1.Liuwei Dihuang Pills improve chemotherapy-induced ovarian injury in mice by promoting the proliferation of female germline stem cells.

This study primarily investigates the effect of Liuwei Dihuang Pills on the activation and proliferation of female germline stem cells(FGSCs) in the ovaries and cortex of mice with premature ovarian failure(POF), and how it improves ovarian function. ICR mice were randomly divided into the control group, model group, Liuwei Dihuang Pills group, Liuwei Dihuang Pills double-dose group, and estradiol valerate group. A mouse model of POF was established by intraperitoneal injection of cyclophosphamide. After successful modeling, the mice were treated with Liuwei Dihuang Pills or estradiol valerate for 28 days. Vaginal smears were prepared to observe the estrous cycle and body weight. After the last administration, mice were sacrificed and sampled. Serum levels of estradiol(E_2), follicle-stimulating hormone(FSH), luteinizing hormone(LH), and anti-Müllerian hormone(AMH) were measured by enzyme-linked immunosorbent assay(ELISA). Hematoxylin-eosin(HE) staining was used to observe ovarian morphology and to count follicles at all stages to evaluate ovarian function. Immunohistochemistry was used to detect the expression of mouse vasa homolog(MVH), a marker of ovarian FGSCs. Immunofluorescence staining, using co-labeling of MVH and proliferating cell nuclear antigen(PCNA), was used to detect the expression and localization of specific markers of FGSCs. Western blot was employed to assess the protein expression of MVH, octamer-binding transcription factor 4(Oct4), and PCNA in the ovaries. The results showed that compared with the control group, the model group exhibited disordered estrous cycles, decreased ovarian index, increased atretic follicles, and a reduced number of follicles at all stages. FSH and LH levels were significantly elevated, while AMH and E_2 levels were significantly reduced, indicating the success of the model. After treatment with Liuwei Dihuang Pills or estradiol valerate, hormone levels improved, the number of atretic follicles decreased, and the number of follicles at all stages increased. MVH marker protein and PCNA proliferative protein expression in ovarian tissue also increased. These results suggest that Liuwei Dihuang Pills regulate estrous cycles and hormone disorders in POF mice, promote the proliferation of FGSCs, improve follicular development in POF mice, and enhance ovarian function.

2.Association relation of C0-C2 Cobb angle and cervical disc herniation.

OBJECTIVE:
Objective To investigate the relationship between cervical disc herniation and C0-C2 Cobb angle.
METHODS:
The clinical data of 301 patients with cervical disc herniation from 2020 to 2024 were retrospectively analyzed. The median value of C0-C2 Cobb angle measurements from 301 patients was used as the boundary, cervical disc herniation patients were divided into two groups, C0-C2 Cobb angle <28.50 group and 151 patients with C0-C2 Cobb angle≥28.50 group. Among them, 150 patients in C0-C2 Cobb angle <28.50 group included 53 males and 97 females, aged 23 to 76 (57.32±12.55) years, with a disease duration of 7 to 19 (13.81±5.32) months;the othor 151 patients with C0-C2 Cobb angle≥28.50 group including 61 males and 90 females, aged 25 to 74 (56.86±12.51) years, with a disease duration of 8 to 18 (14.13±5.56) months. The cervical lordosis angle (C0-C2 Cobb angle and C2-C7 Cobb angle), T1 inclination slope (T1S) and cervical sagittal axial distance (C2-C7 SVA) were measured on the lateral cervical radiographs. The correlation between C0-C2 Cobb angle and cervical disc herniation range, protrusion position, average protrusion size and other parameters was analyzed.
RESULTS:
When the C0-C2 Cobb angle<28.50°, the average protrusion size was (2.21±0.56) mm, the C2-C7 Cobb angle was (19.92±12.06)° and the C2-C7 SVA was (1.10±1.20) mm. When the C0-C2 Cobb angle≥28.50°, the average protrusion size was (2.38±0.60) mm, the C2-C7 Cobb angle was (12.01±13.09 )°, the C2-C7 SVA was (1.53±1.36) mm, and the difference was statistically significant (P<0.05). Between the two groups of patients with C0-C2 Cobb angle < 28.50° and C0-C2 Cobb angle≥28.50°, there were significant differences in the size of C3,4, C4,5, C5,6, C6,7, C7, T1 disc herniation in single segment (P<0.05 ). C0-C2 Cobb angle was correlated with age(r=-0.135, P<0.05 ), C2-C7 Cobb angle (r=-0.382, P<0.01 ), C2-C7 SVA (r=0.293, P<0.01), average protrusion size (r=0.139, P<0.05), and the size of C3,4 (r=0.215, P<0.01 ), C4,5 (r=0.176, P<0.01 ), C5,6 (r=0.144, P<0.05 ), C6,7 (r=0.158, P<0.05 ), C7T1 (r=0.535, P<0.05) disc herniation.
CONCLUSION
There is a positive correlation between C0-C2 Cobb angle and the size of cervical disc herniation. C0-C2 Cobb angle can reflect the degree of cervical disc herniation. Previous studies have shown that the biomechanical changes between C0-C2 Cobb angle, C2-C7 Cobb angle, C2-C7 SVA and cervical extensor muscle group may be risk factors for accelerating cervical disc herniation and this may be one of the mechanisms that C0-C2 Cobb angle is positively correlated with the size of cervical disc herniation.

3.Divergent activation patterns of BRS3 revealed by two Chinese herb-derived agonists.

Bombesin receptor subtype-3 (BRS3) is an orphan G protein-coupled receptor (GPCR) that plays critical roles in energy homeostasis, glucose metabolism, and insulin secretion. Recent structural studies have elucidated BRS3 signaling mechanisms using synthetic ligands, including BA1 and MK-5046. However, the molecular basis of BRS3 activation by bioactive natural compounds and their derivatives, particularly those derived from traditional Chinese medicine, remains unclear. Here, we present high-resolution cryogenic electron microscopy (cryo-EM) structures of the human BRS3-Gq complex in both unliganded and active states bound by two herb-derived compounds (DSO-5a and oridonin), at resolutions of 2.9, 2.8, and 2.9 Å, respectively. These structures display distinct ligand recognition patterns between DSO-5a and oridonin. Although both compounds bind to the orthosteric pocket, they differentially engage the interaction network of BRS3, as demonstrated by mutagenesis studies assessing calcium mobilization and inositol phosphate 1 (IP1) accumulation. These findings enhance our understanding of BRS3 activation and provide valuable insights into the development of small-molecule BRS3 modulators with therapeutic potential.
4.Intraoperative renal collecting system damage increases risks of ipsilateral upper urinary tract stones after partial nephrectomy: a 1∶2 propensity-matched case-control study.

OBJECTIVES:
To investigate the incidence of ipsilateral upper urinary tract stones after partial nephrectomy and its risk factors.
METHODS:
We retrospectively analyzed baseline patient characteristics (age, gender, and body mass index), smoking, alcohol consumption, comorbidities (hypertension, diabetes, hypertriglyceridemia, hyperuricemia, and cardiovascular diseases), preoperative tumor conditions (tumor diameter, multiple foci, location, hemorrhage, necrosis, cystic changes, and endophytic growth), preoperative glomerular filtration rate and intraoperative factors (renal collecting system damage, ischemia time, operation time, surgical approach, and estimated intraoperative blood loss) to identify the risk factors for ipsilateral upper urinary tract stones following partial nephrectomy in our center.
RESULTS:
The overall incidence of upper urinary tract stones following partial nephrectomy was 7.80% (112/1435). The incidence of ipsilateral upper urinary tract stones was significantly higher than those of contralateral stones (4.95% vs 1.46%, P<0.001) and bilateral stones (4.95% vs 1.39%, P<0.001). Intraoperative damage to the renal collecting system was identified as a significant risk factor for ipsilateral upper urinary tract stones (OR=4.550, 95% CI: 2.237-9.252, P<0.001). Diabetes was a probable risk factor for secondary ipsilateral upper urinary tract stones after partial nephrectomy (OR=2.419, 95% CI: 0.973-6.012, P=0.057).
CONCLUSIONS
The incidence of ipsilateral upper urinary tract stones after partial nephrectomy is higher than that of contralateral and bilateral stones. Intraoperative renal collecting system damage is a risk factor for secondary ipsilateral upper urinary tract stones after partial nephrectomy.

5.Dose-effect relationship between dexmedetomidine and ropivacaine during pericapsular nerve group block in elderly patients undergoing total hip arthroplasty

BACKGROUND:Ropivacaine and dexmedetomidine are two commonly used local anesthetic agents and adjuvants in elderly total hip arthroplasty. They provide sedation and analgesic effects during peripheral hip nerve blocks.OBJECTIVE:To explore the dose-effect relationship between dexmedetomidine and ropivacaine in continuous sedation during pericapsular nerve group block in elderly patients undergoing total hip arthroplasty.METHODS:A total of 120 elderly patients who received total hip arthroplasty in Dongguan Hospital,Guangzhou University of Chinese Medicine from January 2021 to January 2023 were selected as study subjects and randomly divided into two groups 1∶1 matching according to propensity matching method. The observation group (n=60) received pericapsular nerve group block by giving 20 mL mixture of ropivacaine and 1 μg/kg dexmedetomidine. The control group (n=60) received received pericapsular nerve group block by giving 20 mL of ropivacaine. The general data,perioperative indexes,postoperative analgesia,hemodynamic indexes,inflammatory factors,stress indexes,postoperative adverse reactions,length of hospital stay,and the occurrence of postoperative complications were compared and analyzed between the two groups. The median effective dose and 95% effective dose concentrations of ropivacaine pericapsular nerve group block were calculated by the Probit method.RESULTS AND CONCLUSION:(1) The postoperative analgesia of the observation group was better than that of the control group,and the visual analog scale score at 6-hour dynamic,12-hour static,12-hour dynamic,24-hour static,24-hour dynamic,48-hour static,48-hour dynamic,as well as the time of first patient-controlled analgesia compression and the number of patient-controlled analgesia compression at 24 and 48 hours postoperatively were statistically different (P<0.05). (2) There were significant differences between the two groups in mean arterial pressure and heart rate at extubation and 12 hours after extubation (P<0.01). (3) The levels of interleukin-6,tumor necrosis factor-α,epinephrine,and noradrenaline in the observation group were significantly lower than those in the control group at 24 and 48 hours after surgery (P<0.01). (4) There was significant difference in the length of hospital stay between the two groups (P<0.01),and no difference was detected in the occurrence of postoperative complications (P>0.05). (5) The median effective dose and 95% effective dose values of dexmedetomidine combined with ropivacaine were 0.164% and 0.188%,respectively,while the median effective dose and 95% effective dose values of ropivacaine alone were 0.194% and 0.276%,respectively. (6) It is concluded that using dexmedetomidine combined with ropivacaine in pericapsular nerve group block can effectively alleviate postoperative analgesia in elderly patients with total hip arthroplasty,help maintain the hemodynamic stability of patients,reduce postoperative inflammation and stress response,and shorten the length of hospital stay. In addition,dexmedetomidine can reduce the dosage of ropivacaine under the condition of effective pericapsular nerve group block reaction.
6.Diagnostic value of masseter muscle thickness fraction measured by ultrasound for dysphagia in patients with Parkinson's disease

Objective To investigate the diagnostic value of masseter muscle thickness fraction(MMTF)measured by ultrasound for the occurrence of dysphagia in patients with Parkinson's disease(PD).Methods A total of 100 patients were selected as the study group,and another 50 healthy individuals who received medical checkups with matched age,gender and body mass index(BMI)in our hospital during the same period were selected as the control group.The demographic data and disease characteristics of PD patients were recorded,and patients were divided into the dysphagia group(n=34)and the non-dysphagia group(n=66)according to the occurrence of dysphagia.The demographic data,and disease characteristics such as Hoehn-Yahr(H&Y)stage,Unified Parkinson's Disease Rating Scale(UPDRS)Ⅲ score,levodopa equivalent daily dose(LEDD)and MMT-related parameters measured by ultrasound were compared between the two groups.The indicators with P<0.05 in univariate analysis were used as independent variables,and the influencing factors of dysphagia in patients with PD were investigated by binary Logistic regression analysis.The diagnostic efficacy of each index for dysphagia in patients with PD was evaluated by constructing receiver operating characteristic(ROC)curves,and the area under the curve(AUC)was calculated.Results The MMT during forceful biting and MMTF were lower in the study group than those in the control group(all P<0.05).Compared with the non-dysphagia group,the dysphagia group was older and had higher H&Y stage and UPDRS III scores(all P<0.05).The MMT during forceful biting and MMTF in the dysphagia group were lower than those in the non-dysphagia group(all P<0.05).Multivariate Logistic regression analysis showed that increased UPDRSⅢ score was the risk factor for dysphagia in patients with PD,and increased MMT during forceful biting and MMTF were protective factors.The ROC curve analysis indicated that the AUC(95%CI)of UPDRS Ⅲ score,MMT during forceful biting and MMTF for diagnosing dysphagia in patients with PD were 0.714(0.615-0.800),0.744(0.647-0.826)and 0.888(0.809-0.942),respectively.The AUC of MMTF for diagnosing dysphagia in patients with PD was higher than those of UPDRS Ⅲscores and MMT during forceful biting(Z values were 2.611 and 2.208,respectively,P<0.05).Conclusion MMTF is an independent influencing factor for dysphagia in patients with PD and can be used as a screening indicator for dysphagic patients.
7.Epidemic Characteristics and disease burden trend of cervical cancer in Guangxi Zhuang Autonomous Region, 2010-2017

Objective:To analyze the changing trend of cervical cancer epidemiological characteristics and disease burden in cancer registration areas of Guangxi Zhuang Autonomous Region (Guangxi) from 2010 to 2017, and to provide scientific basis for the development of cervical cancer prevention and control strategies in Guangxi.Methods:Using descriptive analysis method, based on the incidence and death data of cervical cancer in the tumor registration areas of Guangxi from 2010 to 2017, Crude morbidity, crude mortality, age-standardized morbidity and mortality (referred to as the winning rate), disability adjusted life years (DALYs) rate and the annual percentage change (APC) and average annual percentage change (AAPC) of the above indicators were calculated, and stratified analysis was conducted for urban and rural areas and different age groups.Results:From 2010 to 2017, the crude incidence rate of cervical cancer in Guangxi showed a significant upward trend, rising from 10.31/10 5 in 2010 to 19.94/10 5 in 2017, with an average annual growth rate of 7.9% ( P<0.05). However, after age standardization, the trend of the age-standardized incidence rate of cervical cancer was not statistically significant ( P>0.05). During the same period, the crude mortality rate of cervical cancer increased from 2.69/10 5 to 6.21/10 5, with an average annual growth rate of 13.1% ( P<0.05), and the trend of the age-standardized mortality rate was basically consistent with that of the crude mortality rate. The analysis of urban-rural differences showed that the growth rates of the crude incidence rate and crude mortality rate of cervical cancer in rural areas were higher than those in urban areas from 2010 to 2017 (AAPC incidence rate: 21.3% vs. 2.3%; AAPC mortality rate: 20.1% vs. 8.4%). The analysis of age differences showed that the crude incidence rate and crude mortality rate of cervical cancer in all age groups increased to varying degrees, among which the growth rate of the incidence rate (AAPC=16.2%, P<0.05) and mortality rate (AAPC=14.7%, P<0.05) of cervical cancer in women aged 65 and above was the fastest. In addition, the DALYs rate of cervical cancer in Guangxi increased from 50.6/10 5 in 2010 to 111.0/10 5 in 2017, with an average annual increase of 11.9% ( P<0.05). The growth rate of the DALYs rate in rural areas was higher than that in urban areas, and the growth rate of the DALYs rate in the 50-59 age group was higher than those in other age groups. Conclusions:From 2010 to 2017, the incidence rate, mortality rate and DALYs rate of cervical cancer in Guangxi showed an upward trend. Comprehensive prevention and control measures for cervical cancer, such as improving the early diagnosis and treatment system, promoting the popularization of HPV vaccination and strengthening health education, should be taken to reduce the disease burden of cervical cancer.
8.Epidemiological characteristics and treatment outcome of pulmonary tu-berculosis patients combined with diabetes mellitus in Bijie City from 2017 to 2022

Objective To analyze the epidemiological distribution characteristics and influencing factors of treat-ment outcome of pulmonary tuberculosis(PTB)patients complicated with diabetes mellitus(DM)in Bijie City from 2017 to 2022,and provide reference for formulating prevention and control measures of PTB-DM.Methods The registered management cases of PTB patients combined with DM from the tuberculosis surveillance report informa-tion management system in Bijie City in 2017-2022 were collected.Changing trends in annual registration rates and successful treatment rates were analyzed with Joint-point regression models.Different characteristics between pa-tients with PTB-DM and PTB alone were conducted comparative analysis;the influencing factors of treatment out-come were analyzed by x2 test and binary logistic regression analysis.Results A total of 679 PTB-DM patients were registered in Bijie City from 2017 to 2022,accounting for 1.53%of total PTB patients,the annual registration rate increased from 1.11/100 000 in 2017 to 3.08/100 000 in 2022,with an increasing trend in the annual percentage change(APC=26.67%,95%CI:5.36%-52.29%,t=3.564,P=0.024).In the analysis on characteristics,the proportion of PTB-DM male patients,age ≥45 years and farmers,the ratio of direct consultation to transfer treat-ment,the pathogenicity positive rate,the proportion re-treatment,and rate of delay in consultation were all higher than those of PTB alone;successful treatment rate and proportion of floating population were all lower than PTB alone,difference were all statistically significant(all P<0.05).The successful treatment rate of PTB-DM patients in Bijie City from 2017 to 2022 was 87.80%.Age,occupation,pathogenicity diagnosis results were influencing fac-tors for treatment outcome in patients with PTB-DM,with farmers(OR=3.68,95%CI:1.22-11.09),with pathogenicity positivity(OR=2.84,95%CI:1.24-6.50),and without pathogenic detection result+tuberculous pleurisy(OR=11.35,95%CI:2.16-59.74)being risk factors for successful treatment.Conclusion Although the proportion of PTB-DM in the total PTB patients in Bijie City from 2017 to 2022 was not high,it showed an up-ward trend;it is necessary to attach great importance to male,re-treated,farmer,and pathogenicity positivity co-morbid patients in Bijie City,and strengthen the two-way screening of PTB-DM in high-risk populations.
9.Summary of the best evidence for management of chronic venous disease in the lower extremities

Objective:To retrieve, evaluate, and summarize the evidence on prevention and management of chronic venous disease of the lower extremities, so as to provide reference for clinical practice.Methods:Clinical decisions, guidelines, evidence summaries, and expert consensus on chronic venous diseases of the lower extremities were systematically retrieved from UpToDate, Joanna Briggs Institute Evidence-Based Health Care Center Database, National Institute for Health and Clinical Excellence, Registered Nurses' Association of Ontario, New Zealand Guidelines Group, National Health and Medical Research Council, Guidelines International Network, Medlive, PubMed, Embase, China Biology Medicine disc, China National Knowledge Infrastructure, Wanfang Data, and specialized collaborative websites including the Society for Vascular Surgery, American Vein and Lymphatic Society, American Venous Forum, and European Society for Vascular Surgery. The search period was from the establishment of the database to March 31, 2025.Results:A total of 18 articles were included, including three guidelines, five clinical decisions, four evidence summaries, and six expert consensus. Ultimately, 19 pieces of evidence were integrated from six aspects of diagnosis, clinical-etiology-anatomy-pathophysiology (CEAP) classification and severity assessment, risk factor assessment, prevention, lifestyle modification, stress management, and patient education and follow-up.Conclusions:Evidence included in this study indicates that the diagnosis of chronic venous disease of the lower extremities, along with CEAP classification and severity assessment, has been widely adopted. In clinical practice, emphasis should be placed on assessing risk factors, implementing preventive management and health education for high-risk patients, and conducting long-term follow-up to alleviate symptoms and thereby improve quality of life. It is recommended that clinical nursing staff conduct targeted evidence transformation based on specific clinical contexts to enhance the quality of clinical nursing.
10.Effect of Ad-HIF-1α-Trip on apoptosis of H9c2 cardiomyocytes under high glucose and hypoxic conditions

Objective To investigate the effect of the mutant of adenovirus-mediated HIF-1α triple mutant(Ad-HIF-1α-Trip)on the apoptosis of H9c2 cardiomyocytes under high glucose and hypoxia conditions.Methods H9c2 cardiomyocytes were cultured under hypoxic conditions in vitro and randomly divided into four groups based on glucose concentration and viral transfection status:low oxygen+normal glucose concentration group(LO+NG group),low oxygen+high glucose concentration group(LO+HG group),low oxygen+high glucose+adenoviral null vector group(LO+HG+Ad-Null group),and low oxygen+high glucose+adenovirus HIF-1α triple mutant group(LO+HG+Ad-HIF-1α-Trip group).After 12 hours of hypoxia,HIF-1α,PI3K,and Akt expression were measured via qRT-PCR and Western blot,and apoptosis of H9c2 cardiomyocytes was assessed by flow cytometry.Results HIF-1α,PI3K,and Akt expression decreased in cardiomyocytes under high glucose and hypoxia,while Ad-HIF-1 α-Trip enhanced their expression.Flow cytometry revealed increased apoptosis under high glucose and hypoxia,which was reduced by Ad-HIF-1α-Trip.Conclusion Ad-HIF-1α-Trip upregulates HIF-1α,PI3K,and Akt in H9c2 cardiomyocytes under high glucose and hypoxia,likely reducing apoptosis via PI3K-Akt pathway activation.
