1.Effects of Moluodan Dami Pills on chronic atrophic gastritis rats

AIM To investigate the effects of Moluodan Dami Pills on chronic atrophic gastritis(CAG)rats and their mechanism.METHODS The rat models were randomly divided into the model group,the low-dose group and high-dose Moluodan Dami Pills groups(2.43 g/kg and 4.86 g/kg),and vitamin A group(0.32 g/kg),following the 16 weeks successful induction of CAG by five-factor modeling method,in contrast to another 10 normal rats of the control group.After 8 weeks corresponding administration,the rats of each group had their general physiological status and pH value of gastric juice assessed;their pathological changes of gastric mucosa observed by naked eyes combined with HE staining;their changes of gastrin-secreting cells(G cells)and somatostatin-secreting cells(D cells)in gastric mucosa observed by immunohistochemistry;and their serum levels of pepsinogen Ⅰ/pepsinogen Ⅱ(PG Ⅰ/PG Ⅱ)ratio,TNF-α and IL-6 detected by ELISA.RESULTS Compared with the model group,the groups intervened with Moluodan Damei Pills and vitamin A displayed lower pH values of gastric juice(P<0.05),improved pathological changes of gastric mucosa,increased G and D cells counts(P<0.05,P<0.01),increased ratio of serum PGⅠ/PGⅡ,and decreased levels of IL-6 and TNF-α(P<0.05,P<0.01).CONCLUSION Moluodan Dami Pills can effectively improve the symptoms of CAG rats through its influence on the number and secretion abilityof G and D cells,the levels of serum PG Ⅰ/PG Ⅱ ratio and inflammatory factors.
2.Effects of salidroside pretreatment on high glucose-induced epithelial-mesen-chymal transition in ARPE-19 cells and its mechanism

Objective To explore the effects of salidroside(SAL)on high glucose-induced epithelial-mesenchymal transition in ARPE-19 cells and its mechanism.Methods The appropriate glucose concentration for stimulating ARPE-19 cell proliferation was determined to be 50 mmol·L-1 using the MTT method.ARPE-19 cells were divided into the normal control group(Control group)(cultured with 5 mmol·L-1 D-glucose and 45 mmol·L-1 mannitol),high glucose group(HG group)(cultured with 50 mmol·L-1 glucose),HG+low SAL group,HG+medium SAL group,and HG+high SAL group,with low,medium,and high SAL concentrations of 20 μmol·L-1,80 μmol·L-1,and 320 μmol·L-1,respective-ly.After 4 hours of SAL pretreatment,50 mmol·L-1 glucose was added to treat cells for 48 hours.The cell proliferation rate was detected using the MTT method;the cell migration activity was detected using the scratch experiment;the expres-sion levels of α-smooth muscle actin(α-SMA)and Vimentin in cells were measured using the immunofluorescence;the protein expression levels of α-SMA,Vimentin,fibronectin(FN),collagen type Ⅰ(Col Ⅰ),E-Cadherin,transforming growth factor-β1(TGF-β1),p-Smad2 and p-Smad3 in cells were measured using Western blot.Results Compared with the Control group,the proliferation rate and wound healing percentage of ARPE-19 cells in the HG group increased;the relative expression levels of α-SMA,Vimentin,FN,and Col Ⅰ proteins in the cells increased;the relative expression level of E-cad-herin protein in the cells decreased;the relative expression levels of TGF-β1,p-Smad2,and p-Smad3 proteins in the cells increased,and the differences were statistically significant(all P<0.01).Compared with the HG group,the proliferation rate and wound healing percentage of ARPE-19 cells in the HG+low SAL group,HG+medium SAL group,and HG+high SAL group decreased,the relative expression levels of α-SMA,Vimentin,FN,Col Ⅰ,TGF-β1,p-Smad2,and p-Smad3 pro-teins in the cells decreased in a concentration dependent manner,while the relative expression level of E-cadherin protein increased in a concentration dependent manner,and the differences were statistically significant(all P<0.05).Conclu-sion SAL can reduce the proliferation and migration ability of high glucose-induced ARPE-19 cells and intervene in their epithelial-mesenchymal transition process.This may be related to SAL's inhibition of the TGF-β/Smads signaling pathway in ARPE-19 cells.
3.Latent class analysis of therapeutic effects of Huoxiang Zhengqi Oral Liquid on patients with cold due to wind-cold and dampness stagnation.

The demographic characteristics, drug use, clinical symptoms and other data of the patients treated with Huoxiang Zhengqi Oral Liquid were collected. The latent class analysis(LCA) was performed to reveal the distribution of primary symptoms, and then the doubly robust estimation was employed to analyze the efficacy of different doses of Huoxiang Zhengqi Oral Liquid in different populations. A total of 11 273 patients were enrolled in this study, including 4 347 males and 6 926 females. A total of 15 primary symptoms were included, with the top 3 being dizziness(53.9%), head heaviness(39.6%), and nausea(39.4%). According to the LCA, the population was divided into 3 groups of spleen deficiency and dampness exuberance(5 604 cases, 49.71%), upward harassing of wind-phlegm(4 955 cases, 43.96%), and spleen Qi deficiency(714 cases, 6.33%). Double robust estimation showed that the time to recovery of the patients with spleen Qi deficiency had no significant difference regarding the daily dose. Compared with the daily dose of 20 mL, the daily doses of 40, and 60 mL shortened the time to recovery by 16.67(95%CI[5.23, 28.12]) and 15.94 h(95%C1[7.45, 24.43]) in the patients with upward harassingof wind-phlegm, the daily doses of 30, 40, and 60 mL shortened the time to recovery by 4.42(95%CI[1.06, 7.77]), 13.07(95%CI[1.61, 24.54]), 13.92 h(95%CI[5.14, 22.70]) in the patients with spleen deficiency and dampness exuberance, respectively. The patients with cold due to wind-cold and dampness stagnation had more primary syndromes, and the disease locations were mainly in the head, stomach, and spleen. In clinical practice, the daily dose of Huoxiang Zhengqi Oral Liquid can be increased according to the symptoms of patients, which can shorten the time to recovery.

4.Material Basis and Pharmacological Action of Tongxie Yaofang: A Review

Tongxie Yaofang, also known as Baizhu Shaoyaosan, was first recorded in Danxi's Experiential Therapy (《丹溪心法》) by ZHU Danxi in the Yuan dynasty. It is composed of Atractylodis Macrocephalae Rhizoma, Paeoniae Radix Alba, Citri Reticulatae Pericarpium, and Saposhnikoviae Radix, and serves as the representative prescription for the treatment of painful diarrhea. It has the functions of tonifying the spleen, emolliating the liver, relieving pain, and checking diarrhea, and is mainly used in the treatment of gastrointestinal diseases such as irritable bowel syndrome (IBS) and ulcerative colitis (UC). In addition, it is effective in treating gastrointestinal disorders with mental and psychological abnormalities, as well as obstinate anorexia in children, depression syndrome, and respiratory diseases. Experimental research and clinical practice have shown that Tongxie Yaofang has multi-component, multi-pathway, and multi-target characteristics in the treatment of diseases. The mechanism of Tongxie Yaofang in treating diseases is mainly attributed to anti-inflammation, immune function regulation, intestinal hypersensitivity improvement, emotion regulation, etc. Monoterpene glycosides, flavonoids, chromones, lactones, and other components contained play an important therapeutic role. The research on the systems biology of Tongxie Yaofang, such as metabolomics, proteomics, and network pharmacology, provides a scientific basis for clarifying its mechanism of action and expanding its clinical application. However, there are still some problems to be solved, such as difficulty in combining diseases and syndromes and lack of in-depth systematic research. Through the retrieval and collation of relevant literature, this paper systematically reviewed the material basis, pharmacological effects, and systems biology research of Tongxie Yaofang, aiming to lay a foundation for in-depth research on its mechanism in treating diseases and rational application in clinical practice.
5.Influence of exophthalmos on design of double eyelid incision

Objective:To compare the postoperative effects of double eyelid surgery with different exophthalmos to find its influence on the surgery and necessary changes in preoperative design and during operation.Methods:A total of 50 female patients with single eyelid seeking beauty from June 2021 to March 2022 were selected from the Department of Plastic Surgery, Affiliated Hospital of Qingdao University. The ocular protrusion was measured by HETEL ophthalmostatometer before surgery. Both eyes at 12-15 mm were taken as normal group ( n=26), both eyes at 16-18 mm as mild protrusion group ( n=14) and both eyes at 19-22 mm as severe protrusion group ( n=10). All the patients were treated with double-eyelid surgery by orbital septum and unified postoperative nursing. Results:After six months of follow-up, there was no difference in eyelid width with closed eyes (all P>0.05). The width of double eyelid with open eyes in normal group was smaller than that in mild protrusion group ( F=23.23, all P<0.05), and the width of double eyelid with open eyes in mild protrusion group was smaller than that in severe protrusion group ( F=47.70, all P<0.05). There was no difference in the improvement rate of facial aesthetics among the three groups ( P>0.05). The " feeling of meet" and scar formation in the normal group were less than those in the mild protrusion group ( F=16.92, F=33.45, all P<0.05), and the " feeling of meet" and scar formation in the mild protrusion group were less than those in the severe protrusion group ( F=27.93, F=28.53, all P<0.05). The improvement rate of normal group was higher than that of mild and severe protrusion group (χ 2=7.25, 7.89, all P<0.05). There was no difference in the improvement rate between the mild and severe protrusion groups ( P>0.05). Conclusions:In clinical practice, it is necessary to make corresponding changes in the preoperative design and operation of double eyelid surgery for patients with high eyeball protrusion.
6.Comparison of the application methods and effects of skin flaps in the repair of defects in different parts of the nose according to the concept of aesthetic nosesubunits.

Objective: To analyze the difference of application methods and effects of local flap in small and medium-sized defects of different aesthetic subunits of nose, in order to provide reference for clinical work. Methods: A retrospective analysis was made on 59 patients with external nasal masses and scars who underwent surgical treatment in the Department of Aesthetic Plastic Surgery of the Affiliated Hospital of Qingdao University from July 1, 2021 to January 30, 2022, including 27 females and 32 males, aged 15 to 69 years. Using Likert scale, the repair methods and effects of local flap for nasal soft tissue defects were evaluated and summarized from three aspects of texture, flatness and scar concealment. GraphPad Prism 5.0 software was used for data statistics and analysis. Results: The use of skin flaps to repair small and medium-sized areas of the nose could achieve satisfactory results. For patients with different subunits, in terms of skin flatness and scar concealment degree in the operation area, patients' satisfaction with the dorsal and lateral nasal areas was higher than that of the alar and tip areas, respectively (F=6.40, P=0.001; F=10.57, P<0.001). For patients with different skin flap repair methods, the satisfaction of patients with Z-plasty and Dufourmentel skin flap was higher than that of other skin flap repair methods (F=4.38, P=0.002), and the satisfaction of patients with Dufourmentel skin flap was the highest in the degree of scar concealment (F=2.57, P=0.038). Conclusions: In the small and medium-sized defects of the nose, the use of multiple local flaps can achieve good cosmetic effects and functional recovery. The operator should select the appropriate flap repair method according to the characteristics of different aesthetic subunits of the nose.
7.Phenotype and RHD gene polymorphism of 64 Rh negative individuals

【Objective】 To collect blood samples of 64 RhD negative patients in our hospital for RHD genotyping and phenotype analysis (RhC/c/E/e), and analyze the distribution characteristics of different RHD genotypes. 【Methods】 The RHD gene of RhD negative patients was genotyped by fluorescence quantitative polymerase chain reaction (PCR) method. The Rh phenotype was identified by IgM anti-e, anti-c, anti-C and anti-E, respectively. 【Results】 Forty-two cases of RHD deletion were detected, dominated by ccee phenotype (88.1%); 9 of RHD1227A cases, dominated by Ccee phenotype(77.8%); 8 of RHD-CE(3-9)-D2 cases, dominated by Ccee phenotype (75%); 1 of RHD-CE(3-10)-D2 case with Ccee phenotype, 1 of RHD*711delC case; 1 of RHAG site invalid type were detected. The typing results could not be determined in 2 cases by PCR method. 【Conclusion】 RhD negative patients showed diversity in RHD genotype, dominated by RHD deletion, followed by RHD1227A, RHD-CE(2-9)-D2, RHD-CE(3-10)-D2, RHD*711delC and RHAG site deletions.
8.Serum lipocalin-2 is a potential biomarker for the clinical diagnosis of nonalcoholic steatohepatitis

Background/Aims:
Nonalcoholic steatohepatitis (NASH) is a progressive form of nonalcoholic fatty liver disease (NAFLD) characterized by hepatic steatosis, inflammation, hepatocellular injury, and fibrosis. We aimed to investigate the usefulness of a key biomarker, lipocalin-2 (LCN2), for the detection of NASH progression.
Methods:
A mouse NASH model was established using a high-fat diet and a high-sugar drinking water. Gene expression profile of the NASH model was analyzed using RNA sequencing. Moreover, 360 NAFLD patients (steatosis, 83; NASH, 277), 40 healthy individuals, and 87 patients with alcoholic fatty liver disease were recruited.
Results:
Inflammatory infiltration, focal necrosis in the leaflets, steatosis, and fibrosis were documented in the mouse liver. In total, 504 genes were differentially expressed in the livers of NASH mice, and showed significant functional enrichment in the inflammation-related category. Upregulated liver LCN2 was found to be significantly interactive with various interleukins and toll-like receptors. Serum LCN2 levels were significantly increased in NAFLD patients. Serum LCN2 levels were correlated with steatosis, intralobular inflammation, semiquantitative fibrosis score, and nonalcoholic fatty liver disease activity score. The area under the curve of serum LCN2 was 0.987 with a specificity of 100% and a sensitivity of 93.5% for NASH diagnosis, and 0.977 with almost the same specificity and sensitivity for steatosis.
Conclusions
LCN2 might be involved in the transition from NAFL to NASH by mediating inflammation. Serum LCN2 levels might be a novel biomarker for the diagnosis of NASH.

10.Current situation and influence factors of suicidal self-injurious behavior in teenagers in Guangdong

Objective:To investigate the current situation of suicidal self-injurious behavior in teenagers and analyze its influencing factors. Methods:From January to November 2020, a stratified sampling method was used to investigate 707 teenagers in Guangdong. The “questionnaire of health-related behaviors of Chinese teenagers” compiled by the Chinese Center for Disease Control and Prevention was used anonymously to investigate the current situation of suicidal self-injurious behavior of the teenagers. The chi-square test was used for univariate analysis, and logistic regression was used for multivariate analysis. Results:The incidence rate of suicidal self-injurious behavior was 14.57% (103/707). It was 13.31% (45/338) in males, 15.72% (58/369) in females, 13.83% (48/347) in junior high school students and 15.28% (55/360) in senior high school students.
