1.Application of virtual simulation experiment in the teaching of Laboratory Sciences for Virus Infection Testing

Objective:To investigate the application value of virtual simulation experiment in the teaching of Laboratory Sciences for Virus Infection Testing.Methods:Undergraduate students(n=58)majoring in Health Inspection and Quarantine of grade 2020 were selected as the research objects.The"Virtual simulation experiment of influenza virus isolation and identification in BSL-2 laboratory"was included in the teaching of Laboratory Sciences for Virus Infection Testing.The application effect of the virtual simulation experiment was evaluated by a self-designed questionnaire.Results:A total of 50 students completed the questionnaire effectively.After completing the virtual simulation experiment,96.0%of the students thought that the simulation degree of the experiment was high.Students could understand the instrument composition,internal structure,operation process,and matters needing attention.And 52.0%of the students thought that the experiment had an impact on autonomous learning.Students believed that the experiment could improve their interest in the course(88.0%),increase their mastery of theoretical knowledge(60.0%),and improve their ability to analyze and solve problems(46.0%).And 72.0%of the students thought that the running speed of the virtual simulation experiment was slow.The top three advantages of the experiment evaluated by students were easy operation,no damage to the instrument and no worry about the dangers in the experiment,and helpful to understand the knowledge.The top three disadvantages of this experiment were no improvement in practical ability,no familiarity with the use of instruments,different feelings from the real experiment and too idealized experimental process.And 66.0%of the students thought that virtual simulation experiments should complement traditional experimental teaching.Conclusions:Virtual simulation experiment can carry out dangerous experiments that are difficult to perform in traditional laboratories.It can improve students'mastery of theoretical knowledge,enhance their interest in learning,and improve their ability to analyze and solve problems.However,there are still some problems in its application,which should be combined with traditional experimental teaching to improve the teaching effect.
2.Early diagnosis of acute kidney injury after cardiac surgery and prediction of noninvasive renal blood flow and oxygen metabolism monitoring

Acute kidney injury after cardiac surgery(CSA-AKI) is the most common complication after cardiac surgery. Monitoring, early diagnosis of CSA-AKI and timely intervention are of the great significance to reduce the incidence of CSA-AKI and improve the prognosis of patients. This article mainly reviews the diagnostic criteria of CSA-AKI, new biomarkers of renal injury and the application value of non-invasive renal blood flow and oxygen metabolism monitoring in the early diagnosis of CSA-AKI, in order to provide a basis for early clinical prediction and diagnosis of CSA-AKI through the understanding of early diagnosis and related non-invasive monitoring of CSA-AKI, so as to carry out effective intervention and improve the prognosis of patients undergoing cardiac surgery.
3.Emergency laboratory construction and reflection in infectious hospital under the epidemic situation

In the process of dealing with epidemic situation, medical laboratories have played an important role in disease diagnosis, treatment, efficacy evaluation and prognosis judgment and so on. Beijing Youan Hospital, as the designated hospital of the coronavirus disease 2019 (COVID-19), has set up an emergency laboratory in the capital for the prevention and control of the COVID-19 by carrying out routine tests and virus nucleic acid tests, it provides timely and effective evidence for clinical diagnosis and treatment. To provide ideas and references for the building of the Emergency Laboratory in infectious hospitals. This article discuss how to set up an independent emergency laboratory efficiently, strengthen the cooperation with the Center for Disease Control and Prevention, make the best use of all resources, and share the enlightenment gained in the process of construction.

5.Effect of acupuncture at Renying (ST 9) on blood pressure in patients with ischemic stroke complicated with essential hypertension.

OBJECTIVE:
To observe the effect of acupuncture at Renying (ST 9) on morning blood pressure, daytime peak blood pressure and 24-hour blood pressure load in patients with ischemic stroke complicated with essential hypertension.
METHODS:
A total of 80 patients (3 cases dropped off) were randomized into an observation group (39 cases) and a control group (38 cases). acupuncture and nifedipine were given in the control group. On the basis of treatment in the control group, acupuncture at Renying (ST 9) was applied in the observation group, once a day, 6 times a week for 4 weeks. The changes of morning blood pressure, daytime peak blood pressure and blood pressure load were observed before and after treatment in the two groups.
RESULTS:
Compared before treatment, morning blood pressure, daytime peak blood pressure and blood pressure load after treatment were reduced in the two groups (all <0.05). The change of morning systolic pressure in the observation group was not significant as compared with that in the control group (>0.05); the changes of morning diastolic pressure, daytime peak blood pressure and blood pressure load in the observation group were larger than those in the control group (all <0.05).
CONCLUSION
On the basis of acupuncture and nifedipine, acupuncture at Renying (ST 9) can effectively reduce morning blood pressure, daytime peak blood pressure and blood pressure load in patients with ischemic stroke complicated with essential hypertension.

6. Application of micro-mirror in microsurgical clipping to the intracranial aneurysm

Objective:
To explore the value and disadvantage of micro-mirror in the intracranial aneurysm surgery.
Methods:
Micro-mirror was used to assist microsurgical clipping to 36 intracranial aneurysms in 31 cases, of which 3 were carotid-ophthalmic artery aneurysms, 3 anterior choroidal artery aneurysm, 11 were posterior communicating artery aneurysms, 7 were middle cerebral artery aneurysms, 10 anterior communicating artery or anterior cerebral artery aneurysms, and the others were a posterior cerebral artery aneurysm and a posterior inferior cerebellar artery aneurysm. The micro-mirror was used before and after clipping to observe the anatomic features of necks hidden behind and medial to aneurysms, to visualize surrounding neurovascular structures, and to verify the optimal clipping position. Intraoperative indocyanine green fluorescein angiography confirmed the success of sufficient clipping.
Results:
All aneurysms were clipped successfully. The parent arteries were occluded temporarily in 26 cases, and 9 aneurysms ruptured during the operation. Postoperative follow-up lasted from 2 weeks to 1 year. After operations digital subtraction angiography (DSA) were repeated in 10 cases, computed tomography angiography(CTA) in 21 cases, and no insufficient clipping or occlusion of parent arteries were revealed.
Conclusions
Micro-mirror assisting microsurgical clipping to the intracranial aneurysm is conducive to accurate clipping, and is a convenient and practical operation.
7.Association between dietary microelement intake and metabolic syndrome among elderly males and females in Zhejiang province

Objective To explore the relationship between microelement intake and metabolic syndrome (MS) in elderly males and females in Zhejiang province. Methods Non-parametric Kruskal-Wallis tests were used to compare the intake of microelements by sex among different groups according to the diagnostic criteria of MS in 780 people(404 male,376 female).Logistic regression was used to explore the association between microelements and MS and its components.Results The percentages of inadequate intake of Mn,Zn and Se were relatively high among elderly people of Zhejiang province(males:30.45%,72.52%,75.74%;females:47.34%,33.78%,80.59%,respectively).Microelement intake in males were higher than females(P<0.05).The intake of Fe and Se were higher in the abdominal obesity group than the non-abdominal obesity group among elderly males,the intakes of Fe,Zn,Se in hypertension group were lower, and Se intake was higher in hyperglycemia and hypertriglyceridemia groups. Furthermore, enough intake of Fe(odds ratio OR,0.41;95% confidence interval CI:0.19,0.87)and Se(OR,0.30;95% CI:0.14, 0.63)decreased the risk of hypertension,and high Cu(OR,2.35;95% CI:1.18,4.71)intake increased the risk of hyperglycemia.Among females,Zn intake in the MS group was lower than in the non-MS group;the intakes of Cu, Zn and Se in the hypertension group were lower; the intakes of Fe, Mn and Zn in the hyperglycemia group were lower as well;and Se intake was lower in the low HDL-C group.Again,the high intake of Zn (OR, 3.21; 95% CI: 1.36, 7.59) and Se (OR, 2.79; 95% CI: 1.24, 6.27) increased the risk of abdominal obesity, but moderate intake of Cu (OR, 0.37; 95% CI: 0.19, 0.72) had a protective effect on hypertension. Conclusions The percentages of inadequate microelement intake were relatively high in elderly people of Zhejiang province. There is a relationship between dietary microelements and MS or its components. It is necessary to guide elderly people to adopt reasonable diet by referring to the Chinese dietary reference intake and Dietay Guidelines in order to improve the situation of microelements intake and promote health.
8.Interleukin 1β, 6 and 8 levels around miniscrews during orthodontic tooth movement

40 patients,20 males and 20 females,with 52 miniscrews were included.The concentration of IL-1β,IL-6 and IL-8 in the gingival crevicular fluid around the miniscrews were measured.The results showed that the concentrations of IL-1β,IL-6 and IL-8 increased during orthodintic tooth movement(P <0.05),indicating that orthodontic force had an effect on the cytokines around the miniscrews,which may be related to orthodontic bone resorption.

Objective To evaluate the effect of conversion from mycophenolic acid (MPA) to mizoribine (MZR) in renal transplant recipients with gastrointestinal tract (GI) symptoms.Methods A total of 355 renal transplant recipients with GI symptoms caused by MPA administration were enrolled from April 2015 to March 2017 in 25 different renal transplant centers in China.The symptomatic improvement of GI before (baseline) and after conversion to MZR (1,2,4 weeks) was assessed by each item of GI symptoms indication.In addition,the efficacy and safety of the conversion therapy during 12 months were determined.Results Patients showed improvement in GI symptoms including diarrhea,abdominal pain,abdominal distention and stomachache after conversion to MZR 1,2,4 weeks (P<0.05).In patients with different severity of diarrhea,conversion to MZR therapy significantly improved diarrhea (P<0.05).During 12 months,no patient experienced clinical immune rejection.We did not observe any infections,leucopenia and other serious side effects.Conclusion MZR could markedly improve GI symptoms caused by MPA administration in renal transplant recipients.
10.The application of the cloning and expression of SMCY gender-specific fusion antigens with the corresponding polyclonal antibodies

Objective with the advantages of rapidity in detection protein, We selected the gender-specific amino acid sequence based on human SMCY and SMCX, cloned and expressed SMCY gender-specific fusion antigens. The rabbits were immunized with purified antigens to obtain the polyclonal antibodies. A new method was established for rapidly sex identification of forensic evidence samples by detecting SMCY antigens with the corresponding polyclonal antibodies. Methods We found three differential fragments by analyzing the sequence of human SMCY and SMCX. Then we cloned this three fragments and ligated as a new recombinant.This SMCY gender-specific fusion antigen gene was sub-cloned into pET-28a and expressed in Escherichia coli. The fusion antigen was purified by Ni-NTA column. The rabbits were immunized with purified antigen to produce the specific polyclonal antibodies.The reactivity of the polyclonal antibody was evaluated by ELISA and Western blotting. We developed a colloidal gold test strip for detecting the gender of human samples. Results We successfully selected gender-specific amino acid sequence, the SMCY gender-specific fusion antigen was expressed by prokaryotic expression and the polyclonal antibody was prepared by immunizing rabbit. The results of colloidal gold strip tests showed that there is a significant difference between male and female serums. Conclusion The results showed that the SMCY gender-specific fusion antigen could be recognized by the polyclonal antibody.The colloidal gold strip tests made by SMCY gender-specific fusion antigens and the corresponding polyclonal antibodies could be used for rapidly determining the gender of forensic evidence samples.
