To investigate the effects of Wogonin (WO) on learning and memory impairment, Aβ1-42 was injected intracerebroventricularly to induced a mouse learning and memory impairment model, and D-galactose was injected intraperitoneally to induced a mouse acute aging model. Mice were administered WO (75, 150, or 300 mg/kg) by oral gavage for 28 consecutive days. Cognitive function was assessed using the Morris water maze (MWM), novel object recognition (NOR), and open field tests (OFT). In the Aβ1-42 model, WO treatment (150 and 300 mg/kg) significantly improved the recognition index in the NOR test, while the 150 mg/kg group showed increased target quadrant preference in the MWM test. No changes in the total distance traveled in OFT. In the D-galactose aging model, the 150 mg/kg WO group exhibited increased platform crossings in the MWM test, and all WO doses (75, 150, and 300 mg/kg) enhanced target quadrant preference, with no alterations in spontaneous movement. Western blot analysis revealed that WO significantly attenuated hippocampal apoptosis in both models. These findings suggest that WO ameliorates learning and memory impairment associated with Alzheimer’s disease and aging.

Objective:To explore the value of spectral CT parameters combined with dynamic contrast enhanced magnetic resonance imaging(DCE-MRI) parameters in the short-term efficacy of concurrent chemoradiotherapy for nasopharyngeal carcinoma. Methods: A total of 110 cases with nasopharyngeal carcinoma Ⅲ-Ⅳ staging who received synchronous radiotherapy and chemotherapy at our Hospital from October 2022 to October 2024 were regarded as the study subjects. Complying with the evaluation results after radiotherapy and chemotherapy, they were divided into a complete remission(CR) group of 53 cases and a non CR group of 57 cases. All patients underwent DCE-MRI and energy dispersive CT scans to obtain parameters, such as iodine concentration(IC), volume transfer constant(Ktrans), slope of spectral HU curve(λHU), rate constant(Kep), and normalized iodine concentration(NIC). Logistic regression analysis was used to screen for influencing factors. ROC curve was used to analyze the evaluation value of various parameters. In addition, Z-test was used to compare area under the curve(AUC). Results:The proportion of retropharyngeal lymph node metastasis and λHUvalue in the non CR group were higher than those in the CR group, while Ktrans, Kep, IC value, and NIC value were lower than those in the CR group(P<0.05). Retropharyngeal lymph node metastasis, Ktrans, Kep, IC value, λHUvalue, and NIC value were all influencing factors(P<0.05). The AUC of individual prediction of Ktrans, Kep, IC value, λHUvalue, and NIC value was 0.817, 0.800, 0.785, 0.783, and 0.835, respectively. The AUC of the combination of DCE-MRI parameters, the combination of spectral CT parameters, and the combination of the five parameters were 0.874, 0.900, and 0.980, respectively, the AUC of the combination of the five parameters was significantly higher than the AUC of each indicator alone, the AUC of the combination of DCE-MRI parameters, and the AUC of the combination of spectral CT parameters(P<0.05). Conclusion:The DCE-MRI, and spectral CT parameters (Ktrans, Kep, IC value, λHUvalue, and NIC value)can be used to evaluate concurrent radiotherapy and chemotherapy short-term efficacy for nasopharyngeal carcinoma. And the combination of various parameters can greatly improve the predictive value of efficacy, which has important clinical application value.

In recent decades, the prevalence of hyperuricemia and gout has increased dramatically due to lifestyle changes. The drugs currently recommended for hyperuricemia are associated with adverse reactions that limit their clinical use. In this study, we report that berberine (BBR) is an effective drug candidate for the treatment of hyperuricemia, with its mechanism potentially involving the modulation of gut microbiota and its metabolite, succinic acid. BBR has demonstrated good therapeutic effects in both acute and chronic animal models of hyperuricemia. In a clinical trial, oral administration of BBR for 6 months reduced blood uric acid levels in 22 participants by modulating the gut microbiota, which led to an increase in the abundance of Bacteroides and a decrease in Clostridium sensu stricto_1. Furthermore, Bacteroides fragilis was transplanted into ICR mice, and the results showed that Bacteroides fragilis exerted a therapeutic effect on uric acid similar to that of BBR. Notably, succinic acid, a metabolite of Bacteroides, significantly reduced uric acid levels. Subsequent cell and animal experiments revealed that the intestinal metabolite, succinic acid, regulated the upstream uric acid synthesis pathway in the liver by inhibiting adenosine monophosphate deaminase 2 (AMPD2), an enzyme responsible for converting adenosine monophosphate (AMP) to inosine monophosphate (IMP). This inhibition resulted in a decrease in IMP levels and an increase in phosphate levels. The reduction in IMP led to a decreased downstream production of hypoxanthine, xanthine, and uric acid. BBR also demonstrated excellent renoprotective effects, improving nephropathy associated with hyperuricemia. In summary, BBR has the potential to be an effective treatment for hyperuricemia through the gut-liver axis.

Objective:To investigate the expression of trefoil factor 3 (TFF3) in gastric cancer and its regulation mechanism on the gastric cancer cells proliferation. Methods:The differences in the expression levels of TFF3 in 66 cases of gastric cancer were detected by RT-PCR and immunohistochemical method. SGC-7901 cells were cultured stably. Through the expression of TFF3 gene in silent cells infected with gv248 lentivirus vector (SGC-7901/KD group) and the setting up of control group (SGC-7901/NC group), CCK-8 method, flow cytometry and Transwell method were used to detect the changes of cell activity. SGC-7901/KD or SGC-7901/NC cells were injected into the right armpit of 20 BALB/C nude mice respectively. The expression of VEGF in tumor was detected by immunohistochemical method and Western blot method. Results:The expression level of TFF3 mRNA in gastric cancer tissues (15.31±2.18) was significantly higher than that in adjacent tissues (4.58±1.69) ( P < 0.05). After infection with recombinant lectin virus, the expression level of TFF3 gene in SGC-7901/KD cells was 0.08±0.006, significantly lower than that in the other two groups. After 5 days of culture, the OD value of cells in SGC-7901/KD group was 1.28±0.05, significantly lower than that in SGC-7901/NC group (1.78±0.08). The number of cell colonies formed was also significantly reduced, and the colonies were smaller. Compared with the control group, the proportion and apoptosis rate of SGC-7901/KD cells in G0/G1 phase were significantly higher, while the proportion of cells in G2/M phase was significantly lower. The results of scratch healing test showed that the distance between wound edges of SGC-7901/KD cells was significantly shorter than that of SGC-7901/NC cells. Western blot analysis showed that the relative expression level of VEGF protein in SGC-7901/KD cells (0.98±0.05) was significantly lower than that in SGC-7901/NC cells (2.87±0.17) ( P < 0.05). Compared with SGC7901/NC group, after 2 weeks of injection of SGC-7901/KD cells, the tumor volume of mice was (184.4±50.2) mm 3, significantly smaller than that of SGC7901/NC group (326.8±76.9) mm 3 ( P < 0.05). The expression of VEGF protein in SGC7901/KD group was significantly lower than that in control group ( P < 0.05) . Conclusion:In gastric cancer cells, TFF3 gene silencing can inhibit the growth of gastric cancer cells by inhibiting the expression of VEGF, which is expected to become a new molecular target.

Objective:To explore the diagnostic value of magnetic resonance imaging (MRI) Q-Dixon fat quantification technique in differentiating vertebral metastases from hemangiomas in cancer patients. Methods:A retrospective analysis was conducted on 20 patients with vertebral metastases and 8 with vertebral hemangiomas who underwent vertebral MRI scans at the National Cancer Center/National Clinical Research Center for Cancer/Cancer Hospital, Chinese Academy of Medical Sciences and Peking Union Medical College from December 2021 to December 2023. Two radiologists independently measured the fat fractions (FF) in three areas (the lesion area, the normal area of the same vertebra, and the normal area of an adjacent vertebra) and evaluated the consistency of measurements. Group differences were tested using independent sample t-tests or Mann-Whitney U tests, and diagnostic performance was assessed by plotting the receiver operating characteristic (ROC) curve and calculating the area under the curve (AUC). Results:There was very high inter-observer consistency in the FF measurements across the three regions. The FF in the lesion areas of vertebral metastases group was significantly lower than that in the vertebral hemangioma group (13.8 ± 11.5 vs. 56.5 ± 22.1), there was statistical difference ( P<0.01). There were no significant differences in the FF of normal vertebral areas between the two groups ( P>0.05). ROC curve analysis showed that FF could differentiate vertebral metastases from hemangiomas with an AUC of 0.931, a specificity of 90%, and a sensitivity of 87.5%. Conclusions:The FF measured by the Q-Dixon quantitative fat technique can accurately differentiate between vertebral hemangiomas and vertebral metastases, providing more precise guidance for the diagnosis of vertebral lesions.

ObjectiveBased on the new method of animal model evaluation, this paper summarized and analyzed the characteristics of traditional Chinese medicine(TCM) and Western medicine syndromes in existing autism spectrum disorder(ASD) animal models, and put forward suggestions for improvement. MethodRelevant literature on ASD animal models in China National Knowledge Infrastructure(CNKI) and PubMed were searched. According to the diagnostic standards of traditional Chinese and western medicine, core symptoms and accompanying symptoms were assigned values, and the clinical consistency of the models was comprehensively evaluated. ResultMost ASD model experimental animals were rodents, and the modeling methods included genetic and non-genetic. Domestic research focused on biochemical induction, while foreign research used genetic models more commonly. Among all models, valproic acid induction had the highest clinical consistency, followed by the neuroligin 4(NLGN4) and contactin associated protein like 2(CNTNAP2) gene knockout models. Most modeling methods could meet the characteristics of surface validity and structural validity, but did not clearly distinguish TCM syndromes. Currently, there is no model that has a high degree of clinical agreement between TCM and western medicine at the same time. ConclusionThe existing ASD animal models are mostly constructed under the guidance of western medicine, which lacks the characteristics of TCM syndromes. And the selection of evaluation indicators of western medicine is relatively single, without specifying the types of TCM syndromes. It is recommended to apply TCM intervention factors during the process of model preparation, to improve the evaluation indicators of traditional Chinese and western medicine, and to emphasize the study of on non-human primates, so as to lay a solid foundation for future experiments.

Chinese Journal of Medical Education Research 2024;23(3):382-386
As essential part of teachers' teaching practice, the design and arrangement of assignments is the main carrier of interaction between teachers and students, and is also an important learning way for students. According to the discipline characteristics and teaching modes of histology and embryology courses, we have developed a systematic assignment design model that covers theory, practice, and application with four types of assignments—pre-class assignments, exploratory assignments, practical assignments, and innovative assignments distributed before class, in class, after class, and throughout the teaching process. Behind this assignment design strategy is the concept of learner-centered teaching, which aims to help students learn professional knowledge and develop comprehensive literacy.
