1.A real-time process monitoring scheme for blood center NAT using the median of internal control CT value

Objective: To establish a real-time quality control scheme based on the median (MD-IC) of internal control cycle threshold value in negative samples (NEG-IC-CT), so as to monitor anomalies such as progressive drift in nucleic acid testing system not covered by conventional internal quality control (IQC) in blood center nucleic acid laboratories, and to verify its feasibility. Methods: The internal control CT values of 54 426 negative samples were retrospectively collected. These samples were from four reagent batches of the two new and old equipment sets during the operation of the Wantai nucleic acid testing system in our blood center. The daily median of NEG-IC-CT values was used as the research indicator. Control limits were calculated using median absolute deviation (MAD) to construct the Median-MAD quality control chart. The monitoring performance of this scheme for the operation status of the testing system was simultaneously evaluated. Results: Statistical analysis showed significant differences in NEG-IC-CT value distribution between the new and old equipment sets, as well as between the two different reagent batches of the old equipment (P<0.000 1). The NEG-IC-CT value performance of the two different reagent batches of the new equipment was no significant difference in distribution (P>0.05). This scheme identified three typies of distinct anomalies. The out-of-control events observed with the old equipment in both the O1 and O2 reagent batches suggested potential performance decay due to equipment aging. The unreported change of reagent batch in time of Phase B with new equipment caused a stepwise drift on the quality control chart. In the later stage of Phase A with the new equipment, an alert was triggered, indicating potential quality risks associated with practices such as the mixed use of the remaining reagents and extremely long operator working hours. Conclusion: The realtime quality control scheme based on NEG-IC-CT value established in this study has been preliminarily validated for its monitoring effectiveness in nucleic acid testing in our blood center. This scheme performed well in detecting differences among testing systems and reagent batches, serving as an effective supplement to routine internal quality control. It can provide an intuitive and effective evaluation method for monitoring the performance of the nucleic acid testing process at blood center.

Non-alcoholic fatty liver disease (NAFLD) is one of the most prevalent forms of liver diseases globally. Its progression can lead to cirrhosis and end-stage liver disease, and there is currently a lack of effective pharmacological treatments. Adenosine monophosphate-activated protein kinase (AMPK), as a regulatory hub for maintaining cellular energy homeostasis, can coordinate key cellular processes such as adipogenesis, glucose metabolism, and mitochondrial functions. Its activation exerts metabolic regulatory effects through pathways including inhibiting lipogenesis, enhancing mitochondrial β-oxidation, regulating inflammation and oxidative stress, and promoting autophagy. Accordingly, AMPK emerges as a potential target for the prevention and treatment of NAFLD. Traditional Chinese Medicine (TCM), with low toxicity, high accessibility, and multi-component, multi-target synergistic effects, has demonstrated unique value in NAFLD treatment, particularly showing notable advantages in regulating the AMPK signaling pathway. Sichuan is known as the treasure house of TCM, and the active components of its authentic medicinal materials such as Coptidis Rhizoma not only reflect regional characteristics in AMPK signaling regulation but also form a multi-level metabolic regulatory network through crosstalk with pathways such as sirtuin 1 (SIRT1) and peroxisome proliferator-activated receptor α (PPARα). They can achieve specific regulation by directly activating AMPK and modulating upstream and downstream targets, exerting prominent effects in ameliorating hepatic steatosis and inflammation. This study systematically reviews the research findings on TCM for the prevention and treatment of NAFLD over the past five years, elaborating the mechanisms by which TCM treats NAFLD through regulating the AMPK signaling pathway. It aims to provide new perspectives and references for clinical diagnosis and treatment, basic research, and drug development.
3.Effect of Astragalus polysaccharide on the proliferation of rat intestinal mucosal microvascular endothelial cells by regulating VEGF/VEGFR pathway

This study explored whether Astragalus polysaccharide(APS)can regulate the VEGF/VEGFR signaling pathway to affect the proliferative activity of rat intestinal mucosal microvascu-lar endothelial cells(RIMMVECs).RIMMVECs were isolated from newborn rats,then purified and treated with APS at concentrations of 0.1,1.0,10.0,100.0,1 000.0,and 10 000.0 mg/L.MTT was used to determine the effect of APS on RIMMVECs proliferation and screen for the optimal concentration of APS.Subsequently,flow cytometry was used to detect the changes in cell cycle to evaluate the stage of action of APS on the cell cycle in RIMMVECs.Then,the ELISA was used to detect the changes of VEGFA in cell supernatant to evaluate the potential of cell proliferation and angiogenesis.The changes in fluorescence intensity of Fluo-8AM was observed using fluorescence microscopy to evaluate intracellular Ca2+levels.Finally,Western blot was used to detect the ex-pression of PERK in RIMMVECs to analyze the possible mechanism of APS.The results showed that 100 mg/L APS significantly enhanced the proliferative activity of RIMMVECs,increased the content of VEGFA in the cell supernatant,the intracellular Ca2+levels,and the expression of PERK protein,indicating that APS promotes the proliferation of RIMMVECs,which may be a-chieved by promoting the expression of VEGFA and activating the ERK pathway.

Body weight abnormalities, including overweight, obesity, and underweight, have become a dual public health challenge in Chinese adults: overweight and obesity lead to a variety of chronic complications, while underweight increases the risks of malnutrition, sarcopenia, and organ dysfunction. To systematically address these issues, multidisciplinary experts in endocrinology, sports science, nutrition, and psychiatry from various regions have held multiple weight management seminars. Based on the latest epidemiological data and clinical evidence, they expanded the guideline to include assessment and intervention strategies for underweight, in addition to the core content of obesity management. This guideline outlines the etiological mechanisms, evaluation methods, and multidimensional management strategies for overweight and obesity, covering key areas such as diagnosis and assessment, medical nutrition therapy, exercise prescription, pharmacological intervention, and psychological support. It is intended to provide a scientific and standardized approach to weight management across the adult population, aiming to curb the rising prevalence of obesity, mitigate complications associated with abnormal body weight, and improve nutritional status and overall quality of life.

7. Mechanism of action of formononetin in alleviating allergic asthma through DRP1-NLRP3 signaling pathway

Aim To investigate the mechanism by which formononetin (FN) inhibits mitochondrial dynamic-related protein 1 (DRP1) -NLRP3 axis via intervening the generation of ROS to reduce allergic airway inflammation. Methods In order to establish allergic asthma mouse model, 50 BALB/c mice aged 8 weeks were divided into the control group, model group, FN treatment group and dexamethasone group after ovalbumin (OVA) induction. Airway inflammation and collagen deposition were detected by HampE and Masson staining. Th2 cytokines and superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA), and IgE levels in bronchoalveolar lavage fluid (BALF) were measured by ELISA, ROS in BEAS-2B cells was assessed by DCFH-DA staining, DRP1 expression in lung tissue and BEAS-2B cells was detected by immunohistochemistry and immunofluorescence, and the DRP1-NLRP3 pathway was analyzed by immunoblotting. Results FN treatment could effectively ameliorate the symptoms of asthmatic mouse model, including reducing eosinophil accumulation, airway collagen deposition, decreasing Th2 cytokine and IgE levels, reducing ROS and MDA production, increasing SOD and CAT activities, and regulating DRP1-NLRP3 pathway-related protein expression, thereby relieving inflammation. Conclusion FN ameliorates airway inflammation in asthma by regulating DRP1-NLRP3 pathway.
8.Research on the influence of running gait on sports injuries of lower limb in middle-and long-distance teenage runners

Objective To explore the effect of gait biomechanics on sports injuries in middle-and long-distance teenage runners.Methods Eighteen healthy middle-and long-distance runners were select-ed as subjects,and the biomechanics of their gait during mid-speed running was tested using a three-dimensional infrared motion capture system,a three-dimensional force measurement platform,and a la-ser speed measurement system.After the test,data on sports injuries within one year were collected.Based on the severity of their injuries,the athletes were divided into an injury group,and a non-inju-ry group(healthy group).Their gait spatiotemporal parameters,ground reaction forces,and independent sample t-test for trunk and lower limb joint angles,and lower limb joint power were all analyzed.Re-sults There was no significant difference in the spatiotemporal parameters of the mid-speed running gait and ground reaction force indicators between the two groups(P>0.05).The maximum trunk exten-sion angle of the injury group was significantly greater than the healthy group(P<0.05),while the max-imum trunk flexion angle was significantly smaller than the latter(P<0.05).Moreover,the maximum right flexion angle,hip adduction angle,maximum hip adduction angle,ankle varus angle and ankle internal rotation angle of the injury group were significantly smaller than that of the healthy group,with the ankle internal rotation angle significantly bigger than the latter(P<0.05 for all).Meanwhile,the maximum hip joint extension power of the injury group was significantly lower than the healthy group(P<0.05),while the maximum knee joint abduction power was significantly higher than the latter group(P<0.01).Conclusion When running at medium speed,the spatiotemporal parameters of the sup-porting leg and ground reaction force have a relatively smaller impact on lower limb sports injuries of adolescent middle-and long-distance runners,while a greater degree of trunk tilt back,a smaller right flexion angle,a smaller hip joint adduction of the supporting leg,a smaller internal rotation an-gle when the ankle joint is on the ground,and a higher internal rotation angle when the ankle joint is off the ground make the lower limbs more prone to sports injuries.Moreover,such runners with low-er hip joint extension power and higher knee joint abduction power in their support legs are more like-ly to suffer from lower limb injuries.
9.Correlation between serum 25-hydroxyvitamin D and inflammatory cytokines in neonatal pneumonia

Objective To analyze the correlation between serum 25-hydroxyvitamin D[25(OH)D]and inflammatory cytokines such as interferon γ(IFN-γ),C-reactive protein(CRP)and interleukin-2(IL-2)in neonatal pneumonia.Methods A total of 100 neonates admitted to First Hospital of Qinhuangdao from December 2018 to December 2020 were enrolled in this study.According to the serum 25(OH)D level,they were divided into deficiency group(≤15.0 μg/L,n=18),inadequate group(15.1-20 μg/L,n=42),and adequate group(>20.0 μg/L,n=40).The general and clinical data such as gender,gestational age,serum 25(OH)D level,birth weight,mode of delivery and the levels of IFN-γ,CRP and IL-2 were compared among the three groups.The correlation between vitamin D deficiency and levels of IFN-γ,CRP and IL-2 was analyzed.Results There were no significant differences in gestational age,sex,birth weight,or delivery mode among the three groups(P>0.05).Serum 25(OH)D levels in the three groups were as follows:deficiency group<inadequate group<adequate group,and there were significant differences in the serum 25(OH)D level between groups(P<0.05).The levels of IFN-γ,CRP and IL-2 in the deficiency group were higher than those in the inadequate group and adequate group(P<0.05),and the levels of IFN-γ,CRP and IL-2 in the inadequate group were higher than those in the adequate group(P<0.05).The Pearson correlation analysis showed that serum 25(OH)D level was negatively correlated with IFN-γ,CRP and IL-2 levels in newborns(P<0.05).Conclusion The lower the serum 25(OH)D in newborns,the more deficient the vitamin D and the higher the levels of IFN-γ,CRP and IL-2 are,indicating higher risk of pneumonia.
10.Comparison of different blood vessels as markers in laparoscopic radical resection of right colon cancer

Objective To compare the clinical efficacy and short-term prognosis of laparoscopic radical resection of right colon cancer guided by superior mesenteric artery and superior mesenteric vein.Methods 80 patients with right colon cancer of cT2-4 and/or N0-2M0 admitted from January 2020 to October 2022 were selected as the research objects,and they were randomly divided into observation group and control group,with 40 patients in each group.The observation group was treated with SMA-oriented laparoscopic radical resection of right colon cancer,while the control group was treated with SMV-oriented laparoscopic radical resection of right colon cancer.The curative effect and prognosis of the two groups were compared.Results There was no significant difference between the two groups in general condition,operation time,gastric tube placement time,recovery time of farting,postoperative fasting time,postoperative drainage time,postoperative nutritional index,total incidence of complications and postoperative hospitalization time(P>0.05).The lymph nodes in the observation group were significantly more than those in the control group,and the difference was statistically significant(P<0.05).In the observation group,the lymph nodes in the anterior and left side of superior mesenteric artery were examined(No.D3),and 273 lymph nodes were detected,and Seven patients(17.5% )were diagnosed with D3 metastasis,and 13 lymph nodes were positive(5.2% ).Conclusion Laparoscopic radical resection of right colon cancer guided by superior mesenteric artery,without increasing the incidence of complications and high safety,can more thoroughly clean lymph nodes and reduce tumor recurrence,which is expected to significantly improve the prognosis of patients.
