1.Relationship between gene mutations and clinicopathological characteristics in patients with non-small cell lung cancer based on high-throughput sequencing

Objective To explore the detection rate and distribution characteristics of gene mutations in pa-tients with non-small cell lung cancer,and to analyze their relationship with clinicopathological characteristics.Methods The genetic testing results and clinicopathological data of 213 patients with non-small cell lung cancer who underwent high-throughput genetic testing at the Gene Detection Center,Changchun Cancer Hos-pital from April 2020 to December 2023 were collected.High-throughput sequencing technology was used to detect mutations in 26 genes,and the relationship between the mutation frequency and its distribution and the clinicopathological characteristics of patients was analyzed.Results Among 213 patients with non-small cell lung cancer,192 cases(90.14%)had at least one gene mutation detected.Among them,the genes with rela-tively high mutation frequencies were TP53(60.56%),EGFR(46.48%),KRAS(14.55%),ALK fusion(11.74%),and PIK3CA(8.92%).There were 132 cases(30.28%)of evidence level for Class 1 drugs and 11 cases(2.52%)of evidence level for Class 2 drugs.The incidence of EGFR gene mutations was higher in women,non-smokers and patients with lung adenocarcinoma(P<0.05).TP53 mutations usually occured in women,smokers and patients with stage Ⅳ.ALK mutations were more common in young patients,while KRAS mutations were more frequently seen in male smokers.Conclusion Analyzing the distribution charac-teristics of gene mutations in non-small cell lung cancer and their relationship with clinicopathological charac-teristics can provide a scientific basis for further optimizing genetic testing for patients with non-small cell lung cancer and offer guidance for clinical treatment.
2.Suicidal ideation of freshmen in one university of Tianjin from 2013 to 2020

Objective:
To understand the current situation of suicidal ideation in college students of Tianjin, and to provide reference for suicide prevention, early warning and emergency intervention.
Methods:
A total of 31 596 freshmen from one university in Tianjin were selected to participate in the mental health survey during 2013 to 2020.
Results:
A total of 1 349 freshmen reported suicidal ideation. The incidence of suicidal ideation varied from 2.60% to 6.18% across the years (the overall average annual incidence of suicidal ideation was 4.27%). Significant differences were observed in the incidence of suicidal ideation among freshmen of different origin (4.11% in urban areas, 4.74% in rural areas; χ 2=6.01, P <0.01), whether the only children in the family (4.10% in only children, 4.60% in non only children; χ 2=4.38, P <0.01). During the past five years, the in depth interview showed that the proportion of those with suicidal attempt ranged from 17.09%-33.73% (the total annual average proportion of those with suicidal attempt was 24.84%). Results from electroencephalonquadrantography varied significantly between college students with and without suicidal ideation and those with and without suicidal attempt.
Conclusion
It is necessary to strengthen the screening of college students for suicidal ideation and to identify the risk of students with suicide attempt, so as to provide a better reference for the prevention and intervention of psychological crisis.
3.Current status of anti-reflux surgery of proximal gastrectomy for esophagogastric junction tumor

In recent years, the overall incidence of gastric cancer has been decreasing worldwide, while the incidence of the esophagogastric junction tumor is increasing year by year. With the progress of the diagnostic technology of upper gastrointestinal tumors and the gradual popularization of early cancer screening, the detection rate of early esophagogastric junction tumor keeps increasing. Therefore, in recent years, the clinical application of gastric function preserving surgery is gradually increasing. As an important part of the surgical treatment strategy of esophagogastric junction tumors, proximal gastrectomy has attracted more and more attention with the confirmation of oncological safety. Compared with total gastrectomy, patients after proximal gastrectomy have better nutritional status and quality of life. However, the high incidence of reflux esophagitis after traditional proximal gastrectomy has seriously affected the quality of life of patients, and also hindered the application of proximal gastrectomy in esophagogastric junction tumors. How to reduce the occurrence of reflux esophagitis after proximal gastrectomy by optimizing the method of digestive tract reconstruction has been a big challenge in clinical practice. This article reviews the current methods of anti-reflux surgery for proximal gastrectomy for esophagogastric junction tumors, aiming to provide a reference for choosing a reasonable anti-reflux surgery.

MTT assay, flow cytometry, Transwell assay, and Boyden chamber assay were used to analyze the changes in cell proliferation, migration and invasion. Western blotting was used to detect the changes in ZNF217-related genes in the cells.
RESULTSshRNA-ZNF217 transfection significantly inhibited the expression of ZNF217 in U251 cells and suppressed the cell migration, invasion, growth, and cell cycle transition. ZNF217 knockdown downregulated the expression of pPI3, pAKT, C-Myc, and the mesenchyme biomarker N-cadherin, and stimulated the expression of the epithelium biomarker E-cadherin.
CONCLUSIONZNF217 promotes cell migration, invasion, and growth by activating PI3K/AKT signal to upregulate C-Myc and by modulating the genes associated with epithelial-mesenchymal transition in glioma cells.

8.An experimental study on the relationship between the expression of GFAP and iNOS and the post-traumatic intervals after brain contusion in rats

Objective To observe the relationship between the expression of GFAP and iNOS the post-traumatic intervals after experimental cerebral contusion,in order to attempt to find out a method of timing wound age of brain contusion in the forensic prediction of injury time.Methods After setting up an model of experimental focal cerebral contusion in rats,immunohistochemical method was applied to observing the changes of inducible nitric oxide synthase (iNOS) and glial fibrillary acidic protein (GFAP) staining combined with histology method in rats.The immunostaining results were measured quantitatively with computer imaging analysis system.Results (1) At 3h after injury,GFAP-positive astrocytes around the wounded area were definitely detected.The intensity and the area of GFAP-positive cells increased following the post-traumatic intervals.The intensity peaked dually at 1d and 5d,respectively,followed by reduction at 3d,which revealed two-peak waveforms,The area of GFAP positive cells enhanced significantly at 3h and gradually increased with prolonging survival time,and reached a peak at 7d after injury.(2) At 12h after injury,the iNOS isoform was present and iNOS positive staining cells were noted.The intensity and the area of iNOS positive cells increased following the post-traumatic intervals.The area of positive reached maximal level at 1-3days,gradually subsided after 5d,and maintain a higher lever up to 7d post-injury;but the intensity of individual positive cell gradually increased with prolonging survival time,reached a peak at 5d and gradually subsided after 5d,but the level was still higher than the initial interval.Conclusions Expression of GFAP and iNOS is correlated with post-traumatic intervals after cerebral contusion in rats,suggesting that the expression of iNOS and GFAP may be served as the markers for timing of brain contusion in forensic practice.
9.PATHOLOGICAL STUDY OF SUDDEN CORONARY DEATH——REPORT OF 128 AUTOPSY CASES

This paper reports the results of forensic pathological study of 128 autopsycases of sudden coronary death(SCD).The severity of the coronary arterystenosis was 4 degree in 63 cases,3 degree in 26 cases and 2 degree in 39 cases.The distribution of the artheroscleorosis of 3 and 4 degree was quite extensive. Recent thrombosis in CA was found in 18 cases,hemorrhage in plaques in 17cases.Only 2 cases had acute myocardial infarction.Inflammatory cell infil-tration were found in coronary plaques in 36 cases.Myocardail fibrosis orsmall scar formation were detected in 56 cases.It is suggested that SCD isthe commonest cause of sudden unexpected death.The majority of SCD(61%)were manhood in middle age.Most cases died suddenly during sleep withoutany clear inducements.The characteristics of the pathological changes in theCA and myoc ardium and the pathological diagnosis of SCD were analyzedand discussed.
10.IMMUNOHISTOCHEMICAL STUDY OF MYOGLOBIN DEPLETION FROM MYOCARDIUM IN THE CASES OF SUDDEN CORONARY DEATH

Myoglobin (Mb)depletion from myocardium in the cases of sudden soronary death (SCD) and was firstly studied by an immunhistochemial technique (ABC method) in China. Mb was detected quantitatively using scanning microscope photmeter and the results were analyzed statistically by computer The results showed that there were marked depletion of Mb from myocardium in each case of SCD The Mb depleted arce were multiple, disseminatly and segmentally distributed while no depletion of Mb from myocardium in the cases of control group was seen We suggested that the depletion of Mb can be used as one of the diagnostic criterion in the cases of SCD.
