Dysfunction of the Hippo pathway enables cells to evade contact inhibition and provides advantages for cancerous overgrowth. However, for a significant portion of human cancer, how Hippo signaling is perturbed remains unknown. To answer this question, we performed a genome-wide screening for genes that affect the Hippo pathway in Drosophila and cross-referenced the hit genes with human cancer genome. In our screen, Prosap was identified as a novel regulator of the Hippo pathway that potently affects tissue growth. Interestingly, a mammalian homolog of Prosap, SHANK2, is the most frequently amplified gene on 11q13, a major tumor amplicon in human cancer. Gene amplification profile in this 11q13 amplicon clearly indicates selective pressure for SHANK2 amplification. More importantly, across the human cancer genome, SHANK2 is the most frequently amplified gene that is not located within the Myc amplicon. Further studies in multiple human cell lines confirmed that SHANK2 overexpression causes deregulation of Hippo signaling through competitive binding for a LATS1 activator, and as a potential oncogene, SHANK2 promotes cellular transformation and tumor formation in vivo. In cancer cell lines with deregulated Hippo pathway, depletion of SHANK2 restores Hippo signaling and ceases cellular proliferation. Taken together, these results suggest that SHANK2 is an evolutionarily conserved Hippo pathway regulator, commonly amplified in human cancer and potently promotes cancer. Our study for the first time illustrated oncogenic function of SHANK2, one of the most frequently amplified gene in human cancer. Furthermore, given that in normal adult tissues, SHANK2's expression is largely restricted to the nervous system, SHANK2 may represent an interesting target for anticancer therapy.
2.Statistics and Analysis of 136 Cases of Adverse Events of International Medical Devices.

To explore the law and characteristics of adverse events of medical devices and to provide research methods and basis for reducing the recurrence of similar adverse events, we collect medical devices safety information from five representative countries in the world, and make statistics and analysis on the types of events, the types of management and the causes of events. The results show that among 136 serious adverse events, the top three causes of recall are product design factors, software factors, and component defects. In order to reduce the application risk of medical devices, it is suggested that product designers, operating users and medical institutions should correctly implement the monitoring and evaluation system of medical devices.

3.Professional training needs of caregivers for the elderly in Shaanxi province

Objective? To explore the professional training will and needs of caregivers for the elderly in Shaanxi province and to provide reference for completing the training program for caregivers for the elderly, clarifying the content of training and making relevant policies. Methods? Totally 1 390 caregivers for the elderly from 143 nursing homes in Shaanxi province were selected by convenient sampling from January to September 2017 and investigated with the self-made questionnaire for their will and needs of professional training. χ2 test was used to identify the influencing factors to the caregivers' will of professional training, and the statistically significant variables were included into the Logistic regression model to analyze the influencing factors to professional training of caregivers for the elderly in Shaanxi province. Results?(1) Professional training will:1 085 caregivers (78.1%) were willing to participate in professional training, while 305 caregivers (21.9%) were not willing to do so. Logistic regression analysis showed that educational background, marital status, needs for professional knowledge, certificate of caregivers, effects of previous training and economic income were the influencing factors to the professional training will of caregivers for the elderly in Shaanxi province (P<0.05). (2) Professional training needs: the form of training was mainly short-term training (48.4%), and the cost of training was mostly free of charge or borne by their institutions (68.7%). Their desired form of training was graded training (64.3%), and the content of training focused primarily on daily life care (83.7%) and basic care (82.2%). Conclusions? The proportion of caregivers for the elderly in Shaanxi province who are willing to receive professional training is high. Targeted graded professional training should be provided according to different situations of caregivers so as to improve the service of care for the elderly. The government or nursing homes are advised to provide part-time and short-term professional training which focuses primarily on daily life care and psychological care, complete training plans and meet social needs, without increasing economic burden on caregivers for the elderly.
4.Effect of hierarchical intervention on hypertension prevention and control among middle-aged and elderly people in rural area

Objective To discuss the effect of hierarchical intervention on hypertension prevention and control in rural area,so as to improve awareness rate and health promoting behaviors about the hypertension disease among middle-aged and the elderly in rural area. Methods A gradually deepening intervention scheme was implemented hierarchically of levels of entire population,prehypertension group and hypertension group from March 2014 to February 2016. Results After the intervention, score of health-promoting lifestyle in the research group was(132.86±16.70),significantly higher than that in the control group(113.72±20.86),and also within the group,higher than before the intervention (108.47±16.161)(P < 0.01). Value of blood pressure,regular behavior of measurement and body mass index in the research group were all better than those in the control group and before the intervention (P < 0.05). Awareness rate of knowledge related to hypertension in the research group was significantly higher than those in the control group and before the intervention(P<0.01). Conclusions The gradually deepening hierarchical health education intervention among middle-aged and elderly people in rural communities can prevent and control hypertension by improving awareness rate of the disease and help developing health promoting behaviors.
5.The gender differences and psychological factors for the anal rectal pressure of senile patients with functional constipation

Objective To investigate the gender differences and psychological influencing factors for anal rectal function in senile patients with functional constipation (FC) .Methods The gender differences and correlation of anal rectal function and psychological factors were compared in elderly patients and control group by anorectal manometry and psychological testing methods .Results The anal pressure drops of analog defecation of elderly female and male patients were attenuated and the rectal sensation thresholds increased compared with that of control group ,and the maximal squeezing pressure ,the rectal defecation pressure and anorectal pressure difference of elderly female patients were attenuated compared with that of elderly male patients .The SAS and SDS stand‐ard score of elderly female and male patients were significantly higher than that of control group .The SAS standard score of elderly female patients were higher than elderly male patients .SAS and SDS were negatively correlated with anorectal pressure difference and positively correlated with rectal first sensation volume (FSV) .Conclusion Anorectal dysfunction and psychological factors were involved in the elderly FC pathogenesy ,and they were related .Elderly female patients suffered more severe anxiety and anorectal motility disorders than male patients .
6.Effect ofCoreopsis tinctoria Nutt. from Different Extract Regions on Proliferation and Differentiation of 3T3-L1 Preadipocytes

The 3T3-L1 preadipocytes were used as carriers in the investigation of total extract, n-butanol extract, CB-1 and CB-2 of Coreopsis tinctoria Nutt. on cell proliferation and differentiation. Three groups at different doses were set for each of the four extract regions of C. tinctoria Nutt., respectively. MTT assay was used to detect 3T3-L1cell proliferation by four extract regions of C. tinctoria Nutt. Oil Red O staining was used to analyze the formation and accumulation of cytoplasmic lipid during cell differentiation. The results showed that compared with the control group, there were significant inhibition on cell proliferation when thetotal extract of C. tinctoriaNutt. at 100 μg·mL-1, n-butanol extract at 0.5, 5, and 50 μg·mL-1, CB-1 and CB-2 at 50 μg·mL-1 (P< 0.01). N-butanol extract showed certain dose-dependent manner (r = -0.903). Oil Red O staining showed that compared with the control group, thetotal extract of C. tinctoria Nutt. at 1, 10, 100 μg·mL-1 can obviously inhibit cell differentiation, reduce the formation of cytoplasmic lipid (P< 0.01). N-butanol extract can inhibit cell differentiation in a dose-dependent manner (r= -0.779). CB-1 and CB-2 obviously inhibited cell differentiation at the concentration of 50 μg·mL-1 (P < 0.01). It was concluded that thetotal extract, n-butanol extract, CB-1 and CB-2 of C. tinctoria Nutt. can inhibit the proliferation and differentiation of 3T3-L1 preadipocytes and reduce the formation of cytoplasmic lipid.
7.The exploration of optimum cycle of automatic washer disinfector for endozime auto wash triple plus with advanced proteolytic action

Objective To explore optimum cycle of Automatic Washer Disinfector for endozime auto wash triple plus with advanced proteolytic action to ensure mechanical cleaning quality and improve safety of reusable medical instruments. Methods Visual inspection, STF cleaning efficacy monitoring test and Adenosine Triphosphate (ATP) test were used to verify cleaning quality of the newly updated No.4 automatic washer disinfector, and changed key parameters on weekly basis, such as pre-cleaning temperature, pre-cleaning time, detergent concentration, main cleaning temperature, and main flushing time, recorded cleaning results for three weeks:Group test of initial procedure, Group test 1 of optimization procedure and Group test 2 of optimization procedure.Χ2 test was used to compare differences of the three groups and determine the optimum cycle. Results Group test 2 of optimization procedure had the highest visual inspection and ATP test qualification rate (both were 100.00%), and pass all the STF tests. Cleaning quality was significantly better than the other two groups, and the cycle made full use of the tested detergent. Conclusions Cycle parameters of cleaning machine have great impact on medical instruments cleaning quality; Specific consuming products need to be verified to determine the optimum cleaning cycle.
8.The gender differences of anal rectal function and depression and / or anxiety in elderly patients with constipation

Objective To investigate the gender differences of anal rectal function and depression and/or anxiety in elderly patients with functional constipation (FC).Methods Twenty-seven elder female FC patients (elder female patients group),20 elder male FC patients (elder male FC patients group),28 elder healthy female (elder healthy female group) and 24 elder healthy male (elder healthy male group) were selected.The gender differences and correlation of anal rectal function and psychological factors were compared in elderly patients and control group,by using anorectal manometry and psychological testing methods.Results Compared with elder healthy female group,maximum squeeze pressure (MSP),anorectal pressure decreased in elder healthy male group(P < 0.05 or < 0.01),anal canal overpressure,first sensation volume(FSV),defecating sensation volume(DSV),maximum tolerable volume(MTV) increased in elder female patients group (P< 0.01).Compared with elder male healthy group,anal canal excess pressure increased,anorectal pressure decreased (P< 0.05),and FSV,DSV,MTV increased in elder male patients group (P < 0.01 or < 0.05).MSP,rectum systolic pressure,anorectal pressure in elder female patients group was lower than that in elder male patients group (P < 0.05 or < 0.01),rectal tube excess pressure and rectal sensation threshold was higher than that in elder male patients group,but there was no significant difference (P > 0.05).RRP,rectum systolic pressure,anorectal pressure in elder female healthy group was lower than that in elder male healthy group (P < 0.01).FSV was higher than that in elder male healthy group (P < 0.01).SAS score and SDS score was higher in elder female patients group was higher than that in elder female healthy group (P< 0.01),SAS score and SDS score was higher in elder male patients group was higher than that in elder male healthy group(P< 0.05 or < 0.01).SAS score was negatively correlated with MSP,rectum systolic pressure,anorectal pressure(P < 0.01),was positively correlated with FSV(P < 0.05).SDS score was negatively correlated with anorectal pressure (P < 0.01),was positively correlated with FSV,DSV,MTV (P <0.01).Conclusions Anorectal dys function and psychological factors are involved in the elderly FC pathogenesy,and the two are related.Elderly female patients exist more severe anxiety and anorectal motility disorders than male patients.
9.Clinical significance of detection of serum procalcitonin and high-sensitivity C-reactive protein in AECOPD

Objective To evaluate the clinical significance of detection of serum procalcitonin(PCT),high sensitivity-C-reactive protein(hs-CRP)and leukocyte in acute exacerbation of chronic obstructive pulmonary disease(AECOPD)patients.Methods 45 patients with AECOPD and 50 patients(AECOPD group)with chronic obstructive pulmonary disease(COPD)in slationary phase (COPD group)were selected,and their serum levels of PCT,hs-CRP and leukocyte were tested.Results Before treatment,the con-centrations of PCT and hs-CRP were (3.32±1.81)ng/mL and (22.36±5.03)mg/L in AECOPD group respectively.They were significantly higher than those in COPD group with statistical differences(P<0.01).The positive predictive value of PCT and hs-CRP was 68.9% and 60.0% in AECOPD group respectively,which were significantly higher than those in COPD group(P<0.01).After antibiotic treatment,the leukocyte,PCT,and hs-CRP levels in AECOPD group were significantly lower than those be-fore treatment(P< 0.01).Conclusion In AECOPD patients,PCT and hs-CRP are good diagnostic indexes.
10.Relationship of Arrhythmia and Electrical Parameters after Transcatheter Closure of Ventricular Septal Defect

Objective To study the ralationship of arrhythmias and heart electrical parameters changes after transcatheter closure of ventricular septal defect (VSD) .Method 50 patients had been successfully finnished the transcatheter closure of ventricular septal defect,and then we observed ralationship of arrhythmias and heart electrical parameters changes. Results (1) Compared with the situation of VSD occluder preoperative and postoperative, during follow-up there were 20 arrhythmia cases, including bundle branch block 16 cases, including the complete left bundle branch block 2 cases,complete right bundle branch block 4 cases,a transient third-degree trioventricular conduction block 1 case, most return to normal in the follow-up. 2 Intracavitary electrogram showed before and after transcatheter closure:A-V conduction parameters A-H,H-V value of (75.10 ± 14.34) ms vs (80.67±23.03) ms and (47.9±12.61) ms vs (50.07±15.23) ms,the difference was not statistically significant ( >0.05) . Conclusions (1) Some patients with new ECG changes after transcatheter closure of VSD, manifested as an increase in bundle branch block in a week, but most return to normal,the prognosis is good. (2) Intraoperative A-H,H-V extension is not related with with postoperative slow arrhythmia;(3) Arrhythmia is a common complication after VSD transcatheler closure, to strictly selecte indications,and to avoid too large diameter are the effective measures to reduce the arrhythmias after transcatheter closure of ventricular septal defect (VSD) .
