in urban areas.
10.Antiviral activities of Cholistani plants against common poultry viruses

Herbal medicines are becoming more popular and acceptable day by day due to
their effectiveness, limited side effects, and cost-effectiveness. Cholistani plants are reported
as a rich source of antibacterial, antifungal, antiprotozoal, antioxidant, and anticancer agents.
The current study has evaluated antiviral potential of selected Cholistani plants. The whole
plants were collected, ground and used in extract formation with n-hexane, ethyl acetate and
n-butanol. All the extracts were concentrated by using a rotary evaporator and concentrate
was finally dissolved in an appropriate vol of the same solvent. All of the extracts were tested
for their antiviral potential by using 9-11 days old chick embryonated eggs. Each extract was
tested against the Avian Influenza virus H9N2 strain (AIV), New Castle Disease virus Lasoota
strain (NDV), Infectious bronchitis virus (IBV) and an Infectious bursal disease virus (IBDV).
Hemagglutination test (HA) and Indirect Hemagglutination (IHA) tests were performed for
different viruses. The overall order of the antiviral potential of Cholistani plants against
viruses was NDV>IBV>IBDV>AIV. In terms of antiviral activity from extracts, the order of
activity was n-butanol>ethyl acetate>n-hexane. The medicinal plants Achyranthes aspera,
Neuroda procumbens, Panicum antidotale, Ochthochloa compressa and Suaeda fruticose
were very effective against all four poultry viruses through their extracts. The low IC50
values of these extracts confirm the high antiviral potential against these viruses. It is worth
to mention that Achyranthes aspera was found positive against IBDV through all its extracts
which overcome the problem of unavailability of any known drug against IBDV. In short, the
study proved that Cholistani plants are rich source of antiviral agent and their extracts can be
used as good source of antiviral drugs both in crude and in purified form.
