Objective To explore the feasibility and reference value of allogeneic lung transplantation and postoperative monitoring in miniature pigs for lung transplantation research. Methods Two miniature pigs (R1 and R2) underwent left lung allogeneic transplantation. Complement-dependent cytotoxicity tests and blood cross-matching were performed before surgery. The main operative times and partial pressure of arterial oxygen (PaO2) after opening the pulmonary artery were recorded during surgery. Postoperatively, routine blood tests, biochemical blood indicators and inflammatory factors were detected, and pathological examinations of multiple organs were conducted. Results The complement-dependent cytotoxicity test showed that the survival rate of lymphocytes between donors and recipients was 42.5%-47.3%, and no agglutination reaction occurred in the cross-matching. The first warm ischemia times of D1 and D2 were 17 min and 10 min, respectively, and the cold ischemia times were 246 min and 216 min, respectively. Ultimately, R1 and R2 survived for 1.5 h and 104 h, respectively. Postoperatively, in R1, albumin (ALB) and globulin (GLB) decreased, and alanine aminotransferase increased; in R2, ALB, GLB and aspartate aminotransferase all increased. Urea nitrogen and serum creatinine increased in both recipients. Pathological results showed that in R1, the transplanted lung had partial consolidation with inflammatory cell infiltration, and multiple organs were congested and damaged. In R2, the transplanted lung had severe necrosis with fibrosis, and multiple organs had mild to moderate damage. The expression levels of interleukin-1β and interleukin-6 increased in the transplanted lungs. Conclusions The allogeneic lung transplantation model in miniature pigs may systematically evaluate immunological compatibility, intraoperative function and postoperative organ damage. The data obtained may provide technical references for subsequent lung transplantation research.

Objective To explore the frequency domain characteristics of pedestrian head dynamic response in vehicle collisions and investigate the correlation between frequency domain parameters and time domain injury criteria. Methods Finite element modelling based on human body model was used to simulate the process of pedestrian head-vehicle impacts, and the wavelet packet signal analysis method was employed to obtain the frequency domain response of pedestrian head in the simulations. Results The head energy under impacts at the bonnet area was mainly dispersed in the 0-300 Hz frequency band, while the head energy under impacts at the windshield area was mainly concentrated in the 0-5 Hz frequency band. The peak energy of frequency band for pedestrian head generally increased with the increase of linear and rotational speed, and the influence of rotational speed on the peak energy of frequency band was more significant when the linear speed was higher. The linear correlation R2 between the peak energy of the head frequency band caused by the collision between the bonnet and windshield area and the time-domain criterion for skull injury were 0.85 and 0.61,respectively.But their correlation with the time-domain indicators for brain injury evaluation was relatively low(R2<0.5).Conclusions The frequency domain response characteristics of pedestrian heads are affected by collision speed and location. The peak energy of the frequency band can potentially characterize the risk of skull injury, but the frequency band and concentration of the peak energy in the frequency band are not related to the risk of head injury. This study can provide references for the evaluation of head blunt injury combined with time-frequency response.

Fatty liver is common disease of nutritional metabolism in the perinatal period,character-ized by elevated levels of NEFA in the blood and disorders of lipid metabolism.High concentra-tions of NEFA damage mitochondria and promote the release of reactive oxygen species(ROS).At the same time, lipid peroxidation occurs in lipid accumulation in hepatocytes, producing free radi-cals such as ROS,which leads to oxidative stress in the liver. When the level of intracellular ROS increases,thioredoxin-interacting protein(TXNIP)activates nucleotide-binding oligomerization structure-like protein 3(NLRP3)inflammasomes,and oxidative stress can regulate pyroptosis, but it is unclear whether reactive oxygen species(ROS)produced by oxidative stress in hepatocytes can mediate pyroptosis and induce liver injury in dairy cows through the TXNIP/NLRP3 pathway. In this study, liver tissue samples from healthy dairy cows and fatty liver cows were collected, and NEFA was used to construct a fatty liver cell model, and triglyceride(TG)content and oxidative stress related indicators were detected by kit.Western blot was used to detect the activation of NL-RP3 inflammasomes, the inflammatory pathway of NF-κB and the expression levels of pyroptosis-related proteins.Fluorescence quantitative PCR was used to detect the relative expression level of inflammatory factor mRNA.The results showed that compared with the healthy(control)group, the TG content of fatty liver tissue and fatty liver cells was significantly increased(P<0.05),the activities of SOD and GSH-Px were significantly decreased(P<0.05),and the protein expression levels of TXNIP,NLRP3,GSDMD-N and Caspase-1 were significantly up-regulated(P<0.05).The results of the antioxidant model of fatty hepatocytes using NEFA and antioxidants(NAC)showed that compared with the fatty hepatocyte model, the content of ROS in hepatocytes was sig-nificantly reduced, and oxidative stress,NLRP3 inflammasome activation and pyroptosis were alle-viated.In summary, this study found that when fatty liver disease occurs in dairy cows,ROS pro-duced by oxidative stress in the liver can mediate pyroptosis through the TXNIP/NLRP3 path-way,which can lead to liver injury in fatty liver cows.

7.Research progress of ultrasound microbubbles in diagnosis and treatment of thyroid cancer
The Journal of Practical Medicine 2025;41(3):454-458
Ultrasonic microbubbles are vesicles withgas core stabilized by a shell comprised of lipids,pro-teins,and polymer materials. It can be used not only as a contrast agent to diagnose the disease and monitor the pathological process at the molecular level, but also as a carrier of non-invasive treatment, carrying drugs or genes to the region of interest through blood circulation, thereby exerting therapeutic effects.Compared with traditional diagnosis and treatment methods, ultrasound microbubbles show great potential.Targeted imaging and treatment can identify specific types of tumors in the early stage, and improve the prognosis by reducing the systemic toxicity of chemotherapy drugs while enhancing the anti-tumor efficacy. This provides a new idea and strategy for future diagnosis and treatment of thyroid cancer. This article reviews the application of ultrasound microbubbles in the diagnosis and treatment of thyroid cancer.

Objective:This study aimed to investigate the regulatory role and mechanism of myosin heavy chain 9 (MYH9) in the invasion of Zika virus (ZIKV) into human glioma cells (U251).Methods:Utilizing CRISPR/Cas9 technology, MYH9-knockout U251 cells (U251-MYH9 KD) were constructed. Following ZIKV infection, the protein expression levels, RNA load, and viral titer of ZIKV were detected through western blot (WB), Real-time fluorescence quantitative polymerase chain reaction (qPCR), and plaque formation assays, respectively. The infection efficiency of ZIKV in U251 cells treated with the MYH9 inhibitor blebbistatin was assessed. The binding and internalization efficiency of ZIKV were measured in U251-MYH9 KD cells. The interaction between MYH9 and the ZIKV envelope protein (E) was studied using co-immunoprecipitation (Co-IP). The effects of soluble MYH9 recombinant protein and anti-human MYH9 antibodies on ZIKV infection were evaluated by qPCR and plaque formation assays. Results:It was found that knockout or inhibition of MYH9 significantly suppressed ZIKV infection in U251 cells. MYH9 knockout notably inhibited the binding and internalization of ZIKV in U251 cells. MYH9 interacted with the ZIKV E protein, and both MYH9 recombinant protein and anti-human MYH9 antibodies, by blocking the binding of ZIKV E protein to cell surface MYH9, inhibited ZIKV infection in U251 cells in a dose-dependent manner.Conclusions:MYH9 facilitates ZIKV invasion into U251 cells through interaction with the ZIKV E protein.
