Abstract： Objective　To understand the new HIV-1 infection in MSM (men who have sex with men) sentinel population in Haikou and explore the influencing factors of HIV/AIDS epidemic, so as to provide a basis for AIDS prevention and control. Methods　A continuous cross-sectional study was used to detect HIV-1 infection in the MSM sentinel surveillance population in Haikou City from 2019 to 2023, excluding those with known past positive results and those who had been on antiretroviral therapy for more than six months. The positive cases suitable for new infection testing were examined by HIV-1 restriction antigen affinity enzyme immunization method, and binary non-conditional logistic regression model conducted multivariate analysis of new infections. Results　A total of 2 001 MSM were recruited, with a median age M（P25, P75）of 28 (23, 33) years, 89.8% were recruited from the Internet, 90.8% were single; 79.7% resided in the local area for >2 years, 72.6% were educated to college level or above, and the proportion of household registration in the province was 78.0%. From 2019 to 2023, the HIV-1 positive rate among MSM in Haikou from 2019 to 2023 was 6.85% (137/2 001), with a new infection rate of 2.95% (59/2 001). The estimated new infection rate was 8.61% (95%CI: 6.41-10.81), with no significant trend changes in HIV-1 positivity and new infection rates over the years (P>0.05). Multivariate analysis revealed that, compared to those who lived locally for ≤2 years, those residing locally for >2 years had a lower probability of recent HIV-1 infection (OR=0.473，95%CI: 0.273-0.821). Compared to those who had never received intervention services, those who had received such services had a lower risk of recent HIV-1 infection (OR=0.347, 95%CI:0.197-0.613). Conclusions　In recent years, the Haikou MSM population has shown characteristics of higher education and younger age, and the rate of new HIV-1 infections has not changed significantly. Receiving intervention services is the protection factor to prevent HIV-1 new infections, whereas the mobile MSM population has a higher risk of HIV-1 new infection. Health management of the mobile MSM population should be strengthened to expand the coverage of AIDS intervention services.

This study was designed to explore the effect of DNA methyltransferase 1(DNMT1)on podocyte apoptosis and inflammatory cytokine release induced by high glucose(HG),and analyze the related molecular mechanisms.Podocyte MPC-5 cells were cultured in vitro and divided into control and HG groups.DNMT1 and SOCS1 were either silenced or overexpressed using small RNA interference technology and liposome transfection technology.The expression levels of DNMT1 and SOCS1 genes were measured using qRT-PCR.Apoptosis was assessed by flow cytometry,while ELISA was employed to determine the levels of inflammatory factors such as tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-1β(IL-1β),and monocyte chemoattractant protein-1(MCP-1).Western blot was used to detect the expression of DNMT1,SOCS1 proteins,and the proteins involved in the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathway.Data showed that HG elevated MPC-5 cell apoptosis rate,the level of inflammatory factors and DNMT1 mRNA expression,and the expression of DNMT1,p-JAK2 and p-STAT3 proteins,while reduced SOCS1 mRNA and protein expression(P＜0.05).Both silencing DNMT1 and overexpressing SOCS1 resulted in reduce of MPC-5 cell apoptosis rate,inflammatory factors level,p-JAK2 and p-STAT3 proteins expression(P＜0.05).Additionally,silencing DNMT1 increased SOCS1 mRNA and protein expressions(P＜0.05).Conversely,silencing SOCS1 counteracted the effects of DNMT1 silencing on MPC-5 cell apoptosis,inflammation,p-JAK2 and p-STAT3 proteins expression.Therefore,silencing DNMT1 expression can reduce the apoptosis and inflammation of podocytes induced by HG,and its mechanism may be related to the inhibition of JAK2/STAT3 signaling pathway activation by upregulating SOCS1 expression.

The prevalence of inflammatory bowel disease(IBD),a recurrent,non-specific disease characterized by persistent inflammation of the gastrointestinal system,is rising globally.As high-throughput sequencing,culturomics,and metabolomics have advanced,so has our understanding of the role of gut flora and its metabolites contributed in the development of disease and overall health.This study examined the latest advances in research as well as the mechanism of action of common metabolites of intestinal flora in inflammatory bowel disease.

To rapidly detect porcine rotavirus(PoRV),a one-step detection method was developed based on recombinase-aided amplification(RAA)combined with lateral flow dipstick(LFD)tech-nology.Specific primers and probes were designed according to the conserved sequence of the PoRV VP6 gene,and the amplification system was optimized.The specificity,sensitivity,and re-peatability of the method were thoroughly tested,and its clinical application was evaluated.The re-sults demonstrated that the method could amplify PoRV nucleic acid at 37 ℃ in just 15 min with a minimal detection limit of 22.3 copies/μL.No cross-reactivity was observed with other common diarrhea viruses.The total coincidence rate between RT-RAA-LFD and RT-PCR was 97.0％,with a Kappa coefficient of 0.93(K＞0.75).In summary,the RT-RAA-LFD detection method estab-lished in this study exhibits excellent specificity,sensitivity,and stability,characterized by rapid detection speed and visualization,making it suitable for on-site rapid detection.This method pro-vides a novel technical approach for the rapid diagnosis and epidemiological investigation of PoRV.

Objective To investigate the inhibitory effect of aspirin on liver metastasis of colon cancer in mice and the possible mechanism.Methods A total of 32 BALB/C mice were injected with CT26 colorectal cancer cells to establish colon cancer liver metastatic model,with 3 mice dead,15 mice in control group and 14 mice in experimental group.The control group was given saline 0.2 mL/d,the experimental group were given aspirin 30 mg/kg.The liver weight and the number of metastatic tumors were calculated after 30 days of intervention.HE and CD31 staining was performed by immunohistochemistry to observe the metastasis and angiogenesis.The protein expression of VEGF and cox-2 were analyzed by Western blot.Results The average liver weight and number of liver metastases nodules in the experimental group were significantly lower than those in the control group(P<0.05).Pathological examination showed that the experimental group of mice the number of liver cells and liver tumor angiogenesis were significantly less than the control group(P<0.05).Western blot showed that the expression of VEGF and cox-2 of CT26 cells were down-regulated after treated with aspirin.Conclusion Aspirin can down regulate the protein expression of VEGF and cox-2 protein to inhibit liver metastasis of colon tumor proliferation and angiogenesis,thereby inhibiting metastasis of colon cancer cells,for therapeutic purposes.

Objective:To evaluate the effectiveness of minimal flap surgery in the treatment sub-gingival caries ( class V cavities) . Methods:57 teeth(4 incisors, 13 canines, 38 premolars and 2 molars) with sub-gingival caries in 42 patients were treated by com-bined gingival flap surgery. Sub-gingival caries was fully exposed and the lesion was fully removed, then the deffects were filled with flowing resin. Before operation, 6 and 12 months after treatment, PD, SBI and gingival recession were examined. Results:Success rate of the treatment was 98. 25%. No statistically significant difference of the indexes was found during 12-month-follow up. Conclu-sion:Minimal flap surgery is effective for the treatment of sub-gingival caries ( class V cavities) .

Aim To explore the mechanism of E.coli heat-labile enterotoxin B subunit(LTB)as adjuvant by analysis of cellular proteins interacting with LTB. Methods Whole cell proteins were purified from RAW 264.7 cell after treated with LTB or NaCl 12 h, respectively.The cellular proteins were interacted with LTB and the interacting proteins were purified by pull-down assay and identified by mass spectrography.The LTB interaction proteins were conformed with Western blot and immunofluorescence assay.Results 25 LTB interaction proteins were found,and their interaction network was mapped;four proteins (Jup,Dsp,Ddx5 and Vimentin)were indicated to be related with LTB adjuvant activity;immunofluorescence assay indicated that GM130 interacted with LTB,however,Vimentin had no interaction with LTB in vivo.After treated by LTB,the expression of β-actin was upregulated obvi-ously in RAW 264.7 cell,whereras,Hspd1 did not show any change.Conclusions LTB exerts adjuvant activity through binding to GM1 of immune cells,cau-sing endocytosis and transporting to the Golgi apparatus by vesicles.Then LTB might bind to Jup and affect TCF/LEF activity,regulating the expression of Bcl 2, IL-6,and Runx3.The result is promoted T cell and B cell proliferation,differentiation and activation by se-cretion of cytokines and immunoglobulins.

Objective To evaluate the value of diffusion.weighted imaging(DWI)in the differential diagnosis of benign and malignant breast lesions with Meta.analysis.Methods English and Chinese articles published from 1997 to 2007 were searched in Cochrane library,Pubmed,Ovid.Elsevier.Springer databases and Chinese National Knowledge Infrastructure(CNKI),and selected according to the criteria for diagnostic research recommended by Cochrane Method Group.The characteristics of selected articles including the study background,design,results and diagnostic indicators were analyzed.Statistical analysis was performed with Meta-test version 0.9.Results Six articles were included with a total of 479 lesions.The sensitivity and specificity of DWI for breast 1esions ranged from 64.0% to 92.8% and from 45.8% to 96.7% respectively. All groups had homogeneity.and a fixation effects regression Was developed.The pooled weighted sensitivity and specificity with 95% CI and Az Was 86%(76%-92%).80%(61%-91%)and 92.19% respectively.Sensitivity analysis demonstrated no remarkable change on heterogeneity.Condnsion The value of ADC in DWI can help to differentiate the benign breast lesions from the malignant with good sensitivity and specificity.The further study is needed to assess the clinical aplication of the b-value and diagnostic threshold.
