1.Correlations of brain network functional connectivity alterations with cerebrospinal fluid pathological markers in patients with Alzheimer's disease

Objective:To explore the correlations of brain network functional connectivity (FC) alterations with cerebrospinal fluid (CSF) pathological biomarkers in patients with Alzheimer's disease (AD).Methods:A total of 39 patients with cognitive impairment, admitted to Department of Neurology, Drum Tower Hospital, Affiliated Hospital of Medical School, Nanjing University from January 2020 to December 2022 were recruited; 23 patients were with AD and 16 with non-AD. Clinical data were compared between the 2 groups. Resting-state functional MRI (rs-fMRI) data were collected, and FC differences between brain networks and FC differences within brain networks were compared by independent component analysis. Correlations of FC differences between brain networks and FC differences within brain networks with concentrations of β-amyloid protein 1-42 (Aβ 1-42) and Tau protein in CSF were analyzed. Results:Compared with the non-AD group, AD group had significantly lower Aβ 1-42 in CSF ( P<0.05). Compared with those in the non-AD group, FC alterations between the left frontoparietal network (lFPN) and anterior default mode network (aDMN) and between the visual network (VN) and posterior cingulate cortex (PCC), as well as FC alterations in lFPN, were significantly increased in AD group ( P<0.05). Compared with those in the non-AD group, FC alterations between lFPN and cerebellar network (CEN), and FC alterations in aDMN, sensorimotor network (SMN) and VN were significantly decreased in AD group ( P<0.05). In AD group, FC in SMN was positively correlated with total Tau and phosphorylated-Tau181 in CSF ( P<0.05); FC between VN and PCC was positively correlated with total Tau in CSF ( P<0.05). CSF Aβ 1-42 was positively correlated with FC alterations in aDMN and VN, but negatively correlated with FC in FPN ( P<0.05). Conclusion:In AD patients, characteristic changes in FC within and between multiple brain networks are noted, which are related to changes of Tau protein and Aβ 1-42 in CSF.
2.Effects of different body mass index on perinatal and neonatal outcomes of frozen-thawed embryo transfer in PCOS patients

Objective:To investigate the influence of polycystic ovary syndrome (PCOS) patients with different body mass index (BMI) on perinatal and neonatal outcomes of frozen-thawed embryo transfer.Methods:A retrospective cohort study was performed on the clinical data of patients with PCOS infertility who underwent cryopreservation transplantation in Reproductive Center of the First Affiliated Hospital of Anhui Medical University from 2016 to 2020. The clinical pregnancy was singleton, a total of 1 481 cycles were divided into 4 groups according to BMI value. There were 75 cycles in the underweight group (BMI<18.5 kg/m 2), 793 cycles in the normal weight group (18.5 kg/m 2≤BMI<24.0 kg/m 2), 468 cycles in the overweight group (24.0 kg/m 2≤BMI<28.0 kg/m 2), 145 cycles in the obese group (BMI≥28.0 kg/m 2). The differences of general information, perinatal outcome and neonatal outcome were compared among the four groups. Results:Compared with the overweight group, the normal weight group and the underweight group, the obesity group had the highest early abortion rate [23.4% (34/145) vs. 15.8% (74/468) vs. 14.0% (111/793) vs. 9.3% (7/75), P=0.014], and the lowest live birth rate [68.3% (99/145) vs. 76.7% (359/468) vs. 79.7% (632/793) vs. 88.0% (66/75), P=0.003]. The incidence of gestational diabetes in the obesity group and the overweight group [6.9% (10/145) and 4.5% (21/468)] was higher than that in the normal weight group [2.3% (18/793)] (the obesity group vs. the normal weight group P=0.005, the overweight group vs. the normal weight group P=0.028). The rate of cesarean section in the obesity group and the overweight group [81.8% (81/99), 74.9% (269/359)] was higher than that in the normal weight group [67.6% (427/632)] and the underweight group [57.6% (38/66), the obesity group vs. the normal weight group P=0.005, the obesity group vs. the underweight group P=0.001, the overweight group vs. the normal weight group P=0.015, the overweight group vs. the underweight group P=0.004]. The macrosomia birth rate [18.2% (18/99), 15.6% (56/359)] was also higher than that of the normal weight group [10.1% (64/632)] and the underweight group [6.1% (4/66), the obesity group vs. the normal weight group P=0.018, the obesity group vs. the underweight group P=0.025, the overweight group vs. the normal weight group P=0.011, the overweight group vs. the underweight group P=0.041]. There were no significant differences in late abortion rate, gestational hypertension, ectopic pregnancy and premature birth rate, Apgar score, height and birth defects (all P>0.05). Conclusion:Obesity and overweight affect the perinatal outcomes and neonatal outcomes in patients with PCOS. In clinical work, attention should be paid to the weight management of PCOS patients.

4.Effects of CXCR4 gene transfection on biologicalbehavior of bone marrow mesenchymal stem cells (in vitro)

Aim To observate the effect of chemokine receptor(CXCR4) gene transfection on biological behavior of bone marrow mesenchymal stem cells in vitro.Methods Firstly, bone marrow mesenchymal stem cells were divided into three groups:GFP(transfected GFP into MSCs), CXCR4+(transfected CXCR4+ into MSCs) and CXCR4-(transfected CXCR4-into MSCs) group.Then, their capacity of proliferation, differentiation and migration ability (in vitro) was assessed with immunofluorescence cytochemistry method, flow cytometry assay and Transwell cell chemotaxis test.Results The high or low expression of CXCR4 had no effect on their ability of proliferation and differentiation into lung tissue.Compared with GFP group, however, CXCR4+-MSCs group significantly increased the number of migrating cells, while CXCR4——MSCs group showed no change in the number of migrating cells.Conclusions The proliferation and differentiation capacities are not affected by the high or low expression of CXCR4.The high expression of CXCR4 can significantly enhance the migration ability of MSCs to inflammatory lesions, and the low one has no effect on the migration of the cells.After the transplantation of MSCs, CXCR4′s high expression will access to the lesion area to participate in tissue repairing rapidly and largely, significantly enhancing the therapeutic efficacy.

Objective To study the effects of nasal gingerol gel on motion sickness .Methods The effects of nasal gingerol gel on motion sickness were evaluated by using the rat model of conditioned taste to observe changes in the amount of saccharine water intake , and also the Beagle dog model to observe the latent period of salivation and vomiting, before and after rotational stimulation.Results Before rotational stimulation, there were no significant differences in the amount of saccharine water intake in the rats of the groups . However, following rotational stimulation, the amount of saccharine water intake in the rats of the control group was obviously reduced, as compared with that before stimulation (P <0.01).Before drug treatment, all the Beagle dogs were susceptible to rotational stimulation , displaying early salivation and vomiting .However, after medication, the latent period of salivation and vomiting was all considerably delayed , as compared with the control group(P <0.05).The latent period of salivation of middle and high medication groups were extended to (17.89 ±8.42) min and (18.58 ±9.92) min, respectively.And the laent period of vomition of middle and high medication groups were extended to (25.50 ±12.78) min and (27.47 ±10.25) min.Conclusions Nasal gingerol gel showed certain protective effects against motion sickness .

7.Neurotrophin 3 gene overexpression promotes cholinergic differentiation in neural stem cells

Aim To investigate the effects of neurotro-phin-3 ( NT-3 ) gene overexpression on the differentia-tion into cholinergic neuron of neural stem cells ( NSCs) in vitro and its underlying mechanism. Meth-ods Brain-derived NSCs from newborn mice were iso-lated and cultured in vitro and determined by immuno-fluorescence. The NSCs were divided into three groups: NSCs, GFP-NSCs and NT-3-NSCs groups. The expression of NT-3 was detected by immunofluo-rescence and ELISA. Then, the ability of NSCs on dif-ferentiation into cholinergic neuron was detected by im-munofluorescence and RT-PCR, and the Acetylcholine Assay Kit was used for acetylcholine ( ACh) , and the expression of Hes1 , Mash1 and Ngn1 mRNA was de-termined by RT-PCR. Results The neurosphere dis-played Nestin and Sox 2-postive by immunofluores-cence, suggesting that the cultured cells were NSCs. The proportion of ChAT immunopositive cells was sig-nificantly higher in the NT-3-NSCs group than that in the other two groups ( P <0. 01 ) . Ach secretion in NT-3-NSCs was significantly elevated compared with the other two groups ( P <0. 01 ) . NSCs transfected with NT-3 increased the levels of Mash1 and Ngn1 mR-NA, and decreased the level of Hes1 mRNA ( P <0. 05 ) . Conclusion NT-3 can significantly promote the in vitro differentiation of NSCs into cholinergic neu-rons via probablly inhibiting Notch signaling pathway.
8.Application of intravoxel incoherent motion diffusion-weighted imaging in diagnosis of lung cancer

Objective To explore the diagnostic value of intravoxel incoherent motion diffusion-weighted imaging(IVIM-DWI)in diagnosis of different pathological types of lung cancer.Methods 45 patients were performed traditional MR and multi-b value DWI scan by GE discovery 750 MR.The values of Slow-ADC,Fast-ADC and ffast measured on a AW4.5 workstation were analyzed between the different pathological types of lung cancers.The correlations between these IVIM-DWI parameters and the serum tumor markers of lung cancer were analyzed.The diagnostic efficiency of these parameters were evaluated by receiver operator characteristic curve (ROC).Results 27 cases NSCLC(13 cases squamous carcinoma;14 cases adenocarcinoma )and 18 cases SCLC were finally included in this study.There were significant differences in Slow-ADC values between SCLC group and NSCLC group (P =0.00),the adenocarcinoma group (P=0.03),the squamous carcinoma group(P=0.01).There were no significant difference in Fast-ADC as well as ffast value between any two groups.The AUC of Slow-ADC value was 0.874.There existed negative correlation between squamous cell carcinoma antigen(SCC-Ag)of squamous carcinoma group and Slow-ADC(r=-0.730).Conclusion The Slow-ADC of IVIM-DWI parameters is useful in differential diagnosis of NSCLC and SCLC,which has the largest diagnostic efficiency.The correlation between SCC-Ag and Slow-ADC value has a certain meaning in diagnosing different pathological types of lung cancers.
9.Clinical analysis of imflammatory myofibroblastic tumor of the nasal cavity and paranasal sinus.

OBJECTIVE:
To investigate the clinical features, therapeutic methods and therapeutic efficacy of imflammatory myofibroblastic tumor(IMT)of the nasal cavity and paranasal sinuses.
METHOD:
Clinical data of 14 cases diagnosed as IMT by pathology were retrospectively analyzed. There were 8 males and 6 females,age ranging from 18 to 77 years. 12 cases of them were treated by surgery while 2 cases received postoperative radiotherapy.
RESULT:
All cases were operated. All the patients were followed up for a period of 1 to 7 years after operation and two cases were proved low grade IMT pathologically. Eight cases survived with no recurrence until the last follow-up and 6 cases relapsed, of which 4 cases died and 2 were alive with tumor.
CONCLUSION
IMT of the nasal cavity and paranasal sinuses is very rare. The diagonosis of IMT is based on pathology and immunohistochemistry. Proper diagnosis is essential to avoid mutilating and disfiguring surgical procedures. Radical excision is still the first choice of treatment for IMT of the nasal cavity and paranasal sinuses. Chemotherapy and radiotherapy may not be helpful to prevent recurrence after operation. Due to high recurrence rate, long-term follow up is necessary after operation.

10.Cumulative Analgesic Effect of Electroacupuncture at Sanyinjiao (SP6), Xuanzhong (GB39) and Non-acupoint for Primary Dysmenorrhea:A Comparative Study

Objective To compare the cumulative analgesic effects of electroacupuncture at Sanyinjiao (SP6), Xuanzhong (GB39) and non-acupoint in treating primary dysmenorrhea. Method By adopting a multi-centered randomized controlled study method, 501 patients recruited from Dongzhimen Hospital of Beijing University of Chinese Medicine, China-Japan Friendship Hospital, Beijing Hospital of Traditional Chinese Medicine of Capital Medical University, First Teaching Hospital of Tianjin University of Traditional Chinese Medicine, Huguosi Hospital of Chinese Medicine of Beijing University of Chinese Medicine and the Outpatient of Shandong University of Traditional Chinese Medicine were randomized into a Sanyinjiao group, a Xuanzhong group, and a non-acupoint group, 167 subjects in each group. The electroacupuncture intervention was applied when dysmenorrhea flared up and the Visual Analogue Scale (VAS) ≥40 mm, with frequency at 2/100 Hz and intensity during patient’s endurance, 30 min each time, once a day, and for successive 3 d. Before the first treatment, 30 min after the first treatment, and respectively prior to the second and third treatment, VAS was used to measure the pain intensity. Meanwhile, the Retrospective Symptom Scale (RSS-COX 2) was investigated before the first treatment, right after the removal of needles for the first treatment, before the second and third treatment. Result The decrease of VAS in Sanyinjiao group was more significant than that in Xuanzhong group and non-acupoint group (MD=﹣2.92 mm, P=0.028; MD=﹣3.47 mm, P=0.009), while there was no significant difference between Xuanzhong group and non-acupoint group (MD=﹣0.56 mm, P=0.674); there were no significant differences in comparing the RSS-COX2 total score among the three groups (P=0.086). Conclusion Sanyinjiao (SP6) can produce a more significant cumulative analgesic effect for primary dysmenorrhea patient than Xuanzhong and non-acupoint, and the effects of Xuanzhong and non-acupoit are equivalent.
