1.Experience and enlightenment of safety reporting and learning system for radiotherapy

Radiotherapy,as one of the important means of treating cancer,is more prone to medical errors than other treatment methods due to its involvement of a wide variety of equipment,high demands for personnel collaboration,and relatively complex treatment processes.Therefore,the establishment of an effective reporting and learning system for adverse events of radiotherapy is of great significance for ensuring patients'safety and improving healthcare quality.The International Atomic Energy Agency(IAEA)has established the Safety Reporting And Learning System For Radiotherapy(SAFRON),which constructed comprehensive database and formulated standard procedures of reporting event and analytical methods through collected and analyzed the worldwide cases of adverse event of radiotherapy.At the same time,the SAFRON system emphasizes the cultivation for safety culture,and encourages healthcare professionals to actively report adverse events,and creates a non-punitive learning environment to learn from mistakes and prevent the recurrence of similar incidents.This review introduced the structure and usage method of reporting and learning system of adverse event of SAFRON radiotherapy,so as to understand the using processes,system architecture and security culture of that,which can provide references for the establishment and use of reporting and learning system of adverse event of department,and promote continuous improvement of healthcare quality.
2.Effects of hsa_circ_0019217 on the biological functions of human trophoblast cells and its mechanism in pre-eclampsia

Objectives:To investigate the effect of circular RNA (circRNA) hsa_circ_0019217 on the biological function of chorionic trophoblast cell line (HTR8/SVneo) and its mechanism in the occurrence of pre-eclampsia (PE).Methods:The circRNA expression database was used to analyze the differentially expressed circRNA in placental tissues of PE women. The expression levels of hsa_circ_0019217 and miR-526b-5p were detected by reverse transcription real-time fluorescent quantitative PCR (RT-qPCR), and the subcellular localization of hsa_circ_0019217 was detected by fluorescence in situ hybridization. The proliferation, migration and invasion of trophoblast cells were detected by cell counting kit-8 (CCK-8) assay and transwell assay. Western blot and enzyme-linked immunosorbent assay (ELISA) were used to verify the effects of hsa_circ_0019217, miR-526b-5p and decorin (DCN) on matrix metalloproteinase 2 (MMP2), tissue inhibitor of metalloproteinase 2 (TIMP2), placental growth factor (PlGF) and human chorionic gonadotropin (hCG) protein expression levels. Dual luciferase reporter gene assay and RNA immunoprecipitation (RIP) assay were used to verify the interaction between hsa_circ_0019217, miR-526-5p and DCN.Results:(1) The analysis of circRNA expression database showed that the expression level of hsa_circ_0019217 was significantly increased in the placental tissues of PE women (fold change=67, P<0.05), and it was mainly located in the cytoplasm. (2) Knockdown of hsa_circ_0019217 promoted the proliferation, migration and invasion of HTR8/SVneo cells, increased the expression levels of MMP2 and PlGF, and decreased the expression levels of TIMP2 and hCG in HTR8/SVneo cells. (3) Hsa_circ_0019217 targeted adsorption of miR-526b-5p, and inhibition of miR-526b-5p reduced the proliferation, migration and invasion of HTR8/SVneo cells. The expression levels of MMP2 and PlGF in HTR8/SVneo cells were increased, and the expression levels of TIMP2 and hCG were decreased. Hsa_circ_0019217 knockdown and inhibiting miR-526b-5p could reverse the effect of hsa_circ_0019217 knockdown on promoting the proliferation, migration and invasion of HTR8/SVneo cells, and reversed the effect of hsa_circ_0019217 knockdown on the protein expression levels of MMP2, PlGF, TIMP2 and hCG. (4) miR-526b-5p targeted DCN in HTR8/SVneo cells, hsa_circ_0019217 knockdown reduced the expression level of DCN, and inhibiting miR-526b-5p increased the expression level of DCN. When hsa_circ_0019217 and miR-526b-5p were inhibited simultaneously, there was no significant change in the protein expression level of DCN. (5) Overexpression of miR-526b-5p promoted the proliferation, migration and invasion of HTR8/SVneo cells, while overexpression of DCN inhibited the proliferation, migration and invasion of HTR8/SVneo cells. Simultaneous overexpression of miR-526b-5p and DCN reversed the effects of miR-526b-5p overexpression on cell proliferation, migration and invasion. Overexpression of miR-526b-5p increased the expression levels of MMP2 and PlGF and decreased the expression levels of TIMP2 and hCG in HTR8/SVneo cells. Overexpression of DCN reduced the expression levels of MMP2 and PlGF and increased the expression levels of TIMP2 and hCG. Overexpression of miR-526b-5p and DCN reversed the effects of miR-526b-5p on the expression of these proteins. Conclusion:Hsa_circ_0019217 regulates the expression of DCN by adsorption of miR-526b-5p, thereby affecting the proliferation, migration and invasion of trophoblast cells and regulating the protein expression levels of MMP2, TIMP2, PlGF and hCG, which might be used as a target for early prevention of PE.

5.Study on the fracture resistance of preparation designs of occlusal veneer and fiber posts in pulp-penetrating wedge-shaped defects

Objective :
To investigate the effects of different occlusal veneer preparation designs and fiber posts on the fracture load and failure mode of endodontically treated maxillary premolars with pulp-piercing wedge-shaped defect.
Methods :
60 maxillary first premolars were randomly divided into group A and group B after Root Canal Therapy (RCT) and severe defects(n = 30) .Group A was filled with glass fiber post and resin core,and group B was filled with resin core only.Then the occlusal veneer( G2) ,buccal-occlusal veneer( G3) and buccal-proximal-occlusal veneer ( G4 ) were applied to the isolated teeth.The teeth filled with composite resin ( G1 ) and full crown ( G5) were set as negative and positive control respectively,with 6 samples in each group.The sample was subjected to 5 000 hot and cold cycles and 20 000 vertical compression load aging.Finally,all the samples were loaded with static load until the sample broke at the speed of 0.5mm / min.The fracture load and failure mode of each sample were recorded.
Results :
Both the fracture load in group A and B was G5 >G3 >G2 >G4 >G1,and the fracture load in G2,G3,G4,G5 was significantly higher than that in G1 (P<0. 05) ,and there was no significant difference between group A and B (P>0. 05) .The fracture form of G3 was mostly unbonding of the repair body,which was conducive to re-repair.
Conclusion
Buccal ( occlusal) veneer is the most effective in repairing pulp-piercing wedge-shaped defects.The fiber post could not significantly enhance the fracture load of the pulp-piercing wedge- shaped defects teeth.
6.Effect of Er: YAG laser application on the root canal smear layer removal and dentin microhardness

Objective :
To explore the effects of Er: YAG laser irrigation technology on smear layer removal and
dentin microhardness.
Methods :
A total of 102 ex vivo premolars were selected to amputate the crown , leaving a
root length of 13 mm , after mechanical preparation , there were divided into experimental group (3% sodium hypochlorite , 3% NaOCl) and control group (0. 9% sodium chloride solution , NS) , and received syringe , ultrasound and Er: YAG laser terminal irrigation , which were specifically grouped into NaOCl + syringe , NaOCl + ultrasound , NaOCl + laser, NS + syringe , NS + ultrasound and NS + laser groups ( n = 17 ) . Scanning electron microscopy (SEM) evaluated smear layer removal from the upper, middle and apical parts of the root. The Vickers microhardness tester measured the microhardness before and after irrigation.
Results :
The NaOCl + laser group had the best effect on removing the root canal smear layer, and the amplitude of reducing the microhardness of dentin was the largest. There was no significant difference in the reduction of microhardness compared with the NaOCl + ultrasonic group (P > 0. 05) .
Conclusion
Er: YAG laser⁃activated NaOCl solution improves the clearance of the smear layer
and reduced the microhardness of dentin.
7.User experience of Nursing-Online Practice Promotion Application

Objective:To understand the user experience of Nursing-Online Practice Promotion Application ("N-OPP" APP) .Methods:This study was a cross-sectional study. Using the convenient sampling method, a total of 537 nursing students who had practiced in the Affiliated Hospital of Qingdao University from August to November 2020 and had used "N-OPP" APP were selected as the research objects. The general data questionnaire and the "N-OPP" APP user experience evaluation questionnaire were used for the investigation. A total of 537 questionnaires were distributed in this study and 529 valid questionnaires were recovered, with an effective recovery rate of 98.51%.Results:The total average score of the "N-OPP" APP user experience evaluation questionnaire items of 529 nursing students was (3.89±0.47) , and the average scores of items of operation performance, service performance, user experience and risk were respectively (3.87±0.65) , (4.04±0.60) , (3.90±0.70) and (3.42±0.62) . There were statistically significant differences in the use experience and operation performance dimensions of nursing students from different educational backgrounds, contact time of mobile learning APP, use time and use frequency of "N-OPP" APP ( P<0.05) . The service performance dimension items of nursing students with different educational background and frequency of "N-OPP" APP were compared, and the differences were statistically significant ( P<0.01) . The risk dimension items of nursing students with different "N-OPP" APP use frequencies were compared, and the difference was statistically significant ( P<0.01) . Conclusions:The development of "N-OPP" APP can solve some problems in the traditional teaching and training of clinical nursing skills, help nursing students to consolidate their nursing skills and obtain high recognition from nursing students, which can be used as an auxiliary tool for teaching and training of clinical nursing skills.
8.Proton Magnetic Resonance Spectroscopy Study of Normal Neonatal Brain Development

Objective Single voxel proton spectroscopy technique (1H-MRS)was performed in the healthy newborn brain tissue to investigate the metabolic ratio in the right basal ganglia and the right frontal lobe changes with gestational age, and to improve the understanding of the neonatal brain development. Methods In this study, 69 newborns were selected, including twenty-five premature and forty-four term neonates. By using the Gesell developmental scale to evaluate brain development , they all had normal neurologic outcomes at 12-months corrected age. A point-resolved spectroscopy sequence was used to study the metabolic data obtained from each region of interest. Results N-acetylaspartate: creatine and N-acetylaspartate:choline were positively correlated with gestational age (the right basal ganglia r = 0.27, 0.63, the right frontal lobe r = 0.44, 0.64 respectively, P < 0.05) and choline:creatine was negative (the right basal ganglia r = -0.51,the right frontal lobe r = -0.41, P < 0.05), with a linear regression relationship among variables. In the same gestational age group , the N-acetylaspartate: choline of the right basal ganglia was higher than that of the right frontal lobe , and the metabolic ratios among the anatomic locations were significantly different (P < 0.05). Conclusion Neonatal brain metabolite levels are associated with gestational age , and MRS can monitor newborn brain maturation quantitatively.
9.Case-control studies of the relevant factors among Ningxia Hui and Han prostate cancer groups.

OBJECTIVETo explore the relationship between the relevant factors and prostate cancer among Hui and Han populations.
METHODSThe study involved 267 prostate cancer patients as cases (214 cases from Han population and 53 cases from Hui population) and 534 prostatic hyperplasia patients as controls (428 cases from Han population and 106 cases from Hui population). All the patients were collected from the General Hospital of Ningxia Medical University during January of 2007 to September of 2013. The level of fasting blood glucose (FBG), total cholesterol (TC), triglyceride (TG), total prostate specific antigen (T-PSA), free prostate specific antigen (F-PSA) and free/total prostate specific antigen (F/T-PSA) were collected from the clinical medical records of the patients. Data were analyzed by the conditional logistic regression method, and attributable risk proportion(ARP) was calculated.
RESULTSIn Hui population, the risk of prostate cancer for drinkers was 20.48 times higher than the non-drinkers (35.8% (19/53) to 5.7% (6/106), OR = 20.48, 95% CI: 4.95-84.66). The high level of F-PSA significantly increased the risk of prostate cancer for Hui group (83.0% (44/53) to 55.7% (59/106), OR = 4.27, 95%CI: 1.18-15.43). In contrast, the high TG level decreased the risk of prostate cancer for Hui group (18.9% (10/53) to 20.8% (22/106), OR = 0.24, 95% CI: 0.07-0.83). In Han population, the risk of prostate cancer for smokers was 1.89 times higher than the non-smokers (55.1% (118/214) to 39.7% (170/428), OR = 1.89, 95% CI: 1.28-2.78). Either high level of T-PSA or F-PSA increased the risk of prostate cancer for Han group (86.4% (185/214) to 53.7% (230/428),OR = 2.34, 95%CI:1.22-4.52;85.5% (183/214) to 56.1% (240/428), OR = 2.43, 95% CI:1.29-4.59). However, the high TG level or high ratio of F/T-PSA decreased the risk of prostate cancer (15.4% (33/214) to 18.7% (80/428), OR = 0.59, 95% CI:0.36-0.98; 53.3% (114/214) to 73.4% (314/428), OR = 0.53, 95% CI:0.36-0.78). The APRs of drinking and high level of F-PSA in Han populations were 66.6%, 62.4% in Hui populations , and the APRs of smoking and high level of T-PSA, high level of F-PSA were 33.8%, 71.3%, 67.3% in Han populations.
CONCLUSIONBoth drinking and high level of F-PSA might be the risky factors of prostate cancer while the high TG level might be protective factor for Hui group. However, for Han population, smoking, high T-PSA level, and high F-PSA level might be risky factors for prostate cancer while the high TG level and high ratio of F/T-PSA might be protective factors. In summary, the clinical relevant factors of prostate cancer may play different roles between Hui and Han populations in Ningxia region.
