Objective To investigate the in vitro anti-hepatitis B virus(HBV)effects of icariside Ⅱ(ICS Ⅱ)and its impact on mitochondrial fission.Methods HBV-positive hepatocellular carcinoma HepAD38 cells were used as the cellular model.The cytotoxicity of ICS Ⅱ was assessed via CCK8 assay.The secretion levels of HBV surface antigen(HBsAg)and HBV e antigen(HBeAg),as well as HBV DNA copy numbers,were measured by ELISA and qPCR after treatment with ICS Ⅱ alone or ICS Ⅱ in combination with entecavir(ENT).The effects of ICS Ⅱ on mitochondrial morphology and motility were observed using confocal laser scanning microscopy and transmission electron microscopy(TEM).After ICS Ⅱ treatment,Western blot was performed to analyze the expression levels of key proteins involved in mitochondrial dynamics.Additionally,intracellular reactive oxygen species(ROS)production was evaluated via fluorescence staining.Results The CCK8 assay results showed that ICS Ⅱ treatment at 25 μmol/L had no significant effect on cell proliferation after 72 h.ICS Ⅱ significantly inhibited the secretion levels of HBsAg and HBeAg,with the respective inhibition rates reaching 54.90％and 39.65％(P＜0.05).Additionally,ICS Ⅱ alone reduced HBV DNA copy numbers by 15.19％,while ENT alone achieved a 34.11％inhibition rate.Notably,ICS Ⅱ in combination with ENT reduced HBV DNA copy numbers by 55.81％(P＜0.05).Furthermore,ICS Ⅱ induced mitochondrial shortening and enhanced mitochondrial motility in HepAD38 cells(P＜0.05).ICS Ⅱ significantly increased the expression levels of mitochondrial motility-related proteins,including Mfn1,Fis1,and phosphorylated Drp1(ser 616)(P＜0.05),while no significant changes were observed in the expression levels of Mfn2,total Drp1,or Drp1(ser 637)(P＞0.05).Additionally,ICS Ⅱ significantly suppressed the production of intracellular ROS in HepAD38 cells(P＜0.05).Conclusion ICS Ⅱ inhibits HBV replication in HepAD38 cells,and the underlying mechanism may be associated with the promotion of mitochondrial fission and suppression of ROS production.

AIM: To explore the effects of inflammatory conditions on the pharmacokinetics of methotrexate (MTX) and its related mechanisms. METHODS: The model of adjuvant induced arthritis (AIA) was established. The expression of organic anion transporter 3 (OAT3) in kidney was detected by immunohistochemistry, Western blotting and QPCR. The plasma concentration of MTX was detected by LC-MS/MS, and the pharmacokinetics of MTX after different administration time were compared by isolated rat kidney perfusion, kidney slices, in vitro cell uptake and transport experiments. RESULTS: The expression of OAT3 was significantly increased in the kidneys of AIA rats by immunohistochemistry, Western blotting and QPCR. At the same time, the concentration of MTX was detected by the optimized LC-MS/MS. The results showed that the uptake of MTX in the kidney slices of AIA rats was significantly increased, and Pro could reduce the excretion of MTX by inhibiting OAT3. Furthermore, it was demonstrated in vitro that inflammatory pathology can promote renal excretion of MTX by increasing the expression and functional activity of OAT3.CONCLUSION: Under inflammatory pathological conditions, it can increase the expression of OAT3 in the kidney, enhance its functional activity, accelerate the uptake of MTX by the kidney, and promote the excretion of MTX.

Objective:To investigate the clinicopathological factors predictive of lymph node metastasis (LNM) in undifferentiat-ed early gastric cancer (EGC) and to expand the possibility of endoscopic therapy for treating undifferentiated EGC.Methods:The re-searchers collected the data of 90 undifferentiated EGC patients who had undergone surgery at the Xingtai People's Hospital, Xingtai, China. The relationship between LNM and clinicopathological factors was retrospectively analyzed using univariate and multivariate lo-gistic regression analyses. Results:Univariate analysis showed that tumor size, lymphatic vessel involvement (LVI), and cancer inva-sion depth were the significant and independent risk factors for LNM. The LNM rate was 57.1%in patients with the three clinicopatho-logical risk factors. LNM was not found in patients without the three risk factors. Conclusion:Tumor size, LVI, and invasion depth are independently associated with the presence of LNM in undifferentiated EGC. Endoscopic therapy can be used to treat the patients with-out risk factors.

Aim To investigate the effect of icariin ( ICA) on renal interstitial fibrosis in SHR and explore its mechanism. Methods Fourteen male SHR of 13-week-old were randomly divided into model group( n=7 ) and ICA group ( n=7 ) , and WKY as control group (n=7). One week after adaptive breeding,the rats in the ICA group were given ICA 40 mg·kg-1,ig,bid to 26-week-old. The other rats in the model group and control group were given the same amount of normal sa-line. Then, the morphological changes of the kidney were observed by HE and Masson staining,respective-ly. The contents of plasma aldosterone andⅢcollagen were measured by double antibody sandwich method. The mRNA expressions of TGF-β1 , Smad2 , CTGF and FN were examined by real time RT-PCR. Results Compared with the normal control group, the kidney structures of model group were disordered,the mesang-ial matrix and the tubular interstitial fibrosis were in-creased. The contents of plasma aldosterone and Ⅲcollagen were increased in model group ( P <0. 01 ) . And the mRNA expressions of TGF-β1 , Smad2 , CTGF and FN in kidney tissues were up-regulated in model group( P <0. 01 or P <0. 05 ) . Compared with the model group,the kidney structures were improved and the contents of plasma aldosterone and Ⅲ collagen were reduced, as well as the mRNA expressions of TGF-β1,Smad2,CTGF and FN in kidney tissues were down-regulated(P<0. 01 or P<0. 05)in ICA group. Conclusion ICA may have anti-renal interstitial fi-brosis effect on SHR,and the mechanism might be re-lated to the reduced plasma aldosterone levels and the down-regulated expression of TGF-β1 and Smad2 .

Objective To evaluate the clinical value of speckle tracking imaging in the measurement of two-dimensional strain in patients with myocardial infarction. Methods High frame rate two-dimensional echocardiography were performed in 30 healthy subjects,21patients with anterior myocardial infarction and 15 patients with inferior myocardial infarction. Longitudinal strain (LS), radial velocity (Vs), regional shortening fraction (RFS),radial strain (RS) were measured in the apical two-chamber view and short-axis view(the level of papillary muscle) using two-dimensional strain software. Results Compared with healthy subjects, LS, Vs, RFS, RS decreased in the segments with infarction(P<0.05). LS, Vs, RFS, RS of the two myocardial infarction groups had partly differences(P<0.05). Conclusions Regional wall motion of left ventricle in patients with myocardial infarction can be evaluated accurately with speckle tracking imaging.

Objective To study the effect of CO_2 pneumoperitoneum on animals with intra-abdominal infection.Methods A total of 60 New Zealand white rabbits were used in this study.The animals were divided into two groups (30 in each),and injected with Escherichia coli or bacteroides fragilis into the abdominal cavity to establish models of intra-abdominal infection.In each group, the rabbits were subdivided into laparotomy,pneumoperitoneum,and control subgroups.Before the injection,the peripheral blood of the rabbits in each group was obtained to determine the WBC count and level of C-reactive protein (CRP,by ELISA).After the operation,the tests were repeated for 4 times at days 1,2,4,7 respectively.Afterwards,the animals were killed,and the viscera were harvested for aerobic and anaerobic culture.Results In each of the two groups,the WBC count in the laparotomy subgroup were significantly higher than that in the pneumoperitoneum subgroup at day 1 (P

Objective To evaluate the value of ultrasound-guided catheterization in the treatment of traumatic pericardial tamponade. Methods Nine patients with traumatic pericardial tamponade underwent ultrasound-guided catheterization from March 1998 to September 2003. After the drawing-off of fluid, the catheter was remained in the pericardium to drain the blood. Results The catheterization and drainage gave marked relief from symptoms. Surgical repairs of heart were performed in 4 patients within 4～ 8 h following drainage. Selective operations were required in 2 patients after the improvement of their conditions. Two patients were cured by drainage and 1 patient died of severe brain trauma. Conclusions The technique may produce prompt relief from symptoms of traumatic pericardial tamponade, being a safe and reliable procedure. It may create favorable conditions for further surgical repair or even freedom from operations.

Objective To study the value of sclerotherapy by indwelling intravenous cannula guided by color Doppler flow imaging in the treatment of thyroid cyst. Methods In the guidance of color Doppler flow imaging, the cyst was punctured with intravenous cannula. Then the cyst fluid was drawn out, the cyst cavity irrigated, and the absolute alcohol injected into. Results The treatment was completed successfully in 47 cases, without complications such as bleeding or nerve injuries. The cyst was wiped out within 1 month in 20 cases, within 3 months in 27 cases. All the 47 cases were followed for 6～12 months and no recurrence was seen.Conclusions Puncture guided by color Doppler flow imaging is safe and reliable. Use of absolute alcohol in sclerotherapy is effective.

AIM:To investigate the antiemetic effect of preadministration of Banxia Fuling Capsule(Rhizoma Zingiberis recens,Rhizoma Pinelliae,Poria,etc)(BFC)in pigeons and dogs,and to explore the relationship between antiemetic effect of BFC and 5-hydroxytryptamine(5-HT).METHODS:The pigeons and dogs were pretreated with BFC for 3 days.Acute emesis of pigeons and dogs induced by intravenously injecting cisplatin were observed.The concentraction of 5-HT in the serum,small intestine and brainstem of the animals were determined by spectrofluorimetry.RESULTS:The number and incidence of the emesis decreased in the treatment of BFC in dogs and pigeons;The levels of 5-HT in brainstem and small intestine decreased in pigeons,and so was it in the serum of dogs.CONCLUSION:BFC may inhibit emesis induced by cisplatin in dogs and pigeons,the antiemetic effect of BFC may decrease the synthesis and release of 5-HT in brainstem and small intestine.
