1.Discussion on the Difference of Acupuncture Points Efficacy in Improving Ulcerative Colitis by"Zusanli"or"Neiguan"Based on Transcriptomics Data

Objective To observe the intervention effect of acupuncture at"Zusanli"(ST36)or"Neiguan"(PC6)on the ulcerative colitis(UC)model mice.And to analyze the similarities and differences of gene transcription expression between the diseased colon and the non-diseased heart after acupuncture intervention by using transcriptomics,to explore whether there are efficacy differences between the two acupoints.Methods The UC mouse model was established by inducing dextran sulfate sodium salt(DSS).After successful modeling,the mice were randomly divided into model group,Zusanli group,and Neiguan group,with 9 mice in each group.Zusanli group and Neiguan group intervened with acupuncture at"Zusanli"and"Neiguan",respectively,30 min each time,once a day,for 5 consecutive days.The control and model groups were only bound without acupuncture.Observe the general situation of mice in each group,calculate the disease activity index(DAI),observe the morphological changes of colon tissue by HE staining.Use transcriptome sequencing technology to sequence and biologically analyze the colon tissue of mice in each group,and compare and study whether the two acupoints have differences in acupoint efficacy from the perspective of diseased tissue.At the same time,take the heart tissues of each group for comparative analysis,and discuss whether the effects of the two acupoints are different from the perspective of non-diseased tissue.Results Compared with control group,the DAI score of the model group increased significantly(P<0.001),and HE staining results showed significant pathological damage;after acupuncture at Zusanli or Neiguan,the DAI scores of UC mice were reduced considerably(P<0.05),and the pathological damage was significantly improved.The results of transcriptome sequencing and bioinformatics analysis showed that the effects of acupuncture at Zusanli or Neiguan on the colon of diseased tissue involved multiple genes,and 825 differential genes with similar changes(co-DEGs)were mainly enriched in white blood cells and chemokine pathways;there were 168 differentially expressed genes(ST36 only-DEGs)affected only by Zusanli,which were enriched to gastric acid secretion and other pathways;there were 325 differentially expressed genes(PC6 only-DEGs)only affected by Neiguan,which were enriched in cytokine-cytokine receptor interaction and other pathways.There were 95 co-DEGs in the heart of non-diseased tissue,which were mainly enriched in positive regulation of defense response and response to interleukin-1;there were 28 ST36 only-DEGs;there were114 PC6 only-DEGs,which were enriched in cardiovascular diseases and other pathways.Conclusion Acupuncture of"Zusanli"and"Neiguan"could improve UC mice's symptoms and pathological colon changes.However,through transcriptomic analysis,it was found that the two acupoints had differences in gene transcription effects on the colon of diseased tissue and the heart of non-diseased tissue,suggesting that acupoints with different distribution positions had some similarities in macro efficacy.Still,there were some differences at a micro level.
2.Exploration on Characteristics of Acupoint Efficacy Based on the Self-developed ACU&MOX-DATA Platform

Objective To explore the effects of different acupoints,different target organs,and different interventions on acupoint efficacy based on ACU&MOX-DATA platform;To illustrate and visualize whether the above factors have the characteristics of"specific effect"or"common effect"of acupoint efficacy.Methods The multi-source heterogeneous data were integrated from the original omics data and public omics data.After standardization,differential gene analysis,disease pathology network analysis,and enrichment analysis were performed using Batch Search and Stimulation Mode modules in ACU&MOX-DATA platform under the conditions of different acupoints,different target organs,and different interventions.Results Under the same disease state and the same intervention,there were differences in effects among different acupoints;under the same disease state,the same acupoint and intervention,the responses produced by different target organs were not completely consistent;under the same disease state and acupoint,there were differences in effects among different intervention measures.Conclusion Based on the analysis of ACU&MOX-DATA platform,it is preliminary clear that acupoints,target organs,and interventions are the key factors affecting acupoint efficacy.Meanwhile,the above results have indicated that there are specific or common regulatory characteristics of acupoint efficacy.Applying ACU&MOX-DATA platform to analyze and visualize the critical scientific problems in the field of acupuncture and moxibustion can provide references for deepening acupoint cognition,guiding clinical acupoint selection,and improving clinical efficacy.
3.ScMulti-Omics Technology and its Application in Acupuncture Research in Perspective

Single-cell multimodal omics(scMulti-omics)technology is a cutting-edge biotechnique for identifying cell type specificity and exploring gene regulation of cellular processes.Based on the combined application of two or more single-cell omics,thirteen scMulti-omics patterns of multi-dimensional and multi-scale were formed.In this paper,we proposed specific paths and strategies for constructing the database for scMulti-omics technology,preprocessing scMulti-omics data,and developing a prediction model.Combining scMulti-omics technology with acupuncture is a beneficial exploration for the modernization and advancement of inter-disciplinary Chinese medicine.The establishment of a prediction model of how acupuncture therapy regulates diseases using scMulti-omics data can infer the heterogeneous landscape of cell-level,which is anticipated to profoundly elucidate the multi-dimensional cell regulation mechanism of acupuncture in the treatment of diseases.
4.The relationship between cumulative fasting blood glucose exposure and all-cause mortality

Objective:To investigate the relationship between cumulative fasting blood glucose(cumFPG)exposure and all-cause mortality.Methods:The prospective cohort study included 56 845 subjects of Kailuan Group who participated in physical examinations from 2006 to 2007, 2008 to 2009, and 2010 to 2011 with complete data and a median of 7.77 years of follow up. The end point event was all-cause mortality. The incidence of all-cause mortality was compared in various groups divided by four quartile of cumFPG. The Cox proportional hazards model and natural spline were used to analyze the effect of cumFPG on all-cause mortality.Results:During the average(7.77±1.05)years of follow-up, the incidence density of all-cause mortality was on the rise with the increase of cumFPG(4.93, 5.87, 8.48, and 14.02 per 1 000 person-years), with statistically significant difference by Log- rank test( P<0.001). Cox proportional hazards model showed that after adjusting for potential confounding factors(age, sex, body mass index, hypertension, baseline fasting plasma glucose), the HR value(95% CI)of all-cause mortality in the fourth quartile group was 1.28(1.15-1.42)compared to the first quartile. When cumFPG increased every standard deviation, the risk of all-cause mortality increased by 17%. Natural spline analysis exhibited a similar J curve relationship between cumFPG and all-cause mortality. Conclusion:High cumFPG is a risk factor of all-cause mortality.
5.Study on regulatory mechanism of compatibility of ginseng and gecko dispensing granule on neuroendocrine immune network in model rats with kidney yang deficiency

OBJECTIVE To explore the regulatory mechanism of compatibility of ginseng and gecko dispensing granule on kidney yang deficiency model rats. METHODS Male SD rats were randomly divided into normal group (no modeling ,no administration),model group (modeling,no administration ),Jinkui shenqi pill group (modeling,dose of 2.33 g/kg),ginseng group(modeling,dose of 0.53 g/kg),gecko group (modeling,dose of 0.21 g/kg)and compatibility group (modeling,ginseng 0.53 g/kg and gecko 0.21 g/kg). The body mass and anal temperature of rats were measured at different time points ;the serum levels of cAMP ,cGMP,CRH,ACTH,CORT,T,T3,T4,E2,IgG and IgM were measured ;the pathomorphological changes of adrenal gland ,thyroid gland and testis were observed ;mRNA expression of CRH ,thyroid stimulating hormone releasing hormone (TRH)and gonadotropin releasing hormone (GnRH)in hypothalamus were detected. RESULTS Compared with model group ,the anal temperature ,the levels of cAMP ,CRH,ACTH,CORT,T3,T and cAMP/cGMP ,T/E2 in serum and mRNA expressions of TRH and GnRH in hypothalamus were significantly increased in the compatibility group (P<0.05 or P<0.01);the levels of cGMP,E2 and IgG in serum and mRNA expression of CRH in hypothalamus decreased significantly (P<0.05 or P<0.01); the pathological injuries of adrenal gland ,thyroid gland and testis were all improved. Compared with ginseng or gecko dispensing granules alone ,the anal temperature and T/E 2 of rats in the compatibility group increased significantly ,and mRNA expression of CRH in hypothalamus decreasedsignificantly (P<0.05 or P<0.01). CONCLUSIONS Thecompatibility of ginseng and gecko dispensing granule has a synergistic regulatory effect on kidney yang deficiency model rats , the mechanism of which may be associated with hypothalamus-pituitary-adrenal axis , hypothalamus-pituitary-thyroid axis , hypothalamus-pituitary-gonad axis and neuroendocrine immune network formed by immune function. Compatible drugs are better than single drugs.

Objective:To explore the effect of systolic blood pressure (SBP) trajectories on cancers.Methods:The relevant data of 54, 888 employees of Kailuan (Group) Limited Liability Company who participated in the 3 health examinations from 2006-2007, 2008-2009, 2010-2011 were collected and the new onset cancer cases were recorded. The systolic blood pressure trajectory grouping was carried out using the blood pressure measurement values of the 3 physical examinations. The life table method was used to calculate the incidence of cancer, and the multivariate Cox proportional hazard regression model was used to analyze the influence factors of cancer.Results:According to the systolic blood pressure trajectory, 54, 888 subjects were divided into 5 groups, including 14, 326 in the low-stable group, 25, 630 in the moderate-stable group, 5, 390 in the moderate-increasing group, 6, 438 in the elevated-lowering group, and 3, 104 in the elevated-stable group. A total of 1, 070 new onset cancer occurred during the follow-up period of (4.95±0.53) years. The incidence of cancer in the low-stable group, moderate-stable group, moderate-increasing group, elevated-lowering group and elevated-stable group were 1.3% (177/14, 326), 2.2% (491/25, 360), 3.1% (147/5, 390), 2.7% (156/6, 438) and 3.8% (99/3, 104), respectively, the difference was statistically significant ( P<0.001). After adjusting for gender, age, smoking, drinking, physical exercise, body mass index (BMI), fasting blood glucose, total cholesterol, antihypertensive drugs, hypoglycemic drugs, and lipid-lowering drugs, multivariate Cox regression analysis showed that the systolic blood pressure trajectory was related to the incidence of cancer. 8.Influence of different costal cartilage harvest techniques on donor site pain in rhinoplasty

Objective:To seek a minimally invasive method for costal cartilage harvest by using two different costal cartilage harvest techniques in rhinoplasty and to compare their influence on donor site pain.Methods:Fourty-three female patients who underwent costal cartilage harvest for rhinoplasty from Dec. 2016 to Dec. 2017 were randomly divided into two groups. We harvested the right side seventh costal cartilage in both groups. In control group, we harvested a full thickness segment of costal cartilage in each patient, whereas we harvested a split thickness segment of costal cartilage by preserving the superior strip in experimental group. Donor site pain was evaluated via visual analogue scale (VAS) 6 h, 24 h, 48 h and 72 h after surgery.Results:The VAS scores (mean±standard deviation) at different time points in the control group were (5.515±1.085), (5.250±1.302), (5.315±1.117) and (4.895±1.042). And in the experimental group, they were (2.665±0.713), (2.261±0.642), (1.609±0.398) and (1.383±0.514), respectively. The VAS scores at different time points were significantly higher in the control group than that in the experimental group ( P<0.05). Conclusions:The superior strip preserved costal cartilage harvest technique significantly reduces postoperative donor site pain.
9. Association between sleep duration and brachial-ankle pulse wave velocity

Objective:
To investigate the association between sleep duration and brachial-ankle pulse wave velocity (baPWV).
Methods:
A cross-sectional study method was used to observe 38 604 employees of Kailuan Group who participated in the physical examination and the baPWV test from January 2010 to July 2018. The age was (51.6±11.1) years old. There were 72.4% (27 955/38 604) male participants. According to the sleep duration, subjects were divided into 5 groups including ≤ 5 hours group (3 762 cases),>5 hours and ≤6 hours group (9 585 cases),>6 hours and ≤7 hours group (12 604 cases), >7 hours and ≤8 hours group (11 921 cases) and >8 hours group (732 cases). Multivariate logistic regression model was used to analyze the association between sleep duration and the baPWV.
Results:
The age was (51.6±11.1) years old. There were 72.4% (27 955/38 604) male participants. The prevalence of baPWV≥14 m/s in ≤ 5 hours group, >5 hours and ≤6 hours group, >6 hours and ≤7 hours group, >7 hours and ≤8 hours group, and >8 hours group was 63.5% (2 389/3 762), 58.9% (5 645/9 585), 55.0% (6 926/12 604), 53.3% (6 356/11 921) and 54.8% (401/732) respectively. After adjusting for confounding factors including age, gender, smoking, drinking, physical exercise, snoring, hypertension, diabetes mellitus, dyslipidemia, body mass index≥24 kg/m2, mean arterial pressure, heart rate, and C-reactive protein, the multivariate logistic regression analysis showed that the
10.Evaluation of the reliability and validity of the modified Morisky scale for patient's compliance with moxibustion

Objective:A questionnaire for the compliance with moxibustion was designed based on the 4-item Morisky scale, and the reliability and validity of the questionnaire was investigated.Methods:A modified Morisky scale was designed based on the 4-item Morisky scale for the compliance with moxibustion, and 146 patients having received moxibustion for over 2 weeks were investigated using this scale to evaluate their compliance with moxibustion; the internal equity and the construct validity of the scale were statistically analyzed.Results:The analysis of reliability showed that in item of internal consistency, the Cronbach's α was 0.72 and the split-half coefficient was 0.71, the correlation coefficients between the 4 component scores and the total score ranged 0.67-0.80, and the between-component correlation coefficients ranged 0.24-0.56; the exploratory factor analysis (EFA) totally extracted 1 common factor, and the explicable variation was 55.02%, and the loads of the 4 items were respectively 0.82, 0.81, 0.74 and 0.58.Conclusion:The reliability and validity of the modified Morisky scale for the compliance with moxibustion are acceptable, while several items need further modification and improvement in the expression and content.
