1.Exploring Detoxication Mechanism of Dioscoreae Bulbiferae Rhizoma Processed with Phaseoli Radiati Semen Juice Based on Target Organ Ferroptosis

ObjectiveTo investigate the attenuating effect of Dioscoreae Bulbiferae Rhizoma(DBR) processed with Phaseoli Radiati Semen(PRS) juice, and explore the attenuating mechanism based on ferroptosis of the main toxic target organ. MethodSixty male ICR mice were randomly divided into blank group, DBR group, water roasted DBR group(hereinafter referred to as water group), PRS juice-roasted DBR group 1(DBR-PRS 10∶1, stuffy moistening for 40 min, stir-fried at 130 ℃ for 18 min, hereinafter referred to as group 1), PRS juice-roasted DBR group 2(DBR-PRS 10∶1, stuffy moistening for 80 min, stir-fried at 100 ℃ for 14 min, hereinafter referred to as group 2), PRS juice-roasted DBR group 3(DBR-PRS=20∶3, stuffy moistening for 40 min, stir-fried at 160 ℃ for 14 min, hereinafter referred to as group 3). The raw and processed groups of DBR were gavaged with their corresponding 95% ethanol extract at a dose of 3 g·kg-1·d-1, while the blank group was gavaged with an equal volume of 0.5% sodium carboxymethyl cellulose, once a day for 14 consecutive days. Hematoxylin-eosin(HE) staining was used to observe the histopathological changes of mouse liver. Alanine aminotransferase(ALT) and aspartate aminotransferase(AST) levels in serum, as well as malondialdehyde(MDA), ferrous ions(Fe2+), reduced glutathione(GSH) and superoxide dismutase(SOD) levels in liver tissue were detected by the biochemical detection. Western blot was used to detect the expression of iron key proteins such as ferritin heavy chain 1(FTH1) and glutathione peroxidase 4(GPX4). ResultHE staining results showed that the liver tissue structure of the blank group was clear, the morphology of hepatocytes was normal, the cytoplasms of hepatocytes in the DBR group and water group were loose and vacuolar, with obvious pathological damages, and the pathologic damages of mice in the group 1-3 were significantly improved. Compared with the blank group, the levels of ALT, AST, MDA and Fe2+ in mice from the DBR group were significantly increased(P<0.01), while GSH and SOD levels were significantly reduced(P<0.01), and the protein expression levels of FTH1 and GPX4 were significantly decreased(P<0.01). Compared with the DBR group, the ALT, AST,MDA and Fe2+ levels of mice in the group 1-3 were significantly reduced(P<0.05, P<0.01), the GSH and SOD levels and the protein expression levels of FTH1 and GPX4 were significantly increased(P<0.01). Compared with the water group, the AST and MDA levels of mice in the group 1-3 were significantly reduced(P<0.05, P<0.01), the SOD level significantly increased(P<0.05, P<0.01), the FTH1 protein expression significantly increased(P<0.01), and the serum ALT level of mice in the group 2-3 significantly reduce(P<0.01), Fe2+ level significantly reduced(P<0.01), GSH level significantly increased(P<0.05, P<0.01), and GPX4 protein expression significantly increased(P<0.05, P<0.01). Among the group 1-3, the group 3 had the best detoxification effect. ConclutionProcessing with PRS juice can reduce the liver injury induced by DBR, and the mechanism may be related to the inhibition of ferroptosis in the liver.
2.Research on Wnt2/β⁃catenin pathway changes in liver regeneration and repair in C57BL/6 mice

Objective:
To explore the changing characteristics of Wnt2/β⁃catenin pathway in liver regeneration and repair in C57BL/6 mice.
Methods :
Male C57BL/6 mice were randomly divided into a sham⁃operated group (Sham group) , 1 d post⁃operative group , 2 d post⁃operative group , 4 d post⁃operative group , 6 d post⁃operative group , and 8 d post⁃operative group. Mice in the operated group underwent partial hepatectomy ( PHx) to remove the left and middle lobes of the liver, respectively. Plasma and liver tissues were collected on postoperative days 1 , 2 ,4 ,6 , and 8 , and plasma ALT and AST activity was measured by ALT(alanine aminotransferase) and AST( aspartate aminotransferase) biochemical analysis kits; Ki67 ⁃positive cells were identified by immunohistochemistry in each group;the number of HNF4⁃α and Ki67 double positive cells , the number of LYVE1 and Ki67 double positive cells , and the number of β ⁃catenin transferred into the nucleus were determined by immunofluorescence method. The expression of Wnt2 protein in each group was detected by Western blot , and the time characteristics of its expression during liver regeneration were analyzed.
Results:
Liver weight and liver/body weight ratio peaked on day 6 after PHx and approached the level of the Sham group. After PHx , there was severe liver damage on day 1 , but it had normalized by day 4. On day 2 after PHx , mainly hepatocytes proliferated;and on days 4 and 6 , mainly liver sinusoidal endothelial cells proliferated ,while the Wnt2/β⁃catenin pathway was activated.
Conclusion
The liver has a powerful regenerative repair ability ,which is closely related to the rapid proliferation of hepatocytes and liver sinusoidal endothelial cells , and the activation of the Wnt2/β⁃catenin pathway is activated in the regenerative repair of the mouse liver.
3.Establishment of Elimination Method of Outliers Based on Grubbs Rule and MATLAB Language and Its Application in Ev- aluating Drug Bitterness

OBJECTIVE: To establish the elimination method of outliers based on Grubbs rule and MATLAB language, and to evaluate the effects of it on drug bitterness evaluation. METHODS: Referring to Grubbs rule, the automatic cyclic outliers elimination method based on MATLAB language was established. Totally 20 volunteers were included in single oral taste test (Tetrapanax papyrifer) and multiple oral taste test (10 kinds of medicinal material as T. papyrifer, Changium smyrnioides, Poria cocos, etc.). Seven sensors were selected for electronic tongue test (Clematis armandii). The data of bitterness evaluation in above tests (oral taste test as bitterness value, electronic tongue test as response value of sensors) were used as the data source. Five researchers were selected and adopted table-by-table elimination method based on Grubbs rule (method one), Excel software elimination method based on Grubbs rule (method two) and automatic cyclic outliers elimination method based on Grubbs rule and MATLAB language (method three) to judge and eliminate the outliers. The effects of above three methods were evaluated with the removal time and error rate of outliers as indexes. RESULTS: There were two outliers in the data of bitterness evaluation in single oral taste test; the elimination time of the three methods were(745.400 0±25.904 4),(288.333 3±31.253 1)and(0.000 3±0.000 0)s, respectively; error rates were 20.0%, 0 and 0, respectively. There were six outliers in the data of bitterness evaluation in multiple oral taste test; the elimination time of three methods were (3 693.107 7±75.023 3), (1 494.761 4±53.826 9), (0.005 2±0.000 0)s, respectively; error rates were 10.0%, 4.0%, 0, respectively. There were three outliers in the data of bitterness evaluation in electronic tongue test; the elimination time of three methods were (2 992.673 3±84.117 6), (1 276.367 1±55.024 5), (0.002 3±0.000 0)s, respectively; error rates were 5.7%, 2.9%, 0, respectively. The elimination results of the three methods were consistent. The elimination time of method two was significantly shorter than that of method one (P<0.01); the elimination time of method three was significantly shorter than those of method one and method two (P<0.01). There was no significant difference in error rate of 3 methods (P>0.05). CONCLUSIONS: The automatic cyclic elimination method of outliers based on Grubbs rule and MATLAB language can significantly shorten the elimination time of outliers in data of drug bitterness evaluation, improve the efficiency of data processing, and is suitable for drug bitterness evaluation.
4.Clinical value of the plasma circular RNA hsa_circ_0009024 in the diagnosis and treatment of pulmonary tuberculosis

Objective Detecting plasma level of circular RNA(circRNA)hsa_circ_0009024 in pulmonary tuberculosis(TB)patients,and evaluating its diagnostic value for TB.Methods From January 2016 to December 2016, a hosptial-based, case-control study was performed, which include 90 untreated active pulmonary tuberculosis patients(TB group),75 healthy people(healthy control)and 84 patient with other diseases(other disease group).Plasma level of circRNA hsa_circ_0009024 was detected by real-time quantitative polymerase chain reaction.Furthermore, the 90 patients with TB were divided into different subgroups according to cavity formation and the lung fields involvement: patients without lung cavity(55 cases)vs those with lung cavity(35 cases),patients with involvement of <2 lung fields(49 cases)vs≥2 lung fields(41 cases).Plasma levels of hsa _circ_0009024 of 41 TB patientswere monitored andcomparedbefore and after 3 months anti-TB therapy.The sensitivity and specificity of plasma hsa_circ_0009024 were analyzed by using the receiver operating characteristic(ROC)curve.The comparison between two groups was performed with Mann-Whitney U test and the comparison among multigroupswas conducted with Kruskal-Wallis H test.Results Plasma levels of hsa_circ_0009024 in TB patients[1.98(1.42, 2.71)]were significantly higher than healthy controls[1.03(0.78,1.33)]and other disease groups[1. 13(0.77,1.51)](H=76.58,P<0.0001).Plasma levels of hsa_circ_0009024 in cavity pulmonary TB patients were higher than pulmonary TB patients without cavity(U=392.50,P<0.0001).Plasma levels of hsa_circ_0009024 in TB patients with involvement of ≥2 lung fields were higher than <2 lung fields(U=590.50,P=0.0008).As compared to pre -treatment[2.01(1.41, 2.71)], the plasma hsa_circ_0009024 levels decreased significantly in 3 months[1.22(0.85,1.47)](U=292.50,P<0.0001)after anti-TB therapy.The area under the receiver operating characteristics curve(AUC)of plasma hsa_circ_0009024 in discriminating the patients with TB from normal controls, pneumonia patients and lung cancer patients were 0.841 and 0.811, respectively.Conclusion The hsa_circ_0009024 can be used as a potential biomarker in TB diagnosis and monitoring.

7.Extended endocrine therapy of breast cancer

Endocrine therapy is an important treatment of estrogen receptor (ER) positive breast cancer.However,late relapse after completion of 5-year adjuvant endocrine therapy is still common.Extended endocrine therapy (EET) is a promising strategy to further reduce the risk of recurrence and increase the cure rate in early-stage patients.The decision-making to EET should be individually implemented by weighing the risk of late recurrence,menopausal status,side effects,and patients' preference.Future studies are still needed to identify the patients who are most likely to benefit from EET.
8.Effects of lincRNA-cox2 on the polarization of murine RAW264. 7 macrophages

Objective To investigate the effects of lincRNA-cox2 on the polarization of murine RAW264. 7 macrophages by analyzing the expression of lincRNA-cox2 in RAW264. 7 macrophages of M1 and M2 phenotypes. Methods Murine RAW264. 7 cells were induced by IFN-γand LPS to polarize to M1 phenotype, and were induced by IL-4 to polarize to M2 phenotype. The expression of lincRNA-cox2 in M1 and M2 macrophages were analyzed by real-time quantitative PCR ( RT-PCR) . We designed and synthesized siRNA oligo for lincRNA-cox2 and unrelated sequences. Then the siRNA oligo and NC oligo were transfected into RAW264. 7 cells by LipofectmineTM 2000. The transfected RAW264. 7 cells were induced by IFN-γand LPS or by IL-4 to polarize to M1 or M2 macrophages. Enzyme linked immunosorbent assay ( ELISA) was performed to measure the secretion of IL-10 and IL-12 induced in different conditions. The expression of in-ducible nitric oxide synthase ( iNOS ) , TNF-α, arginase 1 ( Arg-1 ) and found in inflammatory zone 1 (Fizz1) at mRNA level were detected by RT-PCR. The M1 macrophages were transfected with siRNAs to knock down the expression of lincRNA-cox2 for analyzing the biological effects of lincRNA-cox2 on the polar-ization of macrophages. Results The relative expression of lincRNA-cox2 in M1 macrophages was signifi-cantly higher than that in RAW264. 7 cells and M2 macrophages. Compared with the control group, the RAW264. 7 cells transfected with lincRNA-cox2-siRNA showed decreased secretion of IL-12 and inhibited expression of iNOS and TNF-αat mRNA level after IFN-γand LPS induction, but increased secretion of IL-10 and enhanced expression of Arg1 and Fizz1 at mRNA level after IL-4 induction. Transfecting the M1 mac-rophages with lincRNA-cox2-siRNA inhibited the secretion of IL-12, but promoted the secretion of IL-10. Conclusion This study indicated that lincRNA-cox2 was involved in the regulation of macrophage pheno-types by promoting the polarization to M1 macrophages and inhibiting the polarization to M2 macrophages.

10.Analysis on job stress assessment in BSL-3 laboratories

Objective The staffs of biosafety level 3 laboratories (BSL-3) face with the stress of handling highly pathogenic microbs and special laboratory environment.The job stress may result in accidents in the laboratory as negative factor for the risk control.The research may provide support for the control of risk in biosafety laboratories.Methods In order to assess the job stress in the staff in BSL-3 laboratory, we modified “the Chinese simple job stress questionnaire”based on the theory of the JDC mode and ERI mode, and an investigation was carried out.The present study included the staffs (87 employees) from six BSL-3 laboratories located in five provinces ( Shanghai, Zhejiang, Jiangsu, Fujian and Wuhan) .Results Analysis of the data indicates that variables of age, working years, job duties, manipulating of animals, type of microorganisms and transmission route have a significant influence on the level of job stress in BSL-3 laboratory.Conclusion The BSL-3 laboratory staff in higher stress level have the characteristicses:20-39 years old, short work years, regular staff, operating on air-borne microbiology, manipulating of animals and operating on one more microbiology.
