Two patients (patient 1, a 61-year-old male; patient 2, a 58-year-old female) received red yeast rice 6 g once daily orally because of abnormal blood lipids. Patient 1 had schistosomal cirrhosis and cholestatic hepatitis, with triacylglycerol 5.32 mmol/L. After 26 days of oral administration of red yeast rice, the patient developed limb weakness, with creatine kinase (CK) 604 U/L. Red yeast rice was stopped immediately and 3 days later, the patient developed lower limb muscle soreness, with CK 117 748 U/L. After 12 days of treatments with dexamethasone and rehydration, his symptoms gradually disappeared, with CK 79 U/L. Patient 2 had acute hepatitis, with triacylglycerol 2.34 mmol/L. After 24 days of oral administration of red yeast rice, the patient developed weakness and muscle soreness in both lower limbs, with CK 52 222 U/L. Red yeast rice was stopped immediately, and after 12 days of treatments with methylprednisolone and hydration, her symptoms were improved, with CK 210 U/L.

3.Effects of rhPTH (1-34) on adhesion of MC3T3-E1 osteoblasts cultured on titanium surfaces of SLA

Objective To evaluate the effects of the human recombinant parathyroid hormone [rhPTH (1 -34)] on the initial adhesion of MC3T3-E1 osteoblasts cultured on titanium surfaces of sand-blasted large grift acid-etched (SLA). Methods The optimal concentration of the rhPTH(1-34) action was determined by the CCK8 method. Then the MC3T3-E1 cells were divided into experimental groups treated with the optimal concentration of the rhPTH(1-34) medium and control groups treated with blank medium, and inoculated on SLA-treated titanium plates. DAPI immunocytochemistxy, scanning electron microscopy, and RT-PCR were used to count the cell adhesion of MC3T3-E1 cells on the surface of titanium plates. Morphological changes were observed and the expression of the integrin α1, α5, β1 mRNA in MC3T3-E1 cells was determined. Results Compared with the control group, the adherence of osteoblasts was more than that of the control group. The remodeling of actin filaments, the change of cell morphology, and the speed of adhesion and extension on the titanium plate were faster than that of the control group. The expression of the osteoblast integrin subunit α1, α5 and (31 was higher than that of the control group. Conclusion rhPTH (1-34) can promote the adhesion of MC3T3-E1 cells on pure titanium surface.
4.New recognition of the relationship between gallbladder benign lesions and gallbladder cancer

With the development of imaging technology,the detection rate of gallbladder benign lesions has increased year by year.And part of the diseases may evolve into gallbladder cancer through a series of pathophysiologic processes.There are some misunderstandings in the understanding of gallbladder benign lesions for surgeons,so it is difficult for the clinical decision-making.The relationship between gallbladder benign lesions and gallbladder cancer should be correctly understood.Surgeons can neither exaggerate the risk of gallbladder cancer nor miss optimal timing of operation.The key point of the diagnosis and treatment of gallbladder benign lesions should be based on making full use of imaging data and strictly grasp pre-and intraoperative indications,and it is important to identify the malignant transformation of gallbladder benign lesions as soon as possible and carry out standardized treatment.

6.Effect of gypenosides on DMN-induced liver fibrosis in rats.

OBJECTIVETo study the effect of gypenosides on DMN-induced liver fibrosis in rats.
METHODA rat liver fibrosis model was established by injecting DMN intraperitoneally. Four weeks later, model rats were randomly devided into three groups: the model group, the gypenosides treated group (200 mg x kg(-1)) and the colchicine treated group (0.1 mg x kg(-1)), with 10 specimens for each group. After a 2-week treatment, following parameters were observed: (1) last body weight, weight ratio between liver and spleen; (2) content of liver hydroxyproline (Hyp); (3) activity of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), gamma-glutamyltransferase (gamma-GT), content of albumin (Alb) and total bilirubin( TBiL) in serum; (4) liver pathology (Sirius red staining and HE staining); (5) activity of liver superoxide dismutase (SOD), glutathione reduced (GSH), glutathione peroxidase (GSH-Px) and content of liver maleic dialdehyde (MDA).
RESULTThere were classic liver cirrhosis pathological changes in model groups. Compared with the normal group, liver Hyp content, activity of serum ATL, AST, gamma-GT and content of serum TBiL, MDA of model groups significantly increased; content of serum Alb and liver GSH, activity of liver SOD and GSH-Px decreased significantly in model groups. In comparison with the model group, liver cirrhosis remarkable improved in the gypenosides group, content of liver Hyp reduced significantly (P < 0.01), which was equal to the colchicine group. Compared with the model group, liver function parameters improved markedly in the gypenosides group; liver SOD and GSH-Px activities significantly increased; MDA content reduced significantly (P < 0.05).
CONCLUSIONGypenosides shows an effect in treating DMN-induced liver fibrosis in rats.

7.Preliminary study on the relationship between the Toll-like receptor 9 (TLR9) gene polymorphism and condyloma acuminatum

Objective To investigate the relationship between the single nucleotide polymorphisms (SNPs) of TLR9 gene and the occurrence of condyloma acuminatum (CA). Methods Peripheral venous blood was obtained from 63 patients with CA and 23 normal human controls with informed consent. DNA was extracted from the blood samples and subjected to the amplification of TLR9 gene by PCR followed by sequence analysis. Results There were 4 SNPs, i.e., SNP1, SNP2, SNP3 and SNP4 at positions 1174, 1635, 1269 and 1724 of the TLR9 gene, respectively. Of these SNPs, SNP1 was located in intron 1, SNP2, SNP3 and SNP4 in exon 2. The registration number is rs352139 for SNP1, rs352140 for SNP2 in NCBI database. SNP3 and SNP4 were newly discovered positions. The frequency at SNP1 position was 0.690 and 0.609 for allele A in the patients and controls, respectively, 0.309 and 0.391 for allele G, respectively (both P > 0.05). No significant difference was observed between the patients and controls in the frequency of allele A or allele G at position SNP2 (0.302 vs. 0.698, 0.369 vs. 0.630, both P > 0.05). There were 4 haplotypes at the SNP1 and SNP2 positions, including AA, AG, GA and GG, with no significant difference in the frequency between the patients and controls (all P> 0.05). Conclusions There are 4 SNPs including SNP1, SNP2, SNP3 and SNP4 in the TLR9 gene in Guangdong Han population. SNP1 and SNP2 appear unrelated to the liability to CA.

9.Expression of c-kit on amelanotic melanocyte in follicle from vitiligo patients

Objective To analyse preliminarily the role of amelanotic melanocutes (AMMC) and SCF/c-kit signal pathway in the pathogenesis of vitiligo. Methods Three antibodies such as HMB-45, NKl/beteb and c-kit were used to stain sections from scalps from vitiligo patients and the healthy controls. Results There were no HMB-45 positive cells in outer root sheath(ORS) of follicle. NKI/beteb positive cells were small and located in groups at the middle and lower of outer root sheath with their retraction of the dendrites. They only expressed premelanosomal antigens but not melanosomal antigen such as HMB-45. There were no significant difference of AMMC in quantities between vitiligo patients and normal controls (P>0. 05). The expression of c-kit receptors on AMMC in follicle of depigment-ed scalps from vitiligo patients was lower than that in normal contols (P<0. 05). Conclusion Abnormal c-kit expression in AMMC in the follicle of depigmented scalps may play an important role in the pathogenesis of vitiligo.
10.Effect of processing on metabolism of amygdalin from bitter almond in rat.

OBJECTIVETo study the influence of processing on metabolism of the main component of bitter almond-amygdalin in rat.
METHODThe blood was collected at different times after amygdalin given by injection and oral, bitter almond and its processed production given by oral respectively, and then detected by both HPLC and HPLC-MS(n) methods after extraction pretreatment.
RESULTAfter injection, amygdalin was absorbed in prototype to blood rapidly, while the other three kinds of medicine given by oral were all not detected the prototype of amygdalin, but two metabolites were detected which were isomers of prunasin confirmed by mass spectrometry. The metabolic pathway of prunasin in processed bitter almond group was markedly different from the bitter almond group.
CONCLUSIONProcessing has a significant effect on bitter almond metabolic processes in rats.
