Objective:To analyze the differences in determination of fluoride level in drinking water by ion-selective electrode method and high-throughput rapid determination method. Methods:The precision test was carried out by using the two methods to measure two kinds of fluoride standard substances, water samples of external quality control assessment from 2021 to 2023 (two kinds each year) and the fluoride level in three drinking water samples (for 5 times/each sample). Accuracy testing was conducted by measuring the external quality control assessment water samples and the spiked recovery rates drinking water, and water samples were grouped (water fluoride ≤1.00, > 1.00 mg/L) and analyzed according to the "Hygienic Standards for Drinking Water" (GB 5749-85). SPSS 23.0 software was used for statistical analysis of the measurement results.Results:(1) The correlation coefficients ( r) of the working curves of the two methods were both > 0.990, meeting the quality control requirements. (2) In the precision test, when comparing the results of the two methods for detecting two kinds of fluoride standard substances, there was no statistically significant difference ( F = 0.36, 0.15, P = 0.564, 0.707), and the coefficients of variation ( CV) were all < 5%. The CV of the detection results of the external quality control assessment water samples and drinking water samples were < 5%. (3) In the accuracy test, when the fluoride concentration in water was ≤1.00 mg/L, there was no statistically significant difference in the spiked recovery rates between the two methods ( F = 0.49, P = 0.504). When the fluoride concentration in water was > 1.00 mg/L, there was a statistically significant difference in the spiked recovery rates between the two methods ( F = 24.75, P = 0.003). Conclusions:The ion-selective electrode method has the advantages of wide detection range and wide adaptability, while the high-throughput rapid determination method has high accuracy. Testing personnel can weigh and choose the appropriate determination method based on the actual laboratory conditions and sample concentration range.

OBJECTIVE To investigate the expression characteristics and regulatory mechanisms of the circadian clock gene period circadian regulator 1(PER1)and the tumor suppressor gene serine peptidase inhibitor Kazal type 5(SPINK5)in head and neck squamous cell carcinoma(HNSCC),and to elucidate the molecular mechanism by which PER1 regulates SPINK5 transcription via the NF-κB signaling pathway.METHODS Differentially expressed genes in HNSCC were screened using The Cancer Genome Atlas(TCGA)and GSE205155 datasets. The association between SPINK5 expression and patient prognosis was assessed via the GEPIA database.mRNA and protein expression levels of SPINK5 and PER1 in 60 clinical samples were detected by RT-qPCR,immunohistochemistry,and Western blot.PER1 knockdown(using siRNA)and overexpression(via plasmid transfection)were performed in the AMC-HN-8 cell line.Wound healing and colony formation assays were applied to evaluate the effects of PER1,SPINK5,and their interaction on HNSCC cell migration and proliferation.Western blot was utilized to examine the regulatory effect of NF-κB on SPINK5.RESULTS SPINK5 and PER1 were significantly downregulated in HNSCC tissues(all P<0.01),and their low expression was correlated with poor patient prognosis(for SPINK5,HR=0.69,P=0.006 7).A significant positive correlation was observed between PER1 and SPINK5 expression(R2=0.719 2,P=0.001 0).Knockdown and overexpression of PER1 respectively resulted in synchronous alterations in SPINK5 mRNA levels(all P<0.05).PER1 knockdown enhanced cell migration and proliferation(P<0.05),whereas SPINK5 overexpression suppressed these capabilities(P<0.01).Importantly,SPINK5 overexpression reversed the phenotypic changes induced by PER1 knockdown.Mechanistically,PER1 overexpression led to concomitant changes in NF-κB expression, activating the NF-κB pathway and thereby promoting SPINK5 transcription.CONCLUSION PER1 positively regulates SPINK5 transcription via the NF-κB pathway, inhibiting HNSCC cell proliferation and migration. These findings suggest that PER1 and SPINK5 may serve as potential therapeutic targets for HNSCC.

Objective:To investigate the effects of electroheating on pathologic pain, endoplasmic reticulum stress and EGFR/JAK/STAT signaling pathway in rats with knee osteoarthritis. Methods:Totally 40 SPF SD male rats were divided into normal group (10 rats) and modeling group (40 rats) with random number table method. The model of knee osteoarthritis was prepared by injecting sodium iodoacetate into the joint cavity. The successfully modeled rats were divided into model group, sodium hyaluronate group, and electroheating needle group, with 10 rats in each group. After successful modeling, sodium hyaluronate group was injected with 0.1 ml sodium hyaluronate into joint cavity. The electrothermal acupuncture group was treated with electrothermal acupuncture, and the normal group and the model group were given the same volume of normal saline at the same time, once a day, for two weeks. The mechanical pain threshold (PWT) and thermal pain threshold (PWL) in rats were detected. The pathological morphology of cartilage was detected by HE staining. Immunohistochemical staining was used to detect the expressions of glucose regulated protein 78 (GRP78) and C/EBP homologous protein (CHOP) in cartilage tissue. Western blot was used to detect the expressions of EGFR, JAK, and STAT proteins, while RT-PCR was used to detect the mRNA levels of GRP78, CHOP, EGFR, JAK, and STAT in cartilage tissue. Results:The normal group had normal cartilage tissue structure, orderly distribution of synovial cells, neat arrangement of chondrocytes, and no inflammatory cell infiltration. The model group had synovial hyperplasia and hypertrophy, irregular arrangement of synovial cells and a large number of hyperplasia, disordered arrangement of chondrocytes and increased number of inflammatory cell infiltration. Compared with the model group, the PWT and PWL in cartilage tissues of sodium hyaluronate group and electrothermal acupuncture group increased ( P<0.05), the expressions of GRP78 and CHOP in cartilage tissues decreased ( P<0.05), the protein expressions of EGFR, JAK1 and STAT1 in cartilage tissue decreased, and the mRNA expressions of GRP78, CHOP, EGFR, JAK1 and STAT1 in cartilage tissue decreased ( P<0.05). Conclusion:Electroheating needle can improve pathological pain in knee osteoarthritis model rats, and its mechanism may be related to inhibiting endoplasmic reticulum stress and regulating the EGFR/JAK/STAT signaling pathway.
