1.Neuroprotective effect and mechanism of eleutheroside B on Parkinson’s disease model mice by regulating the IKKβ/NF-κB signaling pathway

OBJECTIVE To investigate the neuroprotective effect and mechanism of eleutheroside B (ELB) on Parkinson’s disease (PD) model mice by regulating the IκB kinase β (IKKβ)/nuclear factor-κB (NF-κB) signaling pathway. METHODS Fifty mice were randomly divided into normal control group, model group, positive control group (selegiline hydrochloride, 10 mg/kg), and ELB low-dose and high-dose groups (80, 160 mg/kg), with 10 mice in each group. Each group was given relevant medicine or normal saline intragastrically for 14 consecutive days. Starting from the 10th day of administration, the model group and all administration groups were intraperitoneally injected with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) 30 mg/kg, for five consecutive days to establish the chronic PD model. After the last administration for 24 h, six mice were randomly selected from each group to test their behavioral abilities; detect the levels of interleukin-1β (IL-1β), IL-10, tumor necrosis factor-α (TNF-α) in brain tissue and their mRNA expressions were measured, and positive expression of tyrosine hydroxylase (TH), protein expressions of TH, α -synuclein ( α -syn), ionized calcium-binding adaptor molecule 1 (Iba-1), as well as phosphorylation levels of IKKβ and NF-κB p65 proteins in the brain tissue were detected. The ultrastructure of neurons in substantia nigra was observed. RESULTS Compared with the model group, rotarod endurance time and climbing score of each administration group (except for the ELB low-dose group) were increased significantly ( P <0.05), while the levels and mRNA expressions of IL-1β, TNF-α, α -syn, and Iba-1, as well as phosphorylation levels of IKKβ and NF-κB p65 proteins in brain tissue were decreased significantly (except for TNF-α in the ELB low-dose group). Conversely, the level and mRNA expression of IL-10 (except for the ELB low-dose group), TH positive expression and protein expressions were significantly increased ( P <0.05). Typical neurodegenerative pathological changes, such as neuronal karyopyknosis, mitochondrial swelling and vacuolization, and endoplasmic reticulum dilation, all showed varying degrees of improvement. CONCLUSIONS ELB may exert neuroprotective effects by inhibiting the activation of the IKKβ/NF-κB signaling pathway, alleviating inflammatory responses, reducing abnormal α -syn aggregation and neuronal loss, and further improving motor dysfunction in PD mice.
3.Role of color Doppler echocardiography and electrocardiography in evaluating cardiac function and disease severity in patients with hypertensive heart disease

Objective To evaluate the value of color Doppler echocardiography and electrocardiography in assessing cardiac function and disease severity in patients with hypertensive heart disease.Methods A retrospective analysis was conducted on 205 patients with hypertensive heart disease who were categorized into 3 groups based on the New York Heart Association(NYHA)criteria:class Ⅱ(n=62),class Ⅲ(n=89)and class Ⅳ(n=54).The demographic characteristics(including gender and age)and the results of color Doppler echocardiography and electrocardiography were extracted from the hospital's medical record system for analyzing the relationships of the diagnostic findings from color Doppler echocardiography and electrocardiography with cardiac function and disease severity.Results Electrocardiography revealed statistically significant intergroup differences in QTc interval,QRS voltage,and QRS duration across 3 groups(P<0.05).Among 3 groups,class Ⅳ group exhibited the most pronounced abnormalities with prolonged QTc interval and QRS duration,along with reduced QRS voltage(P<0.05).Compared with class Ⅱ group,class Ⅲ group had longer QTc interval and QRS duration,alongside lower QRS voltage(P<0.05).Color Doppler echocardiography showed that left ventricular systolic diameter,left ventricular diastolic diameter,left atrial diameter,left ventricular posterior wall thickness,interventricular septum thickness,left ventricular ejection fraction,and E/A ratio differed significantly across 3 groups(P<0.05).Class Ⅳ group exhibited the largest left ventricular systolic diameter,left ventricular diastolic diameter and left atrial diameter,the thickest left ventricular posterior wall and interventricular septum,and the lowest left ventricular ejection fraction and E/A ratio(P<0.05).Compared with class Ⅱ group,class Ⅲ group had enlarged left ventricular systolic diameter,left ventricular diastolic diameter and left atrial diameter,thicker left ventricular posterior wall and interventricular septum,and reduced left ventricular ejection fraction and E/A ratio(P<0.05).Spearman correlation analysis found that QTc interval,QRS duration,left ventricular systolic diameter,left ventricular diastolic diameter,left atrial diameter,left ventricular posterior wall thickness,and interventricular septum thickness were positively correlated with cardiac functional impairments in patients with hypertensive heart disease,while QRS voltage,left ventricular ejection fraction,and E/A ratio were negatively correlated with cardiac functional impairments in patients with hypertensive heart disease(P<0.05).Multiple linear regression analysis identified QTc interval,QRS duration,left ventricular end-systolic diameter,left ventricular end-diastolic diameter,left atrial diameter,left ventricular posterior wall thickness,ventricular septum thickness,QRS voltage,left ventricular ejection fraction,and E/A ratio as critical factors for assessment of cardiac function(P<0.05).Conclusion Color Doppler echocardiography and electrocardiography demonstrate significant clinical utility in evaluating cardiac function and disease severity in patients with hypertensive heart disease.

5.Effect of lactobacillus plantarum LB12 on renal calcium oxalate stones in rats

Objective To investigate the effect of Lactobacillus plantarum LB12 on kidney calcium oxalate stones in rats.Methods A total of 24 male SD rats aged 6~8 weeks were randomly divided into four groups:control group,model group,LB12 group,and LB12 intervention group,with 6 rats in each group.The control group and LB12 group had free access to water,and were gavaged with 2 mL of normal saline in the morning.In the evening,the control group received 2 mL of normal saline,while the LB12 group received 2 mL of Lactobacillus plantarum LB12 suspension.The model group and LB12 intervention group were given 1%ethylene glycol in drink-ing water and 2 mL of 2%ammonium chloride solution via gavage in the morning.In the evening,both groups received 2 mL of normal saline and 2 mL of Lactobacillus plantarum LB12 suspension,respectively.After the experiment,the body weight change trend of the rats was recorded,and the levels of urinary calcium,urinary oxalate,serum creatinine,blood urea nitrogen,renal SOD,and MDA were measured.Additionally,H&E staining,VK staining,immunohistochemistry,ROS fluorescence analysis,and Western blot(WB)were performed on kidney tissues.Results Lactobacillus plantarum LB12 demonstrated a 12.34%oxalate degradation rate in vitro.Further animal experiments indicated that Lactobacillus plantarum LB12 alleviated the weight loss trend in the model rats and reduced the levels of urinary calcium,urinary oxalate,serum creatinine,urea nitrogen,and MDA,while increasing SOD levels(P<0.05).Histopathological analysis revealed that the LB12 intervention group had signifi-cantly fewer renal crystals compared to the model group,with lower expressions of IL-6 and ROS(P<0.01).Western blot analysis demonstrated that Lactobacillus plantarum LB12 inhibited the expression of TLR4,NF-κB,and OPN proteins(P<0.01).Conclusions Lactobacillus plantarum LB12 can effectively alleviate the oxidative stress damage caused by calcium oxalate stones to the kidneys and reduce kidney crystal formation in rats.Further-more,it inhibits the expression of TLR4/NF-κB and OPN,making it a potential therapeutic approach for the prevention and treatment of kidney calcium oxalate stones.
6.Epidemiological investigation on diphtheria serum IgG antibody levels in people aged 1-59 years old in Shaanxi province

Objective To investigate the serum diphtheria antibody levels in healthy people aged 1-59 years old in Shaanxi Province.Methods A stratified random sampling method was used to select healthy people aged 1-59 years old from 30 county-level disease surveillance sites in 10 cities of Shaanxi province from November to December 2017.Diphtheria IgG antibody was detected by enzyme-linked immunosorbent assay(ELISA),and the positive rate(IgG≥0.01 IU/mL)and mean geometric concentration(GMC)of diphtheria IgG antibody were analyzed.Results A total of 6 439 subjects completed laboratory testing.The positive rate and GMC of diphtheria IgG antibody were 85.79%and 0.035(0.034-0.036)IU/mL,respectively.The GMC and the positive rate of diphtheria IgG antibody were higher in males than those in females(P<0.01).The positive rate of diphtheria IgG antibody in different cities was 65.25%-98.23%,and the GMC was 0.021(0.018-0.024)IU/mL-0.046(0.037-0.055)IU/mL,the difference between the groups was statistically significant(P<0.01).The GMC of the group with DCV immunization history,the group without DCV immunization history and the group with unknown DCV immunization history were 0.097(0.090-0.105),0.018(0.018-0.019)and 0.027(0.026-0.028)IU/mL,respectively,and the positive rates of diphtheria IgG antibody were 92.74%,80.18%and 85.44%,respectively.There were significant differences between the three groups(P<0.01).Among the 9 age groups,the group with the lowest positive rate of diphtheria IgG antibody was the 40-49 age group(78.39%),and the highest was in the 1-2 age group(95.77%),which decreased with age(χ2=200.612,P<0.01).It showed a downward trend with age(χ2=200.612,P<0.01).The lowest GMC(IU/mL)was in the 40-49 age group[0.017(0.016-0.017)],and the highest was in the 7-9 age group[0.190(0.162-0.226)],showing a downward trend with increasing age(P<0.01).After the whole course of vaccination,the positive rate of diphtheria IgG antibody and the interval between the last dose of vaccination showed a downward trend(P<0.01).Conclusion The diphtheria immune barrier in whole population has been established in Shaanxi province,but the positive rate of diphtheria antibody and GMC in healthy people aged 1-59 years old show a downward trend with age.
7.Effect of triply periodic minimal surfaces structure and ceramic volume fraction on mechanical properties of polymer-infiltrated ceramic network composites fabricated by additive manufactured zirconia and resin

Objective:To investigate the effect of triply periodic minimal surfaces (TPMS) structure and ceramic volume fraction on the mechanical properties of polymer-infiltrated ceramic network (PICN) composite and reveal its strengthening and toughening mechanism.Methods:In this study, TPMS structures with gyroid (G), primitive (P), diamond (D), and ceramic volume fraction (40%, 55%, 70%, 85%) were designed. Porous zirconia scaffolds were prepared using stereolithography technology, and resin was infiltrated into the scaffolds through a vacuum. Then, the PICN composites were obtained after curing. The bending strength, elastic modulus and fracture toughness of PICN were tested using an electronic universal testing machine, with commercial PICN as the control group. The micromorphology of PICN was observed through stereomicroscope and scanning electron microscope. The cytocompatibility of PICN was verified by using cell counting kit, live/dead cell staining and phalloidin staining.Results:The bending strength values of PICN with different ceramic volume fractions ranged from 82.0 MPa to 376.0 MPa, and they gradually increased as the ceramic volume fraction rised. The elastic modulus values of PICN with different ceramic volume fractions ranged from 12.1 GPa to 56.1 GPa. The fracture toughness values of PICN with different ceramic volume fractions ranged from 1.7 MPa·m 1/2 to 6.5 MPa·m 1/2. The bending strength of 85G PICN reached 306.0 MPa, and it had the highest fracture toughness (6.5 MPa·m 1/2) and an appropriate elastic modulus between that of the control group and that of enamel. Under scanning electron microscopy, it could be observed that the cracks branch and deflect at the interface and eventually terminate within the resin phase. After co-culture with PICN, the survival rate of mouse fibroblasts exceeded 80%, indicating that PICN had no cytotoxicity. Conclusions:The PICN composite with TPMS structure can satisfy the mechanical properties and cytocompatibility of dental prosthesis.
8.Changes in cortical electroencephalogram BSR during sevoflurane anesthesia and correlation with cerebral blood perfusion in septic mice

Objective:To evaluate the changes in cortical electroencephalogram (EEG) burst suppression rate (BSR) during sevoflurane anesthesia in septic mice and the correlation with cerebral blood perfusion.Methods:Forty SPF male C57BL/6J mice, aged 8-10 weeks, weighing 22-25 g, were divided into 2 groups ( n=20 each) by the random number table method: sham operation group (Sham group) and cecal ligation perforation group (CLP group). The sepsis model was established by cecal ligation and puncture in anesthetized animals. Mice in both groups inhaled 2% sevoflurane for 2 h. During sevoflurane anesthesia, BSR (30 min as an epoch) on electroencephalogram was recorded, and the cortical cerebral blood perfusion was recorded using the laser speckle flow imaging at 30, 60, 90 and 120 min of anesthesia. Results:Compared with Sham group, the cortical EEG BSR was significantly increased, and the cortical cerebral blood perfusion was decreased during sevoflurane anesthesia in CLP group ( P<0.05). Cortical EEG BSR was negatively correlated with cortical cerebral blood perfusion ( P<0.05). Conclusions:Cortical EEG BSR increases during sevoflurane anesthesia in septic mice, which may be related to decreased cortical cerebral blood perfusion.
9.Gas signaling molecules activate cGAS-STING signaling pathway in tumor immunotherapy

Cancer is one of the major diseases of high morbidity and mortality worldwide,and its therapeutic approaches are facing great challenges.Immunotherapy,especially the activation of innate immunity represented by the cGAS-STING signaling pathway,is the current research hotspot in tumor immunotherapy.Activation of innate immune response by gas therapy is the latest development in tumor therapeutic approaches,especially the use of gas signaling molecules(NOx CO,H2S and SO2)to activate the cGAS-STING signaling pathway to induce intrinsic immunity of the organism,which leads to anti-tumor immunotherapy.Although intrinsic immunity activated by gas signaling molecules plays an important role in tumor immunotherapy,few reviews have been reported on its association with the cGAS-STING signaling mechanism.In this paper,we will comprehensively describe how gas signaling molecules damage the mitochondrial matrix and DNA damage through oxidative/nitrosative stress,thereby activating the cGAS-STING signaling pathway and triggering the innate immune cascade,aiming to summarize the process of activation of anti-tumor immune effects by gas signaling molecules,and to provide more references for the gas therapies in the future anti-tumor immunity research.
10.Safety evaluation of tetravalent meningococcal conjugate vaccine in combination with the inactivated poliomyelitis vaccine and diphtheria-tetanus-acellular pertussis vaccine for infants

Objective:To investigate the safety of the tetravalent meningococcal conjugate vaccine (MPCV-ACYW) in combination with the inactivated poliomyelitis (IPV) vaccine and diphtheria-tetanus-acellular pertussis (DTaP) vaccine for infants aged 3-5 months and provide real-world evidence for the immunization strategy of vaccine combination.Methods:From June to October 2023, a total of 600 3-month-old infants were selected and divided into three groups: control group, mono-vaccination group and combined vaccination group. They were simultaneously or individually vaccinated with MPCV-ACYW, IPV and DTaP vaccines at 3, 4, and 5 months of age, respectively. The incidence rate of adverse reactions within 30 days after each dose was observed.Results:All 600 infants completed at least one vaccination and entered the safety data analysis. The age of the control group (100 infants), the mono-vaccination group (250 infants), and the combination group (250 infants) was (101.20±7.88), (102.26±7.94), and (102.35±7.76) days, respectively. The body lengths were (63.00±3.02), (62.55±3.06), and (63.14±4.02) cm, respectively. The body weights were (6.90±0.77), (6.86±0.94), and (6.99±0.95) kg, respectively. Boys accounted for 49%, 50.4%, and 52.4%, respectively, with no statistically significant differences (all P>0.05). The overall incidence rates of adverse reactions in the control group, mono-vaccination group, and combined vaccination group were 4.00%, 2.80%, and 3.20%, respectively, with systemic adverse reaction rates of 3.00%, 2.40%, and 2.00%. The incidence rates of local adverse reactions were 1.00%, 0.40%, and 1.20%, with no statistically significant differences (all P>0.05). Adverse reactions were mainly grade 1, with incidence rates of grade 1 adverse reactions of 3.00%, 2.00%, and 1.60% in the three groups, and incidence rates of grade 2 adverse reactions of 1.00%, 0.80%, and 1.60%, respectively. No grade 3 or 4 serious adverse reactions occurred, and the differences were not statistically significant (all P>0.05). The adverse reaction symptoms of the three groups were mainly systemic reactions, among which fever and diarrhea symptoms were reported in individual cases in each group, with no statistically significant differences in the incidence rate (all P>0.05). The symptoms of adverse reactions were mostly transient and self-relieved, all of which were cured. Conclusion:The combination of MPCV-ACYW and IPV or DTaP vaccines is safe for infants aged 3-5 months.
