Objective To systematically evaluate the similarities between six-gene-modified pig red blood cells and human red blood cells in multiple dimensions, including cell morphology, hematological parameters, microbial safety and serological compatibility, and to verify the feasibility of using six-gene-modified pig red blood cells as an alternative for blood transfusion in cases of traumatic hemorrhage. Methods Blood samples from O-type human donor, six-gene-modified O-type pig and wild-type O-type pig were collected. The pig gene-modified phenotype was identified by flow cytometry. The morphology was observed using field emission scanning electron microscopy and the diameter was measured. Hematological tests, including blood routine, osmotic fragility test, adenosine triphosphate (ATP) and 2,3-diphosphoglycerate (2,3-DPG) content and partial pressure of oxygen at 50% hemoglobin saturation (P50), were conducted to assess the deformability of red blood cells. Real-time fluorescent quantitative polymerase chain reaction was used to screen for zoonotic viruses, and cross-matching was performed using column gel method, condensation amine method and saline tube method. Results The six-gene-modified pig red blood cells successfully knocked out the α-Gal, Neu5Gc and Sda antigens and expressed hCD55. Wild-type pig red blood cells expressed these three antigens but did not express human complement regulatory proteins. All three types of red blood cells presented a biconcave discoid morphology. The diameter of O-type human red blood cells was larger than that of the six-gene-modified porcine red blood cells and wild-type pig red blood cells (all P < 0.05). The average red blood cell volume and average red blood cell hemoglobin of O-type humans were higher than those of the six-gene-modified pig red blood cells, and the distribution width-variable coefficient of O-type human red blood cells was lower than that of the six-gene-modified pig red blood cells (all P < 0.05). There were no statistically significant differences in the permeability fragility, ATP content, 2,3-DPG content, P50 and red blood cell deformability indicators of the three types of red blood cells (all P > 0.05). The screening for zoonotic viruses in human and animal cells of the six-gene-modified pigs and wild-type pigs was negative. The cross-matching results showed that in the column gel method and condensation amine method, the primary and secondary sides of the six-gene-modified pig red blood cells did not agglutinate with human red blood cells, while the wild-type pig red blood cells agglutinated with human red blood cells in all cross-matching methods. Conclusions The six-gene-modified pig red blood cells are highly similar to human red blood cells in terms of morphology, key hematological parameters, energy metabolism, oxygen-carrying capacity, mechanical properties, microbial safety and serological compatibility, providing an experimental basis for the clinical transformation of xenotransfusion.

Journal of Public Health and Preventive Medicine 2026;37(5):73-76
Objective To analyze the characteristics of stroke incidence among residents in Changsha City from 2022 to 2023, and to provide a scientific basis for formulating stroke prevention and control measures. Methods The data were obtained from routine cardiovascular and cerebrovascular event monitoring data. The incidence rate, constituent ratio, standardized incidence rate, age incidence rate and other indicators were calculated using SPSS21.0 software. The incidence rate between different genders and between urban and rural areas was compared by χ2 tests. Results From 2022 to 2023, a total of 67 400 stroke cases were reported in 9 districts and counties (cities) of Changsha. The incidence rate and the age standardized rate were 327.40/105 and 374.68/105, respectively. The incidence rate of stroke in men was higher than that in women (χ2=2501.312, P<0.001). The incidence rate of stroke in rural areas was higher than that in urban areas (χ2=2501.312, P<0.001). Among the four subtypes of stroke, cerebral infarction was the most common (62 278 cases, 92.40%). With the increase of age, the incidence rate of stroke generally showed an upward trend, with a low incidence in those aged under 40 years old, and a significant upward trend in those aged 40 years and older. The incidence rate of stroke among residents above 85 years old was the highest, reaching 3414.25/105. Conclusion The incidence of stroke in Changsha residents is higher in men than in women, higher in rural areas than in urban areas, and higher with age. Therefore, targeted stroke prevention and control efforts should be carried out in key areas and populations.

BACKGROUND:
Spicy food consumption has been reported to be inversely associated with mortality from multiple diseases. However, the effect of spicy food intake on the incidence of vascular diseases in the Chinese population remains unclear. This study was conducted to explore this association.
METHODS:
This study was performed using the large-scale China Kadoorie Biobank (CKB) prospective cohort of 486,335 participants. The primary outcomes were vascular disease, ischemic heart disease (IHD), major coronary events (MCEs), cerebrovascular disease, stroke, and non-stroke cerebrovascular disease. A Cox proportional hazards regression model was used to assess the association between spicy food consumption and incident vascular diseases. Subgroup analysis was also performed to evaluate the heterogeneity of the association between spicy food consumption and the risk of vascular disease stratified by several basic characteristics. In addition, the joint effects of spicy food consumption and the healthy lifestyle score on the risk of vascular disease were also evaluated, and sensitivity analyses were performed to assess the reliability of the association results.
RESULTS:
During a median follow-up time of 12.1 years, a total of 136,125 patients with vascular disease, 46,689 patients with IHD, 10,097 patients with MCEs, 80,114 patients with cerebrovascular disease, 56,726 patients with stroke, and 40,098 patients with non-stroke cerebrovascular disease were identified. Participants who consumed spicy food 1-2 days/week (hazard ratio [HR] = 0.95, 95% confidence interval [95% CI] = [0.93, 0.97], P <0.001), 3-5 days/week (HR = 0.96, 95% CI = [0.94, 0.99], P = 0.003), and 6-7 days/week (HR = 0.97, 95% CI = [0.95, 0.99], P = 0.002) had a significantly lower risk of vascular disease than those who consumed spicy food less than once a week ( Ptrend <0.001), especially in those who were younger and living in rural areas. Notably, the disease-based subgroup analysis indicated that the inverse associations remained in IHD ( Ptrend = 0.011) and MCEs ( Ptrend = 0.002) risk. Intriguingly, there was an interaction effect between spicy food consumption and the healthy lifestyle score on the risk of IHD ( Pinteraction = 0.037).
CONCLUSIONS
Our findings support an inverse association between spicy food consumption and vascular disease in the Chinese population, which may provide additional dietary guidance for the prevention of vascular diseases.

Ovarian cancer (OC) is a common gynecological malignant tumor in clinical practice. In the early stage,it is often asymptomatic, while in the late stage,it mainly presents with non-specific symptoms such as abdominal distension, poor appetite, and dull abdominal pain. Some patients may also have cachexia such as weight loss and anemia. Early diagnosis is difficult, and the mortality rate ranks first among gynecological malignant tumors, making OC a major challenge in clinical treatment. The classic Chinese medicine formula Guizhi Fulingwan comes from the Jingui Yaolue and has the effects of promoting blood circulation, removing blood stasis, and reducing abdominal lumps. In recent years,it has been widely used to treat OC with good results. This article summarized the clinical application of Guizhi Fulingwan in the treatment of OC from two aspects:The analysis of its basic prescriptions and clinical research. In terms of basic prescriptions, the formula has the ability to promote blood circulation, remove blood stasis, and reduce abdominal lumps. It can exert therapeutic effects considering both water and blood aspects and reduce abdominal lumps, with characteristics of simultaneous Yang warming and heat clearing and parallel supplementation and elimination. Through the methods of "circulation" and "supplementation", it strengthens the body, dispels evil, and eliminates underlying symptoms. In clinical studies,Guizhi Fulingwan can be applied to various stages of patients with OC,which not only promotes the recovery of the body after OC surgery but also can be combined with chemotherapy and immunotherapy to synergistically treat advanced OC and enhance treatment efficacy. In addition, the formula can also alleviate various adverse reactions caused by chemotherapy, with high safety, improve patients' quality of life, prolong survival, and optimize tumor control effects. Based on the above analysis, this article elaborated on the current clinical research status of Guizhi Fulingwan combined with Western medicine in the treatment of OC and proposed suggestions and improvements to address the shortcomings in current clinical research,so as to provide reference for the clinical application of this formula in the treatment of OC and the construction of a combined traditional Chinese and Western medicine treatment model.

Objective To explore the correlation between single-leg standing posture control and ankle plantar flexor muscle stability,so as to provide a new theoretical basis for improving the ability of human posture control. Methods A total of 20 healthy male college students were randomly selected as the experimental subjects. The iBalance tester and training system was used to test the trajectory data of the center of pressure(COP)of the foot standing on one leg.The CON-TREX MJ multi-joint isokinetic test and training system was used to test the moment amplitude data during the contraction of ankle plantar flexor muscles.Univariate repeated measures variance analysis was used to analyze the standard deviation data of ankle plantar flexor moment amplitude between groups. The Pearson correlation coefficient was used to study the correlation. Results The greater the intensity of the muscle stabilization task performed by the ankle plantar flexor muscle, the greater the standard deviation of the moment amplitude. The C90 area was positively correlated with the coefficient of variation(CV)of the 10%maximum voluntary contraction(MVC)moment of ankleplantar flexor muscle(r=0.761,P<0.05)during single-leg standing without interference. The C90 area was positively correlated with the CV(r=0.632,P<0.05)of the 30%MVC moment of ankle plantar flexor muscle during single-leg standing. When the proprioception was interfered during single-leg standing, the C90 area was positively correlated with the CV(r=0.583,P<0.05)of the 20%MVC moment amplitude of ankleplantar flexor muscle. Conclusions With the increasing difficulty of muscle strength stabilization performed by the ankle plantar flexor muscles, muscle stability decreases. There is a positive correlation between ankle plantar flexor strength stability and single-leg standing posture control.Compared with the case without interference, under visual and proprioceptive interference,an additional information input is reduced or disturbed, and it is more difficult to maintain body balance, and the ankle plantar flexor muscle needs a higher muscle stability in the force mode to participate in the posture control of the human body during single-leg standing.

Journal of Medical Biomechanics 2025;40(3):638-645
Objective To classify the foot types of young Chinese males, extract characteristic indicators of foot types, and construct a standard foot type database. Methods Foot type data from 1 483 healthy young male individuals were collected.Spectral clustering algorithm was utilized to categorize foot types, and a deep neural network(DNN)was employed for training the classification model.Layer-wise relevance propagation(LRP)and the correlation coefficient method were combined to extract foot type features, and the differences in various foot type characteristics were compared. Results Spectral clustering yielded 4 distinct foot type categories.Foot type 1 was characterized by a high-arched foot with a prominent big toe, inwardly rotated 5th toe,and a high heel with a wide foot;foot type 2 was characterized by a narrow foot with hallux valgus;foot type 3 was characterized by a low-arched foot with hallux valgus;foot type 4 was characterized by a high-arched foot with a prominent big toe.By integrating interpretable neural networks and the correlation coefficient method, nine indicators were extracted from 27 foot type indicators, including the heel to the sole length, hallux height, navicular bone height, lateral malleolus height, hallux valgus angle,5th toe angle, dorsal foot circumference, heel angle, and longitudinal arch angle. The classification model constructed with these extracted indicators achieved an overall discrimination accuracy rate of 93.67%.Conclusions Chinese young males can be classified into four typical foot types.By examining the rearfoot, midfoot, and forefoot regions, nine key foot morphology parameters, including length, height, circumference, and angle, can be extracted. These parameters provide both theoretical and empirical support for establishing normative data on foot morphology for Chinese young males and for advancing biomechanical researches on the foot and ankle.
