1.Research Progress in the Treatment of Pediatric Lung Diseases with Medicinal Plasters

[Objective]To systematically analyze the application of medicinal plasters in pediatric lung diseases and provide a basis for their clinical use in such conditions.[Methods]By integrating relevant literature and utilizing the Ancient and Modern Medical Case Cloud Platform,this paper focuses on the treatment of pediatric lung diseases with medicinal plasters and summarizes research findings in four aspects:clinical manifestations,expert experience,formulation patterns and mechanisms.[Results]The main pediatric lung diseases treated by medicinal plasters are asthma,recurrent respiratory tract infections,allergic cough and allergic rhinitis.Taking recurrent respiratory tract infections,a common pediatric lung disease as an example,the medicinal properties are mainly neutral and warm,with the main flavors being sweet,pungent and bitter,mostly belong to meridians of the spleen,lungs,stomach and heart.The core formula is composed of Astragalus membranaceus(Huangqi),Atractylodes macrocephala(Baizhu),Saposhnikovia divaricata(Fangfeng),Pseudostellaria heterophylla(Taizishen),Dioscorea opposite(Shanyao),Citrus reticulata(Chenpi),Poria cocos(Fuling),Polygonatum sibiricum(Huangjing),Glycyrrhizae Radix Preparata(Zhigancao),Gallus gallus domesticus gigeria(Jineijin),Crataegus pinnatifida(Shanzha),Ophiopogon japonicus(Maidong),Schisandra chinensis(Wuweizi),Xanthium sibiricum(Cangerzi),Dolichos lablab(Baibiandou)and Cinnamomum cassia(Guizhi).The treatment mechanisms include enhancing lymphocyte function,phagocytosis,immune organ function,and reducing the levels of immunologically active substances.[Conclusion]The application of medicinal plasters in treating pediatric lung diseases is limited to asthma,recurrent respiratory tract infections,allergic cough and allergic rhinitis.The treatment mechanisms are still limited to single medicinal and require further research.

3.Association of expression profiles of miRNA and mRNA in Chinese hamster buccal pouch squamous cell carcinoma

Objective To establish the expression profiles of differentially expressed miRNA and mRNA in oral squamous cell carcinoma(OSCC)and to investigate the roles of miRNA and mRNA associated with the occurrence and development of OSCC. Methods The expression profiles of miRNA and mRNA were constructed using a new generation of high-throughput sequencing techniques. The miRNA and mRNA associated with the occurrence and development of OSCC were predicted by Gene Ontology enrichment analysis and KEGG pathway analysis. Results We successfully constructed the differentially expressed miRNA and mRNA profiles of Chinese hamster buccal pouch squamous cell carcinoma. 11 known and 3 novel significantly differentially expressed miRNAs and 194 differentially expressed mRNAs were found. A miRNA can regulate multiple mRNAs, and multiple miRNAs can control one mRNA. Conclusions Differential expression of miRNA play a an important role in the carcinogenesis and development of OSCC through regulating mRNA and forming a complex regulatory network. It provides theoretical data for the occurrence,pathogenesis,clinical treatment and prognosis of OSCC.
4.Effects of Different Dose Compatibility of Scutellaria baicalensis-Rheum palmatum-Coptis chinensis Based on Sanhuang Xiexin Decoction on Dissolution Rate of Baicalin and Baicalein

OBJECTIVE: To investigate the effects of different dose compatibility of Scutellaria baicalensis-Rheum palmatum-Coptis chinensis based on Sanhuang Xiexin decoction (shorted for Xiexin decoction) on dissolution rate of baicalin and baicalein, and to provide reference for studying chemical mechanism of TCM compound dose composition compatibility. METHODS: HPLC method was used to determine the contents of baicalin and baicalein. By fixing the dose of S. baicalensis (3 g), using the dose of R. palmatum and C. chinensis as factors, dissolution rate of baicalin and baicalein as response value, two-factor and five-level central composite test was designed. The optimal dose compatibility of S. baicalensis-R. Palmatum-C. chinensis was optimized by response surface method, and compared with the dissolution rate of baicalin and baicalein in classic dose Xiexin decoction (S. baicalensis 3 g, R. Palmatum 6 g, C. chinensis 3 g). RESULTS: When the doses of S. baicalensis, R. palmatum, C. chinensis were 3, 1. 76, 0. 17 g, total dissolution rate of baicalin and baicalein was the highest in extract. The average total dissolution rate of baicalin and baicalein in validation test was 21. 89% (RSD=0. 46%, n=3),and the relative error was 2. 88% with the predicted value of 22. 54%. Compared with the classical dose of Xiexin decoction, the total dissolution rate of baicalin and baicalein in the optimum dose of S. baicalensis-R. Palmatum-C. chinensis in extract was increased by 47. 21%. CONCLUSIONS: The different dose compatibility of S. baicalensis-R. Palmatum-C. chinensis based on Xiexin decoction influence the dissolution of baicalin and baicalein to certain extent. When the doses of S. baicalensis, R. palmatum, C. chinensis are 3, 1. 76, 0. 17 g, dissolution rate of baicalin and baicalein are higher than that of classic dose.
5.Survey on aerodontalgia in fighter pilots and relevant factors analysis

Objective To investigate the incidence rate of aerodontalgia in fighter pilots and analyze the related factors . Methods The cross-section study was used . The invasion of aerodontalgia was obtained upon a questionnaire survey from 192 fighter pilots , including the occurrence time ,degree ,and location and the incidence rate was calculated .The flying hours and the dental disease were compared between the pilots with and without aerodontalgia . Results The incidence rate of aerodontalgia in fighter pilots was 4 .17% (8/192) .In 8 aerodontalgia cases 7 pilots had dental disease .But in 184 pilots who felt free of aerodontalgia ,53 pilots (28 .80% ) were diagnosed as dental disease .There were significant difference between the incidence rates (χ2= 109 .634 , P<0 .01) .In 8 pilots with aerodontalgia ,5 cases had toothache during taking-off and 3 landing .The flying altitude was 500-1 000 m .Two of 8 aerodontalgia cases appeared the toothache on maxillary teeth and jaw ,5 on lower jaw and 1 on maxillary teeth .There was no significant difference among them (P>0 .05) .The average flying hours of 8 pilots with aerodontalgia were (2 425 .00 ± 1 445 .93) h while the average flying hours of 184 pilots without aerodontalgia was (1 703 .56 ± 1 331 .08) h .There was no significant difference between them (P>0 .05) . Conclusions The incidence rate of aerodontalgia was higher in the fighter pilots with dental disease .But the incidence rate has no correlation to the location of toothache and flying hours .It is suggested that pilots should conduct oral examinations every 6-12 months in order to detect and treat dental diseases in time .

7.The expression of EGFR,PDCD4,TGF-β1,Smad3 and Smad7 during oral buccal mucosa carcinogenesis in chinese hamster

Objective To study the expression and significance of EGFR, PDCD4, TGF-β1, Smad3, Smad7 in oral normal mucosa, oral simple hyperplasia, oral epithelial dysplasia and oral squamous cell carcinomas(OSCC).Methods Serum levels of EGFR, PDCD4 were measeured with ELISA in Chinese hamster during the oral mucosa carcinogenesis;The expressions of TGF-β1, Smad3, Smad7 in oral normal mucosa, oral simple hyperplasia, oral epithelial dysplasia and OSCC tissues were examined by immunohistochemistry.Results In the process of oral carcinogenesis,the expression level of EGFR increased significantly,while the expression of PDCD4 was decreased, the negatively correlation was evident between expression of these two proteins in ED and SCC.The expression of TGF-β1, Smad7 was higher in OSCC than in oral normal mucosa, oral simple hyperplasia, oral epithelial dysplasia(P < 0.05), while the expression of Smad3 was decreased.Further analysis showed the expression of TGF-β1 was correlated with the expression of Smad7.Conclusions The expressions of EGFR, PDCD4, TGF-β1, Smad3, Smad7 are closely related with the biological behaviors of oral squamous cell cancinoma.This experiment provides a theoretical basis for the study of oral squamous cell carcinomas mechanism research.So it has important significance for the development and prognosis of oral squamous carcinomaare.
8.Expression of apoptosis-related genes caspase-3, caspase-9, Bax and Bcl-2 during oral buccal mucosa carcinogenesis in Chinese hamster

Objective To study the expression and significance of apoptosis-related genes caspase-3,caspase-9,Bax and Bcl-2 in oral normal mucosa,oral simple hyperplasia,oral epithelial dysplasia and oral squamous cell carcinomas (OSCC) in Chinese hamsters.Methods The expressions of mRNA and protein of caspase-3,caspase-9,Bax and Bcl-2 in the oral normal mucosa,oral simple hyperplasia,oral epithelial dysplasia and OSCC tissues in Chinese hamsters were examined by immunohistochemistry and RT-PCR.Results During the process of oral carcinogenesis,the expression of Bcl-2 protein was significantly higher in OSCC than in oral normal mucosa,oral simple hyperplasia,and oral epithelial dysplasia (P<0.05).In dysplastic epithelia,the protein expressions of caspase-3,caspase-9 and Bax were more than that of normal epithelia,and along with the increased dysplasis,the expression level was decreased.Further analysis showed that expression of Bcl-2 was negatively related with the expressions of Bax,caspase-3 and caspase-9 (P<0.05).The result of RT-PCR showed that Bcl-2 was significantly increased in OSCC compared with normal mucosa,while the expressions of caspase-3,caspase-9 and Bax were decreased (P<0.05).Conclusions In the Chinese hamster squamous carcinoma,the expressions of caspase-3,caspase-9 and Bax are reduced and the Bcl-2 expression are increased,indicating that the expressions of caspase-3,caspase-9,Bax and Bcl-2 are closely related with the occurrence and development of oral squamous cell cancinoma.This study can offer some clues for gene therapy of OSCC,or can provide a reference for evaluating the biological characteristics and prognosis of OSCC.
9.Absorption, distribution and excretion of 2-fluorine-6-trifl-uoromethylpyridine in rats by radioactivity isotope tracing method

OBJECTIVE To study the absorption,distribution and excretion of 2-fluorine-6-trifluoromethylpyridine (JJBD) in rats.METHODS [14C] Radioactivity isotope tracing method was used.Male SD rats were ig given a single dose of JJBD 10 and 100 mg·kg-1 (radioactivity:3.7 GBq·kg-1).Concentrations of rat plasma,tissue,feces,urine and bile were determined with a liquid scintillation counting (LSC) analyzer.Toxicokinetics (TK) parameters were fitted using WinNonlin.RESULTS TK parameters of JJBD 10 and 100 mg · kg-1 in male SD rats were as follows:area under the curve (AUC(0-t)) was 22 548±1579 and (203 395±27 586) h·iμg Eq.·L-1,half time (t1/2) was 15.8±1.0 and (14.1±0.9) h,peak time (Tmax) was 4.0±3.0 and (6.0±5.0) h,peak concentration (Cmax) was 1450±355 and (7776±1703) μg Eq.·L-1.JJBD was mainly distributed in fat,livers,kidneys,stomachs and intestinal walls.The concentration of JJBD in most of the tissues reached peak values after 4 h.However,JJBD couldn't be detected in the muscle,thymus gland,brain,gonad or spleen.Excretion rate of JJBD was 43.1% in urine,29.7% in feces and 9.97% in cleaning solution within 0-168 h.JJBD could be excreted through bile at a rate of 28.1% within 0-72 h.CONCLUSION JJBD can be absorbed immediately and excreted slowly in SD rat.There is no accumulation risk.The distribution of JJBD in vivo is very extensive,but cannot go through the blood-brain barrier.JJBD is mostly excreted through feces and urine.
10.Medical fear and influencing factors of school-age children in hospital in Lanzhou area

Objective To study medical fear and its influencing factors in hospitalized school-age children in Lanzhou area.Methods Children's Medical Fear Scale was used to investigate 110 cases of hospitalized school-age children,and the investigation result underwent analysis.Results The total score of children's medical fears was (50.43±7.34),the three items with the highest score among 30 items were surgery,death,being deprived of education,homework delay,surgery and death tied for first.Conclusions Medical fear of school-age children was related with age and gender,female was significantly higher than that of male.No relationship with the family education environment,duration of illness,area of residence.
