1.The value of cuprotosis-related gene CDKN2A in the diagnosis of chronic obstructive pulmonary disease induced by cigarette smoke

Objective To explore the value of cuprotosis-related genes in the diagnosis and disease progression of chronic obstructive pulmonary disease(COPD).Methods Differential gene expression related to COPD was identified through gene expression omnibus series(GSE)10006,GSE11784,GSE19407,and GSE8545 datasets,and compared with previously identified cuprotosis-related genes to identify differentially expressed genes.Weighted gene co-expression network analysis(WGCNA)was then performed on GSE8545 and GSE19407 datasets to explore the correlation between cuprotosis-related genes and COPD phenotypes.GSE20257 dataset was introduced to validate the gene's expression level changes and further investigate the impact of smoking on gene expression levels.Subsequently,the effects of smoking cessation on gene expression levels were explored by using the GSE7895 datasets.GSE162635 dataset was then utilized to investigate the impact of differentially expressed cuprotosis-related genes on the severity of COPD.Results The study identified cyclin-dependent kinase inhibitor 2A(CDKN2A)as a potential cuprotosis-related gene associated with COPD,confirming that smoking upregulated CDKN2A expression in lung tissue,which returns to baseline levels after smoking cessation.Finally,the study confirmed that the expression level of CDKN2A was associated with the prognosis of COPD,increasing with severity.Conclusion The study identifies CDKN2A as a cuprotosis-related gene associated with COPD,and shows promising diagnostic potential and correlation with disease severity.
2.Expression levels of serum IL-6 and NLRP3 in children with bronchial asthma combined with pulmonary infection and its relationship with prognosis

Objective To explore the expression levels of NOD-like receptor thermal protein domain-asso-ciated protein 3(NLRP3)and interleukin-6(IL-6)in children with bronchial asthma combined with pulmona-ry infection and its relationship with prognosis.Methods A sample of 150 children with bronchial asthma combined with pulmonary infection hospitalized in Nanjing University Medical College Teaching Hospital,North Jiangsu People's Hospital from January 2021 to December 2022 were prospectively enrolled as observa-tion group,All children were followed up for one year.Another 164 healthy children underwent health check-ups were concurrently selected as control group.According to the prognosis,the children in the observation group were divided into good prognosis groups(n=92)and poor prognosis groups(n=58).Serum IL-6 and NLRP3 expression levels were detected in all participants,and receiver operating characteristic(ROC)curve was plotted to evaluate the value of IL-6 and NLRP3 in predicting the poor prognosis in children with bronchi-al asthma combined with pulmonary infection.Results Serum IL-6 and NLRP3 expression levels in the obser-vation group were significantly higher than those in the control group(P<0.05).Serum IL-6 and NLRP3 ex-pression levels in the poor prognosis group were higher than those in the good prognosis group(P<0.05).The results of ROC curve analysis showed that the area under the curve of IL-6 alone for predicting the poor prognosis of children with asthma combined with pulmonary infection was 0.772,with a sensitivity of 79.3%and a specificity of 73.9%.The area under the curve(AUC)of NLRP3 alone for predicting the poor prognosis of children with asthma combined with pulmonary infection was 0.735,with a sensitivity of 55.2%and a spe-cificity of 88.0%.The AUC of the combination of IL-6 and NLRP3 for predicting poor prognosis in children with asthma combined with pulmonary infection was 0.851,which was higher than those of IL-6 and NLRP 3 alone for predicting poor prognosis in children(Z=1.563,2.003,P=0.059,0.045),and the sensitivity was 82.8%and specificity was 75.0%,with high predictive value.Conclusion The expression levels of serum IL-6 and NLRP3 is abnormally up-regulated in children with bronchial asthma combined with pulmonary infec-tion,and the two could serve as predictive indicators for poor prognosis.

4.Diagnostic value of photoplethysmography for obstructive sleep apnea syndrome in critically ill patients

Objective To investigate the diagnostic value of photoplethysmography (PPG) in the detection of obstructive sleep apnea syndrome (OSAS) among critically ill patients with respiratory or cardiocerebrovascular diseases accompanied by snoring. Methods A total of 91 critically ill patients with respiratory or cardiocerebrovascular diseases accompanied by snoring, admitted to the internal medicine ward of Subei People's Hospital from January 2019 to October 2023 were enrolled. After informed consent, overnight polysomnography (PSG) and PPG were performed. The consistency of the sleep apnea parameters obtained of the two methods by sensitivity, specificity, positive predictive value, negative predictive value, and the area under the receiver operating characteristic (ROC) curve. 5.Hsa-miR-1293 positively regulates SLC3A2,a disulfide death gene and promotes the development of lung adenocarcinoma

Objective To analyse the gene expression profile microarrays of lung adenocarcinoma patients by bioinformatics methods,to study the mechanisms affecting the development of lung adenocarcinoma,and to explore new therapeutic targets for lung adenocarcinoma.Methods The mRNA and miRNA expression data were downloaded from The Cancer Genome Atlas database,key genes were screened by R language and survival analysis and verified by the survival module of the database,and high-expression pathway enrichment analysis was performed for prognostic related genes.Finally,the correlation between key genes and immune cells was analyzed.Results The key disulfide death gene SLC3A2 and key nuclear activation miRNA(hsa-miR-1293)were identified by screening.SLC3A2 was positively correlated with hsa-miR-1293,and the higher the expression,the worse the prognosis of lung adenocarcinoma patients.The most relevant enrichment pathway for SLC3A2 is aminoacyl transfer RNA biosynthesis.SLC3A2 was positively correlated with 4 kinds of differential immune cells and negatively correlated with 4 kinds of differential immune cells.Conclusion Hsa-miR-1293 positively regulates the expression of SLC3A2,a disulfide death gene,to promote the occurrence and development of lung adenocarcinoma,which may provide ideas for developing new therapeutic targets for lung adenocarcinoma.
6.Clinicopathological features of cystic neutrophilic granulomatous mastitis

Objective:To investigate the etiology, clinical manifestations, clinicopathological features of cystic neutrophil granulomatous mastitis (CNGM).Methods:From Jan 2019 to Dec 2020, 95 CNGM cases diagnosed by biopsy pathology at Chongqing Hospital of Traditional Chinese Medicine and Chongqing Liangping District Hospitol of Traditional Chinese Medicine were reviewed.Results:There were 95 female patients, aged 21 to 50 years, with a median age of 32 years. Laboratory examination showed that 56% (53/95) cases had elevated rheumatoid antibody level, 27 % (26/95) had increased level of serum thyroid antibody, 15% (14/95) had elevated antineutrophil antibody, 35% (33/95) had increased ESR, 38% (36/95) had increased C-reactive protein. The positive rate of Gram-stained bacilli was 82% (78/95). Histology: pyogenic granuloma with lobule of breast as the center, the center of granuloma was cystic vacuole.Immunohistochemistry showed that the inflammatory cells in and around granuloma were mainly CD3 + cells, and CD4 + cells were more than CD8 + cells. Conclusions:The cystic neutrophilie granulo matous mastitis is a rare type of idiopathic granulomatous mastitis. The diagnosis of CNGM is dependent on its specific pathological features.
7.Reference values for urinary flow rate in elderly women: based on a national multicenter study

Objective:To collect data on urinary flow rate in the elderly female population across the country and to analyze the range of reference values.Methods:This study enrolled 333 subjects from July 2020 to June 2022.The study implementation process was divided into two steps.In the first step, subjects completed an electronic questionnaire, which included basic information about the subject, a short form for urinary incontinence, and a scoring form for the symptoms of overactive bladder syndrome.In the second step, the staff introduced the use of a mobile uroflowmetric device and distributed the instrument and materials.Uroflow rate data were automatically uploaded to a cloud database via the mobile phone.Subsequently, two or more physicians specializing in urinary control performed Uroflow rate-qualifying screenings and conducted statistical analyses.Results:A total of 333 subjects were enrolled in the study, and the researchers collected 1375 qualified urine flow rate records using a mobile urine flow rate instrument.The age of the subjects ranged from 60 to 84 years, with a mean age of 69 years.The reference ranges for urinary flow rate were found to be 24.8-26.2 s, with a mean urinary flow rate of 12.2-12.9 ml/s, a maximum urinary flow rate of 22.2-23.4 ml/s, and a time to peak of 8.5-9.7 s. The study observed a tendency for both maximal and mean urinary flow rates to decrease in older women as their age increased(Pearson correlation coefficient: -0.1, P<0.001). Conclusions:The uroflow rate of older women decreases with aging.Specifically, the average uroflow rate of women over 80 years old is lower than that of other age groups.This study aims to establish normal uroflow parameters for uroflowmetry in healthy older women in China.

Objective To evaluate the expression of FUNDC1 and its clinical significance in non-small cell lung cancer. Methods We used TCGA database to analyze the difference of mitochondrial receptors (DRP1, BNIP3, FUNDC1, NIX, RHEB, LC3, OPA1 and MFN1) expression between normal and NSCLC tissues, as well as its effect on the prognosis of NSCLC patients. Immunohistochemistry was used to detect FUNDC1 expression. The correlations between FUNDC1 expression and clinicopathological characteristics, prognosis were evaluated by SPSS 22.0 statistical software. Results FUNDC1 expression was increased in NSCLC tissues, compared with normal tissues. FUNDC1 expression was related to the degree of differentiation and lymph node metastasis, but not to gender, age, pathological type, distant metastasis or TNM classification. The Cox regression analysis showed that FUNDC1 protein expression, lymph node metastasis, differentiation degree were independent prognostic factors of NSCLC. Objective To analyze the mortality risk and evaluate the curative effects of surgery and non-surgery on NSCLC with diameter > 7.0 cm. Methods We collected the data of NSCLC patients with diameter > 7.0 cm from 2010 to 2015 from the SEER database. The 1, 2, 3-year survival rates were analyzed by life table method. Overall survival curve was estimated by Kaplan-Meier method. Univariate and multivariate Cox regression models were used to analyze the independent prognostic factors. Objective To analyze the factors affecting the prognosis of patients with pulmonary sarcomatoid carcinoma (PSC) and construct a nomogram prediction model for the prognosis of PSC patients. Methods Based on the SEER database, 1671 patients diagnosed as PSC from 1988 to 2015 were collected and divided into modeling group and validation group according to the ratio of 7:3. Univariate and multivariate Cox regression analysis were performed in the modeling group to explore independent risk factors affecting the prognosis and construct a nomogram survival prediction model. The consistency index and calibration curve were used for verification in the modeling group and the test module respectively. Results Age, gender, histological type, TNM stage, tumor diameter > 50mm, surgery, radiotherapy and chemotherapy were independent factors that affected the prognosis of PSC patients. The nomogram prediction model was constructed and verified based on independent factors. The C indexes of the modeling group and the test model were 0.790 (95%
