In this study,the similarities and differences of plasma collection and quality control in China and abroad were analyzed by comparing the related regulations,standards,guidelines and literatures.Rational and constructive suggestions were proposed,aiming to optimize domestic plasma management and promote the improvement of plasma-related standards.There was little difference on facilities and safety control process of plasma between China and the developed countries(United States,EU and Japan),However,significant differences existed on plasma station setting,donor screening standards,collection interval,volume limits,plasma testing modes and tests,plasma quarantine standard and utilization of recovered plasma.The United States sets the industry benchmark and is worthy of reference for our country both in plasma collection and quality control.

3.Analysis of iodine nutrition status of children aged 8 to 10 years in Yantai City in 2022

Objective:To understand the iodine nutrition status of 8 - 10 years old children in Yantai City, and to provide a scientific basis for scientific iodine supplementation according to local conditions.Methods:Using multi-stage stratified cluster random sampling method, each county (city and district, hereinafter referred to as county) in Yantai City in 2022 was divided into 5 areas according to east, west, south, north and center, and 1 township (street) was selected as the survey site for each area, and 1 school was selected for each survey site, and 40 non-residential children aged 8 - 10 years old in each elementary school were selected as the target respondents, and the salt samples of edible salt in their homes were collected, and the urine samples of salt iodine and urinary iodine content were measured at random. A sample of salt and a random sample of urine were collected from the children's homes, and the salt iodine and urine iodine content were measured. Some of the children were sampled and their thyroid gland was tested using ultrasound. Each survey site is based on administrative villages (communities); administrative villages with centralized water supply collect one sample of terminal water; administrative villages with decentralized water supply take two wells (or all of them if there are fewer than 10 wells) in each of the five directions: east, west, south, north, and central, and collect water samples from the wells to test the iodine content of the water. Zhifu District and Zhaoyuan City each took 2 canteens of enterprises and institutions and 5 medium-sized and 5 small restaurants to collect salt samples of edible salt and test the salt iodine content.Results:A total of 2 343 children were tested for urinary iodine and salt iodine; the median urinary iodine was 147.10 μg/L, and the median urinary iodine in children from different regions was 173.96, 148.20, 148.05, and 136.04 μg/L, respectively, with statistically significant differences ( H = 15.55, P = 0.001); the median salt iodine was 17.81 mg/kg, and the different regional The median salt iodine was 0.00, 0.00, 20.02, and 22.48 mg/kg, respectively, and the difference was statistically significant ( H = 263.48, P < 0.001); the iodized salt coverage rate (15.34%, 48.37%, 69.63%, and 76.69%) and the consumption rate of qualified iodized salt (13.50%, 39.14%, 61.86%, and 69.17%) were compared in different regions were compared, and the differences were statistically significant (χ 2 = 257.65, 235.64, P < 0.001). A total of 1 419 children underwent ultrasound examination of the thyroid gland, and there were no statistically significant differences when comparing the rates of goiter (2.44%, 2.18%, 3.04%, and 3.12%) and nodule detection (3.90%, 4.90%, 5.89%, and 2.18%) among the children in different regions (χ 2 = 0.85, 6.69, P > 0.05). A total of 2 488 water samples were monitored, and the median water iodine was 3.70 μg/L. When comparing the median water iodine in different regions, the difference was statistically significant ( H = 141.21, P < 0.001). A total of 120 salt samples of edible salt from catering units were tested, and the consumption rate of qualified iodized salt was 62.50%. Conclusion:The consumption rate of qualified iodized salt among children in Yantai was below 90%, but their iodine nutrition was at an appropriate level, suggesting that the iodine intake of children in Yantai may not be entirely dependent on iodized salt.
4.Infection status of Anisakis larvae in the major economic marine products in the Yellow Sea and Bohai Sea from 2016 to 2020

Objective:To learn about the infection status of Anisakis larvae in the major economic marine products in the Yellow Sea and Bohai Sea, and provide baseline data for systematic monitoring of Anisakis and prevention and control of related diseases. Methods:From April 2016 to September 2020, the samples of marine products collected in the surrounding waters of 9 fishing sites in the Yellow Sea and Bohai Sea (Bohai Bay, the middle part of the Yellow Sea and Bohai Sea junction, the southern part of the Yellow Sea and Bohai Sea junction, the northern part of the Yellow Sea and the southern part of the Yellow Sea) in the coastal areas of Yantai City and Weihai City, Shandong Province were dissected and tested for worms. The infection and distribution of Anisakis larvae in different types of samples and different organs in the samples were compared, and the differences of the infection level of Anisakis larvae in marine fish among the surrounding waters of different fishing sites and different sampling sites in China were compared. At the same time, a survey on the awareness of health knowledge of anisakiasis was carried out among the residents near each fishing sites. Results:A total of 708 cases of 5 types of marine products were tested in the Yellow Sea and Bohai Sea, including 581 cases of marine fish, 22 cases of mollusks, 20 cases of echinodermata, 75 cases of crustaceans and 10 cases of shellfish. Anisakis larvae infection was detected only in marine fish (191 cases), and 4 723 Anisakis larvae were found. The infection rate was 32.87% (191/581) and the infection intensity was 24.73(4 723/191) larvae/case. They were mainly distributed in mesentery and intestinal wall (38.96%, 1 840/4 723), coelom (22.04%, 1 041/4 723) and gastric wall (17.95%, 848/4 723). The infection levels of Anisakis larvae in marine fish among the surrounding waters of different fishing sites were compared, the infection rate in the southern part of the Yellow Sea was the highest, and its infection intensity was significantly higher than that in the middle and southern part of the Yellow Sea and Bohai Sea junction ( P < 0.05). The infection levels of Anisakis larvae in marine fish among different sampling sites in China were compared, the infection rates of Zhoushan Port, the fish sold in Jinzhou, Yantai and Shantou were significantly higher than those in the Yellow Sea and Bohai Sea ( P < 0.05), and the infection rates of the fish sold in Dandong and Qingdao were significantly lower than those in the Yellow Sea and Bohai Sea ( P < 0.05). A total of 1 805 residents living near the Yellow Sea and Bohai Sea were investigated on the health knowledge of anisakiasis. Among them, 20.78% (375/1 805) residents had heard of anisakiasis, 15.73% (284/1 805) residents knew how to get it, 12.30% (222/1 805) residents knew the harm of anisakiasis to human body, and 16.68% (301/1 805) residents knew how to prevent it. Conclusions:The marine fish in the Yellow Sea and Bohai Sea are infected by the Anisakis larvae, and the level of infection is relatively high. In the future, we should strengthen the popularization of knowledge on prevention and control of anisakiasis.
5.Distribution and transfusion efficacy of unexpected antibody

【Objective】 To investigate the clinical characteristics and antibody distribution as well as evaluate the transfusion efficacy in unexpected antibody positive patients. 【Methods】 A total of 12 235 patients from January 1, 2022 to March 31, 2023 who hospitalized in our hospital and applied for blood transfusion were selected, and those with unexpected antibody were included. The clinical data, including gender, age, diagnosis, blood type, history of transfusion and pregnancy were collected for antibody distribution analysis. Patients who received transfusion were grouped according to the DAT results and the components of red blood cells transfused, and the Hb values of each group before and after transfusion were compared. 【Results】 Among12 235 patients, 118 were positive for antibody screening, with a prevalence of 0.96%. The antibodies from Rh system were the most common (27.43%, 48/175), followed by MNS system (8.57%, 15/175) and Lewis system (6.29%, 11/175), mainly anti-E (18.29%, 32/175), anti-M (8.00%, 14/175) and anti-Lea (5.71%, 10/175). In addition, 62 transfused patients were divided into group A with suspended red blood cell transfusion and group B with washed red blood cell transfusion for positive DAT, and group C for negative DAT. Hb values (g/L) pre- and post-transfusion were 59.19±15.67 vs 77.52±15.09 in group A, 56.35±14.08 vs 74.44±15.63 in group B, 56.00±12.06 vs 75.00±4.73 in group C, respectively. The Hb values of post-transfusion for three groups were all higher than those of pre-transfusion (P<0.05). 【Conclusion】 Anti-E from Rh system is the most common antibody in patients with unexpected antibody. Appropriate red blood cells transfusion with Hb increases by an average of 6-7 g/L per 1 U of red blood cells indicating good transfusion efficacy. For positive DAT patients, transfusion of suspended red blood cell is feasible.
6.AP2α negatively regulates PDHA1 in cervical cancer cells to promote aggressive features and aerobic glycolysis in vitro and in vivo

Objective:
As a gate-keeper enzyme link, pyruvate dehydrogenase E1 subunit alpha (PDHA1) functions as a key regulator during glycolysis and the mitochondrial citric acid cycle, which has been reported in several tumors. Nevertheless, the effects of PDHA1 on biological behaviors and metabolism remain unclear in cervical cancer (CC) cells. The study aims to explore the PDHA1 effects on glucose metabolism in CC cells and its possible mechanism.
Methods:
We first determined the expression levels of PDHA1 and activating protein 2 alpha (AP2α) as a PDHA1 potential transcription factor. The effects of PDHA1 in vivo were evaluated through a subcutaneous xenograft mouse model. Cell Counting Kit-8 assay, 5-ethynyl-2′-deoxyuridine (EdU) labeling assay, Transwell invasion assay, wound healing assay, Terminal deoxynucleotidyl transferase dUTP nick end labeling assay and flow cytometry were performed in CC cells. Oxygen consumption rate (OCR) levels were determined to reflect aerobic glycolysis level in gastric cancer cells. Reactive oxygen species (ROS) level was measured with 2′, 7′-dichlorofluorescein diacetate kit. The relationship between PDHA1 and AP2α was examined by conducting chromatin immunoprecipitation assay and electrophoretic mobility shift assay.
Results:
In CC tissues and cell lines, PDHA1 was downregulated, while AP2α was upregulated. Overexpression of PDHA1 remarkedly inhibited the proliferation, invasion and migration of CC cells, and tumor growth in vivo, as well as promoted OCR, apoptosis and ROS production. Moreover, AP2α directly bound to PDHA1 within suppressor of cytokine signaling 3 promoter region to negatively regulate PDHA1 expression level. What is more, PDHA1 knockdown could effectively reversed the AP2α silencing-mediated suppressive effects on cell proliferation, invasion, migration, and the promotive effects of AP2α knockdown on OCR, apoptosis and ROS production.
Conclusions
Our findings demonstrate that AP2α negatively regulated PDHA1 via binding to PDHA1 gene promoter to promote malignant CC cell behaviors, which may provide a potential approach for CC therapeutics.
7.Correlation of dark red tongue and metabolic parameters in health checkup population

Objective To study the metabolic parameters in health checkup people with dark red tongue and light red tongue,with different coating nature,coating color or liguliform,to understand the relationship between dark red tongue and human metabolic state.Methods Clinical data of 2 891 individuals who underwent health checkup in our center between Dec.2013 and Aug.2014 were collected,including body mass index,blood pressure,blood glucose,lipid and uric acid.Tongue diagnosis was made by Chinese Medicine practitioners.Results The dark red tongue was more common in males,it was found in 61.58% males and 38.42% females in the dark red tongue group.The average level including body mass index,systolic blood pressure,diastolic blood pressure,blood glucose,total cholesterol,triglyceride,low-density lipoprotein cholesterol and blood uric acid in dark red tongue group were higher than those in light red tongue group (P<0.01).When the dark red tongue had yellow fur-like or thick and greasy coat,the average level of body mass index,blood glucose,total cholesterol,low-density lipoprotein cholesterol and blood uric acid were higher than those of the normal dark red tongue group,the level of high-density lipoprotein cholesterol was lower than that of the normal dark red tongue group.Conclusions Dark red tongue has homogeneous relations with abnormal metabolic parameters,it may be the external expression of metabolic abnormalities;tongue is the external macro manifestation of human body health state.
8.Effect of interferon on HEL cell apoptosis and JAK2 V617F mutation gene expression

Objective To observe the effect of Interferon-α2b on HEL cells (human erythroleukemia cell line) growth, apoptosis and JAK2 V617F mutation gene expression. Methods HEL cells were placed in RPMI1640 containing 10% FBS and incubated in a cell incubator. Cells in the logarithmic growth phasem were collected, adjusting the cell density to 1 × 105/mL for experimental research. The interferon concentration in five groups were 0, 5 × 105, 10 × 105, 50 × 105, 100 × 105 U/L, with different incubation time (0, 24, 72, 120 h), respectively. The cell growth status in different groups was observed in the inverted optical microscope; MTT was used to detect the inhibition of interferon on HEL cell proliferation. Cell apoptosis was detected by flow cytometry. Fluorescence quantitative PCR was used to detect the mutation gene of JAK2 V617F expression. Results Inhibition rates of Interferon on the HEL cell proliferation in 5 × 105 U/L, 10 × 105 U/L, 50 × 105 U/L, 100 × 105 U/L groups were 18.57%, 25.10%, 42.10%, 57.00%, respectively. JAK2 V617F/GAPDH by fluorescence quantitative was 1.556, 1.213, 0.870 respectively under the concentration of interferon 100 × 105 U/L for 24, 72, 120 h. Conclusions Interferon-α2b can inhibit HEL cells proliferation and induce HEL cells apoptosis. Increasing concentration of interferon increases HEL cell apoptosis rate. Interferon can inhibit JAK2 V617F expression of HEL cells in a dose-dependent manner.
9.Fatty liver development and changes of metabolic parameters

Objective To investigate the relationship between fatty liver progress and metabolic changes.Methods A total of 414 patients who had B-mode ultrasonography confirmed fatty liver development during 2 health check-ups within 2 years were enrolled in this study.Paired t and χ2 tests were used to compare body mass index (BMI),alanine aminotransferase (ALT),aspartate aminotransferase (AST),fasting plasma glucose (FPG),serum uric acid (UA),triglycerides (TG),and high-density lipoprotein cholesterol (HDL-C) among the participants.Results BMI,TG,HDL-C,ALT,AST,and UA were significantly increased after fatty liver progressed,although HDL-C was largely decreased.ConclusionsPrevention and treatment of MS as well as early intervention for fatty liver should be important for successful control of fatty liver.
10.The using of tongue diagnosis in fatty liver patients' health management

Objective To find the syndrome tendency of fatty liver people and then providing basis for TCM integrated into fatty liver patients'Health Management by analyzing the characteristics of fatty liver people's tongue and combining them with the investigating results of symptoms.Methods Using 1∶1control study methods,examined 368 cases(184 cases were patients with fatty liver),observed their tongues as well as their symptoms,and recorded results.Results x2 test showed fat tongue,thin and small tongue,teeth-marked tongue,thick fur,greasy fur,fond of cool drink,hate feat,diarrhea after eating cold food,more sweat,lethargy,heavy four limbs,more flatus were significantly increase in fatty liver people(x27·580,11.740,23.700,8.666,10.793,P>0.05).Conclusions The symptoms of patients with fatty liver disease were the reflection of heat and(or)damp-heat.Found the generally pathology rules of patients with fatty liver disease,we could be more reasonable and effective prevention and treatment.
