The clinical application of binocular endoscope relies primarily on the visual system of physicians to create a three-dimensional effect, but it cannot provide accurate depth information. The utilization of 3D reconstruction technology in binocular endoscopy can facilitate the recovery of image depth information, and the application of deep learning-based 3D reconstruction technology can significantly improve the accuracy and real-time performance of reconstruction results, making it widely applicable in the realm of minimally invasive surgery. This paper aims to explore the key technologies and implementation methods of deep learning based 3D reconstruction for binocular endoscopic images, and seeks to outline strategies for enhancing the quality of 3D reconstruction in endoscopic images, providing guidance for sustainable development of binocular endoscopic image reconstruction technology in clinical settings. This will assist in the application of minimally invasive surgery and contribute to meeting the demands of precision medicine.

2.Submicron-sized superantigen biomimetic liposomes with highly efficient pulmonary accumulation to remodel local immune microenvironment for cancer chemoimmunotherapy.

Metastatic lung cancer continues to cause a high number of deaths due to high malignancy and poor prognosis, and the efficacy of typical chemotherapy or immunotherapy is less than ideal due to the low pulmonary accumulation and targeting of therapeutics. Here, a submicron-sized biomimetic liposome was formulated for the lung-targeted co-delivery of bacterial superantigen and paclitaxel. Recombinant staphylococcal enterotoxin C2 (rSEC2), a bacterial superantigen, was expressed with the Escherichia coli system and showed potent immunostimulatory activities to mediate tumor cell death. The submicron-sized (∼800 nm) biomimetic liposomes, namely 4T1 cell membrane-hybrid rSEC2 paclitaxel liposomes (TSPLs), exhibited high lung-accumulation efficiency and tumor homologous effect due to the suitable particle size and membrane hybridization of cancer cell membranes with phospholipids. Intravenous TSPLs remarkably inhibited metastatic lung cancer with limited systemic immune responses. TSPLs reversed the immunosuppressive state and increased the proportion of local CD4+ and CD8+ T cells in the lung; moreover, paclitaxel increased tumor cell apoptosis and reduced tumor burden. In summary, the high lung cancer targeting was achieved by particle size control and cell membrane hybridization, and the highly efficient anticancer effect was achieved by the co-delivery of superantigens and chemotherapeutic drugs.

Cardiac fibrosis is characterized by an elevated amount of extracellular matrix (ECM) within the heart. However, the persistence of cardiac fibrosis ultimately diminishes contractility and precipitates cardiac dysfunction. Circular RNAs (circRNAs) are emerging as important regulators of cardiac fibrosis. Here, we elucidate the functional role of a specific circular RNA CELF1 in cardiac fibrosis and delineate a novel feedback loop mechanism. Functionally, circ-CELF1 was involved in enhancing fibrosis-related markers' expression and promoting the proliferation of cardiac fibroblasts (CFs), thereby exacerbating cardiac fibrosis. Mechanistically, circ-CELF1 reduced the ubiquitination-degradation rate of BRPF3, leading to an elevation of BRPF3 protein levels. Additionally, BRPF3 acted as a modular scaffold for the recruitment of histone acetyltransferase KAT7 to facilitate the induction of H3K14 acetylation within the promoters of the Celf1 gene. Thus, the transcription of Celf1 was dramatically activated, thereby inhibiting the subsequent response of their downstream target gene Smad7 expression to promote cardiac fibrosis. Moreover, Celf1 further promoted Celf1 pre-mRNA transcription and back-splicing, thereby establishing a feedback loop for circ-CELF1 production. Consequently, a novel feedback loop involving CELF1/circ-CELF1/BRPF3/KAT7 was established, suggesting that circ-CELF1 may serve as a potential novel therapeutic target for cardiac fibrosis.

Enamel demineralization, the formation of white spot lesions, is a common issue in clinical orthodontic treatment. The appearance of white spot lesions not only affects the texture and health of dental hard tissues but also impacts the health and aesthetics of teeth after orthodontic treatment. The prevention, diagnosis, and treatment of white spot lesions that occur throughout the orthodontic treatment process involve multiple dental specialties. This expert consensus will focus on providing guiding opinions on the management and prevention of white spot lesions during orthodontic treatment, advocating for proactive prevention, early detection, timely treatment, scientific follow-up, and multidisciplinary management of white spot lesions throughout the orthodontic process, thereby maintaining the dental health of patients during orthodontic treatment.

5.Investigating Causal Relationships Between Serum Trace Elements and Head and Neck Cancers:a Two-Sample Bidirectional Mendelian Randomization Study

[Purpose]To investigate the potential causal relationships between serum levels of trace elements and head and neck cancers.[Methods]Single nucleotide polymorphism(SNP)of oral cancer,oropharyngeal cancer,laryngeal cancer and thyroid cancer,associated with calcium,copper,iron,magnesium,zinc,were obtained from genome-wide association studies(GWAS).A two-sample bidirectional Mendelian randomization(MR)analysis was performed using the inverse variance weighting(IVW)method by calculating odds ratio(OR)and 95%confidence interval(CI).Pleiotropy was assessed using MR-PRESSO and MR-Egger regression,and sensitivity analysis was conducted via the"leave-one-out"method.[Results]IVW analysis revealed a causal association between serum magnesium levels and the incidence of oral cancer(OR=0.976,95%CI:0.956~0.997,P=0.025),also between thyroid cancer and serum calcium levels(OR=1.008,95%CI:1.001~1.015,P=0.023).No significant causal associations were observed between other trace ele-ments and head and neck cancers(all P>0.05).[Conclusion]This MR study suggests that serum magnesium levels serve as a protective factor against oral cancer,while thyroid cancer leads to el-evated serum calcium levels.
6.Paroxetine alleviates dendritic cell and T lymphocyte activation via GRK2-mediated PI3K-AKT signaling in rheumatoid arthritis.

BACKGROUND:
G protein-coupled receptor kinase 2 (GRK2) could participate in the regulation of diverse cells via interacting with non-G-protein-coupled receptors. In the present work, we explored how paroxetine, a GRK2 inhibitor, modulates the differentiation and activation of immune cells in rheumatoid arthritis (RA).
METHODS:
The blood samples of healthy individuals and RA patients were collected between July 2021 and March 2022 from the First Affiliated Hospital of Anhui Medical University. C57BL/6 mice were used to induce the collagen-induced arthritis (CIA) model. Flow cytometry analysis was used to characterize the differentiation and function of dendritic cells (DCs)/T cells. Co-immunoprecipitation was used to explore the specific molecular mechanism.
RESULTS:
In patients with RA, high expression of GRK2 in peripheral blood lymphocytes, accompanied by the increases of phosphatidylinositol 3 kinase (PI3K), protein kinase B (AKT), and mammalian target of rapamycin (mTOR). In animal model, a decrease in regulatory T cells (T regs ), an increase in the cluster of differentiation 8 positive (CD8 + ) T cells, and maturation of DCs were observed. Paroxetine, when used in vitro and in CIA mice, restrained the maturation of DCs and the differentiation of CD8 + T cells, and induced the proportion of T regs . Paroxetine inhibited the secretion of pro-inflammatory cytokines, the expression of C-C motif chemokine receptor 7 in DCs and T cells. Simultaneously, paroxetine upregulated the expression of programmed death ligand 1, and anti-inflammatory cytokines. Additionally, paroxetine inhibited the PI3K-AKT-mTOR metabolic pathway in both DCs and T cells. This was associated with a reduction in mitochondrial membrane potential and changes in the utilization of glucose and lipids, particularly in DCs. Paroxetine reversed PI3K-AKT pathway activation induced by 740 Y-P (a PI3K agonist) through inhibiting the interaction between GRK2 and PI3K in DCs and T cells.
CONCLUSION
Paroxetine exerts an immunosuppressive effect by targeting GRK2, which subsequently inhibits the metabolism-related PI3K-AKT-mTOR pathway of DCs and T cells in RA.

7.Progress on elimination of radioactive nuclides by polysaccharides

Radionuclides can be hazardous by absorbed through the skin, respiratory and digestive tracts. Chelating agents and adsorbents already could effectively remove them, however traditional chelators have side effects such as nephrotoxicity, teratogenicity, and embryotoxicity. As a new type of nuclide adsorbent, polysaccharide has the advantages of safety, biocompatibility, and high clearance rate. In this paper, the main types and perniciousness of radionuclides, and the latest research of polysaccharides in radionuclide removal were summarized. The application of polysaccharide as an effective adsorption molecule for radio nuclides in nuclear wars, nuclear accidents and other sudden nuclear events is promising.
8.The changes of core symptoms and nursing implications for lymphoma patients with chemotherapy

Objective To explore the changes of core symptoms and nursing strategies for lymphoma patients during chemotherapy using a dynamic network analysis model,and provide a reference for precise symptom intervention.Methods A longitudinal study was conducted,and lymphoma patients undergoing chemotherapy in the Department of Lymphoma at a tertiary hospital in Tianjin from September 2021 to December 2023 were conveniently sampled.The Lymphoma Patient Symptom Assessment Scale was used to track symptoms at 3 time points:7 to 14 days after the 1st treatment cycle(T1),the 3rd cycle(T2),and the 6th cycle(T3).Dynamic symptom network analysis,including network relationships,core symptoms,and bridge symptoms,was performed using R software.Results During the research process,135 participants withdrew,resulting in a final inclusion of 865 cases for analysis.Fatigue exhibited the highest incidence rate(34.80%)at T1;alopecia was the most prevalent symptom(58.03%and 53.64%,respectively)at T2 and T3.The T1→T2 dynamic network analysis showed that fever had the highest out-expected influence(EI=0.551)and bridge expected influence(EI=0.225).The T2→T3 dynamic network analysis showed that limb numbness exhibited the highest out-expected influence(EI=0.203)and bridge-expected influence(EI=0.170).Reliability tests indicated that both dynamic networks demonstrated moderate accuracy but suboptimal stability.Conclusion Fatigue was the most common symptom at T1;alopecia was the most prevalent symptom at T2 and T3.Fever was the core symptom and bridging symptom in the T1→T2 symptom network,whereas limb numbness assumed the role of the core symptom and bridging symptom in the T2→T3 symptom network.The clinical practitioners could take high-incidence symptoms and core symptoms as intervention targets to develop precise symptom management strategies for lymphoma patients undergoing chemotherapy.
9.Real experience of continuous renal replacement therapy in ICU patients: a qualitative study

Objective:To explore the real experience of ICU patients during continuous renal replacement therapy (CRRT) and to inform the development of interventions.Methods:This study was a qualitative study. Using the social ecosystem theory as a theoretical framework and purposive sampling method, 15 ICU patients who received CRRT at Beijing Friendship Hospital, Capital Medical University from May 2023 to February 2024 were selected for semi-structured interviews. Colaizzi 7-step analysis was applied to analyze the data.Results:A total of three themes and 11 sub-themes were summarized, including microsystems (symptom improvement, limited mobility and self-care, sleep disorders, hypothermia, emotional overload, hope and gratitude), mesosystems (heavy financial burden, disrupted rhythm of family life, dependence on peer support), and macrosystems (information support needs to be improved, expectation of an open ICU visitation system) .Conclusions:The social ecosystem of patients with CRRT in the ICU is not optimistic. Healthcare professionals should provide appropriate support for patients to facilitate their physical and psychological recovery.
