The purpose of this study was to investigate the damage mechanism of pathogenic E.coli on mouse brain microvascular endothelial cells(BMEC cells)and mouse alveolar macrophages(MH-S cells),as well as the lung and brain of healthy mice.In this study,BMEC cells and MH-S cells were infected with pathogenic E.coli strains, and cell morphological changes were observed.Plate counting method was used to detect the adhesion and invasion ability of the strains to cells and the number of bacteria in the lungs and brains of mice.RT-qPCR was used to detect the ex-pression of TNF-α,IL-1β and IL-6 genes in cells and mouse organs at different time periods.West-ern blot was used to detect the expression of p-NF-κB,p-JAK2 and p-STAT3 proteins related to inflammation in cells and mouse organs after infection. The results showed that the cell culture medium of the infection group was turbid, the cell vision became dark and blurred, some cells shrank and died, and more fragments were produced. The adhesion rate and invasion rate of BMEC cells at 3 h were significantly lower than those at 6 h(P<0.050),and the adhesion rate and inva-sion rate of MH-S cells at 3 h were significantly higher than those at 6 h(P<0.010).Infected mice had a large area of swelling and bleeding in the brain, and the lungs had different degrees of swell-ing and bleeding. The bacterial load in the brain and lung was the highest at 12 h.Compared with the control group, the mRNA expression levels of IL-1β,IL-6 and TNF-α in the infection group were significantly increased at 3 h and 6 h(P<0.050),and the mRNA expression levels of inflam-matory factors in BMEC cells and MH-S cells were the highest at 6 and 3 h,respectively. The mR-NA expression of inflammatory factors in the brain and lung of infected mice showed a trend of in-creasing first and then decreasing with time, with the highest expression at 12 h after infection. The expression levels of p-NF-κB protein in BMEC cells,MH-S cells, lung and brain tissues of mice in the infection group were significantly higher than those in the control group(P<0.001),and the expression levels of p-JAK2 protein and p-STAT3 protein were significantly lower than those in the control group(P<0.050).The above results showed that pathogenic E.coli could adhere and invade BMEC cells and MH-S cells, colonize in lung and brain tissues of mice, promote the expres-sion of NF-κB protein in cells and tissues, inhibit the expression of JAK2 protein and STAT3 pro-tein,and then stimulate cells and tissues to produce inflammatory response.

Objective:To explore the relationship among depression, social belonging and fertility quality of life of infertile women, and to provide a theoretical basis for the development of effective measures to improve the quality of life of infertile women. Methods:From July 2022 to October 2022, a random sample of 400 patients treated for infertility in the Reproductive Assisting Centre of the First Hospital of Anhui Medical University were selected using a general basic questionnaire, the Depression Screening Self-Rating Scale (PHQ-9), the Social Attribution Measurement Questionnaire and the Fertility Quality of Life Scale quality of life questionnaire (FertiQoL) were administered. Spearman correlation was used to analyze the association between the variables, and mediating effects were examined by Bootstrap procedure and PROCESS software. Results:The 400 infertile women had a median depression score of 5, reaching a subclinical mild level of depression, a median score of 12 on the social belonging measure, and a median score of 62.87 on the FertiQoL. Social belonging was negatively correlated with depression ( r=-0.206, P<0.001); social belonging was positively correlated with FertiQoL ( r=0.267, P<0.001); and depression was negatively correlated with FertiQoL ( r=-0.428, P<0.001). Mediation analysis showed that after controlling for relevant variables, the indirect effect of depression on FertiQoL in infertile women was -0.069 0 (95% CI: -0.173 3--0.008 3), and the direct effect was -1.019 1 (95% CI: -1.436 3- -0.601 8), the mediating effect of FertiQoL through social belonging on depression in infertile women accounted for 6.34% of the total effect. Conclusion:Social belonging plays a partial mediating role in depression and FertiQoL among infertile women. Infertile women should be given timely attention to their psychological situation and given more social recognition and support to improve their FertiQoL.

To obtain the optimal preparation technology of Fang-bing nasal inhalant from components of traditional Chinese medicine by central composite design, with an apparatus containing nasal inhalant that simulated the expiration and inspiration of nose, the dissolution in vitro of different optimized inhalant samples designed through central composite design were investigated. The accumulative release of linalool, borneol, menthol was detected with GC. Response surface methodology was used to optimize the conditions of preparation technology by establishing multiple linear regression and second-order quadratic models. Then, deviation was carried out through comparing the observed and predicted values. It was showed that the coefficient of correlation of second-order quadratic model was high. The related coefficient reached 0.999 3, 0.998 0, 0.944 9, separately. The optimum conditions of preparation technology were as following: 84.39% of alcohol concentration, the weight of starch 1.45 g and the weight of carmellose sodium (CMC-Na for short) 1.22 g. The deviations between observed and predicated values showed -0.36%, 1.52%, 2.40%, separately. In this experiment, the established model can describe the good relation between factors and indexes from preparation technology of Fang-bing nasal inhalant and the outcome of prediction is well. This optimal Fang-bing nasal inhalant was used to study its in vivo effect on model rats deprived from sleep and showed sedative and sleep aiding, which will bring an instruction on inhalants of components from traditional Chinese medicine.
