Recently, continuing medical education has made progress under the support of national policies. However, the exploration of continuing medical education has overlooked the concept of non-technical skills. The lack of training in non-technical skills has become increasingly prominent, which seriously restricts the development of medical education. Drawing on the developmental trajectory and curriculum design experiences of non-technical skill training in Europe and the United States, this study examines the current situation and the problems of non-technical skill training in China. By establishing an effective curriculum support system to enhance engagement, improving the training process for non-technical skills to optimize the curriculum structure, and focusing on faculty cultivation targets to establish admission standards, this study aims to promote the steady and healthy advancement of non-technical skill training, and achieve a "win-win" situation in terms of patient safety and the sustainable development of continuing medical education.

Objective To search for and verify the genetic factors that lead to familial thoracic aortic aneurysm and dissection. Methods Patients with a family history were screened from clinical cases of thoracic aortic aneurysm and dissection, and their relevant clinical data were collected. After extracting the whole-genome DNA from the proband's blood samples, gene panel testing was carried out,and first-generation sequencing was used to verify the blood samples of the proband's immediate and collateral relatives.Meanwhile, relevant cell experiments were designed.First, the ACTA2-Leu195 * point mutation plasmid was constructed, and then the mutant plasmid and wild-type ACTA 2 were transfected into cells through expression plasmids respectively. After 48 hours,RNA and total protein were extracted, and the expression levels were detected by qPCR and Western blotting, respectively. Results Through high-throughput sequencing, the mutation site of c.583delC in the ACTA2 gene was found, and it was also confirmed that the heterozygous mutation at this site in the family was closely related to familial thoracic aortic aneurysm and dissection. The results of cell experiments showed that the mRNA and protein expression levels in the group transfected with the ACTA2-Leu195 * mutant plasmid were significantly decreased compared with those of the wild type, suggesting that the mutant could not express proteins normally. Conclusion Through clinical family studies and cell-level experiments,a new mutation site in ACTA2 leading to familial thoracic aortic aneurysm and dissection has been discovered, which provides a new approach for the screening, detection and clinical treatment of familial thoracic aortic aneurysm and dissection.

Chinese Journal of Cancer Biotherapy 2025;32(6):587-593
Objective:To investigate the effects of cyclic adenosine monophosphate response element binding protein(CREB)on the malignant biological behaviors of prostate cancer PC3 cells. Methods:Prostate cancer PC3 cells were routinely cultured. The overexpression control plasmid(vector),CREB overexpression plasmid(CREB-oe),knockdown control sequence(si-NC),and si-CREB sequence were transfected into PC3 cells using transfection reagents, namely vector,CREB-oe,si-NC,and si-CREB groups.Scratch wound healing assay,Transwell chamber assay, and flow cytometry were performed to evaluate cell migration, invasion, and cell cycle distribution,respectively.PC3 cells with CREB knockout were constructed using CRISPR/Cas9 technology, and a xenograft tumor model was employed to evaluate the impact of CREB knockout on tumor growth in vivo. Results:CREB was successfully knocked down or overexpressed in PC3 cells(all P<0.01).CREB overexpression significantly promoted, while its knockdown significantly inhibited the migration and invasion of PC3 cells(all P<0.01).Overexpression of CREB promoted the transition of PC3 cells into the S phase, whereas knockdown of CREB induced G1 phase arrest(all P<0.01).PC3 cells with CREB knockout were successfully constructed, and CREB knockout significantly inhibited the growth of PC3 cell-transplanted tumors(P<0.01).Conclusion:Knockdown or knockout of CREB inhibits migration and invasion of PC3 cells and induces G1 phase arrest, thereby suppressing tumor growth.

Objective To compare the effects of three nickel-titanium file systems,ProTaper Next,TruNatomy, and VDW.RO-TATE,on the morphology of the Danger Zone of the mesial buccal root of mandibular first molar after root canal preparation using Mi-cro-CT and 3D printing technology. Methods 3D-printed mandibular first molars were selected and designed for purpose. They were randomly divided into three groups according to the used NiTi files(n=20).Micro-CT was used to scan the cross-sectional images of the Danger Zone 2 mm below the furcation of the mesial buccal root before and after root canal preparation. The changes in the root ca-nal wall thickness, root canal volume, surface area,cross-sectional area, and root canal transportation in the Danger Zone were ob-served.Statistical analysis was performed using one-way ANOVA(P<0.05).Results Statistically significant differences were observed before and after root canal preparation in the Danger Zone among the three groups(P<0.05).Among the three groups, the PTN files caused the largest change before and after preparation, followed by VDW files, and the TRU files had the smallest change. Conclusion The study highlights TruNatomy's conservative shaping capacity, advocating its use in minimally invasive endodontics, whereas Pro-Taper Next may be reserved for cases requiring aggressive canal preparation.
