1.Effects of tanshinone ⅡA on TNBS-induced mouse model of chronic colitis through PXR/NF-κB signaling pathway

AIM To investigate the therapeutic mechanism of tanshinone ⅡA in a mouse model of chronic colitis induced by trinitrobenzene sulfonic acid(TNBS).METHODS The BALB/c mice were randomly divided into the control group,the model group,and the low-dose and high-dose tanshinone ⅡA groups(10,20 mg/kg).Chronic inflammatory bowel disease(IBD)was induced in the model and tanshinone ⅡA groups by epicutaneous application of 3.75 mg TNBS(dissolved in 48%ethanol),followed by intrarectal administration of TNBS(0.75,1.5 and 2.25 mg in 40%ethanol)on days 7,14 and 21.Starting on day 7 post-modeling,the mice underwent their 14-day consecutive dosing of corresponding drugs by gavage.The mice had their disease activity index(DAI)assessed;their colon length and weight measured;and their levels of inflammatory factors IFN-γ and TNF-α in the colon mucosa detected by ELISA.The wild-type and PXR-/-mice were randomly divided into the control group,the model group,and the tanshinone ⅡA group(20 mg/kg).After modeling and drug administration using the aforementioned method,Masson staining was used to assess the intestinal fibrosis;immunohistochemistry was employed to detect the colon expression of ZO-1 and Occludin proteins;and immunofluorescence was used to detect the colon expression of NF-κB p65.RESULTS Tanshinone ⅡA(20 mg/kg)reduced DAI scores,colon weight/length ratio,and the colon levels of IFN-γ and TNF-α of the mouse models(P<0.05,P<0.01).Compared with the WT control group,the WT model group and PXR-/-control group exhibited increased colon histopathological scores and fibrosis areas(P<0.01),decreased protein expressions of ZO-1 and Occludin(P<0.01),and increased expression of p-NF-κB p65(P<0.01).Compared with the WT model group,the WT tanshinone ⅡA group showed reduced colon weight/length ratio,histopathological scores,and fibrosis areas(P<0.01);increased protein expressions of ZO-1 and Occludin(P<0.05,P<0.01);and decreased expression of p-NF-κB p65(P<0.01).However,tanshinone ⅡA showed no significant therapeutic effect upon PXR-/-model mice(P>0.05).CONCLUSION Tanshinone ⅡA(20 mg/kg)can effectively alleviate TNBS-induced chronic colitis in mice,and this protective effect may be exerted by the modulation of PXR/NF-κB signaling pathway.
2.Study on the changes of common bile duct pressure after laparoscopic common bile duct exploration in patients with common bile duct stones

Objective To study the change of common bile duct pressure after laparoscopic common bile duct exploration in patients with common bile duct stones.Methods Clinical data of 17 patients with common bile duct stones treated in our hospital from January 2022 to December 2022 were collected.All patients underwent laparoscopic common bile duct exploration and common bile duct pressure measurement.The patients were followed up for 8 weeks after operation,and the common bile duct pressure was measured by indwelling T tube at 2,3,4 and 8 weeks.Results Among the 17 patients,1 patient showed persistent high common bile duct pressure due to residual common bile duct stones.In the remaining 16 patients,there were no significant difference between the initial common bile duct pressure(19.7 cm H2O)and the common bile duct pressure at postoperative(17.0 cm H2O)and week 1(15.5cmH2O)(P>0.05).There were significant differences between the initial common bile duct pressure and the common bile duct pressure at week 2(13.5 cm H2O),3(12.2 cm H2O),4(11.1 cm H2O)and 8(10.0cmH2O)(P<0.05).Conclusion Compared to initial common bile duct pressure,the pressure of common bile duct began to decrease significantly from 2 weeks after laparoscopic common bile duct exploration in patients with common bile duct stones.
3.Epidemiological and genetic characteristics of school influenza outbreaks in Changzhou from 2021 to 2024

Objective:To characterize the etiological and genetic features of pediatric influenza outbreaks in Changzhou between 2021 and 2024,with the goal of informing evidence-based prevention strategies and guiding effective management of influenza outbreaks in school settings.Methods:During the period of 2021 to 2024,throat swabs of influenza-like cases from school outbreaks in Changzhou were collected. These samples underwent real-time reverse transcription-polymerase chain reaction(RT-PCR)testing and virus isolation. Epidemiological data were integrated to conduct pathogenetic analysis. The HA genes of isolated strains were amplified and sequenced to perform genetic characterization.Results:Between 2021 and 2024,a total of 256 influenza outbreaks were reported in schools in Changzhou. A total of 3 201 specimens were collected,of which 2 245 were tested positive for influenza viruses,resulting in a positivity rate of 70.13%. The outbreak season was primarily concentrated from December to February each year,with settings predominantly distributed in primary schools(accounting for 73.83%). The predominant epidemic strains were influenza A viruses,including 118 outbreaks caused by H1N1 and 104 by H3N2. A total of 74 influenza virus strains were successfully isolated from positive specimens,and sequencing of the hemagglutinin(HA)gene was completed. Phylogenetic analysis revealed that certain B/Victoria lineage strains(e.g.,B/Changzhou/01/2021)clustered closely with the vaccine strain B/Austria/3594/17(bootstrap support:99%). Among influenza H1N1 strains,multiple isolates from 2023—2024 clustered within the same major branch as A/Victoria/4897/2022(bootstrap support:100%). In contrast,the H3N2 strains exhibited a complex evolutionary pattern,showing variable genetic distances to vaccine strains from different years(e.g.,A/Massachusetts/18/2022,A/Darwin/6/2021);some isolates were closely related to vaccine strains,while others were more distantly related and scattered across the phylogenetic tree.Conclusions:The influenza outbreak situation in schools was severe and has significant public health implications. Continuous surveillance is essential,and preventive strategies should be promptly adjusted based on the epidemiological and genetic characteristics of circulating strains.
5.Gender differences and age-related changes in body composition of Miao adults in Guangxi Rongshui

Objective To investigate gender differences and age-related changes in body composition(BC)among Miao adults in Rongshui,Guangxi Province,and to provide the basis for assessing nutritional status and health.Methods With informed consent,630 Miao adults(218 males,412 females)were randomly selected for this study.Body composition was assessed using bioelectrical impedance analysis(BIA).Results Weight,fat-free mass,muscle mass,trunk muscle mass,limb muscle mass,waist-to-hip ratio(WHR),body water,presumtion of bone mass and protein were significantly higher in males than in females.And the fat mass,trunk fat mass,limb fat mass,visceral fat content,subcutaneous fat content and percentage of body fat were significantly higher in females than in males.According to the evaluation of body mass index(BMI)and WHR,the proportion of overweight and obesity of Miao adults was higher than the average level of Miao residents,and their obesity was characterized by central obesity.With age,weight,fat mass,muscle mass,fat-free mass,limb muscle mass,limb fat mass,subcutaneous fat content,percentage of body fat,body water,presumtion of bone mass,and protein of Rongshui Miao adults showed a gradual decreasing trend,while visceral fat content and WHR increased progressively.BMI in male Miao adults,along with BMI,fat mass,trunk fat mass,subcutaneous fat content,percentage of body fat,and body water in female Miao adults,showed a trend of increasing followed by decreasing,peaking at the age of 40-49 years.Conclusion The body composition of Miao adults in Rongshui,Guangxi,exhibits significant gender differences and age-related variation change patterns,which may increase the risk of sarcopenia and metabolic diseases with aging.
6.Development and psychometric evaluation of a scale for assessing science popularization competency in traditional Chinese medicine

Objective:To develop a scale for assessing science popularization competency in traditional Chinese medicine (TCM) and to evaluate its reliability and validity.Methods:Based on Lasswell's "5W" communication theory, the scale was constructed through literature review, expert consultation, and a pilot survey. Using convenient sampling, 450 TCM-related professionals were selected from two ClassⅢ Grade ATCM hospitals and their affiliated medical alliance community health institutions in Hunan Province. A total of 450 questionnaires were distributed and 446 valid responses were collected (valid response rate: 99.11%). Of these, 222 questionnaires were used for item analysis, reliability testing, and exploratory factor analysis (EFA) ; the remaining 224 were used for confirmatory factor analysis (CFA) .Results:The final TCM Science Popularization Competency Scale comprised 26 items under 6 dimensions: namely popular science cognition; science content creation ability; communication ability; professional values and personality traits; quality of science communication products; evaluation and feedback of communication effectiveness. EFA extracted six common factors, explaining 86.85% of total variance, with all item factor loadings > 0.500. The overall Cronbach's α coefficient for the scale was 0.972, with subscale values all > 0.800. Split-half reliability was 0.948. CFA showed good model fit across indices, indicating high reliability and validity.Conclusions:The TCM Science Popularization Competency Scale demonstrates good psychometric properties and can be used to assess the science popularization competency of TCM professionals. It offers a scientific tool to support the dissemination of TCM knowledge and contribute to the goals of the Healthy China initiative.

8.Clinical application effect of an innovative adjustable butterfly needle puncture protection kit for TIVAP

Background and Aims:Totally implantable venous access ports(TIVAP)are widely used in patients requiring long-term intravenous therapy.Traditional butterfly needle puncture fixation methods have limitations,including low success rates,increased pain,and risk of needle-stick injury.This study aimed to design an adjustable puncture protection kit for butterfly needles and evaluate its clinical utility using a simulated device.Methods:A prospective randomized controlled trial was conducted with 70 patients implanted with upper arm ports in the Hematology Department of Xiangya Hospital,Central South University,from January to December 2024.The patients were divided into a study group and a control group,with 35 cases in each,using a randomized block design.The study group underwent puncture with the simulated adjustable protection kit,while the control group used the traditional finger fixation method.Outcomes compared included first-attempt success rate,vertical puncture rate,pain score,puncture time,and complication rate.Results:The baseline characteristics of the two groups were balanced.The study group had significantly higher first-attempt puncture success rate and vertical puncture rate than the control group(94.3%vs.77.1%;91.4%vs.57.1%,both P<0.05).In the experimental group compared with the control group,pain scores were lower(1.80±1.13 vs.2.94±1.33,P<0.05),and puncture time was shorter[(31.31±9.05)s vs.(41.80±23.97)s,P<0.05].There was no significant difference in the incidence of puncture-related complications between the two groups(2.9%vs.14.3%,P>0.05).Conclusion:The simulated adjustable butterfly needle puncture protection kit effectively improves puncture success,enhances efficiency,reduces patient pain,and demonstrates good clinical safety.This innovative design provides a promising solution for reducing needle-stick injury risks and optimizing port puncture procedures,although larger,multicenter,and long-term studies are warranted.
