1.Longitudinal cohort study on pubertal development trajectories of testicular and breast development among children

Objective:
To characterize longitudinal trajectories of testicular development in boys and breast development in girls, so as to provide reference data for understanding patterns of pubertal sexual maturation.
Methods:
Based on the Shanghai Pudong New Area Cohort Study on Growth, Development and Health in Children and Adolescents, a baseline survey was conducted in 2020 using a mult stage cluster random sampling method. A total of 2 184 children who completed all follow ups during the primary school period from 13 elementary schools in Pudong New Area,Shanghai,with annual follow ups during 2021-2025. Testicular volume and Tanner stage of breast development were assessed by professional physicians using standardized visual inspection and palpation. The age distribution of testicular volume and breast development was fitted by using cumulative link mixed models and Turnbull s nonparametric maximum likelihood estimation method.
Results:
Median ages for testicular volumes of 2, 3, 4 and 5 mL in boys were 7.07, 9.24, 10.29, and 11.57 years old, respectively. Median ages for Tanner breast stages Ⅱ, Ⅲ, Ⅳ, and Ⅴ in girls were 8.55 , 10.17, 11.18, and 13.78 years old, respectively. Based on overweight and obesity, stratified analysis showed that earlier pubertal onset among overweight/obesity children, and the key milestones for pubertal initiation were testicular volume reaching 4 mL in boys and breast Tanner II in girls for 10.29, 10.83; 8.18, 9.00 years.
Conclusion
Overweight and obesity are associated with earlier pubertal initiation,but there are certain gender and developmental stage specific patterns.
2.Clinical distribution and trend of drug resistance of Klebsiella pneumoniae isolates from a three-A hospital of Suzhou from 2019 to 2023

OBJECTIVE To investigate the clinical distribution and dynamic change of drug resistance of K.pneu-moniae and carbapenem-resistant Klebsiella pneumoniae(CRKP)isolated from a three-A hospital of Suzhou so as to provide scientific bases for prevention and control of hospital-associated infections and reasonable application of antibiotics.METHODS The K.pneumoniae and CRKP strains that were isolated from the submitted specimens were collected from the patients who treated in the Affiliated Suzhou Hospital of Nanjing Medical University from 2019 to 2023.The clinical characteristics of the patients with infection and the trend of drug resistance were statis-tically analyzed.RESULTS Totally 5631 strains of K.pneumoniae were isolated,1205(21.40%)of which were CRKP,and the isolation rate of CRKP showed an upward trend in the five years(x2=236.352,P<0.001).Among the K.pneumoniae isolates,51.59%were isolated from sputum,13.51%from urine;19.43%were isolated from intensive care unit(ICU),7.64%from emergency department,and 7.19%from respiratory department.There were significant differences in gender,age and season between the patients detected with CRKP and the patients detected with non-CRKP(P<0.05).The drug resistance rates of the K.pneumoniae strains to cephalosporins,quinolones and carbapenems con-tinuously increased from 2019 to 2023(P<0.001),the drug resistance rate to imipenem increased from 11.69%to 34.24%,meropenem from 10.92%to 34.24%.CONCLUSIONS The K.pneumoniae isolates show severe drug re-sistance from 2019 to 2023,and the isolation rate of CRKP strains rises increasingly.It is necessary for the hospi-tal to focus on the continuous monitoring of key populations and departments and optimize the management of an-tibiotics and infection control strategies so as to provide guidance for reasonable clinical use of antibiotics,effective control of transmission of drug-resistant strains and cope with the increasingly severe drug resistance.
3.The Influence of Boundary Condition Changes of Inner and Outer Walls of Osteon on Fluid Flow Characteristics:A Finite Element Study

Objective To explore the characteristics of fluid flow within loaded osteons under different boundary conditions.Methods The COMSOL Multiphysics software was used to establish a three-dimensional(3D)finite element model of osteons with different boundary conditions,and the variation rules of pore pressure and flow velocity of osteons under different inner wall pulsating blood pressures and outer wall elastic constraint conditions were analyzed.Results As the pulsatile blood pressure inside the osteon increased from 0 mmHg(1 mmHg=0.133 kPa)to 300 mmHg,the peak pore pressure within the osteon correspondingly increased from 26 kPa to 68 kPa.As the elastic constraint on the outer wall of osteons changed from being completely elastic to completely constrained,the peak pore pressure within osteons increased from 15 kPa to 26 kPa,and the peak flow velocity increased from 0.04 um/s to 0.07 um/s.Conclusions This study reveals the influence laws of changes in boundary conditions such as the pulsatile blood pressure on the inner wall and the elastic constraint on the outer wall of osteons on fluid flow characteristics within loaded osteons.These findings are conducive to a deeper understanding of the mechanical response mechanisms of bone tissues in both physiological and pathological states,and provide an important theoretical basrs for further researches on bone mechanotransduction.
4.A comparative study of endoscopic super minimally invasive surgery and laparoscopic minimally invasive excision for gastric glomus tumors

Objective:To compare the efficacy and safety of the gastrointestinal endoscopic super minimally invasive surgery (eSMIS) and the laparoscopic minimally invasive procedure (LS) for the treatment of gastric glomus tumors(GGT).Methods:This study retrospectively included 15 patients with GGT who underwent eSMIS at the First Medical Center of Chinese PLA General Hospital from May 2014 to May 2024. Data on demographic characteristics, surgical indexes, efficacy evaluation indexes, postoperative medical disposition, health economics indexes and postoperative laboratory indexes were collected and analyzed, the patients were followed up for survival outcomes and the postoperative quality of life, efficacy and safety of the two groups were compared.Results:The amount of bleeding in the eSMIS group was lower than that in the LS group: 1.25 (0, 3.75) ml vs. 41.56 (10.00, 50.00) ml, and the surgical cost in the eSMIS group was lower than that in the LS group: 10 792.87 (8 424.90, 12 730.30) yuan vs. 21 773.06 (19 940.60, 24 843.10) yuan, there were statistical differences ( P<0.05). The surgical efficacy, postoperative laboratory indicators and medical treatment convenience between the two groups had no statistical differences ( P>0.05). Conclusions:In the choice of surgical procedures for the treatment of GGT, eSMIS is superior to LS in terms of bleeding and surgical cost, and not inferior to LS in other indicators. This suggested that the treatment of GGT with eSMIS is feasible, safe and effective.
5.Expert consensus on weight management for adult solid organ transplant recipients with overweight or obesity(2025 edition)

In recent years,the prevalence of overweight/obesity in solid organ transplant recipients has been rising rapidly.Clinical research evidence on obesity in solid organ transplant recipients has been increasing,but there is a lack of standardized diagnostic and treatment guidelines and comprehensive management pathways to guide clinical practice.Based on this,the Expert Consensus on Weight Management in Overweight or Obese Adult Solid Organ Transplant Recipients(2025 Edition)was organized and written by experts from the Diabetes Professional Committee of the Chinese Research Hospital Association.This consensus mainly provides recommendations on the epidemiological data,diagnostic criteria,screening and assessment methods,management pathways,and intervention strategies for overweight/obesity in solid organ transplant recipients.It aims to further improve the management level of medical workers engaged in the whole process of organ transplantation for the population with overweight/obesity combined with solid organ transplantation,standardize clinical diagnosis and treatment,improve patient prognosis,and enhance quality of life.
6.Effects of LINC00626 on proliferation,apoptosis and drug resistance of colorectal cancer SW480 cells

Aim To investigate the high expression of LINC00626 in colorectal cancer,and explore the effects of LINC00626 on the proliferation,apoptosis,and drug sensitivity of colorectal cancer SW480 cells,as well as its underlying mechanisms.Methods Flu-orescence in situ hybridization(FISH)was used to de-tect the expression levels of LINC00626 in 38 colorec-tal cancer tissues and their corresponding adjacent nor-mal tissues.The JASPAR database was utilized to pre-dict co-expressed genes and their possible binding sites.Cell transfection technology was employed to knockdown LINC00626.Western blot and qRT-PCR techniques were used to verify the transfection efficien-cy.CCK-8 assay,cell apoptosis and necrosis staining,and Western blot were used to detect the changes in the proliferation,apoptosis,drug sensitivity,and ap-optotic proteins of SW480 cells,respectively.Results The FISH results indicated that LINC00626 was highly expressed in colorectal cancer tissues(P<0.05).The expression of LINC00626 was not associat-ed with the age or gender of patients,but was related to the TNM stage and the presence of lymph node me-tastasis($ P<0.05 $).The results of CCK-8 assay and cell apoptosis and necrosis staining showed that af-ter knockdown of LINC00626,the proliferation ability of SW480 cells decreased,the apoptosis level in-creased,and the drug resistance decreased(P<0.05).Western blot results showed that with the de-crease in the expression level of LINC00626,the ex-pression of caspase-3 protein decreased,the expression of cleaved caspase-3 protein increased,and the expres-sion of Bcl-2 protein decreased(P<0.05).Conclu-sions LINC00626 is highly expressed in colorectal cancer and is associated with the TNM stage and the presence of lymph node metastasis.LINC00626 can af-fect the proliferation,apoptosis,and drug sensitivity of SW480 cells and alter the expression of apoptotic pro-teins.
7.Multi-slice spiral CT enhanced scanning for preoperative staging of gastric cancer

Objective To observe the value of multi-slice spiral CT enhanced scanning for preoperative TNM staging of gastric cancer compared with postoperative pathology.Methods A total of 219 patients with gastric cancer confirmed by postoperative pathological examination were retrospectively enrolled.The efficacy of spiral CT enhanced scanning for preoperative TNM staging,and its consistency with postoperative pathology were analyzed.Results Multi-slice spiral CT enhanced scanning detected gastric cancer in 214 cases,with detection rate of 97.72%(214/219)and missed diagnosis rate of 2.28%(5/219).Among these 214 cases,the accuracy of spiral enhanced CT for T,N,M staging of gastric cancer was 91.12%(195/214),88.79%(190/214)and 85.98%(184/214),respectively,which was high to extremely high consistent with that of postoperative pathology,with Kappa value of 0.868,0.844 and 0.701,respectively.Conclusion Multi-slice spiral CT enhanced scanning had high value for preoperative TNM staging of gastric cancer.

9.Effects of circCSPP1 on the proliferation,migration,and invasion of human retinoblastoma Y-79 cells by regulating miR-642a-5p expression

Objective To analyze the effects of circular RNA CSPP1(circCSPP1)and microRNA-642a-5p(miR-642a-5p)on the biological behaviors of retinoblastoma Y-79 cells.Methods A total of 47 patients with retinoblastoma(47 eyes)who were admitted at Zhengzhou Central Hospital from June 2019 to October 2022 were included in this study,and the tumor tissues and paraneoplastic tissues of these patients were collected during surgery.The human retinoblastoma cell line Y-79 was cultured in vitro and then divided into seven groups based on different transfectants:the Con group(nor-mally cultured Y-79 cells),sh-NC group(transfected with sh-NC),sh-circCSPP1 group(transfected with sh-circCSPP1),miR-NC group(transfected with miR-NC),miR-642a-5p group(transfected with miR-642a-5p mimics),sh-circCSPP1+an-ti-miR-NC group(co-transfected with sh-circCSPP1 and anti-miR-NC),and sh-circCSPP1+anti-miR-642a-5p group(co-transfected with sh-circCSPP1 and anti-miR-642a-5p).The real-time quantitative polymerase chain reaction(RT-qPCR)was used to measure the expression level of circCSPP1 and miR-642a-5p in retinoblastoma tissues,paraneoplastic tissues,and Y-79 cells.The proliferation,migration,and invasion of Y-79 cells were assessed via the cell counting kit-8(CCK-8)assay,scratch wound healing assay,and Transwell chamber assay,respectively.The targeting relationship between cir-cCSPP1 and miR-642a-5p was verified by the dual-luciferase reporter assay.Western blot was performed to quantify the ex-pression level of E-cadherin and N-cadherin.Results Compared with paraneoplastic tissues,the expression of circCSPP1 was up-regulated,that of miR-642a-5p was downregulated in retinoblastoma tissues,and the differences were statistically significant(both P<0.05).Compared with the Con group and the sh-NC group,the sh-circCSPP1 group showed a reduced scratch healing rate,decreased invasive cell count,up-regulated relative protein expression of E-cadherin,and down-regu-lated relative protein expression of N-cadherin,and the differences were statistically significant(all P<0.05).Compared with the Con group and the miR-NC group,the miR-642a-5p group exhibited a reduced scratch healing rate,decreased in-vasive cell count,up-regulated protein expression of E-cadherin,and down-regulated protein expression of N-cadherin,and the differences were statistically significant(all P<0.05).The dual luciferase reporter assay demonstrated that cir-cCSPP1 negatively regulated the expression level of miR-642a-5p.Compared with the sh-circCSPP1+anti-miR-NC group,the sh-circCSPP1+anti-miR-642a-5p group displayed an increased scratch healing rate,increased invasive cell count,down-regulated protein expression of E-cadherin,and up-regulated protein expression of N-cadherin,and the differences were statistically significant(all P<0.05).Conclusion Silencing circCSPP1 may suppress the proliferation,migration,and invasion of Y-79 cells by up-regulating the expression of miR-642a-5p,suggesting that circCSPP1 may become a poten-tial therapeutic target for retinoblastoma.
