Elemene is widely recognized as an effective anti-cancer compound and is routinely administered in Chinese clinical settings for the management of several solid tumors, including non-small cell lung cancer (NSCLC). However, its detailed molecular mechanism has not been adequately demonstrated. In this research, it was demonstrated that elemene effectively curtailed NSCLC growth in the patient-derived xenograft (PDX) model. Mechanistically, employing high-throughput screening techniques and subsequent biochemical validations such as microscale thermophoresis (MST), microRNA-145-5p (miR-145-5p) was pinpointed as a critical target through which elemene exerts its anti-tumor effects. Interestingly, elemene serves as a binding stabilizer for miR-145-5p, demonstrating a strong binding affinity (dissociation constant (K D) = 0.39 ± 0.17 μg/mL) and preventing its degradation both in vitro and in vivo, while not interfering with the synthesis of the primary microRNA transcripts (pri-miRNAs) and precursor miRNAs (pre-miRNAs). The stabilization of miR-145-5p by elemene resulted in an increased level of this miRNA, subsequently suppressing NSCLC progression through the miR-145-5p/mitogen-activated protein kinase kinase kinase 3 (MAP3K3)/nuclear factor kappaB (NF-κB) pathway. Our findings provide a new perspective on revealing the interaction patterns between clinical anti-tumor drugs and miRNAs.

Glioma is the most common primary malignant tumor of the central nervous system in adults. Despite the standard treatment of surgery combined with radiotherapy and chemotherapy, the prognosis for high-grade glioma patients remains poor, highlighting the urgent need to further explore its pathogenesis and develop new therapeutic strategies. This article reviews the research progress in the field of glioma in China in 2024, covering tumorigenesis mechanisms, tumor immune microenvironment composition, advances in imaging techniques and novel imaging agents, improvements in surgical approaches, mechanisms of radio- and chemoresistance, and explorations of new therapeutic modalities. These studies provide a solid theoretical foundation for advancing clinical diagnosis and treatment of gliomas and may offer new opportunities to improve patient outcomes.
4.Cinnamaldehyde Attenuates Hepatic Fibrosis in NAFLD Rats by Inhibiting the SphK1/S1P Signaling Pathway

Objective To investigate the therapeutic effects and mechanisms of cinnamaldehyde(CA)on hepatic fibrosis in non-alcoholic fatty liver disease(NAFLD)rats.Methods A NAFLD rat model was established using a high-fat diet.Successfully modeled rats were randomly divided into:model group,low-and high-dose CA groups,and high-dose CA+K6PC-5[a sphingosine kinase-1(SphK1)activator]group,with healthy rats as normal controls.After intervention,serum lipid profiles[total cholesterol(TC),triglycerides(TG),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C)]and liver function markers[alanine aminotransferase(ALT),aspartate aminotransferase(AST)]were measured using an automated biochemical analyzer.Serum inflammatory cytokines(TNF-α,IL-6,IFN-γ)were quantified by ELISA.Hepatic histopathology was evaluated via hematoxylin-eosin(HE)and Masson staining.Immunohistochemistry detected fibrosis-related proteins(COL1A1,α-SMA),while Western Blot analyzed SphK1 and sphingosine-1-phosphate(S1P)expression.Results Compared with the normal group,the model group showed significant destruction of hepatic lobule structure,hepatocyte swelling and degeneration,disordered cell arrangement,abundant lipid vacuoles in cytoplasm,obvious inflammatory cell infiltration,extensive blue collagen fiber deposition,and severe fibrosis.Moreover,the levels of TC,TG,LDL-C,ALT,AST,TNF-α,IL-6,IFN-γ,COL1A1,α-SMA,SphK1,and S1P were significantly increased,while HDL-C levels were decreased,with statistically significance(P<0.05).Compared with the model group,both low-and high-dose CA groups demonstrated alleviated liver histopathological changes,reduced blue collagen fiber deposition,and improved fibrosis.These groups also showed significantly decreased levels of TC,TG,LDL-C,ALT,AST,TNF-α,IL-6,IFN-γ,COL1A1,α-SMA,SphK1,and S1P,along with increased HDL-C levels(P<0.05).However,compared with the high-dose CA group,the high-dose CA+K6PC-5 group exhibited more severe destruction of hepatic lobule structure,aggravated hepatocyte swelling and degeneration,dramatically increased cytoplasmic lipid vacuoles,significant inflammatory cell infiltration,increased blue collagen fiber deposition,and worsened fibrosis.This group also had significantly elevated levels of TC,TG,LDL-C,ALT,AST,TNF-α,IL-6,IFN-γ,COL1A1,α-SMA,SphK1,and S1P,along with reduced HDL-C levels(P<0.05).Conclusion CA alleviates hepatic fibrosis in NAFLD rats by inhibiting the SphK1/S1P signaling pathway.
5.Realationship between personality impulsivity and aggressivity in adolescents: the effecting path of sibling relationships

BackgroundIn China, the structure shift from just one-child family to both one-child and more-than-one-child families is happening. Exploring how the sibling relationships effect between adolescent personality impulsivity and aggressivity is of great significance for promoting adolescent mental health as well as maintaining social harmony and stability. ObjectiveTo investigate the effecting path of sibling relationships between personality impulsivity and aggressivity in adolescents, so as to provide references for the prevention of violent and aggressive behavior in adolescents. MethodsFrom February to April, 2023, a total of 1 200 students with sibling relationships from 12 primary and secondary schools in a county of Sichuan province were included by random sampling. Barratt Impulsivity Scale (BIS-11), Chinese Version of Buss & Perry Aggression Questionnaire (AQ-CV), and Sibling Relationship Questionnaire (SRQ) were used for cross-sectional investigation. Pearson Correlation Analysis was used to analyze the correlation between the scores of these scales. Bootstrap method was used to test the effecting path of sibling relationships between personality impulsivity and aggressivity. ResultsThe total score of BIS-11 was positively correlated with that of AQ-CV as well as the scores of conflict and competition dimensions in SRQ (r=0.485、0.276、0.280,P<0.01), while negatively correlated with the score of warmth/intimacy dimension in SRQ (r=-0.383, P<0.01). The total score of AQ-CV was positively correlated with the scores of conflict and competition dimensions in SRQ (r=0.387, 0.340, P<0.01), while negatively correlated with the score of warmth/intimacy dimension in SRQ (r=-0.304, P<0.01). Within SRQ, negative correlations could be observed between the score of warmth/intimacy and scores of conflict and competition (r=-0.307, -0.375, P<0.01), whereas positive correlation could be observed between the score of conflict and that of competition (r=0.267, P<0.01). The total effect of personality impulsivity level on aggressivity level in adolescent was 0.480 (P<0.01). Sibling relationships played a mediating role between personality impulsivity and aggressivity in adolescents. Meanwhile, the indirect effect values of warmth/intimacy, conflict and competition were 0.054, 0.075 and 0.062, with the effect values accounting for 11.21%, 15.70% and 12.93%, respectively. ConclusionThe personality impulsivity of adolescents can directly affect their aggressivity, and sibling relationships may act as an important channel connecting personality impulsivity and aggressivity. 6.Signal mining and analysis of selinexor adverse events

OBJECTIVE To mine the adverse drug event (ADE) signals of selinexor, and to provide reference for its clinical safety medication. METHODS ADE data for selinexor reported from July 3rd, 2019 to March 31st, 2023 were collected from the FDA adverse event reporting system (FAERS). Data mining was performed by using the reporting odds ratio (ROR) and proportional reporting ratio (PRR) methods, and categorization statistics were performed by using the system organ class (SOC) and preferred term (PT) from drug ADE terminology set in the MedDRA (version 26.0). RESULTS A total of 3 084 ADE reports were obtained for selinexor, with a total of 134 ADE-positive signals. Among the reported genders, there were 127 males and 124 females, with a predominant age of ≥65 years old (4.12%); the United States had the highest number of reports (96.53%), with consumers being the main reporters (77.27%); severe ADR was mainly characterized by hospitalization/prolonged hospitalization (26.26%), followed by death (17.15%). The top 3 ADE in the list of frequency were nausea (1 162 times), fatigue (790 times) and anorexia (610 times), all of which were mentioned in the selinexor’s instructions. The top 3 signals in the list of strength were device-associated bacteremia (ROR=115.07, PRR=114.94), blepharospasm dysfunction (ROR=106.70, PRR=106.54), and salmonella sepsis (ROR=99.90, PRR=99.81), all of which were not mentioned in the selinexor’s instructions. CONCLUSIONS In addition to the ADE of nausea mentioned in the instruction manual, attention should also paid to device-associated bacteremia, blepharospasm dysfunction, salmonella sepsis, and other ADE not mentioned in the instruction manual when using selinexor in clinical practice; weekly rechecking of the patient’s blood routine should be done to monitor the patient’s blood indexes, symptoms of infection, and so on, to ensure that the safety of 1661962346@qq.com drug use.
7.Investigation and epidemiological analysis of chronic diseases and comorbidities in hospitalized patients

Objective·To investigate the current situation and distribution characteristics of chronic comorbidities,and to provide reference for further improving the self-management of comorbidities and implementing the whole course and all-round management of comorbidity.Methods·Two thousand and forty-five inpatients in the Department of Geriatrics,Renji Hospital,Shanghai Jiao Tong University School of Medicine were enrolled in this study from December 2020 to February 2023.The general vital signs,routine laboratory examination and disease status were collected.The epidemiological distribution characteristics of chronic diseases and comorbidities were analyzed.Results·The incidence of chronic diseases in the surveyed population was 99.6%,and the incidence of comorbidities was 94.2%.The top 5 chronic diseases were hypertension(43.68%),diabetes mellitus(24.81%),malignant tumor(21.48%),hyperlipidemia(18.38%)and coronary heart disease(11.99%).The detection rates of hypertension,diabetes mellitus,coronary heart disease,chronic obstructive pulmonary disease,stoke and chronic kidney disease in males were significantly higher than those in females(P<0.05).The proportion of patients with 5 chronic diseases was the highest(11.99%),followed by 7 chronic diseases(10.26%)and 6 chronic diseases(10.04%).Among the patients of different ages,the comorbidity rate was the highest in the patients aged 50-59 years(27.78%).In different age groups,patients aged 50 to 59 with 2 chronic diseases had the highest incidence of comorbidity,which was as high as 40.82%.Although the overall proportion of comorbidities among male patients(95.37%)was higher than that among females(93.77%),there was no statistically significant difference(P=0.125).However,the proportions of male patients with 2 and 5 chronic diseases were 70.41%and 60.63%,respectively,which were significantly higher than those of female patients(29.59%and 39.37%).The correlations between coronary heart disease and diabetes mellitus,hypertension and coronary heart disease,hypertension and diabetes mellitus were higher(r=0.24,r=0.27,r=0.35,all P<0.05).Conclusion·The prevalence of chronic diseases and comorbidities is high in the middle-aged and elderly population,and the number of comorbidities increases significantly with the increase of age.
8.An observational study of symptomatic changes in patients with nasopharyngeal carcinoma following nimotuzumab administration one week before radiotherapy

Objective:To prospectively observe the changes of tumor-related symptoms in patients with nasopharyngeal carcinoma following the administration of nimotuzumab one week before radiotherapy.Methods:Non-metastatic nasopharyngeal carcinoma patients with positive epidermal growth factor receptor (EGFR) expression and symptoms caused by the primary lesion or metastatic cervical lymph nodes admitted to Cancer Hospital of Chinese Academy of Medical Sciences were prospectively recruited. Investigators recorded tumor-related symptoms in recruited patients one day before the first administration of nimotuzumab (D0) and conducted follow-up visits from day 2 to day 7 after the first administration (D2-D7) to document symptom changes. All recruited patients were asked to assess tumor-related symptoms on D0 and D7 by visual analogue scale (VAS) scores. VAS scores were analyzed by paired t-test. Results:From June 2020 to April 2023, a total of 21 patients met the inclusion criteria. The median age was 49 years (range: 27-69 years), with a male-to-female ratio of 1.3:1. Among the patients, 17 patients (81%) received concurrent nimotuzumab for 8 cycles, 7 cycles for 3 cases (14%), and 6 cycles for 1 case (5%), respectively. All patients completed symptom assessments as required. The overall response rate of symptoms after the first administration of nimotuzumab was 62%, with response rates of 4/6、5/8、4/10、4/10、4/11、3/11 for tinnitus, headache, aural fullness, secondary pain caused by neck mass, nasal bleeding, and nasal obstruction, respectively. The VAS scores for overall symptoms were significantly decreased after the administration of nimotuzumab one week before radiotherapy ( P<0.001), with the most significant decrease in VAS scores for tinnitus, aural fullness, and headache. Conclusion:The administration of nimotuzumab one week before radiotherapy significantly alleviates tumor-related symptoms in patients with nasopharyngeal carcinoma, particularly in alleviating tinnitus, aural fullness, and headache.
9.Comparison of Transoral Anterior Jefferson-Fracture Reduction Plate and Posterior Screw-Rod Fixation in C1-Ring Osteosynthesis for Unstable Atlas Fractures

Objective:
To compare the clinical outcomes of transoral anterior Jefferson-fracture reduction plate (JeRP) and posterior screw rod (PSR) surgery for unstable atlas fractures via C1-ring osteosynthesis.
Methods:
From June 2009 to June 2022, 49 consecutive patients with unstable atlas fractures were treated by transoral anterior JeRP fixation (JeRP group) or PSR fixation (PSR group) and followed up at General Hospital of Southern Theatre Command of PLA; 30 males and 19 females were included. The visual analogue scale (VAS) score, Neck Disability Index (NDI), distance to anterior arch fracture (DAAF), distance to posterior arch fracture (DPAF), lateral mass displacement (LMD), Redlund-Johnell value, postoperative complications, and fracture healing rate were retrospectively collected and statistically analyzed.
Results:
Compared with that in the PSR group, the bleeding volume in the JeRP group was lower, and the length of hospital stay was longer. The VAS scores and NDIs of both groups were significantly improved after surgery. The postoperative DAAF and DPAF were significantly smaller after surgery in both groups. Compared with the significantly shorter DPAF in the PSR group, the JeRP group had a smaller DAAF, shorter LMDs and larger Redlund-Johnell value postoperatively and at the final follow-up. The fracture healing rate at 3 months after surgery was significantly greater in the JeRP group (p < 0.05).
Conclusion
Both C1-ring osteosynthesis procedures for treating unstable atlas fractures yield satisfactory clinical outcomes. Transoral anterior JeRP fixation is more effective than PSR fixation for holistic fracture reduction and short-term fracture healing, but the hospital stay is longer.
10.Development of a mobile personnel radiation protection equipment

Objective To design a mobile personnel radiation protection equipment for operation in environments with high radiation such as spent fuel reprocessing plants, to achieve simultaneous protection against γ radiation, neutron radiation, and radioactive aerosol, to reduce the internal and external exposure dose of radioactive workers, and to meet the requirement of operation for two hours. Methods The core parts of the mobile personnel radiation protection equipment included a shielding chamber and a respiratory maintenance system. An automated chassis was used for the movement and lifting of the shielding chamber. MCNP software was used to simulate and calculate the protective effects of shielding chamber made of different materials and material thicknesses. Experimental verification of the shielding chamber design was conducted. Mathematical models were established to describe the variations in the content of various gases in the chamber with personnel operation time. A respiratory maintenance system, a harmful gas absorption device, and an automated mobile chassis were designed. Results The shielding chamber made of polyethylene with a thickness of 80 mm achieved an 80% neutron shielding rate. The respiratory maintenance system could support workers for 2 hours of operation inside the equipment. The mobile chassis allowed operation of the equipment with one person. Conclusion This mobile personnel radiation protection equipment can solve the problem in simultaneous protection against γ radiation, neutron radiation, and radioactive aerosol. The equipment can provide radiation protection for radioactive workers, reduce exposure dose, and reduce personnel burden. This system provides technical means for the operation and maintenance of equipment in high-radiation sites such as spent fuel reprocessing plants.
